Abstract

Mosaic disease outbreaks on chili (Capsicum annum) associated with Cucumber Mosaic Virus (CMV) is serious worldwide. The use of resistant cultivars is an effective control to reduce the loss of yield caused by virus’s infections. The objective of this study is to detect the resistant characters of CMV in chili using OPH-17 and OPA-20 RAPD primers. The inoculation of CMV in PSPT-C11, C17-PSPT, ICPN 7 # 3 and 1 Chilibangi 1 varieties cause systemic mosaic symptoms vary from mild mosaic to the mosaics with malformations. PSPT-C11 varietiy showed rather resistant, while the varieties PSPT-C17 and Chilibangi 1 are susceptible, and ICPN 7 # 3 is very susceptible. OPH-17 and OPA-20 primers could not be used to determine the resistance differences against CMV in PSPT-C11, C17-PSPT, ICPN 7 # 3 and Chilibangi 1 varieties.

Abstract

Abstract

Abstract

A research regarding “Runner-tip culture of strawberry (Fragaria x ananassa Duch) Grown on Several Shoot-induction Medium” has been investigated. The objective of the research was to find out the most suitable medium for shoot production from runner-tip culture of strawberry at establishment step of micropropagation. The research was laid out in a Completely Randomized Design, 4 treatments of medium type for shoot induction and 10 replication, each was represented by one (1) bottle with 6-8 explants. The treatments were summarized as follows: T1 = MS ; T2 = MS + 2 ppm BAP + 0.01 ppm NAA; T3 = MS + 1 ppm of TDZ; T4 = WPM + 2 ppm BAP + 0.01 ppm NAA. The parameters observed were days of the bud emergence, the average number of shoots per explant, and the average number of leaves per explant. It can be concluded that among medium used in the current research, the medium of MS added with 1 ppm thidiazuron (TDZ) was the most suitable medium for shoot production of strawberry from the explant of runner-tips. The treatment was resulted in the earliest time of bud emergence, and producing the highest number of shoots and leaves.

Abstract

The aim of this study was to investigate genetic diversity and relationship of guava (local name: jambu biji) genotypes grown in Bali, Indonesia, based on Random Amplified Polymorphic DNA (RAPD) markers. Twelve guava cultivars namely Jambu biji ‘Australia’, ‘Bangkok Merah’, ‘Bangkok Putih’, ‘Cokorde’, ‘Dadu 1’, ‘Dadu 2’, ‘Getas Merah’, ‘Kamboja’, ‘Kristal’, ‘Pipit’, ‘Susu’ and ‘Variegata’ were collected from nine Regencies in Bali using survey and explorative method. Ten decamers RAPD primers were employed to distinguish between the 12 cultivars and determine their genetic relationships. A dendogram was constructed using coefficient dissimilarity analysis based on phylogenetic analysis using parsimony (PAUP). The twelve cultivars were grouped into 2 main clusters and five smaller clusters. Variation between genotypes of guava local will be good sources for future crop improvemen.

Abstract

Tomato (Lycopersicum esculentum Mill.) is a fruit with many benefits. Plant growth regulators (PGRs) prove effective in improving the quality and production of tomatoes. Gibberellic Acid/GA is a PGR that stimulates the process of flowering, parthenocarpy, and carbohydrate mobilization during germination and influences other plant physiological aspects. However, because synthetic GA is costly, numerous natural ingredients have been proposed as its source, for example, bamboo shoots, vegetable ferns, corns, and shallots that can improve the quality of tomatoes. This study aimed to determine the role of natural GA from various plant extracts on the quality of tomatoes, providing recommendations for the best extract to improve the quality of tomatoes. It was conducted in the experimental garden at the Agronomy and Horticulture Laboratory, Physiology Laboratory of the Faculty of Agriculture, and Food Analysis Laboratory of the Faculty of Agricultural Technology, Udayana University. This research was compiled using a Randomized Group Design consisting of one factor, i.e., the type of natural extract with five settings and four repetitions. The tomato plant treatments were E0 = no extract sprayed, BE = sprayed with bamboo shoot extract, VE = vegetable fern extract, OE = onion extract, and CE = corn extract. The results showed that gibberellins from natural ingredients significantly affected fruit diameter, fruit thickness, and total sugar content of tomatoes. The best tomatoes were obtained by applying onion extract as a source of natural gibberellins.
