Abstract

Sweet potato varieties (Ipomoea batatas) Pucuk Hitam Panyalaian and Madu Pucuk Hitam Panyalaian have the potential to be a superior commodity in West Sumatra, but the information of genetic diversity is very limited. Data on the diversity of sweet potato germplasm is beneficial in the selection of plants to obtain superior cultivars in plant breeding. Aim: The aim of the study was to find specific primers for the study of sweet potato genetic markers using Ipomoea batatas chloroplast genes matK. Methods: DNA extraction from sweet potato young leaves; designing forward and reverse primers Ipomoea batatas chloroplast genes matK; PCR; sequencing; bioinformatics analysis and species identification by comparing the NCBI database. Results: Plant DNA barcoding PCR using designed primers matK were successfully resulting single DNA band in different amplicon size in some samples. This indicating that the designed primers used were able to distinguish variation in one species.

Abstract

Snack types of chips, biscuits are sold using packaging. The snack packaging contains important information related to the nutritional component of the snack, expiration date, product halalness and other important information that is important for buyers to know. Elementary school students have insufficient attention and knowledge related to the information on snack packaging so that it can result in adverse conditions for elementary school students if they consume snacks that are not good for health. As a preventive step, it is necessary to educate elementary school children, one of which is knowledge of how to identify the meaning of symbols and information found on snack packages. The methods used in this activity are lectures and discussions by conveying information to elementary school students about the information found on snack packages and demonstrating several pictures and ways to identify symbols found on snack packages. The results of the evaluation showed that the students enthusiastically participated in this activity. In addition, there is also an increase in students' knowledge and ability to explain the symbols and information found on snack packaging.

Abstract

Rice is the main food source for Indonesians. The demand for rice to meet people's needs increases every year due to population growth and efforts to improve nutrition by the community. One of the problems that can affect rice production is drought. Drought is an environmental condition when plants do not get enough water to grow and develop optimally, which can cause a decrease in rice production. To find out how rice growth responds to drought, a study was carried out by giving drought simulation treatments using polyethylene glycol (PEG) on several rice varieties. This study used a completely randomized design that was arranged in a factorial manner with two factors. The first factor was the rice varieties (Harum, Situbagendit, Rosna) the second factor was 0% and 20% PEG concentration. The data obtained were then analyzed statistically using a two-way ANOVA test, and if the results were significantly different, then proceed with Duncan's test at the 5% level. The results showed that the drought simulation treatment had a negative effect on rice growth. Drought simulation treatment using 20% PEG resulted in a decrease in Kadar air relatif (KAR), root length, plant height, and root dry weight of rice. The highest decrease in KAR was found in the sensitive rice variety (Rosna), which was 43.42%. The highest average root length (7.99 cm) was on the sitabaendit variety, and the lowest (5.61) was on the rosna variety. The highest average crown height (17.32 cm) and the lowest (6.61) were on the rosna variety.

Abstract

Abstract Rice is a plant that has high economic value. According to FAO, Indonesia is the third largest rice consuming country in the world. However, rice will become stunted and its growth will be stunted if there is a lack of phosphorus. The PHT1 gene plays an important role in plant growth and development, because this gene plays a role in taking phosphate from the soil. The PHT1 gene is transcribed when there is drought, salt stress, and nutrition in plants. Genetic variation in a population will affect the survival of an individual. This study used the restriction enzyme ScaI. This study aims to analyze genetic variation in the PHT1 gene sequence in NCBI Popset 2400280979 rice using RFLP in silico. The results showed that there was genetic variation in the rice PHT1 gene sequence and two allele variations present in 24 rice gene sequences using the restriction enzyme ScaI.
