Abstract

Isolation of steroid-alkaloid compounds from Banyan leaf (Ficus benjamina L.) has been carried out. As much as 27.13 g of concentrated ethanol extract was resulted from 1.0 kg dried powders of Banyan leaf that was macerated using ethanol as a solvent. Further, this extract was partitioned with n-hexane to obtain 11.39 g concentrated extract of n-hexane. Ethanol extract was dissolved in ethanol-water (7:3), then was evaporated leaving water extract. Futhermore these water extract was partitioned with chloroform to obtain 0.7 g concentrated chloroform extract which was dark green and 2.21 g concentrated water extract which was green.Separations of n-hexane extract containing steroids using column chromatography (stationary phase : silica gel 60 and mobile phase : chloroform-ethylacetate (4:1)) resulted in fourteen fractions with F8 fraction being the most positive toward Liebermann-Burchard reagent, but this fraction contains at least in five components. Purification of the F8 fraction using TLC preparative resulted in six fractions (F8.1, F8.2, F8.3, F8.4, and F8.5) with F8.1 fraction being the most active steroid.The infrared spectrophotometer analysis suggested that the isolate (F8.1 fraction) was a steroid-alkaloid compound having functional groups including O-H stretching at 3448.72 cm-1 that was supported by one band C-OH streching at 1381.03 cm-1, the band of C-H aldehyde group at 2924.09 and 2854.65 cm-1 and the carbonyl group C=O of aldehyde at 1735.9 cm-1. Absorption at 2337.72 cm-1 indicated C?N stretching which was supported by N-H aliphatic and C-N aliphatic groups at 1172.72 and 1111.00 cm-1 respectively. The other groups were C=C aliphatic (1558.48 cm-1), C-H aliphatic bending (1458.18 cm-1), and C-O-C stretching (1257.59 cm-1). The gas chromatogram and mass spectra showed an isomer compound with Mr of 393 and molecular formula of C24H27O4N which was an alkaloide.

Abstract

Studies on biosorption of chromium (III) on NaOH-activated biosorbent of coconut coir fibre have been carried out. These studies included the determination of optimum concentration of NaOH for activating the biosorbent, the determination of biosorbent acidity, the optimum pH, the contact time of biosorption, and the adsorption isotherm type.The result showed that the NaOH optimum concentration for activation of coconut fiber was 1,2 M. The surface acidity of BBN and B00 were of 9,99 ± 0,17 and 3,17 ± 0,21 mmol/g respectively. The surface specific of BBN and B00 were of 20,21 and 19,14 m2/g respectively. The optimum pH of biosorption Cr(III) on BBN and B00 biosorbent was 3 and the contact time were 30, and 120 minutes. The type of isotherm adsorption for the biosorption of Cr(III) on BBN and B00 biosorbent was isotherm S.

Abstract

?-L-Arabinofuranosidases (EC 3.2.1.55) catalyze the hydrolysis ?-L-arabinofuranosidic bonds and act synergistically with other hemicellulases enzymes for the complete hydrolysis of hemicelluloses. Yeast Saccharomyces cerevisiae that considered as Generally Recognized As Safe (GRAS) was chosen for expression and secretion of thermostable ?-L-arabinofuranosidase (AbfA) from Geobacillus thermoleovorans IT-08 termofilik dengan teknologi DNA rekombinan. The extracellular enzyme from the secretion result in this recombinant yeast was precipitated with ammonium sulphate fractionation. Base on the result of measurement of the enzyme activity and the concentration of total protein showed that the specific activity of AbfA enzyme was the lowest at 40% saturation of ammonium sulfat, and the highest at 80% saturation of ammonium sulfat.

Abstract

The potency of Fourier transformed infrared spectroscopy (FTIR) combined with chemometric method for determining the total flavonoid content of tempuyung (Sonchus arvensis L) has been investigated. Partial least squares regression (PLSR) was used to build a prediction model based on the relationship between concentration of total flavonoids obtained from the reference method (AlCl3) and FTIR spectrum. Application of PLSR in the determination of total flavonoids content in tempuyung gave moderate value for goodness of fit of the model with r calibration = 0974, r validation = 0742, Standard Error Calibration = 0023, Root Mean Square Error Calibration = 0023, Standard Error Prediction = 0078, Root Mean Square Prediction Error = 0076, and bias = -0001 using FTIR derivative spectrum segment 1. From these results, combination of FTIR spectrum and PLSR can be used for the prediction of total flavonoids content in tempuyung.

Abstract

Cation exchanger have been prepared by graft copolymerization of acrylic acid (AA) onto a low density polyethylene (LDPE) with “grafting-on” method using Fenton reagent (H2O2/Fe2+) as initiator under nitrogen atmosphere at 280C. The percentage of grafting was determined by the gravimetric method. The percentage of grafting was influenced by monomer concentration, initiator concentration (H2O2), and the duration of grafting. The optimum conditions were obtained at 15% (v/v), 0.2 mL, and 8 hours for the monomer concentration, the volume of initiator H2O2 30%, and the duration of grafting respectively.Analysis of the PE-g-AA was conducted using Fourier Transform Infrared (FTIR) spectroscopy to confirm graft copolymerization which revealed the existence of new absorption peak at 1712,79 cm-1 and 2661,77 cm-1 assigned to C=O and O-H respectively. Characteristic of PE-g-AA film was tested by water uptake capability and cation exchange capacity toward Cu2+. The two characteristics were increased by percent grafting.

Abstract

The potency of chitin/chitosan isolated from shrimp waste as coagulant to reduce turbidity of water have been studied in this research. The chitin/chitosan was isolated through some processes including deproteination, demineralization and deacetylation.The chitin/chitosan obtained was characterized using FTIR and the degree of deacetylation was calculated based on FTIR spectra. The deacetylation degree was found 64.86%, which mean the chitin mixed with chitosan or crude chitosan. In which using concentration 0.5% alumina can only reduce the turbidity of water by 54.21% while chitin/chitosan can reduce up to 90.37% the turbidity of water.

Abstract

Lipid content and fatty acid composition of seaweed Ulva reticulata Forsskal obtained from Segara Sanur Beach have been analyzed. Lipid was extracted from this seaweed using soxhlet extractor with chloroform:methanol (2:1) as solvent, while the composition of fatty acid was analysed using gas chromatrography accomplished by a mixture of 37 standard fatty acids (ST37AL85.D01). It was found that Ulva reticulata Forsskal contained (1,41±0,04)% (w/w) of crude lipids. The crude lipid was composed of 11 major fatty acids including pentadecanoic acid (12,08%), heptadecanoic acid (40,00%), cis-10-heptadecenoic acid (4,37%), oleic acid (1,77%), linolelaidic acid (4,31%), linoleic acid (13,01%), arachidic acid (4,71%), g-linolenic acid (2,27%), heneicosanoic acid (7,58%), cis-11,14-eicosedienoic acid (8,19%), and cis-8,11,14-eicosetrienoic acid (1,71%).

Abstract

Changes in consumption behaviour to instant food can cause various health problems, such as obesity, dislipidemia, and atheroschlerosis. A study was conducted to investigate sardinella longiceps oil as anti dislipidemia through increasing HDL-C levels.This is a true experimental with randomized pre and posttest control group designs, emplyoying 50 Wistar rats grouped into five groups, i.e. control group (0% fish oil), treatment 1 group (10% fish oil), treatment 2 group (15% fish oil), treatment 3 group (20% fish oil), and treatment 4 group (25% sardinella longiceps oil). This research observed that intake of 20% sardinella longiceps oil resulted in the higest significant increase of HDL-C of 22.78 % (from 55.97 ±1.25 to 68.72 ± 6.25 mg/dL) for dislipidemia study. Therefore, it can be concluded that intake of sardinella longiceps oil in Wistar rat acts as anti dislipidemia by increasing the HDL-C levels. Further research is required to determine whether the application of sardinella longiceps on human will result in a simillar way, as anti inflammation and anti dislipidemia.

Abstract

This paper reports the process conducted to maintain a high quality of GC responses in obtaining reliabledata on steroid analyses of environmental samples. Works including setting up the Limit of Detection (LOD) andLimit of Quantitation (LOQ), maintaining linearity, controlling the reproducibility, and determining its reliability aredescribed. It is evident that in this work the LOD of the GC is 0.0002 ng (by 1 mL injection of 0.2 mgL-1 samples) andthe LOQ is 0.001 ng (by 1 mL injection of 1 mgL-1 samples), which means it is capable of detecting much lowerconcentrations of steroid compounds if the samples undergo 2000 times pre-concentration during preparation. Underthe range investigated the GC provides reliable quantitative linear responses to all steroid compounds.

Abstract

The objective of this research is to study the potential use microorganism which is identified as Enterobacter aerogenes isolated from sludge of textile wastewater treatment plant. Reactor was operated to treat remazol red RB azo dyes under aerobic condition. The performance of the bioreactor was evaluated by monitoring color removal within various pH of the medium, dye concentrations and incubation times. The degradation products were characterized by UV–vis techniques. The result showed, Enterobacter aerogenes bacteria was able to successfully decolourize 75 mg/L of remazol red RB azo dyes under aerobic condition, at pH 7-8 and for seven days incubation with biodegradation efficiency of up to 87,9%.
