Abstract

An experiment was carried out to recognize the effect of long equilibration time on the quality of spermatozoa and to obtain optimal equilibration time in the process cryopreservation of landrace pig semen. Semen was obtained from a landrace boar aged 2-3 years using glove hand method. The semen was than evaluated macroscopically and microscopically, good quality semen was diluted in a basic diluent, than kept for two hours at a temperature of 27-28oC. The semen were then centrifuged at 2000 rpm for 15 minutes, the supernatant was discarced and the pellets was re-diluted into basic diluent with a ratio of 1:1. Then, semen was diluted in treatment diluent with modified durasperm + 6% lactose + 4% glycerol with a concentration of 500 x 106/0.5 mL. Furthermore, the semen was packaged in 0.5 ml straws, then equilibrated at a temperature of 3-5oC appropriate to treatment, with 1 hour equilibration (T1), 2 hours equilibration (T2) and 3 hours equilibration (T3). After equilibration, the straws was frozen horizontally 5 cm above the liquid N2 vapor for 10 minutes at a temperature of ± -110oC and then stored in a liquid N2 container (-196oC). To test frozen semen, it was done by stored frozen semen at 37oC for 30 seconds. The results showed that the highest quality of spermatozoa (P <0.05) was found in T2 and T3 when compared to T1 (P> 0.05), with the motility value was T2: 28.13 ± 2.39%; T3: 25.00 ± 4.08%; and T1 18,15±3,23%. It can be concluded that 2 hours and 3 hours equilibration time was able to maintain the quality of frozen semen landrace boars and the statistically was not significant.

Abstract

This study aims to determine the quality of beef meatballs processed with red sorghum flour as a substitute for tapioca flour during the shelf life. This study used a completely randomized design with 5 treatments and 4 replications. The treatments consisted of TS0 = 100% tapioca, TS25 = 25% red sorghum flour, TS50 = 50% red sorghum flour, TS75 = 75% red sorghum flour; TS100 = 100% red sorghum flour. Furthermore, the variation of storage time is 1 day and 30 days. The variables observed were organoleptic (color, aroma, taste), TPC, fat oxidation and elasticity. The results of statistical analysis showed that the use of red sorghum flour as a substitute for tapioca flour up to 100% did not change the color of the meatballs at 1 day of storage, but at 30 days of storage the color of the meatballs darkened, the aroma of the meatballs became on either 1 day or 30 days of storage (P <0.01), the meatball taste score at 1 day of storage decreased and at 30 days the meatball taste score was between very dislike to dislike (P<0.05), the elasticity score decreased from very chewy to not chewy score (P< 0.01), while at 30 days of storage the elasticity score of the meatballs was the same, namely not chewy. The treatment had a very significant effect (P<0.01) on TPC and fat oxidation. This study concluded that the higher the percentage of replacement of red sorghum flour decreased the organoleptic qualities such as darkening of the color, the decrease in aroma, taste and elasticity of the meatballs, the TPC (Total Plate Count) number. and fat oxidation increased from 1 day to 30 days of storage.

Abstract

One of the factors that support the success of reproduction in sheep is the quality of the semen. This study aims to analyze the effect of ejaculation frequency on the quality of ram semen. This study used four local rams two years old. All rams have collected the semen using an artificial vagina five times a day with three replications. The experimental parameters were macroscopic (volume, pH, consistency, and color) and microscopic (mass movement, motility, viability, concentration, normal sperm morphology, and sperm intact plasma membrane). The data analysis used a randomized block design (RBD) with SPSS version 20. The results showed that the frequency of ejaculation in rams shows color and consistency in the 1st ejaculate, which was creamier and thicker than the 4th and 5th ejaculate. Mass movement, sperm concentration per mL, and total sperm concentration in ejaculate 1st and 2nd were higher than in 5th ejaculate. The potential of artificial insemination (AI) in the 1st and 2nd ejaculate was higher than in the 5th ejaculate. The study concluded that the frequency of ejaculation affects mass movement, sperm concentration, and the potential straw production for AI in local sheep.
