Abstract

The study was designed to investigate the optimation of preparation of transdermal patch sodium diclofenac matrix type. In this study, matrix-type base containing diclofenac sodium were prepared using polymeric combination of EC N-20  and PVP K-30 in different ratios ( 9:1 (Formula I); 8:2 (Formula II); 7:3 (Formula III)) and 6:4 (Formula IV). The moisture content was calculated as a difference between initial and final weight with respect to final weight. The homogeneity of patch surface was determined using Scanning Electron Microscope (SEM) Released test using dissolution tester were carried out in 500 mL of  phosphate buffer salin pH 7.4 ± 0.05 at temperature 37 ± 0.5°C with speed of swirl 50 rpm. The concentration of sodium diclofenac released was determined using spectrophotometric method at l 276 nm. Data was analyzed by statistic programmed of SPSS using one way analysis of variance with degree of believed 95% (α = 0.05).  It is concluded that the polymeric combination EC N-20 and PVP K-30 in composition 7:3 (Formula III) was the best formulation. ABSTRAK Telah dilakukan penelitian tentang optimasi sediaan transdermal patch natrium diklofenak tipe matriks. Pada penelitian ini, basis matriks mengandung natrium  diklofenak dibuat dengan kombinasi polimer  etilselulose (EC) N-20  dan polivinilpirolidon (PVP) K-30 dengan berbagai perbandingan yaitu 9:1 (Formula I); 8:2 (Formula II); 7:3 (Formula III)) dan 6:4 (Formula IV). Moisture content (MC) ditentukan dengan cara berat awal dikurangi berat akhir dan dibagi dengan berat akhir. Homogenitas permukaan patch ditentukan dengan menggunakan Scanning Electron Microscope (SEM). Uji pelepasan natrium diklofenak dilakukan dengan alat disolusi dan menggunakan dapar fosfat salin  pH 7,4 ± 0.05 sebanyak 500,0 mL, kecepatan pengadukan 50 rpm dan percobaan dilakukan pada suhu 37 ± 0,5°C. Kadar natrium diklofenak yang terlepas ditentukan dengan metoda spektrofotometer pada panjang gelombang (l) 276 nm. Data yang diperoleh dianalisa secara statistik menggunakan program SPSS dengan derajat kepercayaan 95% (α = 0,05).  Kesimpulan yang didapatkan adalah komposisi polimer EC N-20 dan PVP K-30 dengan perbandingan  7:3 (Formula III) adalah formula yang terbaik.

Abstract

The research has been done for the isolation and identification of lactic acid bacteria (LAB) from mango (Mangifera indica L.). This research aimed to isolated of lactic acid bacteria that is in mango (Mangifera indica L.) and determine the characteristics of lactic acid bacteria isolate (LAB) of mango (Mangifera indica L.). The method used is spoiled  technique of mango (Mangifera indica L.) and isolation using selective media MRS Broth and MRS Agar. The identification isolate of lactic acid bacteria (LAB) used methods macroscopically and microscopically with indirect coloring, gram staining and used biochemical with katalase testing. The results obtained in the form of characteristic isolate of lactic acid bacteria displayed form of bacteria with circle, smooth surface, curve, entire side and white. The microscopically displayed stick form of bacteria and purple with gram coloring

Abstract

Indonesia biodiversity has the second highest number of indigenous medical plants after the amazon rain forest, meanwhile Indonesian people usually used plants as a medicine source and some medicine plants have been developed as modern drugs. One of medicine plant in Indonesia that usually used as a refreshment was grass jelly (Stephania capitata (Blume) Spreng). This plant frequently found in Sumatra, Borneo and Java island and the leaf of grass jelly has been reported as a medicine against stomach complaint (diarrhea) and fevers. This research was designed to investigate the secondary metabolites and the efficacy of leaf extract of grass jelly as anti diarrhea. The leaves extraction was done by using maceration with ethanol solvent at room temperature. Meyer reagent, dagendroff reagent, methanol, magnesium band, hydrochloric acid, iron (III) chloride, anhydrous acetic acid, sulfuric acid, chloroform were used for identification of secondary metabolites from grass jelly leaves. For potency anti diarrhea, animals were divided into six group: diarrhea group, treatment group with loperamid HCl and treatment group of leaf extract of grass jelly. The results showed that the leaf extract of grass jelly contain saponin, phenol, sterol and triterpen and the treatment with leaf extract of grass jelly significantly decreasing time duration of diarrhea.

Abstract

The sodium diclofenac realeased from transdermal patch of combination of EC N–20 and PVP K–30 with ratio 6:4 with plasticizer PEG 400 5% (formula 1), PEG 400 10% (formula 2) and PEG 400 20% (formula 3) was investigated. The percentage of moisture contents was calculated as a difference between initial and final weight with respect to final weight. The homogeneity of patch surface was determined using fluorescence microscope and scanning electron microscope (SEM). Released test using dissolution apparatus were carried out in 500 mL of phosphate buffer saline pH 7.4 ± 0.05 at temperature 37 ± 0.5°C with speed of swirl 50 rpm. Results were analyzed by statistic programmed of SPSS using one way analysis of variance with degree of believed 95% (α = 0,05), showed that that the combination of EC N–20 and PVP K–30 at ratio 6:4 with plasticizer PEG 400 5% was the best choice for manufacturing transdermal patch based on physicochemical and the release profile. ABSTRAK Telah dilakukan penelitian pelepasan sodium diklofenak dari transdemal Patch kombinasi matriks EC N–20 dan PVP K–30 dengan rasio 6:4 dan plasticizier PEG 400 5 % (formula 1), PEG 400 10% (formula 2), dan PEG 400 20 %. Persentase kadar air dihitung sebagai perbedaan antara berat awal dan akhir. Homogenitas dari permukaan Patch di determinasi menggunakan Mikroskop flourescence dan SEM. Uji pelepasan dilakukan menggunakan alat uji dissolusi dalam 500 mL larutan salin fosfat buffer pH 7.4 ± 0.05 pada suhu 37 ± 0.5°C dengan kecepatan pengadukan 50 rpm. Hasil di analisis menggunakan program statistik SPSS analisis one way variance (ANAVA) dengan tingkat kepercayaan 95% (α = 0,05), menunjukkan bahwa kombinasi dari EC N–20 and PVP K–30 pada rasio 6:4 dengan plasticizer PEG 400 5% merupakan pilihan terbaik untuk industri Patch transdemal berdasarkan fisikokimia dan profil pelepasan.

Abstract

ABSTRAK Pelepasan natrium diklofenak dari Sediaan Transdermal Patch Tipe Matriks dengan kombinasi polimer etil selulosa (EC) N-20 dan polivinilpirolidon (PVP) K-30 telah diteliti. Pada penelitian ini sediaan transdermal patch tipe matriks yang mengandung natrium diklofenak dibuat dengan kombinasi polimer etil selulosa (EC) N-20 dan polivinilpirolidon (PVP) K-30 pada perbandingan yang berbeda ( 9:1 (Formula I); 8:2 (Formula II); dan 7:3 (Formula III)). Persentase (%) moisture content dihitung dari perbedaan berat relatif pada penimbangan akhir. Uji homogenitas permukaan patch dilakukan dengan menggunakan metode Scanning Electron Microscopy (SEM). Uji disolusi dengan media larutan dapar fosfat salin pH 7,4 ± 0.05 sebanyak 500,0 mL yang terlebih dahulu dipanaskan sampai mencapai temperatur percobaan 37 ± 0.5°C diaduk pada kecepatan 50 rpm. Hasil dianalisis menggunakan SPSS dengan metode ANOVA one way dengan tingkat kepercayaan 95% (a = 0,05). Berdasarkan penelitian ini, penggunaan polimer kombinasi antara etil selulosa (EC) N-20 dan polivinilpirolidon (PVP) K-30 dapat meningkatkan fluks pelepasan natrium diklofenak dari sediaan patch natrium diklofenak. Perbandingan 7:3 dari etil selulosa (EC) N-20 dan polivinilpirolidon (PVP) K-30 adalah kombinasi yang terbaik untuk sediaan patch natrium diklofenak karena konsistensi bentuk fisik serta nilai fluks pelepasan dan penetrasi yang dihasilkan lebih besar dari perbandingan lainnya. Abstract

A research has been conducted on the antibacterial activity of the santol leaves extract (S.koetjape Merr.) against S.aureus and E.coli. The santol leaves extracts were prepared by maceration used methanol as solvent and the methanol extract was then fractionated. The antibacterial activity was determined by measured the diameter of inhibition zones in difusi method. The result showed that santol leaves extracts and fractions has antibacterial activity against S.aureus and E.coli. The effective concentration as an antibacterial of methanol extracts is at 10%, and of n-hexane fractions is at 5%. Keywords : S.koetjape Merr, Santol leaves, Antibacteria Abstrak Telah dilakukan penelitian uji aktivitas antibakteri ekstrak daun kecapi (S.koetjape Merr.) terhadap S.aureus dan E.coli. Ekstrak daun kecapi dibuat secara maserasi dengan pelarut metanol, lalu difraksinasi. Uji aktivitas antibakteri ekstrak daun kecapi diukur berdasarkan luas daerah zoa bunuh pertumbuhan bakteri dengan metode difusi agar. Hasil uji menunjukkan bahwa ekstrak dan fraksi daun kecapi memiliki aktivitas antibakteri terhadap bakteri S.aureus dan E.coli. Konsentrasi efektif antibakteri ekstrak metanol adalah 10% dan fraksi n-heksana adalah 5%. Kata Kunci : S.koetjape Merr, Daun Kecapi, Antibakteri

Abstract

Each plant is host to one or more endophytes contain bacteria and fungi. Endophytic fungi could potentially have an activity and produce secondary metabolites same as its host. Yacon leaf (Smallanthus sonchifolius) has been known to have antimicrobial activity that allow the endophytic fungi also has potential as antimicrobial. The purpose of this research was to isolated the endophytic fungi from Yacon leaf which has activity as antifungal and can produce secondary metabolites same as its host. The method that used to test the activity of endophytic fungi are using direct planting method, whereas the identification of secondary metabolites using Thin Layer Chromatography (TLC) and spray reagent. The results were obtained that the endophytic fungi has an activity as antifungal against Candida utilis, Candida albicans and Malassezia Purpur, along with the presence of secondary metabolites that same as its host.Key words: endophytic fungi, Smallanthus sonchifolius, antifungal, Thin Layer Chromatography (TLC)ABSTRAKSetiap tanaman mengandung satu atau lebih mikroba endofit yang terdiri dari bakteri dan jamur. Jamur endofit berpotensi memiliki aktivitas dan menghasilkan metabolit sekunder yang sama seperti inangnya. Daun Yakon (Smallanthus sonchifolius) telah diketahui memiliki aktivitas sebagai antimikroba sehingga memungkinkan adanya jamur endofit yang juga berpotensi sebagai antimikroba. Tujuan dari penelitian ini adalah didapatkan isolat jamur endofit dari daun Yakon yang memiliki aktivitas sebagai antijamur dan dapat menghasilkan metabolit sekunder yang sama seperti inangnya. Metode yang digunakan untuk menguji aktivitas isolat jamur endofit adalah menggunakan metode tanam langsung, sedangkan identifikasi metabolit sekunder menggunakan metode Kromatografi Lapis Tipis (KLT) dan pereaksi semprot. Hasil penelitian yang diperoleh adalah adanya isolat jamur endofit yang memiliki aktivitas sebagai antijamur terhadap jamur Candida utilis, Candida albicans, dan Malassezia purpur, serta adanya metabolit sekunder yang sama seperti inangnya.Kata kunci: Jamur endofit, Smallanthus sonchifolius, antijamur, Kromatografi Lapis Tipis (KLT)

Abstract

Sunscreen can protect the skin from the UV radiation. One of the plants that have the potential to be utilized as active ingredients of sunscreen lotion is miana leaves (Coleus atropurpureus Benth). Lotion made in 3 formulas F1, F2 and F3. Lotion tested its stability with a freeze-thaw method, and evaluated organoleptic, homogeneity, pH, viscosity and scatterness and tested sunscreen activity of lotion preparation. The preparation shows good homogeneity, and has oleum rosae odor and a thick consistency with the lotion color is brown (F1), dark brown (F2) and slightly greenish brown (F3). Spreading, viscosity, and pH of the lotion meet the Indonesia Standart requirement. The lotion formula comes in the category of fast tanning (F2) and extra protection (F3) sunscreen.

Abstract

Telah dilakukan penelitian mengenai isolasi dan identifikasi bakteri asam laktat (BAL) dari buah mangga (Mangifera indica L.). Penelitian ini bertujuan untuk memperoleh isolat bakteri asam laktat yang ada di dalam buah mangga (Mangifera indica L.) dan mengetahui karakteristik isolat bakteri asam laktat (BAL) dari buah mangga (Mangifera indica L.). Metode yang digunakan yaitu teknik pembusukan buah mangga (Mangifera indica L.) dan isolasi dengan menggunakan media selektif MRS Broth dan MRS Agar. Di identifikasi isolat bakteri asam laktat (BAL) secara makroskopik dan mikroskopik dengan pewarnaan tak langsung dan pewarnaan gram serta secara biokimia dengan uji katalase. Hasil yang di peroleh berupa karakteristik isolat bakteri asam laktat yang memperlihatkan bentuk bakteri bulat dengan permukaan halus, elevasi cembung, tepi entire dan berwarna putih susu dan dari pengamatan mikroskopik memperlihatkan bentuk bakteri batang dan berwarna ungu dengan pewarnaan gram.

Abstract

Daun Miana (Coleus atropurpureus Benth) merupakan salah satu tanaman tradisional yang digunakan sebagai obat untuk menyembuhkan beberapa penyakit, seperti batuk, perangsang nafsu makan, dan penetralisir racun (antitoksik). Penelitian ini bertujuan untuk mengetahui aktivitas antioksidan fraksi etil asetat daun miana. Daun miana (Coleus atropurpureus Benth) diekstraksi dengan pelarut etanol menggunakan metode maserasi kemudian difraksinasi secara bertahap menggunakan pelarut n-heksana dan etil asetat. Uji aktivitas antioksidan menggunakan metode radikal 2,2-difenil-1-pikrilhidrazi (DPPH), dimana absorbansi yang diukur pada ?maks 516,4 nm dengan menggunakan alat spektrofotometer UV-Vis. Hasil penelitian menunjukkan bahwa fraksi etil asetat daun miana memiliki aktivitas antioksidan terhadap radikal DPPH dengan nilai IC50 33,768 ppm
