Abstract

This research was conducted to qualitatively analyze the phytochemical content of kepok banana (Musa paradisiaca) peel extract and the effectivity of the extract as antibacterial against Aeromonas hydrophila. Fresh yellow color banana peels were washed, drained, then cut into pieces, crushed and dried at room temperature. Extraction was carried out by maceration method with 70% and 95% ethanol as solvent. A total of 90 g was put in an elemeyer glass then added with solvent until all the banana peels were submerged. The immersion time was 3x24 hours at room temperature while stirring occasionally so that the extraction occurs properly. Furthermore, the extract was filtered with Whatman paper no. 42 to separate the residue from the filtrate. The filtrate was then concentrated using a rotary evaporator. Phytochemical examined consisted of alkaloid, flavonoid, sternoid, triterpenoid, saponin, and tannin. Inhibition test against A. hydrophila was carried out by disc diffusion method using 5 different extract concentrations as treatments, namely A: 0%, B:10%, C:20%, D:30%, E:40%, F:50%. In the inhibition test, the paper disc was dipped in each treatment and then placed on agar media previously stocked with bacteria. The inhibitory zone was measured based on the clear area formed around the disc and observed at 24, 48, and 72 hours. Phytochemical test found that phenol and tannin were only detected in extracts with 95% ethanol while terpenoids and steroids were only found in  ethanol70%.  Flavonoid was detected in the extract using either ethanol70% and 95%. The results of the inhibitory test showed that the kepok banana peel extract could inhibit the growth of A. hydrophila bacteria after 48 hours of incubation. Inhibition zones formed at both 48 hours and 72 hours tended to increase in line with the increase in concentration even though the inhibitory power produced was categorized as "medium".

Abstract

This study aimed to obtain the amount of organic waste, nitrogen and phosphorus that wasted to  lake and measuring water quality parameters, temperature, dissolved oxygen (DO), acidity (pH) and dissolved solids (TDS) in the area of fixed net cages at Lake Tondano in the villages of Eris, Talikuran and Kaima. This research was carried out in Lake Tondano, Eris, Talikuran and Kaima villages . The time of the study was carried out from March to July 2022. This study used     a descriptive exploration method. Data obtained included primary and secondary data. Primary data otained from interview results and parameter measurements of the water quality in the field (in situ). Secondary data obtained through various resources such as journals and the results of previous studies related to the topic under study. The data of organic waste, nitrogen, and phosphorus were analyzed descriptively using Schmittou method. The results of analyzing the parameters quality of water was presented in the form of tables and graphs then compared with the class of water quality standards in Government Regulation Number 22, 2021 regarding the Implementation of Environmental Protection and Management. The results showed that the amount of organic waste in Eris Village was 874.74 tons/year, Talikuran village 170.52 tons/year, and kaima village 105.80 tons/year, the amount of nitrogen waste in Eris village is 49.86 tons/year, Talikuran village 9.67 tons/year, and kaima village 6.00 tons/year, the amount of phosphorus waste in Eris Village is 17.24 tons/year, Talikuran Village 3.35 tons/year and Kaima Village 2.08 tons/year. The temperature in Eris Village is in the range of 27.4-29.5 °C, Talikuran Village is 26.5-30.3 °C and   Kaima Village is 26.6-30 °C, DO in Eris Village is 5.25-6, 74 mg/L, Talikuran Village 3.76-6.69 mg/L and Kaima Village 4.86-7.18 mg/L, pH in Eris Village was in the range of 7.2-7.9, Talikuran Village 7.1-7.8 and Kaima Village 7,3-7,8, TDS in Eris Village is in the range of 0.104-0.114 g/L, Talikuran Village is 0.117-0.160 g/L and Kaima Village is 0.104-0.114 g/L. The measured water quality parameter data still meets the water quality standard value.

Abstract

The purpose of this study was to find out the survival rates of betta fish larvae (cupang) that were given life feed Alona sp., boiled egg yolk and commercial feed for 21 days. Data analysis was performed by one-way ANOVA, and continued with BNJ (Tukey) test. The result of the survival rate is as follow treatment with life feed Alona sp. 56%, treatment with boiled egg yolk 26%, and treatment with commercial feed 0%. The results of statistical tests show there are significant differences.  Significant value p <.0002 (<.05). Water quality parameter measured during the study was temperature of 26 ºC.Keywords: betta fish larvae, life feed, Alona sp., survival rateABSTRAKTujuan penelitian ini adalah untuk mendapatkan laju kelangsungan hidup larva ikan betta (cupang) yang dibeerikan pakan alami Alona sp., rebusan kuning telur dan pakan komersil selama 21 hari.  Analisis data dilakukan dengan ANOVA one way, dan dilanjutkan dengan uji BNJ (Tukey).  Hasil laju kelangsungan hidup adalah sebagai berikut, perlakuan dengan pakan alami Alona sp. 56%, perlakuan dengan rebusan kuning telur 26%, dan perlakuan dengan pakan komersil 0%.  Hasil uji statistik menunjukan adanya perbedaan yang nyata.  Nilai signifikan p <.0002 (<.05).  Parameter kualitas air yang diukur selama penelitian adalah suhu 26ºC.Keywords: larva ikan betta, pakan alami, Alona sp., kelangsungan hidup

Abstract

The objective of research was to isolate probiotic bacteria from catfish intestine and to examine its effects on the growth and resistance of carp against Aeromonas hydrophila.  The probiotic isolated from intestine was grown on MRS agar.  Fish used in this research was juvenile of carp with an average weight of 5.5 g obtained from Freshwater Aquaculture Board at Tatelu Village, North Sulawesi Province.  Before running the experiment, the juveniles were adapted for one weeks in 15 glass aquaria at a density of 15 individuals each.  After acclimatization, the fish were fed diet supplemented with probiotic at different concentrations namely 1x109 , 1x108 , 1x107,  1x106 cfu/mL for three weeks. The fish were fed two times a dayat 08.00 am and 17.00 pm with a dose of 5% of body weight per day. At the end of feeding, the fish were challenged with A. hydrophila.  Data collected included average growth rate, absolute growth, feed efficiency, and food conversion ratio.  Research results found that the addition of probiotic into feed was  able to increase growth and feed efficiency and reduce food conversion ratio as well (p<0.01). The best growth, feed efficiency and food conversion ratio were obtained in fish fed diet added with probiotic bacteria at 1x108 cfu/mL.  The highest survival rate was also observed in fish probiotic diet containing 1x108 cfu/mL.  As conclusion, probiotic bacteria isolated from catfish intestine was potential to improve growth, feed efficiency, reduce food conversion ratio and increase resistance of fish against bacterial pathogen.Keywords: probiotic, carp, Aeromonas hydrophila, feed efficiency, food conversion ratioABSTRAKPenelitian ini bertujuan untuk mengisolasi bakteri probiotik dari usus ikan lele dan menguji pengaruh pemberian probiotik dari usus ikan lele terhadap pertumbuhan dan resistensi ikan mas terhadap infeksi Aeromonas hydrophilla. Sumber probiotik diisolasi dari usus ikan lele menggunakan media MRS (de Man Rogosa Sharpe) dan bakteri Aeromonas hydrophilla ditumbuhkan pada media TSA (Tryptic Soy Agar). ikan uji diambil dari Balai Budidaya Air Tawar Tatelu , Provinsi Sulawesi Utara . ikan diaklimatisasi selama seminggu dalam 15 akuarium dengan kepadatan 15 ekor/akuarium dan berat awal rata-rata 5,5 g. Setelah diaklimatisasi ikan diberi pakan yang ditambahkan bakteri probiotik sebagai perlakuan dengan konsentrasi berbeda yaitu 1x109 , 1x108 , 1x107,  1x106 cfu/mL selama empat minggu sebanyak 5% /berat tubuh/hari dengan frekuensi pemberian pakan dua kali sehari yaitu jam 08.00 pagi dan jam 17.00 sore. Setelah diberi perlakuan maka diuji tantang dengan bakteri Aeromonas hydrophilla.  Data yang dikumpulkan terdiri dari laju pertumbuhan harian, pertumbuhan mutlak, efesiensi pakan, konversi pakan dan uji tantang terhadap bakteri patogen. Hasil penelitian mendapatkan bahwa penambahan bakteri probiotik dalam pakan mampu meningkatkan pertumbuhan, efesiensi pakan dan menurunkan konversi pakan (p<0.01). Pertumbuhan, efesiensi pakan dan konversi pakan yang terbaik dicapai pada ikan yang diberi pakan dengan penambahan bakteri probiotik 1x108 cfu/mL.  Kelangsungan hidup ikan mas yang diuji tantang dengan bakteri patogen yang paling tinggi dicapai pada ikan yang diberi pakan 1x108 cfu/mL. Sebagai kesimpulan bahwa bakteri probiotik yang diisolasi dari usus ikan lele berpotensi meningkatkan pertumbuhan, efesiensi pakan, menurunkan konversi pakan serta meningkatkan sistem kekebalan tubuh pada ikan mas.Kata Kunci : Probiotik, Ikan Mas, Aeromonas hydrophilla, Pertumbuhan, Efesiensi Pakan, Konversi Pakan

Abstract

The fishery sector which has great profit prospects can still be developed by cultivating fish better by paying attention to the condition of the water quality. In fish farming, it is very important to study water quality conditions because water quality parameters are a limiting and life-supporting factor for the types of aquatic biota cultivated in a waters including tilapia (Oreochromis niloticus. This research was conducted to assess water quality, both physical, chemical, and aquatic biology of Lake Tondano to support the feasibility test activities of Lake Tondano waters for Cage Netting Cage cultivation media. The research method used was purposive sampling. Primary data comes from primary data which is the result of measurements of physico-chemical parameters of water quality directly in the field (in situ) and supported by secondary data on several physical and chemical parameters of Lake Tondano waters as well as data on water quality of the cultivation media with the Netting Cage system. The physical and chemical parameters of Lake Tondano waters at the three stations were analyzed using the suitability scoring method for the location of Tondano Lake waters to be used as a maintenance medium fish with a Stuck Net Containment system. The results of the study concluded that the physical and chemical quality of water in Urongo waters was suitable for net cage culture.
