Abstract

The existence of cave dwelling bats of karst area need to be conserved, because bats have important roles for the ecosystem both inside the cave  as well as outside the cave. The purpose of  this study was to observe  structural respiratory adaptation of the cave bats . This study was conducted from July 2013 to November  2013 at three caves in  Karst  Gudawang, Bogor Regency, West Java. In each cave where the bats roosting, the temperature, also the humidity, and  oxygen level were measured. Samples were caught from every bat roost during the day when the bats were staying in their roosts.  Lungs and trachea  removed from body and  were  made preparations for histology. The lung and trachea  was observed under the microscope. The data were analyzed using ANOVA. The result showed:  The ratio of  lung weight/body weight  increased by the decrease of oxygen levels. This tendency also revealed  in diameter of trachea/body weight  change

Abstract

Traditional medicine "Subur Kandungan" is a medicine that is widely used and produced in Madura. The problem to develop Madura’s herb is raw material that hasn’t the standardized medicine used for Madura’s traditional medicine, because that raw materials are mostly purchased from traditional markets. This led to the standardization of Madura’s traditional medicine is difficult to do. This condition causes didn’t known active compounds contained in the Madura’s medicine. This study was conducted to determine the content of phytochemical compounds from ethanol extract medicine "Subur Kandungan" as well as antioxidant activity. Based on the results, it was concluded that the class of chemical compounds contained in the Madura’s traditional medicine "Subur Kandungan" are classified as a flavonoids and alkaloids, while the antioxidant activity of extracts of the medicine " Subur Kandungan " are expressed in IC50 was at 90.19 ppm classified as a strong antioxidant.

Abstract

Background: An anthracnose disease caused by pathogenic fungal Colletotrichum capsici has been attacking the cayenne plants either harvested or has not been harvested. This disease must be handled appropriately and quickly because it can reduce the production of chili up to 90%. Recently, anthracnose disease prevention still use chemical fungicide that if applied for long time will cause new impact for environment. Objective: Trichoderma and Bacillus cereus endophytes may be used as antagonistic agents for C. capsici pathogens because they have various antibiotic compounds. Methods: This research uses experimental method. The stages of this study include sterilization of tools and materials, preparation of culture media of fungal and bacteria, rejuvenation of endophytic microbe culture Trichoderma sp. and Bacillus cereus, rejuvenation of  C. capsici pathogen, antagonistic test in vitro using dual culture method. Result and conclusion: The results of in vitro antagonistic tests showed that inhibition percentage of Trichoderma treatment (96%) and combination treatment Trichoderma and B. cereus (97%) is not significantly different. While in B. cereus treatment (11, 88%) significantly different with all of treatments. Endophytes are shown by its dominating growth in petri dishes than C. capsici pathogen or B. cereus endophytes. Furthermore,for endophytes Trichoderma sp continued on in vivo test because it was most effective.The result for incubation period is 3 days after inoculation compared with negative control 2 days. For disease incidence 100%, and for disease intensity that is 61,25% compared with negative control equal to 88,75%.

Abstract

Hypertension is an abnormal increase in blood pressure. Regulating blood and cardiovascular function have correlated with the ACE pathway. To decrease blood pressure can use the ACE inhibitor. This paper aims to predict potential of Caffeic Acid as anti-hypertension by blocking the ACE pathway. The method in this research used in silico study. The protein was obtained from Protein Data Bank (PDB) and the ligand was obtained from PubChem. Molecular docking was performed by using HEX and visualization analysis was analyzed by using Discovery Studio. The interaction of caffeic acid and ACE has a functional as anti-hypertension roles. The evidence by twelve amino acid, which bind with the caffeic acid (ASP377, ASN277, ASN285, GLU376, ALA170, ASN167, ASN374, THR372, THR166, CYS370, GLU162 and PRO163). The chemistry bond was formed are hydrogen bond, van der Waals and electrostatics in amino residue ASP377. This binding could stop the synthesis of AT-I to AT-II which pathway to hypertension. Caffeic acid has a potential role as anti-hypertension by inhibiting ACE.

Abstract

S. oleosa belongs to the Sapindaceae family and has phytochemicals phenolic acid so that it has enormous benefits in the process antioxidant. The presence of antioxidant activity in cells can affect the defense of the cell protein membrane, so that the ability of cells to perform division is optimal. In vitro this is mostly done by adding hormones to cell culture media or natural compounds to increase cell proliferation. Every population of cell that has undergone division during the proliferation process to confluent is an indication that the cell is aging. This study was an experimental study was designed using a completely randomized design (CRD) with 5 treatments and 5 replications as follows: K0: granulose cells and culture media without treatment (0 µl), P1: granulose cells and culture media with 1% kesambi leaf extract. (30 µl), P2: granulose cells and culture media given kesambi leaf extract 1.5% (45 µl), P3: granulose cells and culture media given kesambi leaf extract 2% (60 µl) and P4: granulosa cells and The culture media was given 2.5% (75 µl) to the leaves extract., the characteristics of goat ovarian granulosa cells by giving kesambi (Scheichera oleosa) leaf extract at different passages were at the level of confluence showing the highest value in passage I, namely treatment P1. Meanwhile, the level of viability shows the highest value in section I, namely the P4 treatment

Abstract

Pineapple Liquid Waste has high organic material  and still unfulfilled required quality standard. Processing occurred at the waste processing installation with pond system (Lagoon) required a wide space and keep long time so that less efficient. Based on this problem, it needs an environmentally sound waste management by utilizing bioprocess technology using indigenous bacteria’s ability in degrading organic pollutant called Bioremediation. The objective of this research was to get isolated indigenous bacteria, enables to decrease organic material. It was divided into four stages. First, performed characteristic observation of isolated bacteria that potentially decrease organic material, secondly decomposition capability test in vitro, and thirdly scale bioremediation test pilot plan, fourthly bioremediation test in ex situ at the microbiology laboratory of Malang State University. Data were analyzed by Varian analysis to test the presence or absence of differences and regression test to analyze the link between variables. First phase result: 15 isolated indigenous bacteria.. Potential consortia bacteria consisting of more effective 4 species, indicated by 4 consortia bacteria decreased BOD in 6 days. Starter bacteria with a consortia K1’s combination (ABCD), Volume: 5% (v/v) and 6 days were the most effective incubation time in decreasing BOD. Second phase result: 1) A significant link among the number of bacteria, a. Bacillus cereus, b. Acinetobacter baumanni, c.Bacillus subtilis, d. Pseudomonas pseudomallei and not abcd against organic material with contribution of 72,7% and negatively correlated, 2) Media treatment and incubation time gave distinctive outcome against organic material,  3) there was a link between organic material organic acid concentration decrease would decrease BOD  at scale bioremediation test Pilot plan, and ex situ scale. Threshold BOD achievement limit value on 10th day (75) quality standard: 75. In accordance with ministry decree LH No. 05 year 2007 industrial wastewater quality standard, suggested to industry to use this study in processing its waste.Keywords: organic material, isolated indigenous bacteria, pineapple liquid waste.

Abstract

The aim of the current study was to know if adenocarcinoma mamma tumor can be appear at the stomach of F1 hybrid mouse (C3HxAJ) that innoculated adenocarcinoma mamma's tumor via antravenous, intraperitoneal, and subcutaneus. Methods : The study was experimentaly  using  posttest  control  group  design,  to  examine  the  pylorus  mucosa thickness, the number of metaplasia, hyperplasia, and inflammatory cell. Result : There were no metaplasia and hyperplasia but inflammatory cell has been found. Analize of the pylorus mucosa thickness and the number of inflammatory cell by Oneway ANOVA showed there was no significant different of control group and manipulated group (intravenous, intraperitoneal and subcutaneus) (p0.05). Conclution : Adenocarcinoma mamma's tumor has been not found at the stomach of F1 hybrid mouse (C3HxAJ) that innoculated with via intravenous, intraperotoneal and subcutaneus.

Abstract

Hepatitis virus merupakan masalah kesehatan global yang menyebabkan mortalitas dan morbiditas jutaan orang di dunia, baik dewasa maupun anak-anak. Hepatitis virus akut terbanyak disebabkan oleh virus hepatitis A, sedangkan virus hepatitis B menyebabkan hepatitis akut dan kronis. Diagnosa hepatitis virus ditegakkan berdasarkan pemeriksaaan laboratorium karena secara klinis banyak kasus yang tidak menunjukkan gejala. Deteksi virus hepatitis dilakukan secara langsung dengan pemeriksaan antigen virus, maupun secara tidak langsung dengan pemeriksaan antibody virus. Deteksi ini dilakukan berdasarkan patogenesa virus dan struktur virus hepatitis A dan B. Deteksi ini bermanfaat untuk mengetahui perjalanan penyakitdan status infeksius penderita

Abstract

Selection of traditional medicine is often considered as a choice since the affordable price and low side effects than chemical drugs. One of the well-known herbal medicine produced and consumed in java is Madura Herbal Medicine (MHM). MHM "Subur Kandungan" is one of the original medicinal products which is widely known in inducing fertility, but has not been scientifically standardized so make this many doubt for its usefulness. With the standardization and scientific assessment, MHM ought to be accepted in the modern medical system as well as lead in increasing both society revenues and public health. MHM "Subur Kandungan" consist of garlic (Allium sativum L.), Sweet Flag (Acorus calamus L.), and Intersection rhizome mango (Curcuma mangga Val.) is widely recognized as efficacious herbal medicine for inducing fertility. To determine the potency of those three plants, these were extracted by ethanol and continued by testing the antioxidant potential (DPPH) both individually and mixed. Assessments of phytochemical and antimicrobial potency were discussed. Test results showed that antioxidant activity represented by DPPH IC50 values of ethanol extract of A. sativum, A. calamus, C. mangga  and a mixture of those are 383.56, 335.68, 32.48, 27.773 µg/ml (ppm) respectively. Allium sativum and A.calamus are weak antioxidants as having IC50 in the range of 250-500 ppm, while the C. mangga and a mixture of those have strong antioxidant content as having IC50 of less than 50 ppm. Allegedly due to the high antioxidant content of the content of steroids, phenols, tannins, flavonoids, glycosides, diterpene, triterpenes, and alcaloids in A. calamus, also contains steroids, flavonoids, glycosides, diterpene, triterpenes and alaloids on A. sativum and also the presence of terpenoids as compound intersection primary contained in C. mangga respectively.

Abstract

The purpose of this research was to determine the effect of polysaccharide krestin (PSK) on the testicular protein profiles and testosterone levels of Mus musculus with variety of dosages. This research used a completely randomized design. It were devide into four treatment group i.e. control group, PSK treatment at a dose of  15, 30, 60 mg/kgBW. Each group had six replications. Testicular proteins were isolated by flushing technique and analized by SDS-PAGE. Testosterone levels were analized using ELISA technique at wavelength 450 nm. Protein bands analysis showed that there were no diversification between four treatments. Molecular weight of protein bands were 87, 63, 57, 35, and 30 kDa. The results of research showed that the testosterone levels at dosage 60 mg/kgBW had significanly different with control, PSK treatment of 15 and 30 mg/kgBW. PSK treatment of  60 mg/kgBW had lowest level at dosage, i.e. 25946.8 ρg/mL. It can be concluded that giving variety of dosages of polysaccharide krestin did not affect to testicular protein profiles but giving effect to testosterone levels of Mus musculus.
