Abstract

Background:Sesbania grandiflora L flower has high antioxidant activity. This activity is caused by phytochemical ingredients of Sesbania grandiflora L flower, such as vitamin C, flavonoids and tannin. The aims of this research were to know the hepatoprotector effect of Sesbania grandiflora L flowers extract to the liver cell structure damage of mice caused by over reheated palm oil and to find out dose escalation of Sesbania grandiflora L flowers extract that can enhance the effect. Methods:This research was laboratory experimental study with the post-test only controlled group design. The tested animal used in this experiment were male mice, Swiss webster strain.The research population were 30 male mice divided into 5 groups, each group consisted of 6 mice. The negative control group (KKn) was given distilled water, while the positive control group (KKp) was given over reheated palm oil. The 1st treatment group (KP1), the 2nd treatment group (KP2) and the 3rd treatment group (KP3), all, were given Over Reheated Palm Oil as much as 0.06 ml/ 20 gr BW andSesbania grandiflora L flowers extract with dose 7 mg, 14 mg and 28 mg per 20 gr BW of mice, consecutively. The liver histological images were assessed by the sum of the cell nuclear pyknosis, karyorrhexis, and karyolysis. Data were analyzed by using One-Way ANOVA test and Post Hoc Test (LSD) (?=0.05). Results:One-Way ANOVA test showed that p value was 0.000 (p < 0.05).The results of the post hoc multiple comparisons test showed the significant differences between group KKn-KKp, KKn-KP1, KKn-KP2, KKn-KP3, KKp-KP1, KKp-KP2, KKp-KP3, KP1-KP2, KP1-KP3, and KP2-KP3 Conclusions:Sesbania grandiflora L flowers extract has a protective effect on liver cell structure damage of mice (Mus musculus) caused by over reheated palm oil and the escalated dose of Sesbania grandiflora L flowers extract can enhance the effect. Abstract

Background: Gentamicin may cause nephrotoxicity and inhibits blood creatinine excretion. Xanthone from mangosteen pericarp has antioxidant and antiinflammation effects and can potentially protect kidney from inflammatory-mediated gentamicins nephrotoxicity. Gentamicin-induced renal damage can be identified from the elevation of blood creatinine levels. The purpose of this research was to find the potential of mangosteen pericarp extract to prevent the elevation of blood creatinine levels. Methods: This was an experimental research with pre and posttest controlled group design. Samples were 30 male Wistar-strain rats, 2-3 months old with approximately 200 gram of weight. Samples were randomly divided into 3 groups:control group (KN) administered Na CMC 0.5% orally, negative control group (K-) administered gentamicin 100 mg/kgBW intraperitoneally, and treatment group (KD) administered gentamicin 100 mg/kgBW intraperitoneally and 125 mg/kgBW mangosteen pericarp extract orally. Blood creatinine levels were assessed before treatment and 10 days after treatment by spectrophotometer. Data collected were analysed by Kruskal-Wallis and Mann-Whitney tests. Results: There was significant difference of blood creatinine levels before and after treatment among the three groups (p<0.001). Mann-Whitney tests showed statistically significant differences of creatinine level between KN-K- and KD-K- (p<0.001). No significant difference of blood creatinine levels was found for KN-KD comparison(p=0.06). Conclusion: Mangosteen pericarp extract at dose of 125 mg/kgBW prevented the elevation of blood creatinine levels in Wistar rats induced by gentamicin. Abstract

Background: Fatigue is a condition characterized by a decrease in work efficiency and endurance. BCAA capsule contains of three kinds of essential amino acids, those were leucine 2275 mg, isoleucine 1137.5 mg, and valine 1137.5 mg, each of which plays an important role in protein synthesis and energy system for activity. This study aimed to examine the effect of supplement administration of BCAA to reduce fatigue levels in medical students of Sebelas Maret University (UNS). Methods: This was an experimental research with pretest-posttest group design. It was conducted at Faculty of Medicine UNS on December 13th, 2012. Subjects (n=40) were randomly selected from list of current medical students at preclinical level. Subjects who had been fasting for eight hours, then, were randomly arranged into two groups: (1) control group whose fatigue levels were measured twice in resting condition at10-minute interval, and (2) treatment group whose fatigue levels were measured before and 10 minutes after BCAA administration. Fatigue referred to decreasing concentration determined by reaction time to light stimuli. Pretest-postest fatigue levels for each group were analyzed using paired sample t-tests, whereas posttest fatigue levels between the two groups were analyzed using independent sample t-test. Data analysis were performed using SPSS 17.0 for Windows. Results: Mean fatigue levels of control group at first and second measurement, each, were 270.115.3 and 255.510.9 millisecond. While mean fatigue levels of treatment group at pretest and after BCAA administration were 268.9 14.8 and 211.738.8 millisecond, consecutively. Analysis of paired sample t test of pretest-postest fatique levels in both groups, each, showed significant difference (p = 0.001 for control group and p < 0.001 for treatment group), while analysis of independent t test of posttest fatigue levels between the two groups showed that the posttest fatigue levels between the two groups also differed significantly (p <0.001). Conclusions: BCAA administration significantly reduced fatigue levels in medical students of UNS. Fatigue levels were significantly shorter in subjects administered BCAA supplement than those of control group who did not take BCAA supplement. Keywords: BCAA suplement, fatigue

Abstract

Background : The purpose of this research was to know the relationship between obesity and irregularity menstrual cycle. Methods: This was an analytic observational study with cross sectional approach, using purposive sampling technique. The samples were 76 women, 20 35 years-old from Minomartani Village, Ngaglik, Sleman. Body Mass Index (BMI) data were obtained by measuring the height and weight, while the type of menstrual cyle by interview, on April 2012. Data were analyzed by Chi Square and Odds Ratio test. Results: Thirty women had BMI more than 25 kg/m2 (obese) and fourty six women had BMI between 18.5-24.9 kg/m2 (normal). The Chi Square test showed the value of p was 0.035 (p < 0.05), while the Odds Ratio test showed that value of OR was 3.154 (OR>1). Conclusions: There was significant relationship between obesity and irregularity menstrual cycle in fertile women in Minomartani village, Ngaglik, Sleman. Keywords: Obesity, Menstrual Cycle

Abstract

Introduction : Broccoli is known to contain antioxidants such as vitaminC, flavonoids and sulforaphane. Antioxidants can protect liver from damage caused by free radicals. This study aims to prove that broccoli extract protect liver cell from damage in mice induced by paracetamol in toxic dose and an increased doses of broccoli extract enhance the effects of protection to liver cell in mice. Methods : This study was an experimental laboratory with post test only controlled group design. Samples (30 mice) were taken by incidental sampling. Samples divided randomly into 5 groups, each group consisted of 6 mice. K1 was given distilled water. K2 was given paracetamol in toxic dose. KP1, KP2 and KP3 were given broccoli extract dose I, II and dose III respectively for 14 days and also were given paracetamol in toxic dose on day 12, 13 and 14. On day 15, the livers of mice were taken for making preparations with HE staining. The liver cell damages were observed by counting the nuclei of cells undergoing pyknosis, karyorrhexis and karyolysis. Data were analyzed by Oneway ANOVA and Post Hoc LSD analysis test (? = 0.05). Result : K2 had the most damaged cells (157.17 7.834 cells) and the least damaged cells were at K1 (1.50 1.049 cells). Oneway ANOVA test result was p = 0.00 (p < 0.05). Result of Post Hoc LSD analysis test showed a significant difference between groups on all pairs with p = 0.00 (p < 0.05). Conclusions : Broccoli extract protect liver cells from damage in mice induced by paracetamol. Increased doses of broccoli extract were proven to increase the protection effect on liver cells. Abstract

Background: This stydy is to know the effect of acupuncture at Baihui (GV20) acupoint and electroacupuncture at Zusanli (ST36) acupoint to WBC level in stress model white rats resulting from chronic intermittent noise. Methods: This study was an experimental with pre and post test group designs. Subjects were 32 male white rats (Rattus norvegicus), wistar strain samples and grouped into 2 groups. KB (the noise group) were induced with chronic intermittent noise and KBA (acupuncture-noise group) were induced with chronic intermittent noise and treated with acupuncture at Baihui (GV 20) acupoint and electroacupuncture at Zusanli (ST36) acupoint. Subjects was treated for 28 days. The collected data were analyzed using independent samples t-test, paired-samples t-test, alternative test with Wilcoxon test, and Mann-whitney test. Results: Mean KB (pre test), KB (post test), KBA (pre test), and KBA (post test) were 12,952.502,845.52/uL; 8,151.881,978.19/uL; 12,493.132,584.90/uL; and 8,942.502,591.93/uL, respectively. The statistic analysis showed that there was a significant difference of white blood cell count between KB (pre test) and KB (post test) as well as between KBA (pre test) and KBA (post test) but there was no significant difference between KB (pre test) and KBA (pre test) as well as beetween KB (post test) and KBA (post test). Conclusion: There is no effect of acupuncture at Baihui (GV20) acupoint and electroacupuncture at Zusanli (ST36) acupoint against WBC level of white rats model stress resulting from chronic intermittent noise. Abstract

Background: Star fruit is a good source of natural antioxidants which can prevent oxidative damages and modulate reactive free-radicals. However, no study had been carried out so far on the hepatoprotective effect of star fruit. This research aimed to study the hepatoprotective effect of star fruit juice on SGPT level in paracetamol- intoxicated rats. Methods: This was a laboratory experimental study with posttest only controlled group design. A total of 30 white male, Wistar rats (Rattus norvegicus), aged 2-3 months and weighed 160-200 grams was assigned into five groups. Normal group and negative control group were administered distilled water, while positive control group was administered Curcuma (3.6 mg/200 g bw). The treatment group 1 and 2 were administered star fruit juice at concentration of 50% and 100%, successively. This study was performed for 14 days. In the last 3 days, all groups except normal group were induced by paracetamol at toxic dose (291.6 mg/200 g BW) an hour after the treatment of star fruit juice (for treatment group 1 and 2) was done. Blood samples were collected from retro-orbitalis venous to measure SGPT levels. The data were analyzed statistically using one-way ANOVA followed by LSD test. Results: The analysis of SGPT with one-way ANOVA showed a significant difference across all groups. Further, the LSD tests of SGPT level showed significant differences between negative control group (182.5 U/L 14.0) and the two star fruit groups (p values <0.001). SGPT mean in treatment group 1 (87.0 U/L 14.1) and treatment group 2 (91.7 U/L 14.9) also differed from those of positive control (67.1 U/L 5.5, p=0.008) and normal group (53.8 U/L 8.6, p=0.002). There was no significant difference between two groups treated with star fruit juice (p=0.511). Conclusions: Star fruit juice at concentration of 50% and 100% significantly inhibited the elevation of SGPT levels in paracetamol-intoxicated rats. Abstract

Introduction: Kersen leaves are known to contain various kinds of active substances one of which is a flavonoid, a type of flavonoid that is quersetin have a function as lowering uric acid levels through inhibition of the enzyme xanthine oxidase. This study aims to find out Kersen leaves (Muntingia calabura) Ethyl Acetate Extract Effect on Blood Uric Acid Levels in Mice (Mus muculus) Male with Hyperuricaemia Model. Methods: This study was an experimental laboratory with post test only control group design. A sample of male mice, aged 2-3 months, body weight 25-40 grams. The subjects were male mice 24 individuals, were divided into six groups randomly. Group I as normal control, group II as negative control received caffeine 0.42 mg and 0.5ml chicken liver juice orally. Group III, IV, V and VI received respectively allopurinol treatment 0.2 mg (positive control), kersen leaf extract consecutive doses 12.74 mg, 25.48 mg and 50.96 mg it orally, and induced caffeine 0.42 mg and chicken liver juice 0.5ml orally 1 hour after treatment. Measurement of uric acid levels using a GCU test strip, blood was taken from the mice tail one hour after the induction of hyperuricemia. Data were analyzed using one-way ANOVA followed post hoc LSD test Results: The result showed blood uric acid levels throughout the test animals was significantly different (p?0.05). The test results, post hoc LSD test showed that the positive control group and group IV, V and VI significantly different from the negative groups (p?0.05) while there was a significant difference in the group IV, V and VI (p ? 0.05) with the normal control group and positive control group. Conclusion: All doses can lower uric acid levels of mice significantly. And all three doses did not differ significantly of uric acid levels compare to therapeutic doses of allopurinol. Abstract

Background: Guava leaves contain tannins and saponins that have been known to have anthelmintic effect. This study aimed to determine the effect of guava leaf extract toward mortality of Ascaris suum, Goeze in vitro Methods: The study was a laboratory experimental research using the post-test only controlled group design. Subjects were adult Ascaris suum, Goeze. The sampling technique used was purposive sampling. Subjects were divided into 7 groups, each group consisting of 5 worms, replication performed 4 times. Saline solution was used in negative control group, pirantel pamoate 5 mg/ml was used in positive control group, while the treatment group used guava leaf extract concentration of 20%, 40%, 60%, 80%, and 100%. Worms were immersed in the test solution at 25 mL and was incubated at 37?. Observations were made every 1 hour for 7 hours and counted the number of dead worms for 7 hours. Data were analyzed with Kruskal-Wallis test, followed by Mann Whitney test and probit analysis. Results: Total number of dead worms after 7 hours of observation in the negative control group was 0, guava leaf extract concentration of 20%, 40%, 60%, 80%, and 100% respectively by 4, 7, 12, 20, and 20, and in the positive control group was 20. Kruskal Wallis test results showed a significant difference, while Mann Whitney test showed some significant and unsignificant differences among those groups (p <0.05 and p> 0.05). Probit analysis showed LC50 and LC90 respectively at concentration 54.320% and 74.516%.. Conclusions: Based on this study, it can be concluded that the guava leaf extract affects mortality of Ascaris suum, Goeze in vitro, the increased concentration of extract increases the number of deaths of worms. Abstract

Background: Data of APOBEC3B gene deletion status in Indonesia are limited. Moreover, information aboutAPOBEC3B gene deletion status in Multidrug-Resistant Tuberculosis (MDR-TB) patients in Dr. Moewardi General Hospital in Surakarta was unknown. This study was aimed to know the APOBEC3B gene deletion status in MDR-TB patients in Dr. Moewardi General Hospital in Surakarta. In the future, this research was expected can be used as asupporting examination in chronic tuberculosis patients which tend to progressively become MDR-TB. Methods: Blood samples from 43 MDR-TB patients in Dr. Moewardi General Hospital in Surakarta were collected, DNA isolation was performed for all samples, followed by a single Polymerase Chain Reaction (PCR)to detectthe presentation of APOBEC3B gene deletion genotype. Results: APOBEC3B gene deletion genotype was detected in81.4%(35/43) samples. Conclusions: The APOBEC3B gene deletion genotype was frequently found in MDR-TB patients in Dr. Moewardi General Hospital in Surakarta. Keywords: APOBEC3B, deletion, MDR-TB
