Abstract

The research aimed to investigate the free radical scavenging activity of ethyl acetate fraction of roselle calyces infusion fermented by Lactobacillus casei strain Shirota against free radical agent, DPPH (2,2-di-phenyl-2-picrylhydrazyl). In sequence, the roselle calyces were extracted by infusion method with concentration at 5% w/v. The infusion was fermented using Lactobacillus casei strain Shirota during 3x24 hours at 37⁰C. As regard the fermentation process, the result of that was extracted using ethyl acetate. Free radical scavenging activity was measured using DPPH method with unfermented-ethyl acetate fraction as control negative. As the result, both fermented and unfermented-ethyl acetate fraction exhibited free radical scavenging activity at 370.68 ppm and 685.49 ppm, respectively. Abstract

Isoflavon merupakan komponen polifenol utama dalam kacang kedelai. Isoflavon kedelai utamanya dalam bentuk glikosida (Genistin dan Daidzin) dibanding bentuk aglikonnya (Genistein dan Daidzein). Tujuan penelitian ini adalah untuk mengetahui pengaruh metode ekstraksi terhadap kandungan isoflavonnya (Genistein, Daidzein, Genistin, Daidzin). Isoflavon kedelai diekstraksi dengan beberapa cara, yaitu: 1) kacang kedelai tanpa kulit ari diblender dengan air panas, 2). Kacang kedelai tanpa kulit ari diblender dengan etanol 70 % , 3) kacang kedelai utuh diekstraksi secara maserasi dengan aseton 70 %, dan 4) kacang kedelai utuh diekstraksi secara maserasi dengan metanol.. Perbandingan kacang kedelai dan cairan penyari 1:10. Ekstrak yang diperoleh dianalisis kadar isoflavonnya menggunakan kromatografi cair kecepatan tinggi. Hasil penelitian menunjukkan total isoflavon tertinggi (dihitung sebagai total genistein, daidzein, genistin, daidzin) ada pada kacang kedelai utuh yang diekstraksi dengan metanol.Kata Kunci : Metode Ekstraksi, Isoflavon, Kedelai (Glycine max L.), Ultra Fast Liquid Chromatography

Abstract

The emergence of antibiotic-resistant Pseudomonas aeruginosa is one of main health issues in global communities. To overcome such threat, the discovery of novel antibacterial agents is indispensable. This study aimed to evaluate the in vivo antipseudomonal activity of Ulva reticulata extract in Drosophila model of infection. Ethanolic extract of Ulva reticulata was prepared using maceration method and the extract was subsequently assessed for its in vivo antibacterial effect against P. aeruginosa using survival assay, bacterial load enumeration, and gene expression analysis in the wildtype Drosophila. Survival and bacterial load analysis were further performed in a similar fashion on the mutant flies devoid of component responsible in the activation of immune responses against P. aeruginosa. Decline in the survival of infected host accompanied by augmentation of bacterial proliferation was documented in the wildtype Drosophila upon infection with P. aeruginosa. These phenotypic events were further amplified in immune-deficient mutant Drosophila. Nevertheless, improvement of host survivorship and reduction of bacterial burden were demonstrated in both wildtype or immune-deficient mutant flies upon treatment with Ulva reticulata extract after bacterial challenge. Our data demonstrated in vivo antipseudomonal activity of Ulva reticulata extract and thus provide a valuable information about its future potential for health promotion.

Abstract

The rise of antibiotic-resistant Staphylococcus aureus-related clinical cases is an alarming chronicle for global communities. This research was conducted to examine the antistaphylococcal effect of roselle (Hibiscus sabdariffa L.) calyx fractions in the Drosophila model. In the infection experiment, wild-type and immunodeficient Drosophila were pricked with S. aureus and subsequently subjected to fly survivorship and colony-forming assays, in the presence or absence of roselle calyx fractions. The Involvement of immune stimulation in the host antibacterial protection was assessed in vitro using cell-based luciferase reporter assay and in vivo using RT-qPCR analysis on adult flies. A declining rate of fly survivorship and augmentation of bacterial growth were observable in S. aureus-infected wild-type flies but subject to improvement in the presence of roselle calyx fractions. Cell-based analysis revealed the absence of host immune stimulation via Drosophila Toll pathway and roselle calyx fractions-treated immune-deficient flies lacking for components in the Toll pathway were protected from infection-induced early death phenotype and harbored reduced number of S. aureus colonies. Overall, our data confirmed the in vivo anti-staphylococcal activity of roselle calyx fractions in Drosophila infection model and such protective signature was devoid of host immune stimulation.

Abstract

Saat ini pemerintah tengah menggalakan penggunaan bahan tanaman untuk pemeliharaan kesehatan. Untuk mencapai tujuan tersebut salah satu cara adalah melalui ibu-ibu kader PKK.  Kelurahan Bontolebang merupakan salah satu kelurahan dari kecamatan Galesong Utara yang diprogramkan oleh pemda Kabupaten Takalar sebagai desa sehat. Kendala yang dihadapi oleh khalayak sasaran adalah masih kurangnya pengetahuan tentang manfaat tanaman obat  dan cara  mengolahnya  menjadi sediaan jamu sederhana. Metode yang digunakan adalah: Penyuluhan tentang tanaman obat dan cara mengolahnya serta pelatihan pembuatan sediaan-sediaan jamu sederhana.    Kegiatan pengabdian telah dilaksanakan tanggal 14 September 2019 diikuti lebih dari  50 orang ibu-ibu kader PKK Kelurahan Bontolebang  serta ibu-ibu PKK dari Kecamatan Galesong utara lainnya.    Hasil pelaksanaan memberikan nilai tambah pengetahuan dari khalayak sasaran tentang pengolahan jamu sederhana dan alat-alat peraga utama yang digunakan disumbangkan ke khalayak sasaran

Abstract

Penyakit degeneratif seperti  hipertensi, hiperurisemia,  hiperkolesterolemia, and diabetes mellitus, saat ini masih termasuk prevalensi tinggi  di Indonesia, di usia produktif maupun non produktif. Tujuan tulisan ini ingin mengulas potensi kelopak bunga rosella(Hibiscus sabdariffa L.) sebagai minuman kesehatan  alternatif pencegah penyakit-penyakit degenerative. Metode yang digunakan dalam tulisan ini adalah menyaring beberapa penelitian terkait kelopak bunga rosella melalui penelusuran artikel di google scholar dan membuat protipe minuman berbasis kelopak bunga rosella. Hasil penelusuran diperoleh  beberapa penelitian preklinik dan klinik menunjukkan bahwa ekstrak kelopak bunga rosella dapat menurunkan hipertensi, hiperurisemia,  hiperkolesterolemia, dan diabetes.  Kandungan bioaktif dalam  kelopak bunga rosella yang memiliki aktivitas antioksidan, dan secara in vivo dan klinik mampu menurunkan  tekanan darah, asam urat,  kolesterol darah dan  glukosa darah adalah utamanya senyawa antosianin, flavonoid, dan asam fenolat. Hasil  penerapan teknologi yang diajukan dalam pembuatan minuman rosella ini adalah dengan dibuat dalam bentuk serbuk/granul instant  dari ekstrak air kelopak bunga rosella dan sirup  rosella. Diharapkan dengan adanya produk minuman kesehatan ini, dapat menambah inovasi produk berbasis herbal.

Abstract

Bligo fruit (Benincasa hispida Thunb) is one of medicinal plants that used to cure typhoid fever which caused by Salmonella typhi bacteria. The aim of this research is to find ont the effect of ethanol concentration as a solvent from Bligo fruit (Benincasa hispida Thunb) extracts as an antibacterial, especially to Salmonella typhi. This simplicia was being extracted with 96% ethanol, 70% ethanol, and 50% ethanol by using maceration method for 3x24 hours. The antibacterial activity test was done by diffusion of agar method to the MHA medium by obserring the resistor zone which formed from 96% ethanol, 70% ethanol and 50% ethanol. The diametres that have been obtained are 6,815 mm for 96% ethanol, 25,223 mm for 70% ethanol while for 50% ethanol there is no resisten zone can be seen. From the result canbe conclude that the best concentration of ethanol that has a great potencial as resistor to Salmonella typhi bacteria is the Bligo fruit (Benincasa hispida Thunb) extracts of 70% ethanol.Key words: Bligo fruit (Benincasa hispida Thunb), Antibacterial, Salmonella typhi.

Abstract

Pembuatan pasta gigi ini dilatar belakangi oleh keinginan peneliti agar memudahkan masyarakat menggunakan bahan alami katekin teh hijau yang hasilnya langsung di uji efektifitasnya terhadap daya hambat bakteri kariogenik. Penelitian ini bertujuan untuk melakukan pembuatan prodak pasta gigi katekin teh hijau dengan formula yang sesuai dengan hasil uji dan melakukan pengujian daya hambat pasta gigi katekin teh hijau terhadap bakteri streptococcus mutans dan lactobacillus acidophilus. Pembuatan pasta gigi katekin teh hijau didahului dengan pembuatan ekstrak katekin dan uji konsentrasi hambat minimal (KHM) dan uji konsentrasi bunuh minimal (KBH) menggunakan metode dilusi cair.Peracikan formula dan pembuatan pasta gigi katekin teh hijau dilakukan sesuai hasil uji KHM (MIC). Pasta gigi katekin teh hijau ini dilakukan pengukuran kadar total flavonoid dan total polifenol, selanjutnya pengukuran uji daya hambat pasta gigi katekin teh hijau dilakukan dengan metode difusi menggunakan well (sumuran) sebagai reservoar sampel uji terhadap bakteri streptococus mutans dan lactobacilus acidophilus. Hasil diperoleh prodak pasta gigi katekin teh hijau dengan kandungan kadar flavonoid 77% dan kadar polifenol 41%, sedangkan hasil uji diameter hambatan pasta gigi katekin teh hijau terhadap bakteri streptococcus mutans 17,2 mm dan bakteri lactobacillus acidophilus 19,6 mm. Hasil ini menunjukkan bahwa prodak katekin teh hijau ini sangat efektif digunakan sehari-hari sebagai perawatan gigi dan juga sebagai antibakteri kariogenik. Making Green Tea Catechin Toothpaste and Inhibition Test Against Streptococcus Mutans and Acidophilus Lactobaccilus. The manufacture of this toothpaste is motivated by the desire of researchers to facilitate the public to use natural ingredients of green tea catechins whose result effectiveness against cariogenic bacteria was directly tested inhibition. This study is to find the right formula composition for greentea catechins toothpaste based on the result of the inhibition properties of greentea cathecins against tostreptococcus mutans dan lactobacillus acidophilus. The manufacture of greentea catechins toothpaste was after making catechins extracts and testing minimal inhibition concentration (MIC) and minimal bactericidal concentration (MBC) using liquid dilution method. The toothpaste was then made according to the result of MIC. The toothpaste’s flavonoid and polyphenol total contain was measured; then the inhibition property of the toothpaste to was measured using diffusion method with wells as the sample reservoar for streptococus mutans dan lactobacilus acidophilus. It is found that the toothpaste contains 77% flavonoid and 41% polyphenol, and the diameter of the inhibition property of green tea catechyn is 17.2 mm for streptococus mutans and 19.6 mm for lactobacilus acidophilus. This result shows that greentea catechyn toothpaste is very effective for daily oral care which is antibacterial.

Abstract

More than half of the Indonesian people use herbal medicines to maintain their health, including ones that are served immediately after preparation or also known as jamu gendong. This plant-based medicine is prepared traditionally by boiling using different ratios of leaves to water and heating time. The study was designed to determine the heating time and the fresh leaf-to-water ratio that exhibited the highest antioxidant and antifungal activities against Candida albicans. Fresh betel leaves were extracted by boiling and infusion methods, with the ratio of 1:5 and 1:10 and the heating time of 5 and 10 minutes for boiling and 15 minutes for infusion. The excess water in the resulting extract was removed using a freeze dryer, then the total phenolic content was determined by Folin-Ciocalteu spectrophotometry. Meanwhile, the antioxidant and antifungal activities were tested by the DPPH method and microdilution, respectively. The results showed that betel leaf extract with the highest yield and polyphenol content was obtained by infusion with the sample-to-water ratio of 1:10, and this was positively correlated with antioxidant activity against DPPH (IC50= 17.4 ppm) and antifungal effect against Candida albicans (MIC= 0.5%).

Abstract

The emergence of pathogenic bacteria with β-lactam antibiotics-resistant profile has threatened the continued use of such antibiotics in the future. This research was conducted to investigate the antimicrobial activity of green tea ethanol extract (GTE) and its ability to improve the antibacterial action of several β-lactam antibiotics against Enterobacter cloacae clinical isolates. The simplicia of green tea was extracted by sonication for 30 minutes using 50% ethanol solvent, and the total phenolic content of the GTE was subsequently determined. Next, the GTE used in testing against two clinical isolates of E. cloacae was obtained from the Pathology Laboratory of Wahidin Sudiro Husodo Hospital in Makassar. The sensitivity of bacteria to GTE was confirmed using the agar diffusion method, the Vitek® rapid method, and the double-disk synergistic test. Antibacterial activity of antibiotics, GTE, and combination of antibiotics with GTE were then tested against clinical isolates of E. cloacae using the checkerboard microdilution assay. The results showed that GTE contained 51.64 ± 0.21 % measured as gallic acid equivalent and 37.95 + 5.17 % Epigallocatechin gallate (EGCG). The confirmatory test results indicated that one clinical isolate of E. cloacae (code 13/04) was resistant to amoxicillin-clavulanate but did not produce an extended-spectrum β-lactamase (ESBL). Another clinical E. cloacae isolate (code 275B/06) was indicated to produce ESBL and demonstrated to yield resistance to amoxicillin-clavulanate and cefotaxime. The minimum inhibitory concentration of GTE against the two clinical isolates of E. cloacae was >8000 ppm (8 mg/ml). In conclusion, GTE could not increase the antibacterial activity of amoxicillin and cefotaxime, but it was sufficient to improve the activity of imipenem against the tested isolates of E. cloacae.
