Abstract

Mystacoleucus marginatus is a type of freshwater fish that is spread in various countries with tropical climates, one of which is Indonesia. This fish can be found in the Brantas River Basin (DAS), East Java. In its development, identification of fish species needs to be done in detail and more deeply, so that the correct identification results can be obtained. This study aims to identify genetic and phylogenetic characteristics of Mystacoleucus marginatus in the Brantas River Basin based on the Cytochrome Oxidase C Subunit I (COI) gene. Test samples were taken from 11 individuals of Mystacoleucus marginatus in the Brantas watershed, from June to August 2018. The results showed that Mystacoleucus marginatus in the Brantas watershed had 2 Haplotypes with Haplotype (Hd)=0.96 diversity values and nucleotides diversity (Pi)=0.00357. These results indicate that the population in the Brantas watershed is interconnected between one location and another sampling location, this is supported by a phylogenetic topology of DNA barcodes showing a monophyletic clade, as well as a haplotype distribution indicating that the sample area is genetics are not isolated from one another. So that the type of Mystacoleucus marginatus scattered along the Brantas watershed is still in one species, which is indicated by the low level of genetic variation. Genetic identification efforts must be carried out continuously so that appropriate information can be obtained for the diversity of fish species, especially endemic fish.

Abstract

Study was conducted to estimate live weight of Indonesian Local-Thoroughbred racing horse using their chest girth, body length and body volume represented by chest girth and body length dimensions. Data on animal live weight (LW), body length (BL), chest girth (CG) and body volume were collected from male horses (n=188) kept by the owner club members of North Sulawesi Racing Horse. Animal body volume was calculated using cylinder volume formula with CG and BL as component factors of the formula. Data were classified on animal age basis of five groups. Regression analysis was applied for LW with all linear body measurements. Age significantly influenced all body measurements. Simple regression can be recommended to predict live weight of this animal based on body volume with their average age groups as follows: Live weight (Y in kg unit) = - 0.002+1.16 body volume (dm3) with determination coefficient (R2) of 0.98. The multiple regression can also be recommended to estimate accurately live weight of this animal at age average groups as follows: Live weight (Y in kg unit) = - 0.11 - 2.74 chest girth (X1 in cm unit) + 4.92 body length (X2 in cm unit) with R2 of 0.98.

Abstract

Diabetes is a disorder in the metabolism of carbohydrates, lipids and proteins with various causes. From the previous research, GPR43 is a potent clinical target for the treatment of type 2 diabetes. It can be activated by Short Chain Fatty Acid (SCFA) which is an organic fatty acid produced through fermentation of dietary fiber by bacteria in the distal intestine. The aim of this study was to analyze the effect of C680T GPR43 Gene Variations to Its Interaction with Short Chain Fatty Acid (SCFA) in silico. The study used 8 sequences of GPR43: ID AF024690, AC002511, EU432114, BC095535, BC096198, BC096199, BC096200, BC096201 and  data of ligand (SCFA): acetic acid (CID 176), Butyric acid (CID 264) and Propionic acid (CID 1032). GPR43 was modelled using I-TASSER, Sequence and structural alignments were conducted using Bioedit and Superpose V.10, respectively. The Docking process was done using PyRx and molecular interaction was analyzed using Discovery Studio 2016. The Result showed that three types of SCFA bind to GPR43 variants T and GPR43 variant M obtained a similar pattern. The binding affinity from the largest to the smallest is butyric acid > propionic acid > acetic acid. GPR43 variant T has greater affinity to the SCFA than the GPR43 variant M. There is no differences of preference between GPR43 variant T and GPR43 variant M to bind SCFA.

Abstract

Necrotizing enterocolitis (NEC) is inflammatory state of intestinal tissue which mostly occurred in preterm neonates and associated with ischemia and inflammation. This study was aimed to investigate calprotectin level (inflammatory marker) and intestinal fatty acid binding protein (I-FABP) (ischemia marker) in preterm neonates with NEC. This research was designed as cross sectional which involve 32 preterm neonates divided into 2 groups as follows: NEC group (n=16 subjects) and control group (n=16 subjects). Diagnosis of NEC was established by clinical and radiological signs (abdominal distension and pneumatosis). Fecal calprotectin and urinary I-FABP were measured using ELISA method. Results showed that fecal calprotectin and urinary I-FABP was significantly higher in NEC group as compared to control group (Mann-Whitney test, p<0.05). Both calprotectin and I-FABP was positively correlated with NEC state (Spearman correlation test, p=0.000, r=0.866). Moreover, calprotectin and I-FABP level was positively correlated with grade (Bell’s classification) and type (Gordon’s classification) of NEC (Spearman correlation test, p<0.05). We concluded that calprotectin and I-FABP level was higher in NEC group. Moreover, I-FABP but not calprotectin, were correlated with grade and type of NEC.

Abstract

Mesua ferrea L. (Ceylon ironwood, Calophyllaceae) is a popular medicinal plant with a long history in South and Southeast Asia folk medicines. In this study, we report the phytochemical analysis, FTIR fingerprint, as well as antimicrobial and antioxidant activities of the crude extract of leaves of M. ferrea and its fractions. The dried leaves of M. ferrea were extracted with ethanol by the re-maceration method. The crude extract was further partitioned in ethanol-water and ethyl acetate to obtain ethanol and ethyl acetate fractions, respectively. The identification of compounds in the extract and fractions was conducted according to the standard phytochemical screening method. Fourier-transform infrared spectroscopy (FTIR) was utilized to record their metabolites fingerprint. The antimicrobial activity was evaluated with the disk diffusion method against Escherichia coli, Bacillus subtilis, Candida albicans, and Saccharomyces cerevisiae. The antioxidant activity assay was conducted with 2,2-diphenyl-1-picrylhydrazyl (DPPH) free radical scavenging method. The crude extract of M. ferrea leaves and it fractions contained saponins, phenolic compounds, flavonoids, and terpenoids. The FTIR data supported the presence of terpenoids and glycosides in those samples as well. The extract and fractions demonstrated considerable antimicrobial activity against all tested bacteria and fungi, with MIC values ranged 3.9-31.3 μg/ml.  The ethyl acetate fraction of M. ferrea leaves showed a moderate antioxidant activity with an IC50 value of 49.19 μg/ml.

Abstract

Critical conditions as one of the main focused in hospital care. This condition required rapid, comprehensive and quality management that carried out by nurses especially in the treatment of cardiovascular disease. This condition caused pressure on the nurse and causes stress. This stress can be overcome by complementary and alternative therapy in the form of mindfulness meditation. The purpose of this study was to identify the effect of mindfulness meditation therapy in reducing stress on nurses in ICU and ICCU. This study is quasy experiment research design through one group pretest posttest approach with respondents were 14. The results of this study indicated that there was decreased in stress before and after intervention with p-value=0000 and stress before and 1 week after intervention with p-value=0.001. Mindfulness therapy can reduce nurse stress.

Abstract

Urinary Tract Infection (UTI) is a common infectious disease. Urine culture as a gold standard is less practical and time- consuming. Examination of urine dipstick and microscopic is less accurate. Procalcitonin (PCT), IL-6 and Heparin Binding Protein (HBP) have known increase after infection, which potentially used for early detection of UTI. The aim of this study was to determine the diagnostic performance of urine IL-6, HBP, PCT, leukocyturia and bacteriuria level as biomarkers of UTI. The study was conducted on 51 UTI patients with positive urine culture, 16 UTI patients with negative urine culture, and 16 healthy control.  Urine IL-6, HBP, and PCT level were determined using ELISA. Leukocyturia and bacteriuria were measured using automated flowcytometry and fluorescent dye analyzers.  The gold standard for UTI diagnosis was urine culture. The sensitivity and specificity of urine PCT, IL-6, HBP, leukocyturia and bacteriuria were 60.78% and 65.62%; 72.55% and 56.25%; 96.07% and 50.00%; 80.39% and 78.12%; 80.39% and 59.09% respectively. The AUC of urine PCT, IL-6, HBP, leukocyturia and bacteriuria were 0.600; 0.605; 0.666; 0.877 and 0.830 respectively. For combinations of two biomarkers, the best sensitivity and specificity was demonstrated by urine PCT and leukocyturia.

Abstract

Today, the high incidence of disasters requires nurses to increase their role as disaster nursing in emergency response. This is in accordance with standards of the international council of nursing (ICN) that the nurse as a front line in providing treatment of disaster victims. One of the way to do in these situations is to improve the early preparedness in nursing students through disaster education, so that this plan needs a new method for effective and innovative disaster learning. One of the recommendation learning method is simulation with Tabletop Disaster Exercise (TDE). The purpose of this study was to analyze the effect of TDE learning for undergraduate nursing student’s knowledge in providing the management of victims in disaster response simulations. This study uses quasy experimental with pre-post test and control group design. The samples are 36 respondents were divided into two groups there are treatment and control groups so that each consisting of 18 respondents. The treatment group were given a TDE simulation, while the control group was given standard simulation and module. The results using paired t-test significantly showed that the differences value of knowledge before and after intervention with difference Pre-test 18 respondents mean 23.89±9.00 and Post test 18 respondents mean 81.67±8.04 Difference  Mean (CI 95%) = 57.78 (53.85-61.71) in each group indicated by the value (p = 0.000). The Effect Simulation Standards Knowledge Nursing Students with difference Pre test 18 respondents mean 26.39±9.04 and Post test 18  respondents mean 70.28±10.07 Difference  Mean (CI 95%) = 43.89 (38.59-49.18). The results from unpaired t-test to identify the difference of knowledge between the two groups after intervention has a significance value (p = 0.000). This means that there is significant difference of knowledge between TDE simulation and simulation.
