Abstract

Telmisartan (TMS) has a compact structure and is interlocked between crystal units. As a result, the substance has a high electrostatic force, which causes TMS to sintering when compressed into tablets. One method of overcoming sintering on TMS tablets is to add a disintegrant to the compressed tablet. The disintegrants used are derived from starch groups: Starch 1500® (S1500) and sodium starch glycolate (SSG), as well as microcrystalline cellulose (MCC) and croscarmellose sodium (CCS) groups. The purpose of this study was to examine the effects of several disintegrants on the dissolution and physicochemical properties of TMS with various treatments. TMS was varied with each disintegrant at a ratio of 1:9 % b/b. The treatment of TMS binary mixtures with various disintegrants includes physical mixtures, milled mixtures, compressed mixtures, and crushed compressed mixtures. Characterization and evaluation include PXRD and SEM testing as well as dissolution test. The characterization of PXRD and SEM in various treatments showed the effect of physicochemical properties on the TMS binary mixture with various disintegrants. The combination of TMS:CCS with a ratio of (1:9) resulted in the highest dissolution rate in all treatments. The treatment of the milled mixture produced the highest dissolution with DP60 minutes, which was around 55.86±2.47%. The addition of disintegrants to TMS with various treatments may reduce sintering but is not sufficient to meet the requirements for TMS dissolution, which dissolves within 30 minutes in phosphate buffer medium pH 7.5 with Q>75%.

Abstract

Moringa leaves contain macronutrients and micronutrients, potential to be developed into cereal products in the form of granules using the wet granulation method. The type of binder and the concentration of the suspending agent are the main factors that affect the physical characteristics and proximate content of cereals. In this research, 6 formulas of cereal granules were optimized with various types of binders (arrowroot flour, sorghum flour, cornstarch) and variations in the concentration of xanthan gum (0.5% and 1.0%). This study aims to obtain a granule formula of Moringa leaf cereal that meets the requirements of physical, chemical, proximate, and heavy metal characteristics. The results of the evaluation of the granule characteristics showed that all Moringa leaf cereal formulas had good flow characteristics. The results of the evaluation of cereal characteristics after reconstitution showed that cereal granules dispersed quickly (12.58–16.45 seconds), viscosity 73.75 -245 cps, pseudoplastic flow characteristics, and met the pH requirements of food products (5.00-6.00). The results of proximate analysis and metal content showed that all cereal formulas met the SNI requirements for cereal milk (SNI 01-4270-1996) in terms of protein, carbohydrate, ash, lead (Pb), and copper (Cu) content parameters. The most optimal cereal formula is formula 2 because it shows physical characteristics, fat content, carbohydrates, protein, and heavy metals that meet the requirements, as well as crude fiber content (0.76%) which is closest to the requirements (0.7%).

Abstract

The composition of the intestinal microbiome of neonates can be identified from meconium and feces by Next-Generation Sequencing (NGS) technology. However, the yield of microbiome DNA of meconium and feces has its own challenges due to the consistency and the high content of PCR inhibitors in these samples. This study aims to optimize the yield of microbiome DNA from meconium sample and feces of pre-term neonates. The DNA yield was obtained by applying certain optimized parameters, i.e., considering the replication and condition of the sample, using a particular kit for DNA extraction, and modifying the DNA elution of the column purification. The genomic DNA obtained was quantified and confirmed using Polymerase Chain Reaction. Results showed that the best DNA yield was achieved by replicating the number of samples twice in the pre-extraction stage, working on fresh meconium and feces samples instead, and suspended the sample in ddH2O prior to extraction process as observed on agarose gel visualization with UV trans-illuminator, as well as in quantitative measurement by a nano spectrophotometer. The best extraction process was using MP Biomedical FastDNA Spin Kit for Soil, in addition to the use of an elution buffer in a smaller volume, resulting in a higher concentration and purity of DNA. In conclusion, we were able to obtain an optimized yet reliable DNA yields, especially from meconium, which fulfilled the quality and quantity requirement for further sequencing process of microbiome.
