Abstract

Rhodamin B is dangerous for body due to its ability to improve oxidative stress on various reproduction tissues on female. However, there is no significant scientific proof that Rhodamin B exposure influences VEGF and the number of arteriole on endometrium. VEGF and arteriole is crucial to angiogenesis process of endometrium. Abstract

Objective: To determine the effect of Bryophyllum pinnatum treatment in modulating immune response and the pregnancy outcomes of pregnant pristane-induced lupus mice model.Methods: Sixteen Balb/c mice were intraperitoneally injected with single 0.5 cc pristane to induce lupus manifestations. After 12 weeks of injection, mice were mated and considered as gestational day 0 (GD0). Mice were divided into 4 groups based on the dosages of Bryophyllum pinnatum: control (no treatment), B1 (10.5 mg/kg), B2 (21 mg/kg), and B3 (42 mg/kg). The treatment was given orally every day started from GD9 until 9 days. At the end of the study, blood pressure and fetal size were measured. Serum anti-dsDNA and urine albumin levels were measured by ELISA. Spleen T helper (Th) and mature B cells percentages were measured by flow cytometry.Results: Administration of Bryophyllum pinnatum reduced the percentages of Th1 (p=0.006), Th2 (p=0.005), Th17 (p=0.000), and mature B cells (p=0.007) in dose-dependent manner. B1 and B2 had significantly lower of systolic blood pressure compared to control (p=0.026 and p=0.022 respectively). Significantly lower of anti-dsDNA levels were found in B1 group compared to control (p=0.014). However, no significantly different of urine albumin levels were found between groups. Bryophyllum pinnatum also significantly increased the fetus body weight in dose-dependent manner (p=0.000).Conclusion: Treatment of Bryophyllum pinnatum could improve the pregnancy outcome and modulate the immune response in pregnant pristane-induced lupus mice. Therefore, Bryophyllum pinnatum is a potential herb which can be developed as an immunosuppressive agent in the future.

Abstract

Kurangnya pengetahuan masyarakat tentang COVID-19, lupus, reumatik, dan alergi mempengaruhi keberhasilan terapi, penurunan angka kejadian, dan pencegahan kekambuhan. Tujuan pengabdian masyarakat adalah membangun desa-desa binaan tanggap COVID-19, lupus, reumatik, dan alergi di Malang oleh Tim Kelompok Kajian Lupus, Autoimun, Reumatik, dan Alergi (LAURA) Universitas Brawijaya. Warga desa binaan diberikan penyuluhan berupa seminar awam dan pelatihan tentang pertolongan awal pada penyakit COVID-19, lupus, reumatik, dan alergi, kemudian diminta mengisi kuesioner posttest untuk mengukur pemahaman. Desa-desa binaan diberikan thermo-gun dan wastafel untuk menerapkan protokol kesehatan. Satu bulan kemudian dievaluasi adanya kejadian COVID-19, lupus, reumatik, dan alergi di desa binaan. Hasil evaluasi menunjukkan rata-rata warga desa binaan memahami 78,3% materi yang diberikan dan menerapkan protokol kesehatan sesuai yang diajarkan saat penyuluhan. Dilaporkan tidak ada kejadian COVID-19 serta kekambuhan lupus, reumatik, dan alergi dalam satu bulan terakhir kegiatan. Kesimpulan: pembangunan desa-desa binaan di Malang meningkatkan tanggap warga terhadap COVID-19, lupus, reumatik, dan alergi.

Abstract

Centella Asiatica (Linn) Urban is known as Gotu Kola. Centella Asiatica (CA) is rich in micro and macronutrients. The aim of this study was to investigate the effect of ethanol extract of CA on bone ossification and body length in zebrafish larva stunting model at 9 dpf. This study used zebrafish at 2 hpf (hour post-fertilization) - 9 dpf (day post-fertilization). The population of 300 larvae divided into 5 groups consisting of control group, rotenone group (exposed by 12.5 ppb of rotenone) and 3 rotenone+CA groups that exposed to CA extract for 4, 5 and 6 days,  respectively. The CA extract was obtained by maceration method with ethanol solvent. The results showed that rotenone 12,5 ppb able to inhibit the growth of larvae >2SD of body length and decrease bone ossification at rotenone group, were significantly different from the control group. Administration of CA extract was increased expression of cartilage at rotenone+CA5 as well rotenone+CA6 group and increase expression of bone at the rotenone+CA5 group and also increase body length rotenone+CA groups significantly different from rotenone group. It can be concluded that the period of CA extract exposure can correct the length of the body reaching 99.6% at 9 dpf and increased bone ossification in a time-dependent manner.Centella asiatica (Linn) Urban dikenal dengan nama pegagan. Centella Asiatica (CA) kaya akan mikro maupun makro nutrisi yang diperlukan bagi tubuh terutama masa pertumbuhan. Penelitian ini bertujuan mengetahui efek ekstrak etanol CA terhadap osifikasi tulang dan panjang badan larva zebrafish model stunting yang diinduksi rotenone pada 9 hari post fertilisasi. Penelitian ini menggunakan zebrafish mulai 2 hpf (hour post fertilisation) - 9 dpf (day post fertilisation), populasi larva sejumlah 300 yang dibagi 5 kelompok yang terdiri dari kelompok kontrol, kelompok rotenon (dipapar rotenone 12,5 ppb) dan 3 kelompok rotenone+CA yang diberikan pegagan selama 4, 5 dan 6 hari secara berurutan. Ekstrak CA diperoleh melalui  metode maserasi dengan pelarut etanol. Hasil penelitian menunjukkan bahwa rotenone dapat menghambat pertumbuhan panjang larva >2SD dan menurunkan osifikasi tulang pada kelompok rotenon secara signifikan dibanding kontrol. Pemberian ekstrak CA dapat meningkatkan ekspresi tulang rawan kelompok rotenone+CA5 maupun rotenone+CA6 dan meningkatkan ekspresi tulang keras kelompok rotenone+CA5 serta meningkatkan panjang badan kelompok rotenone_CA secara signifikan dibanding kelompok rotenone. Dapat disimpulkan bahwa lamanya pemberian ekstrak CA dapat meningkatkan osifikasi tulang dan meningkatkan panjang badan mencapai 99.6% pada 9 dpf.

Abstract

Turmeric (Curcuma longa) is widely used as herbal medicine, not an exception by pregnant women. Turmeric consumption by expectant mothers requires standard dose, because of its antiangiogenic effect could be harmful on placentation process and embryonic development. This experiment was undertaken to determine the effect of different concentrations of turmeric decoctum to the expression of Vascular Endothelial Growth Factor Receptor-2 (VEGFR-2) and Angiopoietin 1 (Ang-1) on the 48-hours-old chicken embryo. In this study, turmeric was extracted using decoction method to mimic the common method as adopted by people. The turmeric decoctum were freeze dried into a powder form and was used in preparing the stock solution for 200 ppm (P1), 300 ppm (P2), and 400 ppm (P3) as experimental treatments. The control group (P0) received 2% DMSO without turmeric decoctum. These were administered on the yolk sack of 16 hours incubation of fertile chicken egg by number of 200 µL. After 48 hours incubation, the expression of VEGFR-2 and Ang-1 on the chicken embryo were counted by ImageJ software. The results revealed that there is no significant effect of turmeric decoctum to the expression of VEGFR-2 and Ang-1. This suggested that turmeric decoctum was safe up to 400 ppm on chicken embryo.

Abstract

This study aims to determine the effect of giving Centella asiatica on incised wound healing through the treatment of Centella asiatica on white rats (Rattus norvegicus). This study uses Experimental method in the form of a post-test only group design to determine the effect of Centella asiatica ethanol extract on the healing time of incised wound in white rats (Rattus norvogicus). The sample used in this study was 20 white rats divided into five treatment groups. Data analysis used quantitative analysis with helped of SPSS 29 software. The data test consisted of normality, homogeneity, and one way ANOVA test. The result showed 10% Povidone iodine, vaseline, 10% ointment of Centella asiatica ethanol extract, 15% ointment of Centella asiatica ethanol extract, and 25% ointment of Centella asiatica ethanol extract had the same effect on incised wound healing in white rats (Rattus norvogicus). Intervention with 10% Povidone iodine had average healing time of 12,75 days; intervention with 10% ointment of Centella asiatica ethanol extract had average healing time of 10 days; intervention with 15% ointment of Centella asiatica ethanol extract had average healing time of 8,75 days; and intervention with 25% ethanol extract Centella asiatica had average healing time of 12,75 days. The normaly test shows that the entire group has a significant value of p>0,05, so it says that the data is normally distributed. The homogeneity test shows an overall significance of p>0,05, so the data is nuanced and homogeneous. The ANOVA test showed a significance of p = 0,152, so the value of p >0,05, meant that there was so significant difference between groups. This study proved that the application of Centella asiatica ethanol extract ointment can be used as an alternative in the treatment of incised wound, such as post-section caesarean wound care.

Abstract

Background: Neurodegenerative diseases are the main cause of morbidity and disability in the elderly. SIRT1 activation has been gaining popularity as novel treatment target. Cinnamon is known to possess neuroprotective abilities, however the mechanism in which it protects the brain is still limited. Objective: This research aimed to determine the interaction between several cinnamon active compounds with SIRT1 Methods: We used in-silico method to determine the molecular interactions between cinnamon main compounds as the ligands to target protein SIRT1. SIRT1 3D structure was retrieved from the Protein Data Bank and 4 ligands (Cinnamaldehyde, Caffeic Acid, Epicatechin, and Trigonelline) structures were obtained from PubChem web server, and we used Resveratrol as positive control ligand. SwissADME, Pyrx, Pymol, and Biovia Discovery Studio software were utilized in this research Results: All four ligands fulfilled Lipinski Rule of 5 criteria therefore they are suitable for oral administration. It was discovered in this study that epicathecin had higher binding affinity than the control ligand Resveratrol and interacted with SIRT1 in the similar amino acid residue as Resveratrol did. The binding pocket interaction between all ligands and SIRT1 are the same. Conclusion: Epicathecin, as one of the main cinnamon compounds, may possess neuroprotective properties by interacting with SIRT1. We pproposed that further research be implemented to investigate epicathecin biological effects on SIRT1 in vitro or in vivo.

Abstract

Background: Brain is an organ that is prone to oxidative stress and subsequent apoptosis due to high aerobic metabolism and relatively low antioxidants, especially under hyperglycemic condition. Cinnamomum burmanii (CB) is a species that is abundant in Indonesia, therefore it is of special concern for researchers to identify the anti-apoptotic effect of CB. Objective: This study was initiated to determine the effect of CB extract on the inhibition of brain apoptosis in zebrafish embryos exposed to high glucose and to investigate its anti-apoptosis mechanism by molecular docking approach. Methods: Molecular docking was conducted to determine the interaction between several CB extracts main constituents with target protein procaspase-9, compared to control ligand Saxagliptin. Zebrafish embryos were used to assess the effect of 4% glucose exposure and three doses of CB extract treatment (1.25, 5, and 10 µg/ml) on apoptosis in brain region. High-glucose condition in zebrafish embryo was confirmed with overexpression of Phosphoenolpyruvate carboxykinase (PEPCK). Apoptosis was evaluated by performing acridine orange (AO) staining and quantified by ImageJ software. Results: Molecular docking study indicated that main CB compounds, namely epicatechin, displayed stronger molecular interactions with procaspase-9 compared to control ligand Saxagliptin. There were increased numbers of apoptotic cells seen around brain region in glucose-treated group. Meanwhile, supplementation of CB extract at dose of 10 µg/ml resulted in decreased amount of apoptotic cells in brain region. Conclusion: The results suggest that CB extract protects from hyperglycemic-induced apoptosis in zebrafish embryos brain by modulating procaspase-9.

Abstract

The pathogenesis of Lupus Nephritis (LN) is still unclear, causing difficulties in diagnosis and therapeutic management. Recent studies have suggested that microRNAs (miRNAs) may play an important role in the pathogenesis of LN. However, the role of miRNA-451a in LN has not been widely studied and known, and there is controversy in the results of studies that have been done. This study aimed to analyze the expression of miRNA-451a in LN and lupus without nephritis and to evaluate its correlation with hemoglobin levels, leukocyte count, leukocyturia, and erythrocyturia. Samples from 45 lupus participants were collected, consisting of 22 with LN and 23 without nephritis. The expression of microRNA 451a was determined by reverse transcriptase real-time PCR. Renal disorder criteria based on ACR criteria. Statistical analysis using Mann-Whitney, Spearman correlation, ROC curve, and table 2X2. MiRNA-451a expression in LN (median 0.31; range 0.06-7.21) was lower than in lupus without nephritis (median 1.31; range 0.07-13.26) with a significant difference (p=0.041). The cut-off value for comparing the two groups was 0.7531 with an accuracy of 75%. The correlation between miRNA 451a expression and hemoglobin levels, blood leukocyte count, erythrocyturia, and leukocyturia showed no significant correlation (p > 0.05). MicroRNA-451a expression in lupus nephritis was lower than in lupus without nephritis.
