Abstract

This research aimed to determine the optimal doses of honey addition on the sperm dilution to optimize spermatozoa motility, fertilization and hatchability of Patin Siam  eggs (Pangasius hypophthalmus).  The experimental design used was Complete Random Design (RAL).  Diluent solution consisted of a physiological NaCl 0.9% and honey.  Treatment doses of honey in aqueous diluent sperm tested as treatments were A (0 mL), B (0.2 mL), C (0.4 mL) and D (0.6 mL), each with three replications.  Observations made included spermatozoa motility, fertilization and eggs hatchability.  The research results showed that the addition of honey had a significant influence on motility,  fertilization, and eggs hatchability. This study concluded that the addition of honey in sperm diluent solution with a dose of 0.6 mL of honey in 99.4 mL of NaCl could increase the spermatozoa motility and  fertilization of Patin siam. Abstract

The aim of the experiment was to find out the survival rate of the sea cucumber (Stichopus hermanni) seeds generated from transverse-fission method, and cultured in the waters of Talengen gulf, Sangihe Islands Region.  Twenty five specimens of sea cucumber were collected around the waters of Talengen gulf and Pananuareng bay, the Region of Sangihe Islands.  The specimens were transported by boat to the field-research area of Nusa Utara Polytechnic, as the site of the experiment.  The specimens were kept individually in 25 plastic mesh cages, where each cage measured 40cmx40cmx40cm.    The individual body length of the specimens was measured  and then  was transversely  cut at the middle of the body antero-posteriorly.  The procedure was conducted underwater to prevent  expelling of the visceral organs.  25 individuals were applied transverse-fission,  hence 50 new individuals were generated, which were composed of 25 Anterior individuals (A) and 25 Posterior-individuals (P).   After transverse fission,  the Anterior- individuals (A) and Posterior-individuals (P), generated from the same individual were put back to their previous cage.  The new individuals were cautiously controlled, and at the interval of 6 hours observation on the sealing process of the open wound and the regenerating process were carried out. The result showed that the survival rate of the sea cucumbers generated from transverse-fission method, was as high as 70.83%.  The Posterior-individuals (P) had higher survival rate (75.66%) than Anterior-individuals (66%).   The result also showed that A and P individuals generated from larger individuals had lower survival rate than those from smaller individuals.   Small individuals sizing 6-14cm had highest survival rate, which was 100%, while larger individuals sizing 14-22cm and 22-30cm had lower survival rate which were 85.5%  and  27% respectively.     The sealing of the open wounded took averagely 4 days, while the process of the regeneration took around 4 weeks. Keywords: sea-cucumber, fission, survival-rate

Abstract

The study aimed to identify species of reef fishes, to obtain indexes of diversity  and dominance, eveness and to analyze the comparison of communities between the two study sites by Sorensen similarity index. Data was gathered in-situ with underwater recordings at a depth of 6 meters assisted by diving apparatus, and using fish visual census method. Subsequent data were identified using  fish identifikaction guide and WoRMS (Word Register of Marine Science) online identification applications. The number of species and specimens of reef fishes from each study site after bein identified were then analyzed  the diversity, dominance and similarity of the community between the two study sites. The coral reef fishes identification results that obtained from each study site  were:  20 families, 86 spesies and 362 specimen at  Malalayang beach, and 15 families, 55 spesies and 217 specimen at Meras beach. The value of the calculated index o community structure obtained by H’ 3.42 (Malalayang) and 2.91 (meras) has been shown the ecological condition of coral reef ecosystem which is still stable even though diversity is moderate, but inversely with low dominance value (D=0.08). The coral reef fishes in both study sites were very diverse and there was no dominant species. Eveness of community between two study sites high.The result of community similarity analysis using Sorensen Index was 49 % (<50 %) indicates that there was no community similarity of coral reef fishes between Malalayang Beach and Meras Beach which was allegedly caused by natural factors and or anthropogenic one in both research sites.Keywords : coral reef fishes community, diversity, dominance, community similarity

Abstract

The objective of research was to isolate the probiotic bacteria from intestine of carp.  Isolation was performed by weighing 1 g of intestine, crushed and inserted in a reaction tube containing 9 mL of NaCl then centrifuged. Supernatant was taken and spread over the agar medium by scatter method. The bacteria-stocked medium was then incubated in an incubator at 28°C for 24-48 hours. The growing colonies were re-isolated on the agar medium to obtain pure colonies. The bacteria were then identified by morphological observation, resulting pigment and gram staining. Five nile tilapia (sized 8 – 10 cm) were used in pathogenicity test and placed in the aquarium. Fish were injected with 0.2 ml of Lactobacillus solution containing 1x107 CFU/mL via intraperitoneal injection. The results showed that the bacteria Lactobacillus sp had rod-shaped characteristics, white pigment and gram positive and  not pathogenic, which means it can be used as probiotic for aquaculture.Keywords : Lactobacillus sp, carp, probiotic, pathogenicity test

Abstract

Generally, this study was intended to examine the problems or implementation of turtle conservation in Talise, Gangga and Bangka Islands and specifically with a number of objectives to: know the introduction of sea turtle, to know sea turtle fishing and turtle exploitation, to know the knowledge of the people about turtle as a protected animal. Survey results on Talise Island, Gangga and Bangka showed that people who had seen turtles directly: Talise Island 94%, Gangga 58%, and Bangka 92%. Local residents of Talise, Gangga and Bangka Islands know well the turtles even see directly in their habitat and spawning time. In Talise, Gangga and Bangka Islands,  turtles are still commonly caught, traded and consumed by local people.  People's awareness about turtles as protected animals is lacking.  Keywords: Turtle, Talise Island, Gangga and Bangka

Abstract

The purpose of this study was to analyze the use of cattle dung on the growth and biomass of silk worms. The experiment was conducted at Freshwater Aquaculture Center (BPBAT) Tatelu, Tatelu Village, Dimembe Subdistrict, North Minahasa Regency, North Sulawesi Province, about 35 km from Manado city. The media used was pig manure, chicken manure, cow dung, and fine mud added with EM4. The method used was complete randomized design (RAL) with 4 treatments including A. 500 g of pig manure, 500 g of chicken manure 500 g of cow dung, and 500 g of fine mud; B. 600 g of pig manure, 400 g of chicken manure 500 g of cow dung, 500 g of fine mud; C. 700 g of pig manure, 300 g of chicken manure, 500 g of cow dung, 500 g of fine mud; and K (control) was 2000 g of fine mudinand, each with 3 replications. Water quality parameters measured during the study were temperature, pH, DO, nitrate and nitrite. The results showed that there was a very significant effect on growth but not for the value of silk worm biomass. The highest growth was found in treatment A that was 38 g, while for the highest biomass also in treatment A namely 1.5 g / cm3. Water quality parameters during the study were 24.3-25.4° C, pH 7,1-7,3; DO 2.7-5,7 ppm; nitrate 1.1-1.4 ppm; nitrite 0.011-0.201 ppm.Keywords:  Cattle dung, growth, biomass, Tubifex sp., aquaculture

Abstract

The objective of this study was to determine the minimum concentration of extract in inhibiting motile motile aeromonad septicaemia. The material used was the stems and leaves of the plant P. grandiflora as much as 10 g. The crushed simplicia was then macerated with aquades with a ratio of 1:10,  then filtered with filter paper and, put in an Erlenmeyer, and evaporated in an incubator at temperature of 30°C. The sensitivity test was performed by placing a paper disc containing extract on the agar medium that had been scratched with bacterial colonies. Clear zone formed around the paper disc was compared with standard classification table for clarifying the inhibition level.  The results showed that stem and leaf extract had inhibition zone against Aeromonas hydrophila bacteria that caused motile aeromonad septicaemia. The best result was obtain at concentration of extract 3.25 mg / L) with an average diameter of 22.67 mm and for leaf extract had a moderate to strong zone for all treatments. These proved that stem and leaf extract of P. grandiflora plant had antibacterial activity and can be used as control of MAS disease caused by A. hydrophila bacteria. Abstract

The aim of this research was to study the effect of Binahong extract on the growth of Nile tilapia. A number of 225 fish measuring 10-12 cm in length with an average weight of 15 g was obtained from Fishery Development Center and Training, Tateli. The fish were put  in oxygenated plastic bags and transported to Laboratory of Aquaculture Technology. The fish was kept in aquarium equipped with an aerator with a density of 15 fish per aquaria. The test material used was the stem of Binahong plant stem, macerated with alcohol 70% for 3 days. The extract was then filtered with whatman filter paper and extract was concentrated using a Rotary Evaporator. The extract was further diluted to obtain  concentrations of 12.5, 25, 50, and 100 mg / mL Binahong  extract as treatment was given to the fish after mixed first into the pellet. The fish was fed with treatment feed for 30 days. Control feed was not supplemented with binahong extract. Growth data was measured at day 30 after treatment. The results showed that the extract of binahong had significant effect on fish growth (p <0.01) with the best result achieved at concentration of 100 mg/mL. In conclusion, supplementation of  binahong extract into fish feed could improve growth of  fish in which the best growth was obtained in fish fed diet containing 100 mg/mL of binahong extract.Keywords: Medicinal plants, tilapia, Anredera cordifolia, growth, maceration

Abstract

The objectives of this research were to study the profile of freshwater fish culture in Dimembe District, Analyse the prospects for the development of aquaculture in Dimembe District,  determine cultivation development strategy that is suitable with the condition and potential of Dimembe District. The method used in this research was descriptive using data in the last 5 years. Data were analysed using quantitative and qualitative descriptive analysis and SWOT analysis. Fish farmers in Dimembe District amounted to 226 people and the location of water quality supports the business of freshwater fish cultivation of indigo and carp in Dimembe District. Internal factors included business location, water quality, local government support, existance of BPBAT. Factors weaknesses were availability of less deed, and lack of human resources. External factors affecting the development of fish farming in Dimembe District consist of opportunity factor that were high fish selling value, high market share, big business opportunity, government policy. Treat factors: theft and security. Strategy steps for developing fish farming business in Dimembe District could be conducted by increasing the production of tilapia fish and goldfish, providing training on freshwater fishery cultivation and maintainng the quality of fish farming products, making agreement among farmers in maintaining the security of pond cultivation. Keywords: Dimembe District, freshwater cultivation, fish farmers, SWOT analysis

Abstract

The importance of seagrasses in terms of ecological and economic benefits are well-known. This paper describes the status of research on seagrasses conducted so far, with relevance to the North Sulawesi region. Most of the studies done in Indonesia, especially in North Sulawesi, have focused on species richness and biodiversity as well as basic biological questions such as biomass and morphometric differences of certain seagrass species between sampling localities. While several studies have been conducted, there is a general need for Indonesian marine scientists to expand research areas related to food-web dynamics and inter-connectivity of major fishery and nursery habitats (mangroves-seagrass-coral reefs) with the aid of modern tools (e.g. molecular approach).   Keywords: benefits, biology, ecology, economic, seagrass
