Abstract

Orientation: This  study  examined  the  possibility of using  yam bean as a natural estrogen for mice in their premenopausal stage.Research Purpose: This research aimed for doing examination to the  phytoestrogen contains of Yam Bean, its dynamic in the serum and also its potential to the  female white mice.Motivation for the study: This research provides the benefit of consuming yam bean to increase the estrogen in mice body. Estrogen hormone mostly used by people especially woman to prevent and medicinal of the disease. Yam Bean is a group of phytoestrogen plant because it contains isoflavonoid with chemical structure that similar with estrogen hormone.Reasearch design, Approach, and Method: This research using pattern experiment approach of Complete Random  Design with treatment giving of Yam Bean and daidzein synthesis compound for 24 white mice from the kind of Sprague Dawley in the age of 5 months for 24 days. Daidzein dosage that given based daidzein level in the 1.5 ml of Yam Bean distillate. Blood taking conducted in the 3 fractions (8th, 16th, and 24th hour after treatment). Dissection and uterus organ taking conducted in the 25th day, blood smear making of tissue with HE coloring. Daidzein level test is analyzed with HPLC method. Structure alteration of uterus tissue analyzed descriptively to the endometrial, miometrial and  uterus mucosa layer.Main Findings: Research result shows that daidzein level in serum with Yam Bean distillate giving in the fraction 1, 2, and 3 are lower than daidzein synthesis compound giving. Endometrial tissue of uterus having proliferation, and also uterine gland proliferation in the treatment giving of Yam Bean distillate. The conclusion shows that Yam Bean can improve daidzein level that has potential as natural estrogen.Contribution/Value-add: The process leads other researcher to do the next experiment related to yam bean as a natural estrogen and its possibility of using yam bean as a natural estrogen for human.

Abstract

Hormon estrogen sering digunakan oleh masyarakat khusnya wanita untuk pencegahan dan pengobatan penyakit. Bengkuang merupakan kelompok tanaman fitoestrogen karena mengandung senyawa isoflavon dengan struktur kimia mirip hormon estrogen. Tujuan penelitian adalah melakukan pengujian terhadap kandungan fitoestrogen umbi bengkuang, dinamikanya dalam serum serta potensinya pada tikus butih betina. Penelitian menggunakan pendekatan eksperimen pola Rancangan Acak Lengkap dengan perlakuan pemberian umbi bengkuang dan senyawa sintetis daidzein pada 24 ekor tikus putih jenis Sprague Dawley umur 5 bulan selama 24 hari. Takaran daidzein yang diberikan berdasarkan kadar daidzein pada 1,5 ml perasan umbi bengkuang. Pengambilan darah dilakukan pada 3 fraksi (jam ke-8, 16, dan 24 setelah perlakuan). Pembedahan dan pengambilan organ uterus dilakukan pada hari ke-25, pembuatan preparat jaringan dengan pewarnaan HE. Pengujian kadar daidzein dianalisis dengan metode HPLC.  Perubahan  struktur jaringan uterus dianalisis secara diskriptif terhadap lapisan endometrium, miometrium dan mukosa uterus.  Hasil penelitian menunjukkan kadar daidzein dalam serum dengan pemberian perasan umbi bengkuang pada fraksi 1, 2, dan 3  lebih rendah daripada pemberian senyawa sintetis daidzein. Jaringan endometrium uterus mengalami proliferasi, serta proliferasi kelenjar uterina pada perlakuan pemberian perasan umbi bengkuang. Kesimpulan menunjukkan umbi bengkuang dapat meningkatkan kadar  daidzein yang berpotensi sebagai estrogen alami.   Kata kunci: Umbi Bengkuang; Fitoestrogen; Daidzein; Uterus

Abstract

ABSTRAK Usaha untuk mendapatkan sarana kontrasepsi pria sebenarnya telah banyak dilakukan dengan memanfaatkan bahan alami yang berasal dari tanaman. Salah satunya adalah tanaman famili Leguminoceae yang mengandung senyawa genistein. Genistein sebagai salah satu senyawa derivat isoflavon mempuyai struktur kimia mirip dengan 17?-estradiol yang bersifat seperti hormon steroid estrogen, yang mampu menyebabkan kerusakan pada sistem reproduksi jantan sebagai salah satu indikator digunakannya genistein sebagai senyawa antifertilitas. Penelitian ini bertujuan untuk mengetahui pengaruh genistein terhadap sistem reproduksi mencit jantan (Mus musculus). Penelitian ini menggunakan pendekatan kuantitatif eksperimental, yang rancangannya mengikuti Rancangan Acak Kelompok. Variabel bebas adalah dosis genistein 0 mg/g, 0,0035 mg/g, 0,0042 mg/g, dan 0,0049 mg/g. Variabel terikatnya adalah sel-sel germinal dalam tubulus seminiferus testis, morfologi spermatozoa, viabilitas spermatozoa, dan kadar hormon testosteron. Data sel-sel germinal dalam tubulus seminiferus testis kadar hormon testosteron dianalisis menggunakan Analisis Varians Satu Jalan (One Way ANOVA) dengan tingkat signifikansi 5%. Selanjutnya dilakukan uji Post Hoc dengan Least Significant Difference (LSD) 5%. Data morfologi dan viabilitas spermatozoa dinyatakan dalam prosentase. Hasil penelitian menunjukkan bahwa ada pengaruh genistein pada sistem reproduksi mencit jantan (Mus musculus). Genistein yang diberikan dengan dosis 0,0035 mg/g; 0,0042 mg/g; dan 0,0049 mg/g berpengaruh terhadap jumlah sel germinal, morfologi dan viabilitas spermatozoa. Kadar hormon testosteron berbeda nyata pada dosis 0,0049 mg/g. Kata kunci : genistein, sistem reproduksi jantan

Abstract

Tumbuhan obat dapat dimanfaatkan dan dikembangkan sebagai jamu. Wilayah Desa Winong Kabupaten Madiun terletak di lereng pegunungan Wilis merupakan Desa dengan keragaman tumbuhan. Masyarakat belum mengerti dan memahami adanya keragaman dan potensi tumbuhan lokal. Tujuan kegiatan untuk meningkatkan pengertian dan pemahaman masyarakat mengenai potensi tumbuhan lokal untuk dikembangkan menjadi jamu. Metode pelaksanaan kegiatan melalui tahapan; 1) observasi, 2) penyuluhan, dan 3) demontrasi pembuatan jamu. Berdasarkan hasil angket diperoleh data 87% peserta antusias mengikuti kegiatan, 90% berminat menggunakan tumbuhan lokal sebagai obat atau jamu, 80% berniat menggunakan tumbuhan lokal sebagai obat atau jamu, 86% berminat menanam tumbuhan obat di pekarangan rumah, 90% materi sosialisasi dan demonstrasi telah disampaikan kepada masyarakat, 90% mengerti dan memahami materi, 100% puas mengikuti kegiatan, dan 100% kehadiran masyarakat mengikuti kegiatan. Berdasarkan data yang diperoleh, kegiatan sosialisasi pemanfaatan tumbuhan lokal sebagai jamu dapat ditindaklanjuti untuk pemberdayaan masyarakat dalam mengolah jamu sehingga dapat meningkatkan perekonomian.

Abstract

The use of local wisdom as learning resources has been widely known to be able to create meaningful learning. The purposes of this study were to 1) produce module based on freshwater fish anatomy as local wisdom in Ngebel Lake, Ponorogo and 2) find the quality of the module produced. The Research and Development (R&D) was designed based on the Thiagarajan model which consisted of defining, designing, developing, and disseminating stages. The data were collected using interview guidelines, validation sheets, and students’ response questionnaire. The validation results of the media, material, and language experts showed that the module was very valid with the percentages were 86.5%, 88.2%, and 85.3% respectively. In addition, the student responses reached 87.9% (very good). In conclusion, the fish anatomy learning module based on local wisdom is appropriate to be used as learning resource.

Abstract

Genitri (Elaeocarpus sphaericus Schum.) is an endemic plant of Indonesia that has not been widely cultivated and utilized. The use of genitri is usually as road shade, firewood, seeds for jewelry. The results showed that the diversity of chemical compounds genitri had potential for health. The aim of the study was to analyze the potential of genitri to leukocytes and lung tissue structure of wistar rats induced by influenza virus. The study used an experimental approach, with experimental animals in the form of 24 male wistar rats in 4 treatment groups, P0 negative control, P1 influenza virus induction (positive control), P2 influenza virus induction and given genitri leaf, and P3 influenza virus induction and given genitry seeds. Virus induction on day 0 and day 14. Leaves and seeds were given on the 3rd day. Surgery was performed on day 5 and day 33. The number of leukocytes were calculated using microhematocrit, and identified leukocytes and lung tissue structure staining with HE staining. The results showed that genitry seeds could increase the number of leukocytes up to 13,000 cells/µL, the size of the alveoli diameter was 31.48 m, the size of the alveolar septum was 12.02 m. Abstract

The research aims to identify the insects in rice plants in the district Pilangkenceng and Kare Madiun. Research method using qualitative descriptive and in accordance with what is on the field. Data were collected by observation. Insects captured in two ways: directly and using flay arrest of the net. The results showed that the insects found in rice plant in the district of Pilangkenceng order Orthoptera, Hemiptera, Odonata, Hymenoptera, Lepidoptera, Coleoptera and Diptera. Insects found in the District of Kare consists of the Order Orthoptera, Odonata, Lepidoptera, Coleoptera and Homoptera.

Abstract

Buluharjo Village, Magetan Regency is a rural area, the majority of which are horticultural crops. Shallots are one of the commodities in the region. The productivity of shallots for the last three years has decreased due to the reduction in agricultural land caused by the construction of housing projects. The decline in shallot production is caused by the lack of land and the presence of plant pests such as worms, caterpillars, fungi and bacteria. The use of high chemical fertilizers, improper selection of seeds makes the quantity and quality of shallot production not optimal. The purpose of the activity is to provide training on red onion cultivation with verticulture techniques as a solution for using narrow land. The method used is pre survey, survey, socialization and training. The activity was carried out for 6 months. The result of the activity is an increase in understanding of modern agricultural techniques, namely verticulture in the field of red onion farming. Public understanding of modern agricultural systems, and increasing the productivity of shallots. The community is very enthusiastic and actively involved in applying vertical culture techniques. The community can apply the shallot verticulture technique on narrow land.

Abstract

The objective of this study is to establish the effect of inoculum concentration and incubation time of protease activity on pigeon pea beans fermented by Aspergillus niger. The study comprises two independent variables: the inoculum concentration of Aspergillus niger, which consists of four levels (0 mL/g; 0.2 mL/g; 0.4 mL/g, and 0.6 mL/g), and six levels of incubation time (0 h, 24 h, 48 h, 72 h, 96 h, and 120 h). The obtained data were tested using two-way analysis of variance (ANOVA) followed by an LSD test with a significance level of 5%. The results show that the inoculum concentration of Aspergillus niger and incubation time do affect the activity of the protease enzyme. The highest enzyme activity of 0.298 U/ml was obtained at an inoculum concentration of Aspergillus niger of 0.4 mL/g, with an incubation time of 96 h (P2K4).

Abstract

Permasalahan dalam pembelajaran tematik integratif di sekolah dasar yaitu mata pelajaran masih diajarkan secara terpisah, tidak adanya media perantara yang mendukung proses pembelajaran karena hanya terpaku pada buku pegangan dan belum dikembangkan aktivitas bermain sambil belajar. Penelitian pengembangan ini bertujuan untuk menghasilkan media pembelajaran flashcard tematik berbasis permainan tradisional yang layak untuk pembelajaran tematik integratif kelas IV sub tema Lingkungan Tempat Tinggalku. Metode penelitian menggunakan model 4D (define, design, develop and disseminate). Media diujikan pada 20 siswa kelas IV SDN Gunungsari Madiun. Instrumen pengumpulan data yang digunakan dalam tahap eksplorasi yaitu observasi dan wawancara, sedangkan dalam tahap pengembangan berupa angket. Hasil penelitian menunjukkan bahwa: (1) hasil validasi dari dua validator ahli aspek media dan materi menunjukkan bahwa media sangat layak digunakan dalam pembelajaran, (2) hasil respon siswa skala kecil dan luas menunjukkan bahwa media sangat layak dengan persentase 100% dan (3) hasil belajar sebelum dan setelah diberi perlakuan menunjukkan perbedaan yang signifikan, uji hipotesis pembelajaran 2 diperoleh thitung = 8,376 > ttabel =2,093, dalam pembelajaran 5 diperoleh =7,264 > =2,093. Hasil penelitian menunjukkan bahwa media pembelajaran flashcardtematik berbasis permainan tradisional sangat layak digunakan untuk kelas IV sub tema Lingkungan Tempat Tinggalku.
