Abstract

ABSTRAKPenyakit pullorum merupakan penyakit yang sering menginfeksi ayam yang disebabkan oleh Salmonella pullorum. Penelitian ini bertujuan untuk mengidentifikasi ada tidaknya penyakit pullorum pada ayam petelur periode grower di peternakan ayam kabupaten Sidrap. Pengambilan sampel menggunakan metode purposive sampling. Sampel serum dan organ diambil dari 19 peternakan ayam di Batu-Batu. Sebanyak 57 sampel serum diuji dengan uji aglutinasi. Diperoleh sebanyak 26 sampel serum yang positif dan dilanjutkan dengan pemeriksaan makroskopik. Pemeriksaan makroskopik ditemukan adanya hepatomegali, lesi berwarna kuning-pucat, konsistensi lunak, dan nodul-nodul di permukaan hati serta adanya kardomegali. Hasil penelitian ini menunjukkan bahwa peternakan ayam layer periode grower di kabupaten Sidrap telah terinfeksi Salmonella pullorum dangan gambaran patologis yang bervariasi.  Kata kunci: ayam petelur, identifikasi, Salmonella pullorum, makroskopikIdentification of Salmonella Pullorum in Grower Period of Laying Hens By Agglutinated and Macroscopic Test in Layer Farms of Sidrap RegencyABSTRACTPullorum is a bacterial disease that commonly infected the chicken infected by Salmonella pullorum. This research aimed to identify Salmonella pullorum in the growing period of laying hen in Batu-Batu. The research used a purposive sampling method. The organ and serum samples were collected from 19 farms. Agglutination test for serum and macroscopic test for organ was performed. There were 26 samples that tested positive from 57 serum samples. There were 26 organ samples of macroscopic treatment that resulted in liver damages in the forms of swelling, pale yellow lesions, soft consistency, and nodules on the surface. The heart also experienced changes through swelling, soft consistency, uneven surface, and a mixture of fluid and fibrin inside the pericardium. It can be concluded that layer farms in Sidrap districts have been infected by Salmonella pullorum affecting the growing period.Keywords : laying hens, identification, Salmonella pullorum, macroscopic

Abstract

Pterygoplichthys spp or commonly referred as Pleco is originated from Amazon River in South American Continent.  The aim of this study is to isolate and identify the pathogenic gram-positive bacteria in pleco's kidney (Pterygoplichthys pardalis) and to observe the pleco's kidney histopathology colonized by gram-positive pathogenic bacteria in Sidenreng and Lapompakka Lakes. The samples employed in this experiment includes 10 plecos with each 5 samples represented each lake. The isolation and identification of positive gram bacteria were performed through bacterial culture produced from pleco's kidney swab test on blood agar using gram staining followed by biochemical testing with Vitek 2 compact system. From the results, there were 4 gram-positive bacteria successfully isolated, one of them was Enterococcus faecalis which is the suspected pathogen. Specimen preparation for kidney histopathology was carried out by 10% neutral buffered formalin (NBF) fixation, graded ethanol series dehydration, paraffin wax embedding, 4 μm sectioning, Hematoxylin and Eosin Staining and after that, the specimen was observed. Data analysis employed qualitative descriptive method. From the observation, damages such as necrosis, inflammatory cell infiltration, hemorrhage, and hypertrophy were identified from the pleco's kidney. Kidney damages were most probably caused by the infection by gram-positive pathogenic bacteria Enterococcus faecalis. Keywords : Sidenreng Lake, Lapompakka Lake, Kidney, Histopathology, Plecos.

Abstract

Amazon Sailfin Catfish (Pterygoplicththys pardalis) or commonly known as Plecos is originated from Amazon River in South America Continent. The aims of this study are to identify and isolate pathogenic gram-negative bacteria in the liver of the plecos from Lapompakka and Sidenreng Lakes and to observe the histopathological changes in liver identified with gram-negative bacteria infection. 10 samples were collected from both lakes with each 5 representing each lake. Identification and Isolation of gram-negative bacteria was performed through culture of pleco's liver swab results in MacConkey media using gram staining method and were continued by biochemical testing using VITEK 2 System. The acquired results indicated that there were 8 different species of gram-negative bacteria isolated and all of them were pathogenic to the plecos. The species of bacteria includes Klebsiella pneumoniae, E.coli, proteus sp., Acinetobacter baumannii, Aeromonas sobria, Enterobacterb cloacae complex, Citrobacter freundii, Enterobacter aerogenes. Specimen preparation for liver histopathology was carried out by 10% neutral buffered formalin (NBF) fixation, graded ethanol series dehydration, paraffin wax embedding, 4 μm sectioning, Hematoxylin and Eosin Staining. After that, the specimen was observed under a microscope. Data analysis employed qualitative descriptive method. From the observation, liver damages such as necrosis, inflammatory cell infiltration, hemorrhage, and cell hyperthrophy were identified. Abstract

Newcastle Disease is a contagious disease caused by Avian Paramyxovirus and infects different types of poultries including broiler. Vaccination as a preventive effort against the ND virus could employ both attenuated and inactivated vaccines. This study aims to identify the effect of administering live-attenuated LaSota and B1 ND vaccines against broiler antibody titers. This experiment employed 54 DOCs and was divided into 3 groups of treatments. Vaccination was performed at the age of 3 days old through eye drop administration. Blood specimens were taken from vena brachialis at the age of 7, 14, 21 days old. Hemagglutination Inhibition (HI) assay was analyzed using One-Way Analysis of Variance (ANOVA) followed by Least Significant Different (LSD) test if the probability is significant (P<0.05). The data of Immune Percentage is presented using descriptive quantitative analysis.   The research results demonstrated that live-attenuated LaSota ND vaccination at the age of 3 days could sustain and induce immunity until the age of 21 days while B1 ND vaccination at the age of 3 days could only sustain immune protection until the age of 14 days. Live-attenuated LaSota and B1 ND vaccines did not have any significant effect on the broiler antibody titers.   Keywords: Broiler, Newcastle Disease (ND), LaSota vaccine, B1 vaccine, Hemagglutination test, Hemagglutination Inhibition Test

Abstract

Adaptogenic activity was related to antioxidant activity which can reduce the number of free radicals, which in turn will reduce stress and improve body balance. Mangosteen rind (Garcinia mangostana L.) with a large antioxidant content promises to be Adaptogen. This study aims to examine the Adaptogenic effects of Ethanol Extract of mangosteen peel (Garcinia Mangostana L.) Using the Swimming Endurance Test (SET) in Mice. Simplicia of Mangosteen Fruit extracted by maceration using 96% ethanol, then tested its Adaptogen activity based on swimming endurance test. The group consisted of negative control (NaCMC 1%), positive control (Vit C. 100 mg / kg BB) and 3 groups treatment of ethanol extract of mangosteen peel ranging from 100, 200 and 400 mg / kg BB where all groups were given oral 1x1 oral administration for 7 days and on the 8th day, a SET test were performed. After that, data analysis was performed statistically. The results showed that the ethanol extract of mangosteen peel had the ability to be Adaptogenic where mice swimming in all groups given mangosteen peel extract was longer than the negative group with a significant difference (p <0.05). Ethanol extract of mangosteen peel showed a strong adaptogenic antistress effect in mice based on increased swimming time ability of mice.

Abstract

Assessment of meat physical quality is the way to choosing meat. Pre slaughter time is one method to restorate livestock (including poultry) prior to deductions so that animals do not wage for too long at the time of slaughter. This study aims to determine the effect of pre slaughter fasting on the quality of meat produced. This study using 24 samples of broiler chickens. The sample is divided into 4 groups where group 1 without treatment; group 2 4 hours of fasting; group 3 8 hours of fasting; and group 4 12 hours of fasting. The parameters include specific gravity, pH, color, tenderness, and water holding capacity. The result of this study found that fasting on broiler chickens up to 12 hours did not give a significant effect on specific gravity, tenderness, and water holding capacity. Pre slaughter fasting until 12 hours only gives effect to pH and color of meat. The fasting time is 8 - 12 hours before slaughter not recommended because it has the biggest pH possible to get pale, soft, exudative (PSE) .

Abstract

Catfish is one of the fish that has a high economic value so that it becomes one of the export commodities to meet the consumption and protein needs of the community so that it is widely cultivated. In cultivation, catfish can be attacked by various diseases, one of which can be caused by bacteria. The bacteria that often cause death in fish, both marine and freshwater fish, is Yersinia spp. Yersinia spp is a group of gram-negative bacteria, in the form of bacilli and motile and is also reported to be one of the pathogenic diseases that often attack fish. samples were taken on August 12, 2021 from the mouth organs of catfish at the Hasanuddin University Education Animal Clinic. Samples were cultured in Tryptic Soy Agar (TSA) for 24 hours and then purified in Mac Conkey Agar (MCA). The culture results showed the presence of bacterial growth. Follow-up examination using a microscope and showed the presence of red (Gram negative) and bacilli-shaped bacteria.

Abstract

Leptospirosis is a disease caused by Leptospira spp., zoonotic Gram-negative bacteria. This study aimed to determine the handling of a leptospirosis case in a dog at the Pet Vet Clinic, Jakarta. Handling began with anamnesis and continued by examining the patient's physical condition, followed by serological, microscopic, and polymerase chain reaction examinations. An examination revealed that the dog had Leptospirosis. The therapy given was the administration of crystalline procaine and penicillin-G antibiotics. Supportive treatment was given with hepatoprotective, lipotropic, and diuretic supplements. Ringer's lactate was used as fluid therapy. The treatment administered showed positive, progressive results in the patient.
