Abstract

Practicum activities are an important component in the teaching and learning process because practicum activities provide learning to students about the basic concepts of experimentation. During the COVID-19 pandemic, face-to-face practicum learning activities cannot be carried out, so alternative online practicum activities are needed by utilizing learning media that are needed which still consider the objectives of practicum learning to improve students' abilities and skills. The best practices were prepared to know the interests and learning outcomes in online practical lessons for aquatic microbiology courses using the BLAST feature on the NCBI website for bacterial identification and making phylogenetic trees using MEGA software. The best practice was carried out in the odd semester of the 2021/2022 academic year, starting from November to December 2021. The best practice results show that students have a very high and high interest in this online practicum with the highest percentage of very high interest at 45.71%. All students received a complete score with a score above 70 with the highest score of 90. In conclusion, these best practices have a positive impact on online practicum learning so that it can be an alternative to online practicum learning in the next aquatic microbiology course. ABSTRAKKegiatan praktikum merupakan komponen penting dalam proses belajar mengajar karena kegiatan praktikum memberikan pembelajaran kepada mahasiswa mengenai konsep dasar eksperimen. Saat pandemi covid 19 kegiatan pembelajaran praktikum tatap muka tidak dapat dilakukan sehingga diperlukan alternatif kegiatan praktikum daring dengan memanfaatkan media pembelajaran yang sesuai kebutuhan dengan tetap mempertimbangan tujuan pembelajaran praktikum untuk meningkatkan kemampuan dan keterampilan mahasiswa. Best practice disusun dengan tujuan untuk mengetahui minat dan hasil belajar mahasiswa pada pembelajaran praktikum daring mata kuliah mirobiologi perairan menggunakan fitur BLAST pada website NCBI untuk identifikasi bakteri, dan pembuatan pohon filogenetik dengan menggunakan software MEGA. Praktek terbaik dilaksanakan pada semester ganjil tahun pelajaran 2021/2022 yaitu mulai bulan november sampai dengan Desember 2021. Hasil best practice menunjukkan bahwa mahasiswa memiliki minat sangat tinggi dan tinggi pada praktikum daring ini dengan presentase minat sangat tinggi tertinggi sebesar 45.71%. Keseluruhan mahasiswa mendapatkan nilai tuntas dengan nilai diatas 70 dengan nilai tertinggi yaitu 90. Dengan demikian best practices ini berdampak positif pada pembelajaran praktikum daring sehingga dapat menjadi alternatif pembelajaran praktikum daring pada mata kuliah mikrobiologi perairan selanjutnya.

Abstract

Saat ini banyak terjadi wabah penyakit pada ikan yang dapat menyebabkan kematian ikan sehingga menurunkan keuntungan budidaya ikan. Vaksin dapat digunakan untuk meningkatkan sistem imun ikan sehingga dapat digunakan sebagai salah satu alternatif dalam menjaga kesehatan ikan. Beberapa jenis vaksin telah diaplikasikan pada budidaya perikanan dan terbukti efektif dapat mengendalikan penyakit pada ikan. Penelitian ini bertujuan untuk mengetahui jenis-jenis vaksin yang efektif dapat meningkatkan sistem imun spesifik pada ikan untuk pencegahan penyakit pada ikan sehingga dapat meningkatkan produktivitas dan keuntungan budidaya ikan. Metode yang digunakan pada artikel ini adalah literature review yaitu dengan mengumpulkan artikel penelitian yang terdapat di google scholar dengan menggunakan kata kunci vaksin pada ikan. Berdasarkan hasil dari literature review ini diketahui beberapa jenis vaksin mampu meningkatkan sistem imun tubuh sehingga meningkatkan kekebalan tubuh ikan terhadap penyakit.

Abstract

Wild Oryzias javanicus from Tunda Island has potential as a native Indonesian aquatic ornamental fish and a model for research in the laboratory. The preliminary stage for raising wild fish is transporting from nature to the aquacul-ture environment. Generally, to minimize fish stress during transport in a closed system, anesthetics are added to the transport media. Furthermore, good adaptation in the aquaculture environment will lead to the process of reproduction as the ultimate goal of domestication of wild fish. The research objective is to analyze the transportation of closed systems and the rearing of O. javanicus post-transportation from Tunda Island. Research observations were carried out on behavior, survival, the number of eggs, and male: female sex ratio. Transport using an additional anesthetic treatment of 0.4 mL L-1 and without anesthesia, with fish density of 16 L-1. Anesthesia used is the commercial product Ocean Free® Special Arowana Stabilizer. Transportation is carried out for 11 hours. After that, rearing post-transportation is carried out for 16 days. The results showed that the transportation of O. javanicus can use a closed system without the addition of anesthesia with a density of 16 L-1 for 11 hours. In post-transport maintenance, fish begin active swimming on day 5, swim in groups continuously on day 6, start responding to natural food on day 3 and artificial feed on day 7, and produce a final survival of 91.67%. During maintenance, O. javanicus produces 104 eggs, 0.94 ± 0.06 mm in diameter and has an attaching filaments and a non-attaching filaments in the chorion. Oryzias javanicus is an egg depositor that can spawn with a 1: 1 and 1: 2 sex ratio. Abstrak Oryzias javanicus liar asal Pulau Tunda memiliki potensi sebagai ikan hias akuaskap asli Indonesia dan model untuk penelitian di laboratorium. Tahap awal untuk memelihara ikan liar yaitu melakukan pengangkutan dari alam ke lingkungan budi daya. Umumnya, untuk meminimalkan stres ikan selama pengangkutan sistem tertutup, dilakukan penambahan anestesi ke media pengangkutan. Selanjutnya, adaptasi yang baik dalam lingkungan budidaya, akan menyebabkan terjadinya proses reproduksi sebagai tujuan akhir domestikasi ikan liar. Tujuan penelitian yaitu meng-kaji pengangkutan sistem tertutup dan pemeliharaan pascapengangkutan O. javanicus asal Pulau Tunda. Pengamatan penelitian meliputi tingkah laku, sintasan, jumlah telur dan nisbah kelamin jantan : betina. Pengangkutan mengguna-kan perlakuan penambahan anestesi 0,4 mL L-1 dan tanpa anestesi dengan kepadatan 16 ekor L-1. Anestesi yang digunakan yaitu produk komersial Ocean Free® Special Arowana Stabilizer. Pengangkutan dilakukan selama 11 jam. Setelah itu, dilakukan pemeliharaan selama 16 hari. Hasil penelitian menunjukkan bahwa pengangkutan O. javanicus dapat menggunakan sistem tertutup tanpa penambahan anestesi dengan kepadatan 16 ekor L-1 selama 11 jam. Pada pemeliharaan pascapengangkutan, ikan mulai aktif berenang pada hari ke 5, berenang berkelompok secara kontinu pada hari ke 6, mulai merespons pakan alami pada hari ke 3 dan pakan buatan pada hari ke 7, dan menghasilkan sintasan akhir sebesar 91,67%. Selama pemeliharaan, O. javanicus menghasilkan 104 butir telur, diameter 0,94±0,06 mm dan dilengkapi filamen dan fili pada korion. Oryzias javanicus adalah egg depositor yang dapat memijah dengan nisbah kelamin 1:1 dan 1:2.

Abstract

This study was conducted to determine the best duration of hormone addition through the feed to induce the sex ratio of common carp strain Sinyonya. The study was using a completely randomized design with 5 different treatments namely A (control), B (30 days), C (35 days), D (40 days), and E (45 days) and each performed in 3 replications. The fish samples used in this study were a common carp strain Sinyonya larvae 5 days after hatching and still have the egg yolks. A total of 1.500 larvae of fish larvae were used. The fish larvae were given feed that had been mixed with 17a-methyltestosteron hormone. Feed was provided ad hormonal satiation at 5 times a day, (08.00, 10.00, 12.00, 14.00 and 16.00). The average percentage of the male fish with the highest value obtained in treatment C (35 days) is equal to 100.00±0.00. The result showed that the addition of 17a-methyltestosterone hormone on feed for 35 days and 40 days was the best treatment to improve the sex ratio of male of common carp strain Sinyonya.

Abstract

The culture of striped catfish (Pangasianodon hypophthalmus) is overgrowing and facing the problem of relatively high feed prices. The feed prices affect feed grade; the cheaper the feed price, the lower the quality. This low feed quality may improve its quality by supplementing Chlorella vulgaris. Therefore, this study aimed to determine the level of supplementation of C. vulgaris meal in the feed that produces the best growth performance in striped catfish juveniles. A completely randomized design with three treatments and three replications was used in this experiment. The treatments were various levels of C. vulgaris, namely 0%, 1.5%, and 3%. C. vulgaris meal was added to the commercial feed (26-28% protein) according to the treatment with the coating method. The test feed was fed to striped catfish juveniles (6.66 ± 0.04 g in average weight) reared in an aquarium containing 55 L of water at a density of 20 juveniles, at satiation in the morning, afternoon, and evening for 50 days of the rearing period. Survival rate and hepatosomatic indices of the striped catfish were not affected by various supplementation levels of C. vulgaris meal (P>0.05), but the values of feed consumption, specific growth rate, feed conversion, and protein efficiency ratio of the striped catfish (P<0.05). Supplementation of 3.5% C. vulgaris meal in feed resulted in the best growth performance for striped catfish.
