Abstract

Synthetic estrogen hormone that has known by the public is an estrogen that is contained in hormonal contraception or birth control pills. The lack of public knowledge about the action mechanism of oral contraception on the reproductive organs makes people ignoring about the impact of it. This study was conducted to analyze the differences in the induction of estrogen towards the levels of estradiol and histology of the uterus and its utilization as a supplement book of Reproductive System topic in high school. Balb-C female mices were used as the tested animals and Microgynon, the contraceptive pill’s trademark was used as a source of estrogen induces. Blood samples were taken at pro-estrus phase, while the uterus was taken at each phase of the estrous cycle. Measurement of estradiol levels was done by ELISA method, whereas the thickness of endometrium is determined on a longitudinal incision of the uterus by the method of preparation of histological paraffin and HE staining. The supplement book was arranged refers to 4-D models. The results showed that there was a difference between the levels of estradiol-induced estrogen treatment with control, but in endometrial thickness among the control treatment with estrogen did not show the differences significantly. The results also showed that the estrogen induction has no significant effect on endometrium thickness at each phases of estrous cycle. The results of research were used as supplement book and it can be used for enriching knowledge of reproductive topic on high school.

Abstract

Provision of teaching materials in term of module can be an alternative to meet the learning needs of science. The study aimed at describing science module constructed based on Science, Technology, Engineering, Art and Mathematics (STEAM) equipped with valid and effective biotechnology games for learning science in junior high school. This Research and Development (R&D) was developed using the 4D model (Thiagarajan). This research was conducted at MTsN (State of Islamic Secondary School) 2 Jember, involved 30 IX graders. The design used was one group pretest-posttest design as the test of effectiveness. This study used STEAM-based science module equipped with biotechnology games, test, and validation sheets as research instruments. The validation results by experts were converted into categories. The effectivity test data was analyzed using N-gain and paired samples t-test. The average of validation results of material, media, and users was 87.17 (very valid). The effectivity test results showed the increase of students? cognitive learning outcome [N-gain value = 0.72 (high category), t(29) = 9.030, p < 0.05]. Therefore, the module developed is valid and effective in improving the learning of biotechnology science which means that the use of this module is recommended to be implemented in biotechnology learning process.

Abstract

Detection of genes resistant to bacterial leaf blight in rice cultivars from Situbondo and Jember, Indonesia. Xanthomonas oryzae pv. oryzae (Xoo), causing bacterial leaf blight, is a destructive pathogen that significantly affects rice production. The use of resistant varieties is the most effective and economical strategy to reduce the impact of the disease. This study aimed to analyze the genetic basis of resistance to Xoo in rice. The incidence and severity of bacterial leaf blight were assessed in the field through a diagonal random sampling method. PCR was used to detect resistant genes in rice with eight Xa-specific primers. The presence of Xa genes and environmental conditions were statistically analyzed to determine whether the disease incidence and disease severity were related to average temperature and rain intensity. The means for disease incidence and severity at the generative stage were higher than those at the vegetative stage. The 12 rice cultivars were categorized into four groups based on the presence of Xa gene sequences; most cultivars had Xa10 and Xa13, and one cultivar had five Xa genes. Disease incidence and severity were weakly correlated to the presence of resistance genes, as well as to environmental factors. Most rice cultivars from the Regencies of Jember and Situbondo carry Xa10 and Xa13 resistance genes.

Abstract

The traditional sea-picking ceremony is a ritual carried out by the community as a form of gratitude for the abundance of sustenance provided by God, as well as for asking the safety of fishermen. Besides aiming for blessings and safety, the traditional sea picking ceremony also has an important role as a means of communication for fishermen and maintaining harmony among religious communities, therefore this traditional sea picking ceremony must be held once every year. The purpose of this study was to observe the nature and processing of plants and animals at sea picking ceremony and to find out the Use Value (UV) of plants, animals and the natural environment used. Sampling of informants was done by Purposive Sampling and Snowball Sampling; data obtained using semistructured interviews with open ended question types. The results showed there were 66 species from 37 plant families and 3 species from two animal families. While the Use Value (UV) analysis indicates that there were 12 plant species and one animal species that have a high Use Value (UV) which is 1, it shows that the species is most widely used in traditional sea picking ceremonies. Generally, it needs more efforts for further conservation.

Abstract

In 21st century learning, critical thinking skills are the main choice that must be mastered by students. This study aims to determine the effectiveness of local wisdom-based science teaching materials in improving junior high school students’ critical thinking skills. Most science teachers still apply teacher-centered learning, so students tend to be passive. This type of research is quasi-experimental with a pretest-posttest control group design. The study was conducted in the even semester of the 2019/2020 school year. The subjects of the research were grade VII students. Data collection techniques and instruments in the form of tests that include indicators of critical thinking skills. Data analysis used homogeneity test, Kolmogorov Smirnov normality test, independent sample t-test, and n-gain score. The results of the independent sample t-test are sig. (2-tailed) = 0,000<sig. α = 0.05. Students’ critical thinking skills in the experimental class were higher (N-gain score = 39.09%) than in the control class (N-gain score = 12.03%). Based on the study results, it can be concluded that there is a significant influence on the application of local wisdom-based teaching materials in improving the critical thinking skills of middle school students in the moderate category.

Abstract

Foodborne disease caused by Escherichia coli contamination is increasing every year. It also followed by elevating of drug-resistance of E. coli. Bacteriophage can be an alternative for therapy infection.  This study aimed to determine synergism effect of bacteriophage ϕPT1b which has a high rate virulence to E. coli and phage-antibiotics (tetracycline and amoxicillin) synergy. The indigenous bacteria isolates were KR, MJ, KP, PT, PR. Five bacteriophages used namely ϕKR1b, ϕKR2, ϕPT1a, ϕPT1b, and ϕMJ1b. Virulence test was used to determine the ability of each phage in reducing E. coli. Treatment to examine synergism of phage ϕPT1b and antibiotics were P1: amoxicillin, P2: ϕPT1b, P3: ϕPT1b + Amx = 1:1, P4 : ϕPT1b + Amx = 2:1, P5: ϕPT1b + Amx = 1:2, P6 : tetracycline, P2: ϕPT1b, P7: ϕPT1b + Tet = 1:1, P8 : ϕPT1b + Tet = 2:1, and P9: ϕPT1b + Tet = 1:2. The virulence test showed that isolate ϕPT1a with 106 CFU/ml had the highest ability in reducing E. coli. While, the result of synergism test indicated that the synergism of bacteriophage and antibiotics differ significantly (P ≤ 0.05). The best ratios of synergism were 1:1 (ϕPT1b+tetracycline) and 2:1 (ϕPT1b+amoxicilline). In summarize, phage-antibiotic synergy (ϕPT1b with tetracycline/amoxicilline) can reduce the level of antibiotic resistance in isolated E. coli.
