Abstract

The recent study was carried out to determine the effect of Kebar grass extract of N-hexane  (Biophytum petersianum Klotzsch) for 28 days peroral on weight, ADG and weight of vital organs of white rat. Forty male and female rats of Sprague Dawley strain aged 8 weeks were examined. The formulated rats were divided into four groups, group I (control), group II, III, IV (treatment), in which each consisted of five rats. The feed were given as much as 15 g/rat/day, while drinking ad libitum. In the control group, the rats were given solvent extract, group II, III and IV were given fraction of n-hexane of Kebar grass extract at 1 mg/kg BW, 5 mg/kg BW and 10 mg/kg BW peroral, respectively. Test preparation was given daily for 28 days, observation of toxic symptoms was performed every day while weighing was performed once every two weeks. Based on results obtained in this line of research, the findings indicated that Kebar extract administration for 28 days did not cause toxic symptoms in both male and female rats either in dose II, III or IV. Weight and ADG of male and female rats did not indicate significant differences between the control group and the treatment group. Keywords:  kebar grass, rats, subcronic toxicity test, OECD 407

Abstract

ABSTRACT The utilization of manure as organic fertilizer is on the rise, but the coarse form may negatively impact the aesthetics. This community service aims to train the farmers to operate the livestock manure grinder machine. The community service was performed in the Sidomaju livestock group, Panggungharjo village, Sewon, Bantul, Yogyakarta, with 37 farmers. The method for this community service is providing online presentations followed by online and offline workshops. The participation and satisfaction of the participants were measured by direct interaction and analyzed descriptively. The results of this community service showed that all farmers were more enthusiastic to participate in offline activities rather than online ones. All of the participants of the training can operate the livestock manure grinder machine. Based on the results of this activity, it is concluded that the training on the use of grinders for livestock manure machine has been adopted by farmers. Keywords: Manure; Manure grinder; Livestock; Organic manure ABSTRAK Pemanfaatan kotoran ternak sebagai pupuk organik semakin meningkat, tetapi bentuk yang kasar dapat mengurangi estetika. Pengabdian kepada masyarakat ini bertujuan melaksanakan pelatihan penggunaan alat pelembut kotoran ternak. Pengabdian dilakukan di kelompok ternak Sidomaju, desa Panggungharjo, Sewon, Bantul, Yogyakarta yang beranggotakan 37 peternak. Metode kegiatan yang dilakukan adalah dengan presentasi dan pelatihan secara daring dan luring. Keterlibatan dan kepuasan peserta diukur melalui interaksi langsung dan hasilnya dianalisis secara deskriptif. Hasil pengabdian menunjukkan bahwa semua peternak lebih antusias mengikuti kegiatan secara luring. Semua peserta yang hadir dalam pelatihan telah dapat mengoperasionalkan mesin penghalus kotoran ternak. Berdasar hasil kegiatan ini, disimpulkan bahwa pelatihan penggunaan mesin penghalus kotoran ternak telah dapat diadopsi oleh peternak. Abstract

Abstract The potential of seaweed as a functional food ingredient has not been explored. Several studies, the compotition of seaweed can prevent stress in animals such as sedative mechanisms. The sedative effect was decreased of blood pressure. Pregnancy can be a stressor in animals and cause anemia physiologically. This study aims to determine the effect of seaweed extract on the profile hemoglobin and erythrocytes of pregnant mice. In this study used 10 pregnant rats divided into 2 groups, group 1 (control) and group II (treatment with 450 mg/BW extract Sargassum sp). Calculation of erythrocytes, hemoglobin, and thrombocyte measurements were carried out in day 0, 7, and 14. The results showed that there was an increased thrombocyte count in the treatment group even though in both groups there was a decreased the number of erythrocytes and hemoglobin. The results of statistical analysis showed a significant difference (p <0.05) of thrombocyte counts between the control and treatment groups. Based on the results of the study it was found that administration of Sargassum sp. during pregnancy can prevent thrombocytopenia in pregnant rat. Beberapa penelitian telah menyebutkan kandungan rumput laut dapat mencegah stress pada hewan seperti mekanisme sedativa. Efek sedativa yang sering muncul adalah penurunan tekanan darah. Kebuntingan dapat merupakan stressor pada hewan dan secara fisiologi akan menyebabkan anemia. Penelitian ini bertujuan untuk mengetahui efek pemberian ekstrak rumput laut pada gambaran hematologi tikus bunting. Penelitian ini menggunakan 10 ekor tikus bunting yang dibagi menjadi 2 kelompok yaitu kelompok 1 (kontrol) dan kelompok II (perlakuan ektrak rumput laut Sargassum sp 450 mg/kg BB). Pengambilan sampel dilakukan pada hari ke 0 (sebelum kebuntingan), hari ke 7 dan hari ke 14 (akhir kebuntingan). Pengukuran hematologi sampel meliputi perhitungan eritrosit, pengukuran hemoglobin serta trombosit. Hasil penelitian menunjukkan bahwa terjadi peningkatan jumlah trombosit pada kelompok perlakuan meskipun pada kedua kelompok terjadi penurunan jumlah eritrosit dan hemoglobin. Hasil analisis statistik ada perbedaan yang signifikan (P<0,05) jumlah trombosit antara kelompok kontrol dengan perlakuan. Berdasarkan hasil penelitian didapatkan bahwa pemberian ekstrak Sargassum sp selama kebuntingan dapat mencegah terjadinya trombositopenia pada tikus bunting. Abstract

Abstract Day Old Chicken is a living commodity that is widely distributed between islands throughout Indonesia. Transport trip within two to 24 hours, depending on the distance of the enclosure to be addressed. During the Day-Old Chicken journey, a variety of stresses result in stress and also end in death. Strain Day-Old Chicken has different endurance. Physiological stress parameters in poultry by calculating the ratio of Heterophil: Lymphocytes (H: L). This study aims to determine the difference in stress between Day-Old Chicken strain Cobb and Java Super passed through. Research carried out using Day-Old Chicken strain Cobb as many as 100 chicken/box and Java Super 100 chicken /box. The transportation process from Yogyakarta airport (loading) to Makassar airport (landing). Furthermore, Day-Old Chicken was taken four animals randomly before and after it was passed for blood smear preparations. Blood smear preparations with Giemza staining to calculate the H: L ratio. Analysis of H: L ratio data as a comparison of body resistance to stress between Day-Old Chicken strain Cobb and Java Super using Two Way Anova with a 95% confidence level. The results showed that there was no interaction (P> 0.05) between location (loading-landing) and Day-Old Chicken strain different from H: L ratio. The difference in the Day-Old Chicken strain affects the H: L ratio (P <0.05) with the average value of the H: L ratio of Day-Old Chicken strain Java Super higher than the Cobb. The conclusion is that the difference in Day-Old Chicken lines that experience transportation using airplanes affects the body's resistance to stress. Perjalanan pengangkutan dalam waktu dua sampai 24 jam, tergantung jarak kandang yang akan dituju. Rasio Heterofil : Limfosit (H:L) dapat digunakan sebagai parameter awal adanya stress transportasi. Penelitian ini bertujuan untuk mengetahui rasio H:L serta tingkat kematian Day Old Chicken galur Cobb dan galur Jawa Super yang ditransportasikan dari Yogyakarta ke Makassar. Penelitian menggunakan Day Old Chicken galur Cobb 100 ekor/boks dan galur Jawa Super 100 ekor/boks. Proses transportasi dari bandara Yogyakarta (loading) menuju bandara Makassar (landing). Day Old Chicken diambil empat ekor secara acak sebelum dan sesudah ditransportasikan untuk perhitungan rasio H:L. Analisis data rasio H:L sebagai perbandingan ketahanan tubuh terhadap stres antara Day Old Chicken galur Cobb dan galur Jawa Super menggunakan Two Way Anova. Hasil penelitian menunjukkan bahwa tidak ada interaksi (P > 0,05) antara lokasi (loading-landing) dan galur Day Old Chicken berbeda terhadap rasio H:L. Perbedaan galur Day Old Chicken mempengaruhi rasio H:L (P < 0,05) dengan nilai rata-rata rasio H:L Day Old Chicken galur Super Jawa lebih tinggi dibandingkan galur Cobb. Kesimpulan bahwa perbedaan galur Day Old Chicken yang mengalami transportasi menggunakan pesawat terbang mempengaruhi ketahanan tubuh terhadap stres. Tahapan tahun ni menitikberatakan pada upaya peningkatan keterampilan budi daya kambing pada masyarakat Kemiri, kecamatan Tanjungsari, Kabupaten Gunungkidul. Kegiatan dilakukan dengan inisiasi budidaya kambing kacang dan konsultatif. Abstract

Anadara clam shell contains high Zinc. This microelement is recognised as acting natural aromatase blocker (NAB) that capable of boosting testosterone level. The aim of this study was to evaluates the testosterone level of Canaries following Anadara nodifera administration. Adult male Canaries (n = 27) were group into 3 with 9 repetition of each i.e T0 [control, was given by Carboxymethyl Cellulose Sodium (Na-CMC)], T1 (Zinc Sulphate 0.013 mg/30 g BW), and T2 (NAB 0.3 mg/30 g BW). The T2 treatment was NAB or shell powder of Anadara nodifera which was diluted 1:1 in Na-CMC. The birds were treated orally for 21 days. The complete blood cell count was performed in a haemo analyser except the erythrocyte was calculated manually in the Improved Neubauer counting chamber. Testosterone level was measured in ELISA. Subsequently gen CYP19 aromatase in syrinx, brain and testis were identified employing IHC and descriptively analysed. The result indicated significant elevated level of testosterone (p<0.05). The expression of CYP19 aromatase receptor was depressed in syrinx, brain and testis. However there was no significant changes on the blood profile (p>0.05). To conclude, Anadara nodifera at 0.3 mg/30 g BW was capable of acting as NAB to promote testosterone level. This was confirmed by the reduction of CYP19 aromatase gene receptors in syrinx, brain and testis. However NAB treatment was not significantly affecting routine blood profile.

Abstract

Oyster is one of seafood produced in waters, especially in eastern Indonesia. During this time, oyster is known as aphrodisiac agents, an agent that can increase sexual appetite because it can remind testosterone levels and very high nutrient content of zinc, vitamin A iron, calcium, and selenium are also Vitamins A and Vitamin B12. Therefore, the oyster is known as the mineral's potent testosterone-boosting abilities (Matsuda et al., 2003). Zinc is also very important, it can be used as a stimulant of enzymes, hormones and the immune system.Due to the abundance of oysters in Indonesia, only meat of oyster is consumed, while the existing shells are never used to consume and just thrown away. Therefore, a research plan will be made on the use of oyster shell waste, as a precursor of testosterone in Rat (Rattus norvegicus). Since many kinds of oyster, it would be compared among three kinds of oyster shell namely Kerang darah (Anadara granosa), Kerang hijau (Perna viridis) and Kerang keong (Telescopium telescopium).Based on function, Zn inhibits aromatase enzyme which convert estradiol to testosterone as a consequence, testosterone levels will increase (Michell et al. 2012).The purpose of this study were to determine the highest content of Zn among the three types of oyster shell; 2. To measure testosterone levels in rat after given shell powder containing highest Zn.

Abstract

Accumulation lipid peroxidation can lead to the formation of malondialdehyde to generatefree radicals. Antioxidant will minimize free radical related health problems, such as naturalantioxidant from meat of kepel (Stelechocarpus burahol) (Blume) Hook. F. & Thompson) fruits.Antioxidant activities of methanol extracts and acetate ethyl fractions meat of kepel fruits calculatedprofile of malondialdehyde concentration (ng/mL) in CCl4 induced acute liver toxicity in femaleSprague Dawley rats using ELISA competitively method. All of groups induced CCl4 i.p. at 0 h, exceptuntreated and solvent control group, then vitamin C , methanol extracts and acetate ethyl fractionsp.o. once daily for 72 h. Identification of secondary metabolite was using phytochemical screeningand Thin Layer Chromatography (TLC). Liver weighed and calculated relative organ weight (ROW)of liver, then all of data analyzed using statistic tests, One Way ANOVA and Kruskall Wallis. Thisstudy showed methanol extracts can decrease MDA concentration, ALT and AST were more potent(significantly p<0.05) than acetate ethyl fractions and vitamin C. The TLC analysis identified methanolextracts have same polarity with quercetin (Rf = 0,26) and acetate ethyl fractions showed otherflavonoid class, except rutin and quercetin. ROW of acute liver toxicity in rats increased significantlyin CCl4 and vitamin C groups.

Abstract

Degenerative diseases are mostly caused by high free radical concentrations in the body. Kepel fruit(Stelechocarpus burahol) is known to contain flavonoid compounds, a class of compounds that has free radicalscavenging activity in the body and could affect the activity of antioxidant enzymes. There have been reportsthat methanolic extract and ethyl acetate fractions of Kepel fruit have the lowest IC50 values, compared toextracts or fractions which are made using other organic solvents, on in vitro assays so that flavonoids aresuspected as responsible compounds to the free radical scavenging activity. This study aimed to identify thepresence of flavonoid content in methanolic extract and the fraction of ethyl acetate of Kepel fruit and also itseffect on rat alpha Glutathione S-Transferase (α-GST) enzyme concentration for the function to prevent hepaticcell damage due to carbon tetrachloride exposure. The identification of flavonoids content used a series ofcolor chemical reaction tests and thin layer chromatography that used silica gel 60F254 as the solid phase andmixture of ethyl acetate, formic acid, glacial acetic acid, and water (100: 11 : 11: 20 v/v) as the mobile phase.This study used 30 Sprague-Dawley female rats (age 2-3 months) that were grouped into 5 groups as solventcontrol (CMC-Na) group, negative control (CCl4) group, positive control (vitamin C) group, methanolic extractgroup, and ethyl acetate fraction group. All groups were induced with carbon tetrachloride (CCl4) compoundsexcept for the CMC-Na solvent control group. Blood sampling at 0 h, 24 h, and 72 h, while liver sampling at 24h and 72 h, with 3 samples each group. The analysis of rat α-GST enzyme concentrations used theEnzyme-linked Immunosorbent Assay (ELISA) method. Based on the phytochemical test, the methanolicextract and the ethyl acetate fraction of Kepel fruit have been identified contains flavonoids compound. Basedon the in vivo study, the positive control group's α-GST enzyme concentration was decreased, while thenegative control group's α-GST enzyme concentration was increased. Administration of methanolic extractand ethyl acetate fraction also decreased both liver and blood rat α-GST enzyme concentrations, althoughwithout significant correlation, and still could prevent the hepatic cell damage due to carbon tetrachlorideexposure.

Abstract

Thyroxine hormone (T4) was one of the hormones related to metabolism rate. Animal transportation could be stressor interfering metabolism. Sargassum crasifoium was one of brown algae species that contained some important nutrients and also flavonoids. The study aimed at finding out the effect of the Sargassum on the decrase in thyroxine hormone and body weight loss. It used 9 Javanese Randu goats that were classified into 3 treatment groups. Group I served as control, Group II was treated with 450 mg/kg BW of Sargassum estract and Group III was treated with vitamin B1. All of the goats were transported for 12 hours using pickup truck at the speed of 60 km/hour. Blood samples were drawn before the transportation, during loading, 6 hours after the transportation and at the end of the transportation, while the body weight of the goats was weighed before and after the transportation. The results of the study showed that the T4 hormone decreased in the Group I and increased in the Group III during the transportation, while it was stable in the Group II. The biggest percentage of the body weight loss took place to the Group III (0.13%), while the smallest percentage took place to the Group II (0.063%). Statistical analysis showed that the Sargassum extract did not have any significant effect (p>0.05) on both the thyroxine hormone and the body weight loss during the transportation. Based on the results of the study it could be concluded that the Sargassum extract could stabilize the thyroxine hormone during the transportation so that it could prevent the body weight loss
