Abstract

AbstractThis research has done partial purification by fractionation and optimization crystallization of papain from Carica papaya latex with the addition of ammonium sulphate. The enhancement of purification factor was determined by its specific activity. The fractionation results show that papain fraction of Carica papaya can be obtained by adding 40-80% saturated ammonium sulphate, with the highest specific activity, i.e. 2,0819 U/µg and the factor purification increase of 6,27 fold than papain extract. Meanwhile, the highest total activity, i.e. 10,7780 U of papain crystals can be obtained by presipitant agent addition of ammonium sulphate in the level / concentration 80% saturated at 15 °C. Microscopycally papain crystals profile in this condition have cube and tetragonal shape.Key words: crystallization, fractionation, ammonium sulphate, papain

Abstract

The aim of this research was to express the β-xylosidase gene in the pHIS1525/ Bacillus megaterium MS941 system. The xyl gene was amplified from pTP510 and cloned into pHIS1525 in E. coli DH10β. The recombinant plasmid was transformed into B. megaterium MS941 by protoplast transformation. Transformants were selected by growing the recombinant B. megaterium MS941 on solid LB medium containing tetracycline (10 μg/ ml). The expression of β-xylosidase was assayed using 0.2% methylumbelliferyl-β-D-xyloside (MUX) and the proteins were analyzed by SDS-PAGE method. The β-xilosidase activity was determined toward p-nitrophenyl-β-Dxylopyranoside (pNPX) as a substrate and p-nitrofenol releasing was measured by UV/Vis spectrophotometer at λ = 405 nm. This research showed that recombinant B. megaterium MS941 expressed the β-xylosidase gene (xyl) and secreted it into the culture medium. The SDS-PAGE analysis of extracellular protein (culture medium) showed a 60,0 kD protein band. The recombinant Bacillus megaterium MS941 expressed and secreted the β-xilosidase into culture medium 5 hours after adding 5% xylose. The b-xylosidase activity was 0.441 unit/ml toward pNPX as a substrate.

Abstract

Characterization used retriction enzyme on the 1784 bp (base pairs) DNA fragmen of glucoamylase gene (GLUI) of E. fibuligera ITB. R. cc. 64 has been done. The rectriction enzyme usage was Stu 1, Eco RI, Eco RV, Bam HI and Sau 3A. The purpose of this research were: First was to know recognition site of 1784 bp DNA fragmen of glucoamylase gene (GLUI) by the restriction enzyme above. The second was to know homologyst the glucoamylase gene (GLUI) E. fibuligera ITB. R. cc. 64 and the glucoamylase gene (GLUI) Saccharomycopsis fibuligera HUT 7212 (pSf GLUI). The result of amplification glucoamylase gene (GLUI) indicated that 1784 bp DNA fragmen on GLUI locus has succesfully isolated and gave the same size with the positive control pSf GLUI. Analysis of those DNA fragmen by StuI, Eco RV, Eco RI, Bam HI and Sau 3A indicated that 1784 bp of DNA fragmen from E. fibuligera ITB.R.cc.64. has the same result with 1784 bp of DNA fragmen from pSf GLUI. The result of the fragments after incubated by restriction enzymes are as follows: ± 997 bp and 787 bp by Eco RI, 1000 bp and 1780 bp by Bam H) and 850 bp and 760 bp by Sau 3A. Digestion using StuI and Eco RI was failed. To ensure that the DNA fragmen 1784 bp has characteristic as glucoamylase gene, it should be expressed into S. cerevisiae and/or should be determined the nucleotide sequence by DNA sequencing.

Abstract

Mercury reductase is an enzyme that is able to reduce Hg2+ to Hg0 non toxic. This enzyme is usually produced by mercury resistant bacteria. The research wanted to determine the resistance of indigenous Pseudomonas stutzeri isolate toward mercury and to explore the mercury reductase activity which is produced by the bacteria. The results of resistance assay of the Pseudomonas stutzeri toward mercury ion showed that the isolate could survive in media containing HgCl2 up to a concentration of 80 µM. The bacteria could produce mercury reductase optimally at the 24th of fermentation time. The enzyme showed optimum activity at pH 7 and temperature of 45 oC

Abstract

Mercuric reductase is the important enzyme which catalyzes a reduction of a toxic Hg2+ to non-toxic Hg0. The enzyme which has been potentially used as mercury bioremediation agent is produced by mercury resistant bacteria. These research aims are to determinate the resistance level of a local Bacillus sp to HgCl2 in media, to determine the mercuric reductase activity from the bacteria, and to determine the biochemical properties of the mercuric reductase. The Bacillus sp was grown in the Nutrient Broth media with various of  0; 20; 40; 60; 120; and 160 µM HgCl2 to know the response of the bacteria against mercury, The cell growth of Bacillus sp was measured by optical density (OD) method of at λ 600 nm. The mercuric reductase activity was assayed in the solution of MRA (Mercury Reductase Assay), then the oxidized NADPH was observed by the spectrophotometry method at λ340 nm. The result showed that the Bacillus sp has been resistant to media containing mercury at 120 µM, but the microbial growth was decreased by 50% in media containing mercury 80 µM. The Bacillus sp could produce highly the mercuric reductase enzyme at 16 hours of growth time with enzyme activity as 0.574 Unit/µg. The mercuric reductase from the bacteria has an  optimum activity at pH 6 and temperature 37 °C

Abstract

Hand sanitizer merupakan cairan atau gel pembersih tangan yang memiliki sifat sebagai antibakteri dalam menghambat hingga membunuh bakteri. Dengan demikian hand sanitizer lebih efektif dalam membasmi kuman. Berdasarkan penelitian The Centers for Disease Control and Prevention (CDC), hand sanitizer dengan kandungan alkohol minimal 60% dapat berfungsi sebagai antibakteri maupun antivirus. Hand sanitizer pernah menjadi barang langka di puncak pandemi pada tahun 2020, karena pembelian yang masif dari masyarakat Indonesia. Langkanya hand sanitizer di beberapa toko dan apotek membuat harganya menjadi mahal. Kegiatan Pengabdian Kepada Masyarakat ini bertujuan untuk meningkatkan pengetahuan akan pentingnya kesehatan dan kemungkinan penyebaran kuman terutama virus dan bakteri serta upaya untuk mengantisipasinya. Tujuan yang lain yaitu untuk meningkatkan keterampilan masyarakat dalam pembuatan hand sanitizer. Rangkaian kegiatan ini diselenggarakan di Desa Cangkir, Kecamatan Driyorejo, Kabupaten Gresik pada Bulan November-Desember 2021 secara luring dengan menerapkan protokol kesehatan dengan peserta Ibu-Ibu PKK dan Kader PKK. Kegiatan ini dilakukan melalui metode pendampingan dan demonstrasi dengan presentasi materi dan praktik langsung. Kegiatan ini memberikan dampak yang positif bagi masyarakat Desa Cangkir dalam menghadapi Pandemi Covid-19 saat ini. Hand sanitizer is a liquid or gel that has antibacterial properties to inhibit and kill bacteria. Thus, hand sanitizer is more effective in eradicating germs. Based on research from The Centers for Disease Control and Prevention (CDC), alcohol-based hand sanitizers that contain at least 60% alcohol can function as both antibacterial and antivirus. Hand sanitizer was once a scarce item at the height of the pandemic in 2020, due to massive purchases from the Indonesian people. The scarcity of hand sanitizers in some shops and pharmacies makes the price expensive. This Community Service activity aims to increase knowledge of the importance of health and the possibility of spreading germs, especially viruses and bacteria, and efforts to anticipate them. Another aim is to improve community skills in making hand sanitizers. This series of activities were held in Cangkir Village, Driyorejo District, Gresik Regency in November-December 2021 via offline by implementing health protocols with PKK women and PKK Cadres as participants. This activity is carried out through mentoring and demonstration methods with presentations and hands-on practice. This activity has a positive impact on the Cangkir Village community in dealing with the current Covid-19 Pandemic.

Abstract

Natto and Soy Yogurt are fermented food products that are very beneficial for improving health. This community service activity aims to socialize the benefits and make them known to the wider community. However, community service activities during the Covid-19 pandemic cannot be carried out face-to-face and have activities with the community in large numbers. Therefore, this community service activity was carried out through webinar and practical activities. Online seminars was chosen so that the coverage was wider and more people could participate in the pandemic. Face-to-face and community activities are carried out with a limited number of participants, according to health protocols. The level of success, benefit and acceptance of the community from this community service activity is known from the responses of the participants during the activities. Based on the results of the assessment/ response of participants who were present virtually and those present at the location, In general it can be concluded that the PKM activity entitled "socializing the benefits and making natto and soy yogurt through webinars and practices" is going well, but better preparation is still needed.The material presented by the resource person is suitable for the current pandemic conditions, easy to understand, easy to practice and can be developed for home businesses. The Zoom Meeting application can be used as a medium for community service activities in pandemic conditions even though it still cannot reach the wider community.  Keywords: Socialization, natto, soy yogurt, webinar, practical

Abstract

Tanaman obat merupakan jenis tanaman yang memiliki khasiat sebagai obat serta dapat dimanfaatkan sebagai bahan pembuatan obat tradisional yaitu jamu untuk membantu masyarakat dalam pemeliharaan kesehatan, pencegahan penyakit dan perawatan kesehatan terutama di era new normal dan post-pandemi Covid-19. Beberapa khasiat dari obat tradisional yang umumnya dimanfaatkan masyarakat antara lain meringankan gejala flu, batuk, sakit tenggorokan, sakit kepala, nyeri haid, dan lain-lain. Tanaman obat dapat dibudidayakan di lingkungan sekitar tempat tinggal seperti rumah atau pondok pesantren. Kegiatan pengabdian kepada masyarakat (PkM) ini bertujuan untuk memberikan edukasi publik khususnya di lingkungan pondok pesantren mengenai manfaat tanaman obat sebagai upaya untuk meningkatkan pengetahuan, keterampilan, dan kesehatan masyarakat. Rangkaian kegiatan ini diselenggarakan di Pondok Pesantren An-Nidhomiyah pada Bulan Juli 2022. Kegiatan ini diselenggarakan secara luring dengan tetap memenuhi protokol kesehatan. Peserta kegiatan ini yaitu santri putra dan putri pondok pesantren An-Nidhomiyah yang berjumlah 30 peserta. Kegiatan ini dilakukan melalui metode pendampingan dan workshop dengan pemaparan materi, praktik pembuatan obat tradisional, dan penanaman tanaman obat di halaman pondok. Kegiatan ini mendapatkan antusiasme yang besar dari peserta. Hasil survei kepuasan peserta menunjukkan trendline sebesar 87% yang termasuk dalam kategori sangat memuaskan untuk semua aspek, yang berarti sebagian besar peserta sangat puas terhadap kegiatan PkM ini, baik dari segi materi, metode, narasumber, hingga fasilitas yang diberikan. Medicinal plants are plants that have efficacy for medicinal and can be used as ingredients for making traditional medicines, namely herbs, to help the community in health care, disease prevention, and health care, especially in the new normal era and the post-pandemic Covid-19. Some of the benefits of traditional medicine commonly used by the community include relieving symptoms of flu, cough, sore throat, headache, menstrual pain, and others. Medicinal plants can be cultivated around the residence, such as in houses or Islamic boarding schools. This community service activity aims to provide public education, especially in Islamic boarding schools, about the benefits of medicinal plants to improve knowledge, skills, and public health. This activity was held at the An-Nidhomiyah Islamic Boarding School in July 2022. This activity was held offline by implementing health protocols. The participants of this activity were students of the An-Nidhomiyah Islamic boarding school, totaling 30 participants. The activity is carried out through mentoring and workshop methods, including presentations, traditional medicine production practices, and planting medicinal plants in the boarding school yard. This activity received great enthusiasm from the participants. The participant satisfaction survey results show a trendline of 87%, which is included in the very satisfied category for all aspects, which means that most of the participants were very satisfied with this PkM activity in terms of topics, methods, speakers, and facilities provided.

Abstract

Introduction: in general, residents of flats are a low-income group of people. Economic improvement for Rusunawa residents with concrete efforts is needed to be able to alleviate poverty. Purpose: the community service program's touch is aimed at initiating entrepreneurship among the residents of the flats through providing training on making a variety of microbial bioproducts with economic value, targeting PKK Rusunawa Penjaringan Sari, Rungkut-Surabaya mothers. Method: Community service activities are carried out in 3 stages, namely (1) survey and coordination, (2) workshops and training on making bioproducts from microbes, and (3) Evaluation of activities. Results: Community service participants who took part in the workshop on microbial bioproducts experienced an increase in knowledge about the meaning of microbial bioproducts, types and manufacturing technology. The number of participants who scored 60 and above increased by 95% in the post-test compared to the pre-test. The workshop was able to build participants' skills in making three types of microbial bioproducts, namely yogurt, biosurfactant and liquid organic fertilizer. Positive responses in the form of sentences were very useful for the workshop material, many were focused on the results of the feedback. Especially for yogurt bioproducts, almost all participants considered it very tasty, not inferior to commercial products already on the market. This shows an indication of a very high level of satisfaction. Conclusions: the understanding of community service participants on microbial bioproduct material increased through the workshop, indicated by an increasing of the participants who scored above of 60 in the post-test by 95% compared to the pre-test. Community service members' feedback through testimonials showed that 100% of the participants responded that it was very useful, indicating that the participants were very satisfied with the activity. The workshop can equip participants with skills in making yogurt bioproducts, biosurfactants and liquid organic fertilizers, which in the future are expected to be able to initiate entrepreneurship for the participants.
