Abstract

ABSTRAK Biji Nigella sativa telah banyak diketahui khasiat farmakologisnya. Salah satu aktivitas farmakologis senyawa N. sativa adalah sebagai antimikroba. Untuk itu penelitian ini dilaksanakan dengan tujuan mengetahui aktivitas antimikroba N. sativa terhadap flora normal saluran pencernakan manusia agar diketahui dosis terapi yang tepat dan tidak membahayakan keseimbangan flora normal saluran pencernakan. Penelitian ini dilaksanakan secara in vitro dan in vivo. Pengujian in vitro aktivitas antimikroba ekstrak biji N. sativa terhadap strain bakteri probiotik usus dilakukan dengan uji difusi cakram dengan perlakuan pemberian ekstrak N. sativa fraksi n-heksan, etil asetat dan etanol konsentrasi 500 mg/ml; 250 mg/ml; 125 mg/ml; 62,5 mg/ml; 31,25 mg/ml; 15,625 mg/ml. Pengujian secara in vivo dilakukan dengan mencit dengan pemberian kombinasi antibiotik sefadroxil 1,3mg/20g BB mencit dan adjuvan berupa ekstrak N. sativa 1,3mg/20g BB, 2,6 mg/20g BB, dan 3,9mg/20g BB mencit. Hasil pengujian in vitro ekstrak biji N. sativa fraksi n-heksan memiliki potensi antimikroba tertinggi disusul kemudian fraksi etil asetat dan etanol. Daya antimikroba ekstrak biji N. sativa fraksi n-heksan muncul pada konsentrasi 15,63 mg/ml dan semakin kuat pada konsentrasi yang lebih tinggi. Secara in vivo didapatkan hasil dosis adjuvant ekstrak biji N. sativa 3,9 mg/20g BB memunculkan aktivitas antimikroba pada bakteri probiotik sebagai bagian flora normal di saluran cerna mencit. Kata kunci : N. sativa, antimikroba, bakteri probiotik, in vivo, in vitro ABSTRACT Nigella sativa has a lot of pharmacological activities. One of those is the broad range antimicrobial activity. The aims of this research are to find out the antimicrobial activity against probiotic bacteria as normal flora in gastrointestinal track (GIT), in order to determine the apropriate dose which not harm for GIT normal flora. This research was conducted by in vitro and invivo assay. In vitro assay for antimicrobial acitivity using disc diffusion method with n-hexane, ethyl acetate and ethanol fraction of N. sativa extract. The concentration of each fraction were varied into 500 mg/ml; 250 mg/ml; 125 mg/ml; 62.5 mg/ml, 31.25 mg/ml, 15.625 mg/ml, respectively. In vivo assay using mice (Mus musculus) to analyze the difference of the probiotic bacteria viability in the GIT of treated mice and in that of control mice. N. sativa extract at dose of 1.3 mg/20gBW, 2.6 mg/20gBW, and 3.9 mg/20gBW were used as adjuvant with sefadroxil 1,3 mg/20 g BB. The result showed that the n-hexane extract possessing the highest potency as antimicrobial against probiotic bacteria, wich the minimum inhibitory concentration was 15,625 mg/ml. The in vivo assay shows that the dose of 1.3 mg/20gBW of mice has antimicrobial activity and so do the higher doses. Key words: antimicrobial, probiotic bacteria, N. sativa, in vivo, in vitro

Abstract

Amazake is a tradisional sweet non alcoholic Japanesedrink made from fermented rice which is produced from mix of rice koji and boiled rice. In this process production, amylases produced by the fungus Aspergillus oryzae grown in rice koji hydrolyze starch to dextrin and glucose which is called saccharification. The Objective of this research was to understand the influences on amazake which brewed by varying temperature, pH and other factors that affecting the enzyme effects to qualities. This research design with 6 treatments brewed amazake incubated at 60oC, 75oC, 40oC which then sterilized, fourth treatment was incubated at 60oC without sterilized, fifth treatment was within lactic acid addition and sixth treatment, koji was substituted by enzyme agent. Parameter which observed was enzyme activity, sugar content and also organoleptic test that consist of 7 panelist. The parameters which include were sweetness, umami, sourness and aroma. From this research indicate that amazake product which incubated at 60oC have better quality than the others. Amazake which incubated at 60oC have better sweetness, good taste of umami, litlle sour and have good aroma.

Abstract

ABSTRAK Virus merupakan parasit berukuran mikroskopik, menginfeksi sel organisme biologis, dan hanya dapat bereproduksi dalam material hidup. Penelitian ini dilakukan untuk mengetahui ada tidaknya aktivitas antivirus dari ekstrak etanol daun tapak liman (Elephantopus scaber L.) terhadap Virus Avian influenza beserta profil kromatografi lapis tipisnya. Ektrak dibuat dengan cara maserasi menggunakan cairan penyari etanol 96 %. Uji aktifitas antivirus ekstrak etanol daun tapak liman (Elephantopus scaber L.) menggunakan uji hemaglutinasi, uji hemaglutinasi dilakukan untuk mengetahui daya hambat ekstrak etanol daun tapak liman (Elephantopus scaber L.) terhadap virus Avian influenza. Penelitian ini dianalisis dengan Uji T dengan taraf kepercayaan 95%. Hasil analisis golongan senyawa ekstrak etanol daun tapak liman menggunakan kromatografi lapis tipis (KLT). Hasil uji aktifitas antivirus terhadap virus Avian influenza pada konsentrasi dosis 100µg/mL, 10µg/mL, 1µg/mL menunjukkan daya hambat rata-rata tiap konsentrasi, pada konsentrasi 100µg/ml adalah 96,737%, pada konsentrasi 10µg/ml adalah -700%, dan pada konsentrasi 1µg/ml adalah -775%. Hasil pengujian kromatografi lapis tipis (KLT) menunjukkan bahwa ekstrak etanol daun tapak liman (Elephantophus scaber L) mengandung golongan senyawa flavonoid, seskuiterpen lakton dan steroid. Kata kunci : antivirus, ekstrak etanol, tapak liman, Avian Influenza, lapis tipis. ABSTRACT Virus is the microscopic of measured parasite, to infection the organism cell, and only reproducing in material life. The purpose of this research is to know is there antiviral activity or not from the ethanol extract of tapak liman leaves (Elephantopus scaber L.) to Avian Influenza virus including thin layer chromatography profile. The extract was made by maceration method with using ethanol of 96%. Test antiviral activity of ethanol extract tapak liman leaves (Elephantopus scaber L.) using hemaglutinasi test, the test is `performed to determine hemaglutinasi inhibit the ethanol extract of tapak liman leaves (Elephantopus scaber L.) against Avian influenza virus. This research was analyze with the T test with 95% confidence level. The results of group analyse the ethanol extract of tapak liman leaves (Elephantopus scaber L.) compounds tapak liman using thin layer chromatography (KLT). Antiviral activity of test results against Avian influenza virus at the dose concentration of 100μg/ml, 10μg/ml, 1μg/ml shows the average drag each concentration, the concentration of 100μg/ml is 96,737%%, the concentration of 10μg/ml is -700%, and the concentration of 1μg/ml is -775%. The results of thin layer chromatography test (KLT) showed that the ethanol extract of tapak liman leaves (Elephantopus scaber L.) contains a group of flavonoids compound, seskuiterpen lactone and steroid. Abstract

ABSTRAK Telah dilakukan penelitian tentang Evaluasi Manajemen Pengelolaan Obat di Puskesmas Rawat Inap Kabupaten Purbalingga Berdasarkan Tiga Besar Alokasi Dana Pengadaan Obat. Tujuan dari penelitian ini adalah untuk mengevaluasi manajemen pengelolaan obat di Puskesmas Rawat Inap Kabupaten Purbalingga berdasarkan 3 besar alokasi dana pengadaan obat. Penelitian ini dilakukan menggunakan metode deskriptif dengan pendekatan kualitatif dan didukung dengan data kuantitatif. Sampel yang diambil adalah Puskesmas Kalimanah, Puskesmas Bobotsari dan Puskesmas Rembang. Hasil penelitian menunjukkan bahwa Puskesmas dalam melakukan perencanaan tergolong tidak baik. Stok optimum tidak tercatat pada kartu stok dan masih terdapat permintaan obat diluar DOEN untuk Puskesmas Kalimanah sebesar 84,51%, Puskesmas Bobotsari 79,64% dan Puskesmas Rembang 85,35%. Penerimaan obat sudah dilakukan dengan ketentuan yang berlaku. Penyimpanan Psikotropika belum sesuai ketentuan dan belum menjamin mutu obat. Distribusi obat, pengendalian penggunaan, pencatatan dan pelaporan sudah dilakukan sesuai dengan ketentuan. Kata kunci : Evaluasi, Pengelolaan Obat, Puskesmas Rawat Inap, Alokasi Dana Obat. ABSTRACT A research about Evaluation of Drug Management in Public Health Centre with Lodge Care of Purbalingga Based on Three Major of Public Sector Drug. The purpose of this study to evaluate management of drug in Public Health Centre with lodge care of Purbalingga based on three major of public sector drug. This research use descriptive method with approach qualitative which supported with quantitative data. The sample’s of this research are Public Health Centre of Kalimanah, Bobotsari and Rembang. The result of research show that Public Health Centre in planning was not good. Optimum stock is not be noted at stock card and there are still requested of drug outside DOEN for the Public Health Centre of Kalimanah 84,51%, Bobotsari 79,64% and Rembang 85,35%. Acceptance of drug have been done with procedure. Storing of Psikotropika drug is not according the rule and not guarantee the quality of drug. Drug distribution, operation of usage, reporting and record-keeping have been done with procedure. Key words : Evaluation, Drug Management, Public Health Centre with lodge care, Public Sector Drug.

Abstract

ABSTRAK Telah dilakukan uji cemaran logam berat Timbal (Pb) pada tanaman caisin yang ada di lokasi ramai (Pratin, Karangreja, Purbalingga) dan lokasi sepi (Gombong, Belik, Pemalang). Analisis dilakukan dengan metode destruksi kering menggunakan alat Spektrofotometer Serapan Atom (SSA) pada panjang gelombang 283,3 nm. Dari hasil analisis tanaman caisin didapatkan cemaran Pb pada lokasi ramai 4,88 ppm dan lokasi sepi 4,79 ppm. Kadar tersebut melebihi nilai yang ditetapkan oleh BPOM yaitu 2 ppm. Hasil validasi metode analisis yang dilakukan pada uji linearitas (r) sebesar 0,9952. Dengan harga standar Deviasi (SD) dan relatif standar deviasi (RSD), adalah sebesar 2,93x10-4 dan 2,74%. Pada uji batas deteksi dan batas kuantitasidiperoleh nilai sebesar 0,33ppm dan 1,11 ppm. Kata kunci : timbal (Pb), spektrofotometri serapan atom, caisin ABSTRACT Test contaminans of heavy metal Lead (Pb) have been done to the caisin plant in crowded location (Pratin, Karang Reja, Purbalingga) and quiet location (Gombong, Belik, Pemalang). Analysis done with dry destruction method used Atomic Absorption Spektrofotometry (AAS) instrument at wave length 283,3 nm. From result analysis of caisin plant gets contaminant Lead (Pb) in crowded location 4,88 ppm and quiet location 4,79 ppm. The level is over the number which has been determined by BPOM that is 2 ppm. The result of validation method analysis performed on the test that the linearity obtained for (r) 0,9952, with value Standar Deviation (SD) and Relative Standart Deviation (RSD) in presision test are 2,93x10-4 and 2,74%. Abstract

Daun Jati belanda (Guazuma ulmifolia L.) dan bunga rosella (Hibiscus sabdariffa L.) telah digunakan secara tradisional untuk menurunkan kadar kolesterol yang tinggi (hiperkolesterolemia). Kedua herbal ini mengandung senyawa yang dapat menurunkan kadar kolesterol, dengan mekanisme yang mungkin berbeda. Hasil penelitian sebelumnya menunjukkan bahwa kombinasi dari ekstrak daun Jati belanda dan bunga rosella mampu mengurangi konsentrasi plasma kolesterol, trigliserida, LDL, meningkatkan konsentrasi plasma HDL, dan menurunkan berat tubuh. Penelitian ini menggunakan kombinasi kedua ekstrak yang diformulasikan menjadi sediaan tablet hisap. Ekstrak dibuat granul dengan metode granulasi basah dalam tiga formula, yaitu F1, F2, dan F3 dengan komposisi bahan pengisi yang berbeda. Pengisi F1 hanya digunakan laktosa, F2 terdiri dari laktosa dan sukrosa sama banyak dan F3 hanya digunakan sukrosa. Hasil evaluasi granul menunjukkan bahwa semua formula memenuhi persyaratan sifat aliran dan kelembaban. Hasil evaluasi tablet menunjukkan bahwa semua formula memenuhi persyaratan kekerasan, hanya F2 yang memenuhi persyaratan kerapuhan, dan F2 yang paling disukai oleh para relawan. Jati belanda (Guazuma ulmifolia L.) leaves and rosella (Hibiscus sabdariffa L.) flowers have been used traditionally for lowering high levels of cholesterol (hypercholesterolemia). Both of these herbs contain compounds that can decrease high levels cholesterol, possibly by different mechanisms. The results of previous studies showed that combinations of Jati belanda leaves and rosella flowers extracts were able to reduce plasma concentrations of cholesterol, triglycerides, LDL, increased the plasma concentration of HDL, and lowering body weight. In this study, combination of both extract were formulated into antihypercholesterolemia lozenges. First, extracts were made granules by wet granulation method in three formulas, namely F1, F2, and F3 with different filler composition. The filler of F1 was lactose, F2 filler of consisted of lactose and sucrose in the same ratio, and F3 only used sucrose as filler. The result of granule evaluation showed that all formulas comply the requirements of flow properties and humidity. The result of tablets evaluation indicated that all formulas comply the requirements of hardness, but only the F2 comply the requirements of fragility, and F2 is most preferred by volunteers.

Abstract

Intisari Salah satu penggunaan obat tradisional adalah dalam mengatasi gangguan tidur, tumbuhan yang sering digunakan secara empiris oleh masyarakat dan berkhasiat sebagai penenang adalah kangkung (Ipomoea aquatic Forsk) Tanaman dengan marga Ipomoea mengandung suatu senyawa turunan Lisergic acid yang diketahui berkhasiat sebagai halusinergik. Ubi jalar (Ipomoea batatas L) merupakan tumbuhan yang satu marga dengan kangkung, sehingga pada penelitian ini diuji efek sedasi dari daun ubi jalar. Penelitian ini bertujuan untuk membuktikan efek sedasi ekstrak etanol daun ubi jalar pada mencit jantan galur DDY. Jenis metode penelitian yang digunakan yaitu jenis eksperimental dengan rancangan penelitian posttest only control group design dan metode analisis data yang digunakan adalah one way anova. Pada penelitian ini dibuat 6 perlakuan yaitu kontrol positif fenobarbital 54,6 mg/KgBB, kontrol negatif Na CMC 1%, kelompok perlakuan ekstrak etanol daun ubi jalar dosis 95,5 mg/KgBB; 191 mg/KgB; mg/gBB; 382 mg/KgBB dan 573 mg/KgBB dengan menggunakan metode rotarod serta mengamati daya cengkeram, perubahan diameter pupil mata dan reflek balik badan. Hasil uji efek sedasi diketahui bahwa efek sedasi terbesar didapatkan pada dosis ekstrak 573 mg/KgBB. Hasil uji efek sedasi dianalisis dengan anava satu arah dengan taraf kepercayaan 95% dan dilanjutkan dengan uji-t dengan menggunakan uji Tukey HSD. Hasil uji anava menunjukkan terdapat perbedaan yang bermakna antara perlakuan masing-masing dosis dengan kontrol positif fenobarbital dosis 54,6 mg/KgBB. Hasil uji anava satu arah diketahui bahwa efek sedasi ekstrak etanol daun ubi jalar pada dosis 382 mg/KgBB dan dosis 573 mg/KgBB tidak mempunyai perbedaan yang nyata dengan kontrol positif fenobarbital dosis 54,6 mg/KgBB, sementara pada dosis ekstrak etanol daun ubi jalar 95,5 mg/KgBB dan 191 mg/KgBB menunjukkan perbedaan yang bermakna tehadap kontrol positif. Hal ini menunjukan bahwa potensi ekstrak etanol daun ubi jalar pada dosis 382 mg/KgBB dan dosis 573 mg/KgBB setara dengan kontrol positif fenobarbital dosis 54,6 mg/KgB. Ekstrak etanol daun ubi jalar (Ipomoea batatas L) dapat memberikan efek sedasi pada mencit pada dosis 382 mg/KgBB dan 573 mg/KgBB seperti pada kontrol positif fenobarbital dosis 54,6 mg/KgBB Kata kunci: efek sedasi, fenobarbital, ekstrak etanol daun ubi jalar (Ipomoea batatas L) Abstrak One is the use of traditional medicine in treating sleep, plants are often used empirically by the public and efficacious as a sedative is spinach (Ipomoea aquatic Forsk) the genus Ipomoea containing an derivative compounds Lisergic acid known efficacious as halusinergik. Sweet potato (Ipomoea batatas L) is a genus of plants with kale, so this study examined the effects of sedation of sweet potato leaves. This study had aims to prove the sedation effect of ethanol extract of leaves of sweet potato in DDY. Type of research method was experimental study with posttest only control group design and data analysis,the methode used is the one way ANOVA. In this study made 6 positive controls phenobarbital treatment are 54.6 mg/kgBW, negative control 1% Na CMC, the treatment of ethanol extract of sweet potato leaves 95.5 mg/kgBW: 191 mg/kgBW; 382 mg/kgBW and 573 mg/kgBW rotarod method and observing traction changes in pupil diameter and reflexes behind the body. From the test results sedation are known that the largest sedation effect is obtained at a dose of 573 mg/kgBW. The test results are analyzed with ANOVA sedation in one direction with a 95% confidence level and followed by BNT using Tukey HSD test. The test results show ANOVA there are significant differences between treatment of each dosage with a positive control. Smallest Real Differences test results are known that sedative effects of ethanol extract of sweet potato leaves at dosage of 382 mg/kgBW and the dose of 573 mg/kgBW have no significant difference with the positive control, while the dose of ethanol extract of sweet potato leaves 95.5 mg/kgBW and 191 mg/kgBW show a significant difference with positive control. It is shown that the potential of ethanol extract of sweet potato leaves at dosage of 382 mg/kgBW and the dosage of 573 mg/kgBW is equivalent to a positive control. Abstract

ABSTRAK Penelitian ini bertujuan untuk membuktikan bahwa buah mengkudu dalam bentuk ekstrak soxhletasi dapat mempercepat pertumbuhan rambut kelinci jantan. Kandungan buah mengkudu yang diduga mempunyai efek merangsang pertumbuhan rambut adalah alkaloid, saponin dan flavonoid. ekstrak dibuat dengan cara soxhletasi serbuk buah mengkudu menggunakan pelarut etanol 96% kemudian dipekatkan dengan penangas air, selanjutnya digunakan untuk uji pertumbuhan rambut kelinci jantan. Punggung kelinci dicukur persegi dengan sisi 2 cm. Kotak 1 diolesi etanol 96% sebagai kontrol negatif, kotak 2 diolesi ekstrak soxhletasi buah mengkudu konsentrasi 12,5 %, kotak 3 diolesi ekstrak soxhletasi buah mengkudu konsentrasi 25 %, kotak 4 diolesi hair tonic kina sebagai kontrol positif. Pengukuran pertumbuhan rambut dilakukan setiap 3 hari sekali selama 18 hari. Data pertumbuhan rambut rata-rata perhari yang diperoleh kemudian dianalisis dengan anava yang dilanjutkan dengan uji tukey dengan taraf kepercayaan 95 %. Hasil penelitian menunjukkan bahwa ekstrak soxhletasi buah mengkudu dapat mempercepat pertumbuhan rambut kelinci jantan. Kata kunci : Buah mengkudu, ekstrak soxhletasi, pertumbuhan rambut, kelinci jantan ABSTRACT The aim of this study is to prove whether ethanolic extract of Morinda can increase rabbit hair growth. The secondary metabolites of Morinda which stimulate rabbit hair growth are probably alkaloid, saponin and flavonoid. The extract was made by soxhletation of Morinda used ethanol 96 % as solvent, then it was thickened by water bath. The rabbit back was shaved into 2 cm square box. The first area treated ethanol 96 % as negative control, the second area treated with 12,5 % ethanol extract of Morinda, the third area treated with 25 % ethanol extract of Morinda, the fourth area treated with kina hair tonic as positive control. The measurement of hair growth was done every 3 days for 18 days. The daily average of hair growth was analyzed with anova and continued with Tukey test at 95 % significancy level. The result showed that ethanol extract of Morinda can increase growth of rabbit’s hair. Key words: Morinda, soxhletation extract, hair growth, rabbit.
