Abstract

This research was conducted on digestive system of Java carp (Puntius javanicus). Gut of   fish samples was processed for histological preparation, stained with  periodic acid shiff’s (PAS) staining.The results of this research showed that the properties of mucus cells in the digestive tract of Java carp was comprised of goblet cells. The total number of goblet cells in the mid gut was higher  than that of in the foregut and hindgut.  Morphology of goblet cells  in the foregut and hindgut was round in shape but spherical on the midgut.

Abstract

Functional change of agricultural lands into non-agricultural lands will obviously have an impact on the dimin-ished or loss of habitat of the rice field eels (Monopterus albus Zuieuw). The aim of this research was to un-derstand the difference between (1) reproduction parameters (fecundity, gonadal maturity level (GML), and gonadal maturity index (GMI); (2) histological features of female and male gonadal development (oogenesis and spermatogenesis); (3) range of body length in each sexual phase; and (4) range of body length in each GML stage of captured eel from 4 sub-districts of Banyumas regency, Central Java (Ajibarang, Sumpyuh, Banyumas, and Sumbang). The total number of captured subjects were 631 eels; which consisted of 155; 227; 157; and 92 eels from Ajibarang, Sumpyuh, Banyumas, and Sumbang, respectively. We observed oogenesis in the stages of GML-1 (chromatin nucleolar stages and perinucleolar stage); GML-II (cortical alveolar formation stage or globular yolk stage), GML-3 (late globular yolk stage), GML-IV (mature or ripe stage), and follicle atretic stage. Also, we observed male gonadogenesis and spermatogenesis in the intersex. The range of GMI of captured eels was between 0.023 and 0.686. Eel in GML-I, GML-2, GML-3, and GML-IV had GMI within the range of 0.023-0.096; 0.096-0.425; 0.427-0.686; and >0.686, respectively. Eels achieved stage of GML-IV in body length range of 22.6-34.5 cm in Sumpyuh, 22.6-38.5 cm in Ajibarang, and 26.6-34.5 cm in Banyumas. There was no eel with the status of GML-IV in Sumbang, which had the most residential or functionally changed lands in its area. Based on these findings, can be concluded that the condition of rice fields or habitat of eel in functionally changed lands may affect eel’s growth and gonad maturation.

Abstract

Familia Cyprinidae dapat ditemukan di hampir seluruh sungai Indonesia termasuk Sungai Banjaran, Kabupaten Banyumas. Diferensial leukosit bertujuan mengetahui perbedaan persentase sel leukosit. Penelitian ini untuk mengetahui hubungan antara lokasi pengambilan sampel (hulu, tengah, dan hilir) dengan diferensial leukosit dan mengetahui gambaran apusan darah dari beberapa spesies ikan familia Cyprinidae. Pengambilan sampel menggunakan teknik purposive sampling pada tiga stasiun yang berbeda. Data yang diperoleh dianalisis dengan metode Analysis of Variance (ANOVA) dengan tingkat kesalahan 5% dan gambaran hasil pengamatan mikroskopis jenis-jenis leukosit. Hasil penelitian menunjukkan bahwa perbandingan persentase sel lekosit pada Ikan Nilem (Osteochilus vittatus), Ikan Brek (Barbonymus balleroides), dan Ikan Lunjar (Rasbora argyrotaenia) dengan lokasi Hulu, Tengah, Hilir Sungai Banjaran, Banyumas, berbeda secara signifikan (p<0,05). Persentase sel leukosit yang paling tinggi ada pada zona Hilir di Sungai Banjaran, Banyumas yaitu sel neutrofil dan sel monosit. Presentase sel neutrofil Ikan Nilem didapatkan sebesar 9,5%, Ikan Brek sebesar 8,5% dan Ikan Lunjar sebesar 9,4%. Presentase sel monosit pada Ikan Nilem sebesar 8,9%, Ikan Brek sebesar 8,7% dan Ikan Lunjar sebesar 8,5%. Sel neutrofil memiliki sitoplasma bergranula halus dan ditengahnya terdapat inti bersegmen. Sel eosinofil mempunyai sitoplasma berwarna lebih merah, inti yang terletak memanjang di tepi sel, memiliki granula besar. Sel limfosit memiliki sitoplasma berwarna biru gelap dan inti berbentuk bulat hingga oval. Sel monosit memiliki sitoplasma berwarna biru pucat dan bentuk inti bervariasi ada yang berbentuk seperti ginjal hingga yang berbentuk dua lobus besar.

The utilization of food is a process of energy transfers in the food chain and it is represented in the pyramid of numbers. The objective of this research was to analyze the structure community of fish and the guild compositions in Sempor Reservoir. This research applied a survey with purposive random sampling technique with four station in Sempor Reservoir. Sampling at each site was taken 4 replication with interval 1 monts.  The composition and status of the guild is carried out by mesuring the guild based on the position of the mouth, tooth type, gill filter type, and ratio of body length and intestine. Guilds are composed of compositions depicted by a number pyramid based on the number of species per station. The results of this reserach captured 439 individuals consists of 14 species and belong to 6 families that dominated by Cyprinidae. The number and species of fish caught were the most at the Bangkong river inlet (142 individuals) and the lowest at the center of the reservoir (60 individuals). Comparison of the composition of fish guilds at the four stations in the Sempor Reservoir shows that the number of omnivorous and carnivorous fish is obtained more than herbivorous fish. The composition of the guild at Kalianget river inlet (72.9%) and Central reservoir (42.7%) was dominated by carnivorous fish, while at Pengantalan river inlet (52.9%) and Bangkong (58.9%) were dominated by omnivorous fish. The composition of herbivorous fish is at least at all stations so that in general the composition of fish guilds in the Sempor reservoir has not been balanced.Keywords: species composition, guild, pyramid of number,Sempor reservoir  Abstrak. Komunitas secara fungsional menggambarkan fungsi spesies yang ditentukan dengan pemanfaatan sumber makanan (guild). Pemanfaatan makanan dalam rantai makanan merupakan proses transfer energi dan digambarkan dengan piramida jumlah. Tujuan dari penelitian ini adalah mengkaji komposisi dan  tingkat status guild  komunitas ikan yang tertangkap di  perairan waduk Sempor. Penelitian dilakukan dengan metode survei dan teknik purposive random sampling pada 4 stasiun di Waduk Sempor. Sampling dilakukan sebanyak 4 kali tiap bulan. Komposisi dan status guild dengan mengukur guild berdasarkan posisi mulut, tipe gigi, tipe tapis insang, dan rasio panjang tubuh dan usus. Komposisi guild digambarkan dengan piramida jumlah berdasarkan jumlah spesies per stasiun. Hasil penelitian diperoleh 439 individu terdiri dari  8 Familia dan 14 Spesies yang di dominasi familia Cyprinidae. Jumlah dan jenis individu ikan yang tertangkap paling banyak pada inlet sungai Bangkong (142 individu) dan terendah pada bagian tengah waduk (60 individu). Perbandingan komposisi guild ikan pada empat stasiun di waduk Sempor menunjukkan bahwa jumlah ikan omnivora dan karnivora lebih banyak diperoleh daripada ikan herbivora. Komposisi guild pada inlet sungai Kalianget (72,9%) dan Tengah waduk (42,7%) di dominasi oleh  ikan karnivora, sedangkan pada inlet sungai Pengantalan (52,9%) dan Bangkong (58,9%) di dominasi ikan omnivora. Abstract

Fisheries researchers reveal that many species of native wild fish are threatened with extinction due to uncontrolled fishing, things that can threaten their existence in nature, one of which is in the Banjaran River. The aims of this study were: to determine the quality of fish spermatozoa, to determine the gonadal maturity index (IKG), to determine the Gonad Maturity Level (TKG), and to determine the histological description of the male gonads in order to obtain data for conservation purposes. The research was conducted by survey method, the sample was taken by purposive random sampling technique. The results of the study during the March-May period showed that the sperm quality of Nilem Fish was best in May, which had a milt volume of 0.21 ml, a spermatozoa concentration of 2.3454 x109 cells/ml, Motility 3, and viability of 82.55%. In Brek Fish the highest volume of milt was found in May, namely 0.25 ml, the highest concentration of spermatozoa was in April 34.465 x109 cells/ml, with viability 88.62%, and motility 2.9. The best quality of Lunjar fish sperm was found in May which had a milt volume of 0.02 ml, spermatozoa concentration of 22.435 x109 cells/ml, 80% viability, and motility 3. All sperm samples were white with a pH of 7-8. Based on TKG and IKG, it can be seen that many Nilem fish are found spawning in April and May, Brek and Lunjar fish are found spawning every month. Histological structure of male gonads in Nilem, Brek and Lunjar fish had the same composition, in immature fish the spermatogenic cell composition observed was dominated by spermatocytes, spermatids, and spermatogonia, in mature fish the gonads were dominated by spermatozoa cells. and spermatids, whereas in post-spawning fish spermatozoa and spermatogonia were found in large number.
