Abstract

Chromolaena odorata (kirinyu/sufmuti) is a species of weed which utilized as traditional medicine especially for wound healing. This research was conducted to study the histological effect of C.odorata leaf extract to healing process of incision wound in Sprague-Dawley rats. A total of 45 males - 6 weeks of Sprague Dawley rats at 150-200g body weight were utilized in this study. Those rats were divided into five groups, basis ointment, Betadine 10%, C.odorata 5%, C.odorata 10%, and C.odorata 20%. All groups of rats were given a 3 cm long linear incision at dermal depth on the backs. The extract and medication were topically applied once a day for ten days. 3 rats of each group were euthanized in day 2, 6, and 10 after incision. Subsequently, skin tissue was obtained and fixated in 10% buffer formalin for Hematocixylin - Eosin staining. The measured variables were wound healing both macroscopical and microscopical figures from day 0 to day 10 after incision. Data were analyzed descriptively. Microscopic observation showed that various concentrations of C.odorata extracts affected wound healing process. C.odorata Extract is allegedly able to stimulate wound healing by increasing fibroblasts for re-epithelization.

Abstract

The purpose of this research was to identify the cockroach species infesting public buses, determining its distribution and its infestation degree, and analyze the factors causing its infestation. The observation of the existence of cockroach was performed on 30% of buses found in 6 bus pool in Bogor City. Observations was focused on 6 sites inside the bus: 1) seats, 2 floor), 3) gaps, 4) shelves, 5) window, and 6) toilets. Cockroach were manually colletcted put in a labeled plastic pouchs. Observations was done within 30 minutes each bus. The degree of cockroach infestation was scored as zero, light, moderate, high, and very high infestation. The factors causing cockroach infestation in public buses measured using an enclosed questionnaire. A total number of 1626 individuals were collected identified as Blattella germanica cockroach (1353 individuals) and Periplaneta americana cockroach (273 individuals). The highest infestation of Blattella germanica was found in the buses in the pool-6 area with with average amount of 66.4 individuals per bus (very highly infested). While Periplaneta americana was found very highly infesting buses in the pool-5 area with average amount 25.3 individuals per bus. The cockroach infestation spreading in the gap (48.68%) with the highest number for the presence of both species. The shelves found as site with lowest infestation (12.60%). There was a weak correlation between cockroach infestation in the pool bus area with personal biosecurity (r = -0.116), medium relationship with places/equipment biosecurity (r = -0.406), and strong correlation with environmental biosecurity (r = -0.841).

Abstract

Tuna is one of the fishery products which has a high level of consumption in the city of Kupang but are easily damaged. One of the pathogenic bacteria in fish that cause food-borne disease is Salmonella sp. The contamination of Salmonella sp. is caused by inadequate sanitation, poor hygiene and contaminated water used by the merchant. This study aims to determine both the differences in the value of TPC and whether there is the contamination of Salmonella sp. or not. Moreover, this study was to examine quantification of contamination level and different levels of Salmonella sp contamination the difference level of Salmonella sp. contamination in tuna fishes which has been sold in the fish auction place (TPI), traditional markets and retail fish market in Kupang. 54 samples of Tuna were collected from three locations. This research was also supported by the water quality testing which consisted of 15 water samples. The result showed that there was a significant difference on the TPC values with an average level of contamination from the highest to the lowest, started from the fish retailers as many as 1,35 x 105 CFU/g, followed by the traditional markets as many as 5 x 104 CFU/g and TPI as many as 3,5 x 103 CFU/g. The differences of TPC values were due to the personal higyene and sanitation, locations, the equipments as well as the materials used in the post-arrest process, the distribution and the sales. The results from TPC analysis showed that 4 samples were contaminated by the Salmonella sp. The average level of Salmonella sp contamination was from the Tuna sold by the fish relailers as many as 1,6 x 102 followed by the traditional fish markets as many as 1 x 102 and in the TPI as many as 4x10. This study also showed that based on water quality test, positive results were found in 1 sample in TPI, 3 samples in the traditional markets and 4 samples in the fish retailers. In conclusion, the Tunas sold by the fish retailers have the highest TPC value and the highest level of Salmonella sp contamination compare to the Tunas sold in the traditional markets and TPI.

Abstract

Fish has become one of the favorite foodstuffs for Indonesian society. Like the other food commodities of animal origin, fish also has a perishable nature so it takes a good handling and processing, such as salting and drying which the result is known as salted fish. The processing of salted fish requires attention because there is a high risk of food poisoning hazard for food which the growth of normal flora are inhibited due to the enterotoxin of Staphylococcus aureus. This study aims to determine the contamination level of Staphylococcus aureus within the salted fish in the traditional markets in the city of Kupang. There were 18 samples of salted fishes used which consisted of 8 samples from Oesapa traditional market, 5 samples from Oeba traditional market and 5 samples from theInpres Naikoten traditional market. The sample inspection using the Plate Count method in order to spread. The results showed that the contamination level of all the salted fish samples have passed the maximum contamination limit of S. aureus based on the SNI No. 7388 : 2009 (1 x 103 CFU/g) with the value of the contamination level ranged from 3.4 x 103 CFU/g in the salted fish derived from Oeba traditional market to 1.4 x 106 CFU/g in the salted fish derived from the Inpres Naikoten market. The high value of the contamination level is related to several factors such as environment, storage time, sanitation and personal hygiene. Therefore, salted fish traded in traditional markets require attention to be safe and suitable for consumption.

Abstract

Staphylococcus aureus is one causes of wound infection in dogs, occured as secondary infection or trauma. One of the obstacles in the treatment of Staphycoccal infection is their resistance to antibiotics. This study aims to isolate and identify the Staphylococcus aureus in the cases of dog wound as well as testing the bacterial resistance to the antibiotics namely amoxicillin, tetracycline and ampicillin. There are total of 18 samples of dog wound taken by sterile swab and put in a closed test tube containing sterile physiological NaCl and stored in a ice jar. Swab is applied to the surface on Mannitol Salt Agar and observed morphology of the bacterial colonies that grow after incubated for 24 hours at 37 0C. Furthermore, the Gram staining, hemolysis test, biochemical tests including the catalase and coagulation were also tested. Suspect Staphylococcus aureus colonies were further tested for antibiotics sensitivity test using the Kirby Bauer’s method. Results of this experiment showed 8 isolates (44.44%) were positive to Staphylococcus aureus. The isolation of Staphylococcus aureus in the culture of Blood Agar showed that 8 isolates are Beta hemolytic strain. Antibiotics sensitivity test done in duplo revealed that there are 7 isolates of Staphylococcus aureus sensitive and 1 isolate resistant to ampicillin. The sensitivity test to the antibiotic tetracycline showed that 6 isolates sensitive and 2 isolates resistant. The sensitivity test to the antibiotic amoxicillin showed that 7 isolates sensitive and 1 isolates resistant. It can be suggested that in the treatment of wound infection in the dog's skin, antibiotic sensitivity test should be done, because some strains of Staphylococcus aureus has developed resistance to antibiotics.

Abstract

Pigs are one of the potential livestock to breed with the aim of meeting the needs of animal protein, but in fact the level of pig production is not optimal due to the low efficiency of reproduction. One way that can be used to increase the productivity of sows that by superovulation method. This study aimed to determine the effects of PMSG hormone injections to increase the productivity level of sows that given superovulation treatment. 10 sows that have been selected, be divided into two research groups: control group and treatment group. Before the treatment is given in the form of PMSG hormone injections, sows from both study groups, first injected with hormones PGF2α to estrus synchronization purposes. Then, sows in the treatment group are injected by PMSG hormone with the dose of 2 ml/sow before naturally mating with the boar. After the mating process, the gestation period of pregnant sow will be observed until the parturition time. The parameters were litter size and the birth weight of piglets produced from each sows of the two study groups. Sows were injected with PMSG hormone before mating show better productivity levels compared with sows in the control group. Sows were injected with PMSG hormone produce the overall number of piglets are 46 piglets or the litter size are 9 piglets with the average birth weight of 1.20 kg, while in the control group generates total number of piglets are 36 piglets or the number of litter size are 7 piglets with the average birth weight of 1.05kg. Based on statistical tests, the results showed that there’s no significantly different ((P>0.05) for the litter size of each research groups. The results showed that injection of the PMSG hormone as a superovulation agent before mating is able to increasing piglet birth weight but not able to increase the litter size significantly. Increased productivity of sows through superovulation method is to extend the production period of the sow, especially sows with a number of birth five or more times without being replace by new gilt.
