Abstract

Doxorubicin is one of chemotherapeutic agent used for cancer therapy. However, doxorubicin usage causes some adverse effects, such as lymphocyte, CD4+ and CD8+ cells number.  Therefore,  the  co-chemotherapeutic  agent  is  required  to  reduce  the imunosuppression  effect.  Cardamom  (Elettaria  cardamomum)  contains  terpenoid  1,8  cineol. This  research  aimed  to  know  the  effect  of  Elettaria  cardamomum  distillate  (ECD)  in combination with doxorubicin on Sprague Dawley rat’s hematology profile and the amount of CD4+  and  CD8+  cells.  The  experiment  was  done  for  13  days  using  6  groups  of  rats:  I doxorubicin (dox) 15 mg/kg BW; II dox + ECD5 mg/kg BW; III dox + ECD 50 mg/kg BW; IV dox  +  ECD  100  mg/kg  BW;  V  ECD  100  mg/kg  BW;  VI  control  without  treatment.  The hematology  profile  and  the  amount  of  CD4+  and  CD8+  were  counted  before  and  after treatment  using  flowcytometer.  The  results  show  that  ECD  increases  the  amount  of lymphocyte, white blood, CD4+ and CD8+ cells in dose dependent manner in doxorubicin treated rats. Based on the data, it can be concluded that ECD is potential to be developed as immunostimulant agent for chemotherapy.Keywords: Cardamom (Elettaria cardamomum), immunostimulant, hematology, CD4+, CD8+

Abstract

ABSTRACTImmunostimulatory agent has an important role in the treatment of infectious disease caused bypathogen. Roselle calyx (Hibiscus sabdariffa  immune system. The aims of this research was to know immunostimulatory activity of ethyl acetate fraction ofethanolic extract of roselle calyx on lymphocyte cell proliferation swiss strain mice and to identified thechemical compounds of the fraction.Roselle calyx was ext racted by maceration method using ethanol 96% as a solvent. The ethanolicextract was partitioned gradually by n-hexan and ethyl acetate. Series of ethyl acetate fraction were made toconcentration 10, 20, 50, 100, 200, 400 μg/mL and positive control PHA 10 μg/mL for tested to lymphocyte cellculture. Immunostimulatory act ivity test was done according to MTTAssay  Key words : roselle calyx, ethyl acetate fraction, MTTAssay , immunostimulatorymethod. Lymphocyte proliferat ion activity was analyzed statistically to the value of Optical Density (OD) result of the Enzyme LinkedImmunosorbent Assay (ELISA) reader using Kruskal Wallis Test continued by Mann Whitney Test (p<0,05).Identificat ion of the contents phenolic compounds and flavonoids performed by Thin Layer Chromatography(TLC).The result of the research showed that ethyl acetate fraction had lymphocyte cell proliferation activity atthe concentration of 50, 100, 200, and 400 μg/mL. Abstract

Mengkudu (Morinda citrifolia L.) adalah salah satu tanaman berkhasiat obat  yang sudah tidak asing lagi penggunaannya dalam pengobatan. Mengkudu telah diketahui memiliki efek farmakologis, antara lain efek analgesik, antihipertensi, dan imunomodulator. Penelitian ini bertujuan untuk membuktikan bahwa ekstrak n-heksana buah mengkudu dapat menghambat reaksi anafilaksis kutaneus aktif pada tikus Wistar jantan yang diinduksi vaksin hepatitis B. Penelitian ini menggunakan metode anafilaksis kutaneus aktif, kemudian dilanjutkan dengan pengamatan histopatologinya. Uji anafilaksis kutaneus aktif dilakukan selama dua minggu. Hewan uji dibagi menjadi lima kelompok yaitu kelompok : kontrol tanpa perlakuan, perlakuan dengan  ekstrak n-heksana buah mengkudu dosis   50mg, 100mg, dan 200mg/kgBB tikus, dan kelompok yang diberi kromolin dosis 2,16mg/kg BB tikus sebagai kontrol positif. Hewan uji disensitisasi 2 kali (1x setiap minggu) dengan vaksin hepatitis B (dosis 145µg/kgBB tikus) secara subkutan pada bagian punggung. Pada minggu kedua, hewan uji diambil secara acak 4 ekor setiap kelompok untuk dikurbankan dan diambil jaringan kulit beserta jaringan hatinya. Jaringan dibuat sebagai preparat histopatologi dengan pewarnaan hematoxyllin-eosin. Hasil penelitian menunjukkan bahwa pemberian ekstrak n-heksana buah mengkudu dosis 50mg, 100mg, dan 200mg/kgBB pada tikus jantan Wistar yang diinduksi vaksin hepatitis B dapat menghambat reaksi anafilaksis kutaneus aktif.

Abstract

It has been evaluated the influence of fermented horse milk into Balb/c mice on immunity against hepatitis A vaccine. The study employed a complete randomized design using thirty mice which were divided into 3 groups. Each group orally consumed fermented horse milk at dose 0.4 mL/20g body weight, levamisole (as positive control) at dose 0.4 mL/0.45 mg/20g body weight and distilled water (as negative control) at dose 0.4 mL/20g body weight respectively on a daily basis in addition to the standard diet for 46 days. The mice were immunized intraperitoneally (3 times) at d 7, d 28, and d 42, with 0.65 μg/20g body weight of hepatitis A vaccine. Blood samples were collected from plexus retroorbitalis using heparinized capillary one week after first (d 14), second (d 35) and 4 days (d 46) after third immunizations, and then mice were sacrificed to isolate the lymphocytes. The IgM, IgG and IgA titers in the sera and the culture media were measured by ELISA.The IgM (except d 46), IgG (subtype IgG1 (except d 14), IgG2a, IgG2b, IgG3 (except d 35), and IgA titers in the sera significantly higher (p<0.05) than controls. This difference was associated with enhanced in vitro antibodies secretion by cultured lymphocytes isolated from the spleen for IgA. The result suggested that orally fermented horse milk supplementation inhances immunity against hepatitis A vaccine in Balb/c mice. The SDS-PAGE showed that 30-kDa proteins were present in fermented horse milk as well as fermented cow milk, but 7.4-kDa protein present only in fermented horse milk.Key words : fermented horse milk, immunity, hepatitis A vaccine, Balb/c mice.

Abstract

Berdasarkan studi sebelumnya diperoleh bahwa kadar flavonoid total subfraksi etil asetat Myrmecodia tuberosa (non Jack) Bl memiliki korelasi positif terhadap aktivitas proliferasi sel limfosit. Subfraksi V (kelima) menunjukkan nilai indeks stimulasi proliferasi sel limfosit tertinggi serta memiliki kadar flavonoid total terbesar (6,158±1,298) diantara semua subfraksi. Penelitian ini bertujuan mengisolasi dan mengidentifikasi senyawa flavonoid yang dikandung oleh subfraksi V. Metode pemisahan yang dilakukan adalah melakukan hidrolisis subfraksi V, kemudian dikromatografi lapis tipis preparatif dan dianalisis secara spektroskopi. Hasil spektroskopi  UV-Vis isolat memiliki puncak 1 pada panjang gelombang 324 nm dan puncak 2 pada panjang gelombang 284 nm, spektroskopi infra merah memiliki serapan pada 3.443 cm-1. Data 1H-NMR menunjukkan adanya serapan pada ? 6,74; ? 6,73; ? 6,62; ? 6,10; ? 7,82; ? 7,39; doublet of doublet pada ? 1,14, doublet pada ? 3,47 - ? 3,60 dan singlet pada ? 3,65 serta adanya serapan pada ? 17,81; 71,47; 74,19; 74,51 dan ? 76,78 dari spektrum 13C-NMR. Hasil spektroskopi massa terlihat serapan pada m/z 391,10. Berdasarkan data-data yang diperoleh, senyawa hasil isolasi diduga merupakan senyawa isoflavon.

Abstract

Objective: Centella asiatica L.Urban contains triterphenoid saphonin, flavonoid, and pectin that believed to have an immunomodulatory effect toward the Immunoglobulin-G in Wistar strain male mices which Induced by BCG vaccine. A study was conducted to find out the optimum dose and formulation of in enhancing on the level of IgG. It’s therefore expected that Cantella asiatica L.Urban can be used as an immunomodulatory suplement in both animals and human.Methods: Centella asiatica L.Urban were macerated with ethanol 50%. This was an experimental study with the post-test control group design. The samples in this study were 35 Wistar strain male mices which divided into 7 groups: negative control [Aquadest 0,5mL/20g BW]; control immunosupresant [Prednison® 0,06mg/20g BW]; control immunostimulant [Levamisol® 0,445mg/20g BW]; ethanolic extract of Cantella asiatica L.Urban with a dose of 50mg; 100mg; 150mg; 200mg/Kg B.W were administered orally fo 10 day, after the treatments implemented, BCG [Bacillus Calmette Guerin] vaccine was infected intraperitoneally on day 10th, 14th, 21st, and 28th. The most effective doses of ethanolic extract of Cantella asiatica L.Urban was further studied for its effect on hormonal assay using ELISA reader which compared with the effects of Prednison® and Levamisol®. Based on simplex lattice design to find the optimum proportion of coefficients a, b, and ab of the equation Y = a (A) + b (B) + ab (A) (B), where A is lactose and B is amylum.Results : Oral administration of ethanolic extract of Cantella asiatica L.Urban at dose of 50mg and 100mg/Kg B.W increased the immunity which showed elevated levels of IgG in the blood serum in Wistar strain male mices that had been induced by BCG vaccine. The optimum formula capsul of Centella asiatica L.Urban extract that use combination of 80% Amylum and 20% Lactose which produce the optimum parameters of disintegration time (≤15minutes) and CV of weight uniformity (≤5%)with the test results had no significant difference to the prediction results of Simplex Lattice Design [p>0.05].Keywords:  Centella asiatica L.Urban extract, Immunomodulator, simplex lattice design

Abstract

Ginjal merupakan organ vital yang berperan sangat penting dalam mempertahankan kestabilan lingkungan dalam tubuh dengan fungsi utama yaitu filtrasi oleh glomerulus, reabsorbsi dan sekresi tubulus. Kerusakan sel ginjal sampai kematian sel akan menyebabkan fungsi ginjal terganggu. Efek paparan senyawa radioaktif salah satunya dapat menyebabkan terjadinya gangguan sel-sel ginjal. Penelitian ini bertujuan untuk mengetahui pengaruh infusa wortel (Daucus carota L.) dalam mencegah gangguan sel ginjal akibat paparan uranium. Uji proteksi dilakukan dengan membagi 30 tikus jantan dewasa menjadi enam kelompok masing-masing kelompok terdiri dari 5 ekor. Kelompok I tanpa diberi perlakuan, kelompok II diberi uranium 8 ppm dosis 0,01 mL/g BB sebagai kontrol negatif, kelompok III sebagai kontrol positif dengan vitamin C 200 mg/70 kg BB yang diberikan 15 menit sebelum pemajanan uranium, kelompok IV,V, dan VI sebagai kelompok uji proteksi kerusakan sel ginjal diberi infusa wortel (Daucus carota L.) berturut-turut 10%, 20%, 30% dengan dosis 0,01 mL/g BB 15 menit sebelum diberi uranium 8 ppm dosis 0,01 mL/g BB. Lima hari kemudian hewan dikorbankan dan diambil organ ginjalnya untuk dilakukan pemeriksaan makroskopis dan mikroskopis berupa pengamatan kondisi fisik organ ginjal, pengamatan terhadap nekrosis sel ginjal, dan skoring tipe kerusakan untuk menganalisis efek proteksinya. Hasil pemeriksaan histopatologi menyimpulkan bahwa infusa wortel dapat mengurangi kerusakan sel ginjal akibat paparan senyawa radioaktif uranium.Kidney is the vital organ which has an importance role in maintaining environmental stability of the body, with the main functions are glomerulus filtration, reabsorption and secretion by tubulus. The damage of kidney cells will reduce kidney function. If a kidney is exposed to radioactive substance it could deteriorate the kidney. The aim of this research was to find out the effect of carrot infusion used to prevent kidney cells disorder due to uranium exposure. Thirty rats wistar strain divided into 5 groups were used in this study, comprising of 5 rats in each groups throughout the experiment. Group I was a normal control group. Group II was given uranium of 8 ppm 0,01 mL/g BW as negative control group. Group III was given vitamin C of 200 mg/70 kgBW 15 minute prior to uranium exposure. Group IV-VI were given carrot infusion at 10%, 20%, 30% dose of 0,01 mL/g BW 15 minutes prior to uranium exposure. The rats were sacrificed in day-5, to observe the kidney?s damage upon macroscopy and microscopy examination, involving physical condition of the kidney, necrosis of kidney cells and scoring the damage of kidney cells. The microscopic examination toward necrosis of kidney cells showed that carrot infusion can reduce cell damage due to radioactive compound of uranium.
