Abstract

(The effect of heat shock protein 70 (HSP70) SacII gene polymorphism on heat tolerance of West Sumatera local duck) ABSTRACT. The objective of this study was to identify the effect of HSP70/SacII gene polymorphism associated with the thermotolerance trait. Genotyping was performed on 110 local duck from the West Sumatera ducks population (pitalah, bayang, kamang, and payakumbuh). The restriction enzyme used was SacII. Heat stress was done for 1 hour with samples used as much as 24 birds to analyze hematological status. PCR-RFLP and Sanger sequencing assays were used to identify polymorphism. Analysis of polymorphism was conduct by MEGA 7 software and its effect is analyzed with ANOVA using Statistical SAS 9.4 software. The product of amplification was 466 bp. HSP70/SacII recognized one SNP g.1702TC, producing two alleles (T and C) and three genotypes (TT,TC,CC). The HSP70/SacII locus were polymorphic in all population. T allele had a higher frequency than C allele in all populations. The analysis of chi-square (χ2) showed that all local ducks population were fit with Hardy-Weinberg equilibrium. The effect of HSP70/SacII gene on hematological status showed differences (P0,05) of heterophil and H/L ratio with CT genotype show better heat tolerance than other genotypes. HSP70/SacII gene can be used as a marker of heat tolerance in local ducks based on polymorphism and its effect on hematological status.

Abstract

Safety condition of commercial fermented milk that put on the market in banda aceh based on nutritive value and pathogenic bacteriaABSTRACT. Fermented milk has much value primarily for the health of digestive system. However without hygienically and sterilized treatment it can become toxic to consumers. This research was conducted to observe nutritive value, the present of pathogenic bacteria, and the safety condition of commercial fermented milk that put on the market in Banda Aceh. Samples of commercial fermented milk tested were obtained from recognized retailers in Banda Aceh. Factorial completely randomized design with six replications was applied in this research. Variety of fermented milk products (yoghurt and drink yoghurt) was the first factor to be analyzed, and the second one was expired date of the products (the date of production and the date of by expired). The amount of crude protein and lipid, pH level, and the amount of lactic acid bacteria, Coliform and Staphilococcus aureus were the parameter determined. The variety and the expired date of the products influenced the amount of crude protein and lipid, the pH level, the amount of lactic acid bacteria, Coliform and Staphilococcus aureus in the level of highly significant different (P0,01). There was also found in highly significant interaction (P0,01) between factors that influenced the amount of lactic acid bacteria and Staphilococcus aureus. On the other hand, the amount of Coliform was not influenced by those factors. The study also results in decreasing of the amount of crude protein and lipid as well as lactic acid bacteria caused by the ongoing time of storing. The longer fermented milk was stored, the lower the level of crude protein, lipid and lactic acid bacteria presented. The lipid level of fermented milk products those put on the market in Banda Aceh have been complied with the requirement standard of SNI 01-2981-1992. However, crude protein level has not achieved the requirement standard yet. Due to the amount of Staphylococcus aureus present, the fermented milk products tested, in fact, were not saved to be consumed. On the contrary, although Coliform was also present, the products were in the category of saved to be consumed.

Berdasarkan penelitian disimpulkan bahwa seluruh perlakuan tidak mengganggu fermentasi rumen meskipun nilai CH4 meningkat, pemberian kombinasi ekstrak mampu meningkatkan fermentabilitas pakan, dilihat dari peningkatan VFA total dan NH3.(Fermentability of feed supplemented with Ageratum conyzoides leaves and Zingiber officinale extracts on in vitro dairy cow)ABSTRACT. The research was aimed to determine the effects of Ageratum conyzoides leaves (AC) and Zingiber officinale (ZO) extracts on in vitro ruminal fermentation of dairy cow. Materials used were the rumen fluid and the ethanol extract of AC leaves and ZO. The study used a completely randomized design with 4 treatments and 4 replications. The treatments were control (T1), T1 + AC 0,005 ml (T2), T1 + ZO 0,005 ml (T3), and T1 + AC 0,0025 ml + ZO 0,0025 ml (T4). Data were analyzed using ANOVA and continued by Duncan test. The results showed that both AC and ZO supplementation affected total VFA (P0,05), acetate, propionate, butyrate, NH3 and CH4 concentrations (P0,01). However, the ruminal pH and total protein production concentrations were not affected by the supplementation of either AC or ZO alone. The average of all treatment group is ruminal pH was 6.9; and total protein was 167,779 mg/g. The treatments T3 groups had the highest acetate, propionate, butyrate concentrations (18,51; 5,03; 1,91 mMol/l) and had the lowest CH4 (16,53%). T4 treament groups had the highest VFA and NH3 concentrations (195,00 mM and 8,64 mM). In conclusion, the suplementation of both AC and ZO did not adversely affect the ruminal fermentation, although CH4 concentrations increased. Moreover, supplementing both herbals in the diets could improve the fermentability of rumen, indicated by the enhanced production of VFA and NH3.

Abstract

The effect of reduction of maize as starch source on flow rate of pellet in continuous production processABSTRACT. This experience was conducted to determine effect of reduction of maize as starch source on flow rate of pellet in continuous production process. Design of the experiment used was a completely randomized design with 3 treatments and 3 replicates. The data is analyzed by using of ANOVA and if there is a significance among treatments would tested using by contrast orthogonal. The variables observed were angle of repose, loose bulk density, compacted bulk density, and flow rate. The results showed that the reduction of maize as starch source did not give the significant effect on the variable that observed. The analyzed angle of repose showed variation 24.200 to 25.690, loose bulk density showed variation 621.6kg/m3 to 658kg/m3, compacted bulk density showed variation 668.3kg/m3 to 676.8kg/m3, and flow rate showed variation 449.69kg/minute to 491.41kg/minute. Based on the result that the reduction of maize as starch source (corn, sorghum, menir) did not give the significant effect on flow rate of pellet and the pellet treatment with menir has the fastest pellet production time so flowability pellet from pelleter to material hadling (bucket elevator) machine is faster. As higher the values of flow rate, so the time of material movement from bin to the package is shorter.

Abstract

ABSTRACT. The world’s copra meal production amounted to 1.8 million tonnes in 2002 and most of it was produced in Asia. The Philippines and Indonesia contributed approximately 65% of the world’s copra meal production. The main problems of using copra meal in poultry diets are its physical properties along with its nutritional profiles. This study was conducted to determine the physical characteristics and feeding value of copra meal. Physical characteristics were determined by measuring the bulk density and water holding capacity of copra meal and a digestibility study was undertaken to investigate nutrient digestibility, jejunal digesta viscosity and apparent metabolizable energy of copra meal. A total of 28 day old male Ross chicks were given control starter and grower diets from day 1 to 35. From day 36 to 42 , the birds were fed an experimental diet. Faeces were collected for three consecutive days. Jejunal digesta was measured for viscosity and ileal digesta was used for amino acid digestibility measurements.Data indicated that bulk density and water holding capacity of copra meal were poor, being 0.49 g/cm3 and 4.69 g water / g feed respectively. Although the crude proteind and amino acids contents of copra meal were favourable to meet a broiler chicken’s requirements, their digestibilities were low and lysine digestibility being the lowest while arginine digestibility was high. Dry matter, neutral detergent fibre digestibility and apparent metabolizable energy were also low. The low digestibilities of nutrients were not due to the jejunal digesta viscosity as jejunal digesta viscosity was low.
