Abstract

The low learning motivation of junior high school students can be improved through media with lots of colors and pictures. This study aims to develop Webtoon-based comic  electronic  learning  media  on  the  material  of  the human respiratory system to increase the learning motivation of grade VIII junior  high  school  students  that  is  valid,  practical,  and  effective.  Media development uses the ADDIE model strategy. Media validation is based on the assessment of 4 expert validations. The practicality test is based on the opinion of the teacher and student representatives. The effectiveness test was carried out at class VIII SMPN 11 Jember with 32 students. Statistical test ofdata  processing  using  Paired  Sample  t-test.The  results  of  the  research cover the validity test by 4 expert validators with an average percentage of 91.5% with very valid criteria, the practicality test by students has an overall average  percentage  of  91.4%  with  a  very  practical  category,and  the effectivenesstest   with   the   ARCS   questionnaire   before   and   after implementation  has  consistently  increased  the  average  value  in  4  aspects, namely the aspect of gaining attention increase of 0.22, Relevance gets 0.23, Confident  gets  0.20  and  Satisfaction  gets  0.14,  the  difference  in  the  total mean of the ARCS questionnaire is 0.20.Seeing the enthusiasm of students in learning to use comics is very large, it is suggested that electronic comics can be made in other learning materials.Keywords: E-Comic, Webtoon,  Human  Respiratory  System,  Learning Motivation, ADDIE; ARCSRendahnya  motivasi  belajar  siswa  SMP  dapat  ditingkatkan  melalui  media yang  diperbanyak  warna  dan  gambarnya.  Penelitian  ini  bertujuan  untuk mengembangkan media  pembelajaran elektronik komik  berbasis Webtoonpada materi sistem pernapasan manusia untuk meningkatkan motivasi belajar siswa SMP kelas VIII yang valid, praktis dan efektif. Pengembangan media menggunakan  strategi  model  ADDIE.  Validasi  media  didasarkan  pada penilaian 4  validasi  ahli.Uji  kepraktisan  didasrkan  pada  pendapat  guru  dan perwakilan  siswa.  Uji  efektifitas  dilakukan  di  SMPN  11  Jember  kelas  VIII dengan 32 siswa.Uji statistic pengolahan data mengunakan Paired Sample t-Test. Hasil  dari  penelitian  melingkupiuji  validitas oleh  4  ahlivalidator  ahli dengan  persentaserata-ratasebesar  91,5%  dengan  kriteria  sangat  valid,  uji kepraktisan oleh siswa memiliki presentase rerata keseluruhan sebesar 91,4% dengan  kategori sangat  praktis,  dan  Uji  Efektivitasdengan  angket  ARCS sebelum  dan  sesudah  penerapan  mengalami  kenaikan  nilai  rerata  secara berurutan  dalam  4  aspeknya  adalah  aspek Attentionmendapatkan  kenaikan 0.22, Relevancemendapatkan   0.23, Confidentmendapatkan   0.20   dan Satisfactionmendapatkan 0.14, selisih total rerata angket ARCS adalah 0.20.Melihat  antusiasme  siswadalam  pembelajaran  menggunakan  komik  sangat besar,  maka  disarankan  agar  komik  elektronik  dapat  dibuat  pada  materi pembelajaran lainnya

Abstract

 AbstrakSARS-CoV-2 merupakan virus yang menyebabkan Coronavirus Disease 2019 (COVID-19) di seluruh dunia dan sampai saat ini kasus terbaru masih terus dilaporkan. Diagnostic test merupakan hal yang krusial untuk dikembangkan. Prinsip diagnostic test COVID-19 berbasis antigen, yaitu mendeteksi virus SARS-CoV-2 melalui respon antibodi dari penderita. Penelitian ini bertujuan untuk memproduksi antibodi poliklonal menggunakan protein rekombinan RBD-Spike untuk mendeteksi virus SARS-CoV-2 berbasis antibodi. Penelitian dimulai dengan penentuan domain RBD-Spike menggunakan pensejajaran asam amino, dan konstruksi DNA untuk RBD-Spike pada vektor ekspresi pET28a menggunakan sintetik nukleotida. Produksi protein rekombinan RBD-Spike diekspresikan pada sel bakteri Escherichia coli. Purifikasi dilakukan untuk memperoleh protein RBD-Spike dan selanjutnya digunakan sebagai antigen untuk induksi antibodi poliklonal pada kelinci. Hasil penelitian menunjukkan bahwa ekspresi protein rekombinan RBD-Spike SARS-CoV-2 memerlukan induksi IPTG 0,1 mM dan terekspresi dalam bentuk inclusion bodies dengan ukuran 39 kDa. Purifikasi protein RBD-Spike dilakukan menggunaan resin afinitas NiNTA, elektroelusi, dan dialisis. Total protein RBD-Spike yang diperoleh sebanyak 4 mL dengan konsentrasi 10 mg/mL. Analisa Ouchterlony menunjukkan bahwa antibodi poliklonal terdeteksi pada minggu kedua setelah injeksi booster dan analisa spesifitas antibodi terhadap antigen menunjukkan bahwa antibodi poliklonal dapat mendeteksi protein RBD-Spike pada konsentrasi 0,1 µg. Selanjutnya diharapkan antibodi poliklonal dapat digunakan untuk deteksi keberadaan virus SARS-CoV-2 dan dapat dikembangkan untuk kit deteksi berbasis antibodi.AbstractSARS-CoV-2 is the virus that causes Coronavirus Disease 2019 (COVID-19) worldwide and the latest cases are still being reported until now. The diagnostic test is a crucial to be developed. The principle of the antigen-based COVID-19 diagnostic test is to detect the SARS-CoV-2 virus through antibody response from the patients. This study was conducted to produce polyclonal antibodies using recombinant protein RBD-Spike. The research was carried out by determining the RBD-Spike domain using amino acid alignment and constructing the DNA of RBD-Spike to the expression vector of pET28a using nucleotide synthesis. Production of RBD-Spike recombinant protein was expressed in Escherichia coli. Purification was carried out to obtain RBD-Spike protein and used to induce polyclonal antibody ina rabbit. The results showed that the expression of RBD-Spike recombinant protein required induction of IPTG 0.1 mM and was expressed in inclusion bodies with molecular size of 39 kDa. The purification of RBD-Spike protein was carried out using resin affinity, electroelution, and dialysis. The total protein of RBD-Spike obtained was 4 mL with a concentration of 10 mg/mL. Ouchterlony analysis revealed that polyclonal antibody was detected in the second week after booster injection and analysis of antibody specificity showed that polyclonal antibodies detected RBD-Spike protein at the concentration of 0.1µg of RBD-Spike protein. Moreover, it is expected that our polyclonal antibody detect the SARS-CoV-2 virus and can be developed for antibody-based detection kits.

Abstract

Critical thinking skills are needed in the 21st century, so efforts are being made to improve them, one of which is by developing research-based teaching materials, and research into the use of Extremely Low Frequency Electromagnetic waves to increase food security is currently being carried out very intensively. This research aims to describe the development of research-based teaching materials to improve critical thinking skills in the use of Extremely Low Frequency Electromagnetic waves to increase food security. This development research uses the Borg and Gall method, which consists of 10 steps. The subjects of this research were 90 students at the University of Jember. Undergraduate Physics Education students in classes A, B, and C in environmental physics courses and on material on the use of Extremely Low Frequency electromagnetic waves to increase food security. This research uses quantitative analysis. The results of this research state that research-based teaching materials are effective in improving students' critical thinking skills in the material on the use of Extremely Low Frequency Electromagnetic waves to increase food security with a score of 82.33 based on critical thinking indicators.
