Abstract

Penelitian ini dilakukan untuk mengkaji dampak suplementasi 1,25-dihidroksivitami D3, terhadap ekskresi kalsium(Ca) dalam urin dalam akumulas Ca pada tikus Wistar panhisterektomi yang mengkonsumsi teri tawar. Lima belas tikus Wistar betina umur 8 minggu, dibagi 3 kelompok (kontrol, panhisterektomi dan panhisterektom+suplemen 1,25-dihidroksivitan D3) masing-masing 5 tikus. Ketika berumur 19 minggu, masing-masing tikus dimasukkan kandang metabolikin dividu untuk studi balan. Selama studi balan(hari 4 -11 studi balan), setiap pagi dilakukan pengumpulan feses urin dan sisa pakan untuk pemeriksaan Ca. Hasil analisis menunjukkan konsumsi Ca tikus panhisterektomi yang mengkonsumsi 1,25-dihidroksivitami D3 lebih rendah meskipun  tidak berbeda signifikan dibandingkan tikus panhisterektomi yang tidak mengkonsumsi 1,25-dihidroksivitamin D3, ekskresi Ca dalam feses lebih tinggi dan berbeda sangat signifikan (p<0,01), ekskresi Ca dalam urin lebih tinggi dan berbeda signifikan (P<0,05), retensi Ca lebih rendah dan berbeda signifikan (p<0,05)dengan tikus panhisterektorni yang tidak mengkonsumsi 1,25-dihidroksivitaminD3. Dari hasil penelitian dapat disimpulkan bahwa suplemen 1,25-dihidroksivitamin D3, menyebabkan hiperkalsiuria menurunkan retensi Ca dengan demikian menurunkan akumulasi Ca pada tikus Wistar panhisterektomi.Kata kunci: kalsium, 1,25-dihidroksivitamin D3, hiperkalsiuria

Abstract

Leptin is a hormone produced by adipocytes and plays an important role in wound healing. The objectives of this research were to identify leptin and total leukocyte after incision on rat fed high fat diet and zinc topicalapplication. Sixteen male Sprague Dawley rats at 3 months of age were used in this study. Rats were randomly allotted into 4 groups (A,B,C and D) of 4. Rats in group A and B were fed normal diet, while rats in group C and D were fed high fat diet. After 2 months of treatment, skin incision was done at the back side of the rat. Incision wound was then closed with single interrupted suture. The wound of the rats in group A and C was treated with vaseline, while in group B and D was treated with zinc 10%. Three days after surgery, blood was collected from each rat for leptin analysis and total leukocyte. Leptin level and total leukocyte were analyzed statistically using ANOVA for factorial experiment (2x2). The results showed that leptin level was significantly higher in rats fedhigh fat diet compared to that of the rats fed normal diet (P<0.05). There was no significant different on total leukocyte. Therefore, it was concluded that leptin level was affected by diet, while total leukocyte was notaffected neither by diet nor topical application. However, total leukocyte tends to be lowered in the rat with zinc topical application.Keywords: high fat diet, leptin, zinc, total leucocyte, Sprague Dawley rats

Abstract

Suplemen calcitriol memicu pembentukan tulang, namun juga meningkatkan resorpsi tulang. Penelitian ini dilakukan untuk mengkaji efektivitas calcitriol dalam menurunkan risiko osteoporosis tikus ovariektomi. Lima belas tikus Wistar betina umur delapan minggu, dibagi tiga kelompok (kontrol normal KN, kontrol ovariektomi KOV, dan ovariektomi+ calcitriol OVD ) dan diberi perlakuan selama delapan minggu. Pada akhir perlakuan, 100 dilakukan pengambilan darah melalui pleksus retroorbitalis untuk pemeriksaan estradiol, selanjutnya semua tikus dietanasi menggunakan campuran ketamin10% dengan xylaxine 2%. Femur kiri diambil untuk pemeriksaan histopatologis. Hasil analisis menunjukkan tikus KOV mempunyai konsentrasi estradiol yang secara signifikan lebih rendah dibandingkan tikus KN, dan tikus OVD mempunyai konsentrasi estradiol tidak 100 berbeda signifikan dibanding tikus KOV. Gambaran histopatologis epifisis tulang femur distalis tikus OVD 100terlihat mempunyai lebih sedikit jaringan adiposit dan lebih banyak spikulum trabekula dibanding tikus KOV. Dari hasil penelitian disimpulkan bahwa suplementasi calcitriol 100ng/hari selama delapan minggu pada tikus ovariektomi menurunkan risiko osteoporosis.

Abstract

The research on physiological and reproduction status of Bawean deer (Axis kuhlii) in its habitat has been conducted, to understand and to find out as a basic information on Bawean deer (Axis kuhlii) in its habitats abaseline data for wildlife conservation efforts. The deer is categorized as an endangered animal, therefore, more attention was given toward Bawean deer conservation. Habitat changes, loss of habitat, fragmentation and illegalhunting might caused the wild animals become more marginalized and the populations have been declined. Therefore, it is needed to have research as an effort to save Bawean deer in its natural habitat. This research activity covered monitoring on physiological and reproduction status, and also the examination of deer's feces samples was parasite coprological data that serve as a basic consideration data in conservation and determination of Bawean deers health in the nature and/or conservation. The research was done in Bawean Island, Gresik and East Java, by taking faces of Bawean deers in the nature and conservation. Data gathering of physiological and reproduction status were done by examining deer's condition, either directly in conservation or in its habitat, and also by interviewing people around the forest. The data on physiological status, behavior monitoring, reproduction status, faces samples, blood, food analysis, and interview result were analyzed descriptively. Some worm eggs were found during feces examination, those were Strongyl, Strongyloides sp, Trichuris sp, Fasciola gigantica and Oocysta coccidia. The result showed that the same parasites were found in the cattle and goat raised by people around in the vicinity of the forest, meaning, there was an interaction between wildlife andlivestock. According to the result, it is needed to monitor the physiological status of Bawean deers routinely since Bawean deers is classified as endangered species.

Abstract

Normal range of calcium (Ca) level in the blood is maintained by Ca homeostasis in the intestinal, kidney and bone. The objective of this research was to study the response of calcium homeostasis system ovariectomized rats consuming combination of calcitriol and raloxifene by calcium balance study. Twenty five female Wistar rats at 8 weeks of age were divided into five groups of five: normal control (NK), ovariectomy control (OVK), ovariectomy+calcitriol (OVD ), ovariectomy+ raloxifene (OVR), and ovariectomy+calcitriol+ 20raloxifene supplementation (OVDR), of five each. Seven weeks after the surgery, each rats was placed into individual metabolic cages for Ca balance studies (Ca consumption, feces and urine Ca excretion, and intestinalCa absorption). In 5 to 8 days of the balance studies, the remaining food, urine, and feces were collected every day for Ca analyses. The results showed that Ca consumption, feces and urine Ca excretion, and intestinal Ca absorption of OVDR rats were higher than OVK rats. Consumption and feces Ca excretion of OVDR rats were higher than OVK rats which informed the decrease of estrogen hormone. Intestinal Ca absorption of OVDR rats was higher than OVK rats which informed the decrease of parathyroid hormone. Meanwhile, urine Ca excretionof OVDR rats was higher than OVK rats which informed the decrease of parathyroid hormone and estrogen. In conclusions, the response of calcium homeostasis system in ovariectomized rats consuming combination ofcalcitriol and raloxifene for 8 weeks was marked by increasing Ca absorption in intestine and Ca excretion in urine.

Abstract

Calcitriol supplementation in ovariectomized rats decreased bone resorption and increased bone formation, however, it depend on dose. The objective of the research was to study the response of femur bone in osteoporosis rats consuming calcitriol. Thirty female Wistar rats at 8 weeks of age were randomly divided into six groups (sham operated rats as normal control rats/group N and NK, ovariectomized control rats/group Ov and OvK, ovariectomized rats/group OvDand OvE) of five each. All rats were fed standard diet for 8 weeks. At 16 weeks  of age, group N and Ov  were euthanized, the right femur were taken for histopathological  examination. Group NK and OvK were fed a standard diet, group OvD was fed a standard diet +40ng calcitriol;  and group OvE was fed a standard diet+25µg ethynil ethyl estradiol. Treatments were done for six weeks. At the end of study, blood samples were taken from plexus orbitalis medialis for estrogen analysis. All rats were euthanized using ketamine10% and xylazine 2%. Right femur was taken for histopathological examination  using hematoxylin and eosin stain, and immunohistochemistry using monoclonal antibody anti TRAP5b which was detected with streptavidin-biotin.         The results showed that estradiol level of the rats in group OvD was not significantly different compared with the rats in OvK group, however, it was significanly lower compared to the rats in group OvE. Histopathologic figure of right distal femur metaphysis in group OvD was shown lesser adipocyte in the bone marrow and more trabecular bone speculum compared to group  OvK, however, there was more adipocyte in the bone marrow and lesser trabecular bone speculum compared to group OvE. Immunohistochemistry of distal femur metaphysis in group OvD and OvE were revealed tartrate resistant alkaline phosphatase 5b (TRAP5b) expression in trabecular bone, which was located in bone marrow space and trabecular speculum surface as well. Based on the results, it can be concluded that calcitriol 40ng/day supplementation in osteoporosis rats for 6 weeks decreased bone resorption and increased bone formation distal femur metaphysis.

Abstract

Calcitriol supplementation in ovariectomized rats for 8 weeks reduce osteoporosis risk and safe for kidney depend a proper dose. The objective of the research was to study the efectivity of high dose calcitriol for osteoporosis prevention and safe for kidney  in ovariectomized Wistar rats. Fifteen female Wistar rats at 8 weeks of age were randomly divided into three  groups of five which were sham operated rats (KN), ovariectomized rats (KOV), and ovariectomized rats+calcitriol supplementation 200ng/day (OVD200). After 8 weeks of treatment, blood samples were taken from plexus orbitalis medialis for estrogen analysis. All rats were then euthanized, left tibia and left kidney were taken for histopathological examination using hematoxylin and eosin stain. Immunohistochemistry using anti TRAP5b monoclonal antibody was also done for left tibia by streptavidin-biotin.  The results showed that estradiol level of KOV was significantly decreased compared with KN (P<0.05), meanwhile, estradiol levels of OVD200 were not significantly different compared with KOV. Histopathologic figure of kidney in KOV was not different compared with KN. Glomerulus was surrounded by Bowman‘s capsule, proximal and distal convoluted tubulus were lined with typical cuboidal epithelium. Immunohistochemistry result of proximal tibial epiphysis in KOV were shown more tartrate resistant alkaline phosphatase 5b (TRAP5b) expression in trabecular bone, which was located in bone marrow space, and trabecular speculum surface as well. Meanwhile, proximal tibia epiphysis in OVD200 had less tartrate resistant alkaline phosphatase 5b (TRAP5b) expression compared with KOV. Histopathologic figure  of proximal tibia epiphysis in group OVD200 was shown more trabecular bone speculum and less adipocyte in the bone marrow compared with KOV. Histopathologic figure of kidney in OVD200 was showed glomerular  hypertrophy and atrophy, as well as protein deposits in proximal convoluted tubules epithelium.  In conclusion, calcitriol supplementation 200ng/day in ovariectomized rats for 8 weeks prevented osteoporosis risk but trigger urolithiasis. Key words :  : Calcitriol, osteoporosis, ovariectomized rats, urolithiasis

Abstract

High phosphor diet caused acute nephrosis and osteoporosis. 1,25-dihidroksivitamin D3 supplementation doesn’t changed the structure of kidney and effective for osteoporosis treatment in ovariectomized rats. The objective of the research was to study the effectivity of calcitriol for acute nephrosis and osteoporosis treatments in adult Wistar rats induced high phosphor diet. Twenty five female Wistar rats, at the age of 8 weeks were randomly divided into five groups (Nk, N, ODk, OD, and OD40). Rats in Nk and N groups were fed standard died, while rats in ODk, OD, and OD40 group were fed high phosphor diet (Ca:P=0,6%:2,4%) for 8 weeks for acute nephrosis and osteoporosis induction. As control acute nephrosis and osteoporosis, after 8 weeks of induction, blood samples were taken from retro orbital plexus for calcium and phosphor analysis. Nk dan ODk rats were then euthanized, right tibia and kidney were taken for histopathological examination using hematoxylin and eosin stain. The results showed that calcium and phosphor levels of ODk were not significantly different compared with Nk. Histopathological analysis of the kidney and proksimal tibia epiphysis in ODk group were showed acute nephrosis and osteoporosis. For the treatment of acute nephrosis and osteoporosis, OD40 rats was given standar diet and calcitriol 40 ng, meanwhile, N and OD rats were only given fed with standar diet for 5 weeks. At the end of the study, blood samples were taken from retro orbital plexus for calcium and phosphor analysis. N, OD, and OD40 rats were then euthanized, right tibia and kidney were taken for histopathological examination using hematoxylin and eosin stain. The results showed that calcium and phosphor levels of OD were not different compared with N. Calcium levels of OD40 was not different compared with N, meanwhile phosphor levels of OD40 were significantly increased compared with N. Histopathologically, kidney of OD40 was showed improvement that dose to normal. Meanwhile, proximal tibial epiphysis of OD40 were shown more trabecular bone speculum and less adipocyte in the bone marrow of epiphysis compared with OD. In conclusion, calcitriol 40ng/day supplementation in adult rats for 8 weeks was effective for acute nephrosis and osteoporosis treatments.
