Abstract

High size variation of giant freshwater prawn was found in harvest and resulting in low productivity. Marker assisted selection may be useful to generate broodstock that produces progeny with high growth and homogeneity. This study was conducted to obtain growth related molecular marker in giant freshwater prawn. Genomic DNA was extracted from swimming leg (pleiopods) of 10 giant freshwater prawns fifth Generation for existence of SNP identification, consisted of 5 fast growth (FG) and 5 slow growth (SG).  While for SNP confirmation and resolving power of specific primer studies. The pleiopods sample was taken from six generation of 201 giant freshwater prawns, consisted of 129 fast-growth (FG) with 16.06 ± 2.48 g body weight and 72 slow-growth (SG) with 6.05 ± 0.90 g body weight. Oligonucleotide primers were designed according to Gene Bank database of crustacean hyperglycemic hormone (CHH) gene sequence. The amplified DNA fragment was then sequenced. The results of sequencing showed there was one base different in nucleotides of FG and SG prawns. Six set of primers were designed based on those CH gene sequence. PCR analysis resulted one set of primers which showed a specific amplification product of 280 bp for growth. The result of sequence analysis using the basic local alignment search tools showed that the nucleotide sequence of those PCR products had similarity of 99%-100% with CHH gen of M. rosenbergii. Thus, a candidate of growth related molecular marker have been identified for giant freshwater prawn.

Abstract

The study aimed to evaluate the effect of the water application of probiotic Bacillus sp. DMP13 on the growth and stress response of giant freshwater prawn larvae Macrobrachium rosenbergii. The larvae used in this experiment were 1-day-old larvae post-hatching with an average body weight of 2.7±0.6 mg and an average length of 2.1±0.1 mm. The larvae were reared in 2 L plastic tanks with a density of 100 L-1 for 21 days. The study was conducted using 4 treatments with three replications of each, namely A (control without probiotic), B (Bacillus sp. DMP13 with cell concentration of 102 CFU (colony forming unit) mL-1), C (Bacillus sp. DMP13 with 104 CFU mL-1) and D (Bacillus sp. DMP13 with 106 CFU mL-1). Probiotic was applied in the rearing media on days 9th up to 18th with 3 days intervals. The results showed that application of Bacillus sp. DMP13 with a cell density of 102 CFU mL-1 was able significantly increased the absolute larvae weight (29.67±1.88 mg), increased larval survival in stress concentrations of 1,250 μL L-1 of formalin solution (the highest larval survival rate 98.3±2.9%). Treatment with 102 CFU mL-1 probiotic cell addition also increasing the total bacterial viable count in the media (the cell density reached 5.99±0.28 log CFU mL-1). However, other parameters such as the larvae stage index, survival rate, and absolute length of the larvae with probiotic treatments were not significantly different from those of control.

Abstract

Pemeliharaan benih ikan Patin Siam (Pangasionodon hyphophthalmus) memerlukan pergantian air 70-80% dari total volume setiap hari. Sistem resirkulasi menjadi alternatif mengatasi keterbatasan sumber air dan mempertahankan kualitas air pemeliharaan pada pendederan benih patin siam. Tujuan penelitian untuk mengetahui efektivitas filter pada sistem resirkulasi terhadap kualitas air, kelangsungan hidup, dan pertumbuhan panjang benih patin siam yang dipelihara pada wadah akuarium. Rancangan percobaan faktorial, faktor pertama sistem pemeliharaan/jenis filter (taraf pertama jenis pemeliharaan tanpa filter; taraf kedua filter busa pori-bioball-karbon aktif; taraf ketiga dengan filter kapas dacron-batu apung-batu zeolit) dan faktor kedua padat tebar (taraf pertama 10 ekor L-1 dan taraf kedua 12 ekor L-1), masing-masing dengan tiga ulangan. Benih uji patin siam berukuran panjang standar 1,81±0,18 cm dan panjang total 2,26±0,21 cm. Pakan komersial diberikan secara satiasi sebanyak tiga kali dalam satu hari selama pemeliharaan 21 hari. Hasil penelitian menunjukkan bahwa faktor jenis filter berpengaruh terhadap kualitas air (total kelimpahan bakteri; P(0,00)<0,05) dan pertumbuhan panjang benih patin siam, baik laju pertumbuhan panjang total spesifik (P(0,01)<0,05) maupun laju pertumbuhan panjang standar spesifik (P(0,008)<0,05. Sedangkan faktor padat tebar berpengaruh terhadap kualitas air yaitu amonia total (P(0,04)<0,05) dan total kelimpahan bakteri (P(0,05)≤0,05). Interaksi antar faktor (jenis filter*padat tebar) tidak menunjukkan adanya pengaruh yang signifikan terhadap parameter yang diujikan. Jenis filter penyusun yang terbaik adalah kombinasi busa pori-bioball-karbon aktif (R1).

Abstract

Pendederan benih patin siam (Pangasionodon hyphophthalmus) yang dilakukan secara intensif seringkali belum mencapai target yang diharapkan. Kematian ikan diawali dengan stres pada ikan sebagai respon fisiologisnya, energi yang dibutuhkan untuk beradaptasi melawan stres merangsang mobilisasi glukosa ke dalam darah. Salah satu sumber stres bagi ikan pada budidaya adalah kualitas air pemeliharaan. Penelitian bertujuan untuk mengetahui tingkat stres pada benih patin siam terhadap kualitas air pada pemeliharaan sistem resirkulasi melalui pengukuran kadar glukosa darah. Penelitian menggunakan benih uji patin siam dengan bobot awal 62,74±25,17 mg yang dipelihara selama 21 hari menggunakan beberapa sistem resirkulasi dengan jenis filter berbeda (faktor A) pada dua kepadatan tebar yang berbeda (faktor B). Hasil uji statistik menunjukkan peningkatan kadar glukosa darah benih patin siam pada akhir pemeliharaan dipengaruhi oleh faktor A (P(0,003)<0,05). Faktor B dan faktor interaksi keduanya (faktor A*B; jenis filter*padat tebar) tidak mempengaruhi peningkatan kadar glukosa darah. Parameter pertumbuhan bobot tidak dipengaruhi oleh faktor A, faktor B, dan faktor A*B. Laju pertumbuhan bobot spesifik tertinggi 11,18±0,22% pada perlakuan R1.12.

Abstract

Abstract One of the problems in giant prawn cultivation is female giant prawns laying eggs during rearing. They will incubate their eggs for three weeks so that the energy from the expected feed for growth is used for egg development and other reproductive activities. Giving MPA hormone to giant prawns either by injection or oral could inhibit gonadal maturation and increase growth rate. The maturation process of the prawn ovary consists of the activation of some complex cellular mechanisms involving genes that regulate the stages of oocyte development. This study aimed to evaluate the response of gonadal maturity and the expression of the MrvWD-Kazal gene in giant prawns fed with a diet containing MPA hormone. The design used in this study was a completely randomized design with four treatments and five replications. The treatments were feeding a diet added with MPA with a concentration of 0 mg.kg-1 feed as control (K), 50 mg.kg-1 feed (P1), 100 mg.kg-1 feed (P2), and 150 mg.kg-1 feed (P3). MPA hormone at a concentration of 50-150 mg/kg feed could inhibit the gonadal maturation of female prawns. In 100 mg/kg of feed concentration showed the lowest gene expression level, indicating an inhibition of gonadal maturation molecularly. The administration of MPA hormone through the feed is a recommended method of aquaculture because it is more applicable and effective than by injection, even though it has a non-uniform impact on each individual. Highlight Research Administration of MPA hormone through the feed to female giant freshwater prawns can inhibit the level of gonad maturity. Expression of von Willebrand factor D (vWD) – Kazal gene were analyzed in different levels of prawn maturity Gonadosomatic index and gonadal histology confirmed that different levels of administration of MPA hormone through the feed could affect the gonad development. The administration of MPA hormone through the feed is a recommended method for aquaculture because it is more applicable and effective to inhibit early maturation of prawns.
