Abstract

Empirically, societies have utilized neem plant to treat worm infection, scabies, malaria, candidal infection, and allergy. Several researches have indicated that consuming neem leaves could improve antibody production. Our previous research had demonstrated the existence of immunomodulatory component in neem leaves weighted 70 and 100 kDA. The aimed of study was to test the activity immunomodulatory components to Candida albicans. This research was an in vitro study with rats as experimental subject. The rats were divided into three treatment groups and one control group, which were labeled as KO (no treatment), KP1 (inoculated by C. albicans only), KP2 (inoculated by C. albicans and were given the 70 kDA immunomodulatory component), KP3 (inoculated by C. albicans and were given the 100 kDA immunomodulatory component). In day 21, preparations from rats’ tongue were done with immunohistochemistry staining and were analyzed for the activity of TNF-α macrophage phagocytosis and also for calculation of C. albicans colony from tongue swab. The result showed that there was no significant difference between the 70 and 100 kDA immunomodulatory components in increasing activity of TNF-α and macrophage phagocytosis and in decreasing the number of C. albicans colony. It was assumed that those components might actívate NF-κB and AP-1 that play role in protein transcription, which stimulated gene expression like TNF-α, which played role in phagocytosis. In conclusión, 70 dan 100 kDA immunomodulatory components increased the activity of TNF-α and macrophage phagocytosis and also decrease the number of C. albicans colony.

Abstract

ABSTRACTIntroduction: Periodontitis is an inflammatory disease that occurs in tooth-supporting tissues with a high prevalence. Porphyromonas gingivalis is the primary bacteria that cause periodontitis. In the inflammatory process, mononuclear cell, such as macrophages and lymphocytes, infiltrate. Macrophages and lymphocytes play an essential role in the body’s defence against pathogens during inflammation. However, excessively activated macrophages and lymphocytes can cause tissue damage. One of the natural ingredients that can act as an anti-inflammatory is Robusta coffee. This study was aimed to analyse the effect of brewed Robusta coffee on the number of macrophage and lymphocyte cells in the chronic periodontitis rat model. Methods: This research was a laboratory experiment towards 20 rats, which were randomly divided into five groups: the control group (K); group of mice induced by Porphyromonas gingivalis (P5-A); group of mice induced by Porphyromonas gingivalis and administered with brewed robusta coffee (P5-B), euthanised on the 19th day 19; group of mice induced by Porphyromonas gingivalis (P7-A); group of mice induced by Porphyromonas gingivalis and administered with brewed robusta coffee (P7-B), euthanised on the 21st day. Brewed robusta coffee was administered for as much as 3.6 ml during 14 days. Tissue processing and HE staining were performed on the mandibular preparations—calculation of macrophages and lymphocytes in the area around the lower M1 buccal alveolar. The results were then analysed with one-way ANOVA. Results: There was a significant difference in the number of macrophages and lymphocytes between the control and the treatment group (p<0.05). However, there was no significant difference (p>0.05) found in the number of lymphocytes between all treatment groups (P5-A, P5-B, P7-A, and P7-B). Conclusion: Brewed Robusta coffee can reduce the number of macrophage and lymphocyte cells in the chronic periodontitis rat model. Keywords: Brewed Robusta coffee, macrophage, lymphocyte, periodontitis.

Abstract

Oxidative stress in diabetes mellitus triggers activation and increases in platelet counts that play a role in the formation of platelet plugs in the blood clotting process. If excessive blood clots occur, there is a risk of cardiovascular disease. Red seaweed extract contains carotenoids, phenols and their derivatives, fikobilin pigments, polysaccharide sulfates, and vitamins. The biggest phenol compound is flavonoids which are compounds that can produce platelet antia-gregation effects. The purpose of this study was to examine the effect of giving red seaweed extract to platelet counts in diabetic mice. The study group consisted of the treatment group of red seaweed extract, normal control, positive control and negative controls. The dosage of seaweed extract as much as 10 mg / 20 grBB is given in condition. The calculation of platelet count is carried out directly with the Brecher Cronkite method using a calculated room Improved Neubauer. The One Way ANOVA test results showed a significance value (p <0.05), that is, the group of diabetic mice given red seaweed extract had a lower platelet count of 280000 μl compared to diabetic mice given metformin as much as 379000 μl. The decrease is due to flavonoids, saponins, alkaloids, β carotene, vitamins and minerals able to stabilize the production of megakaryocytes and platelets, prevent the destruction of pancreatic β cells and prevent free radicals. It was concluded that red seaweed extract can reduce platelet counts of diabetic mice near normal values.
