Abstract

Antioksidan merupakan senyawa yang berguna mengatasi kerusakan oksidatif akibat radikal bebas dalam tubuh sehingga turut berperan mencegah berbagai macam penyakit. Penelitian ini bertujuan untuk mengetahui aktivitas antioksidan dari beberapa tumbuhan obat Indonesia. Sebanyak 37 ekstrak metanol tumbuhan obat diuji aktivitas antioksidannya secara in vitro menggunakan metode peredaman radikal bebas 2,2-diphenyl-1-picrylhydrazyl (DPPH) dan vitamin C sebagai kontrol positif. Hasil pengujian menunjukkan 29 ekstrak memiliki aktivitas diatas 50% pada konsentrasi 1000 ppm. 5 ekstrak dengan aktivitas tertinggi antara lain ekstrak kulit batang Sapium baccatum dan Leucosyke capitellata, ekstrak daun Ardisia crispa, Glochidion cauliflorum, dan Glochidion superbum.

Abstract

This study aim to examine the aphrodisiac activity of ethanol extract from the tail meat of porcupine (Hystrix javanica F. Cuvier, 1823) that is traditionally believed by people in Java (Indonesia) could enhance male vitality and sexual perfor-mance. Twenty sexually inexperienced male mice were randomly divided into four groups and paired with artificially estrus female mice after one hour of drug an extract administration. Two doses of ethanol extract; 150 and 750 mg/kg were administered (p.o) to male mice. Sildenafil citrate was used as the positive control while 5% Tween 80 solution used as the negative control. Sexual behavior parameters such as mounting and intromission were observed for three hours of mating. Male mice treated with ethanol extract of porcupine tail meat at the dose 750 mg/kg BW showed higher mounting and intromission frequency compared to the group of ethanol extract dose 150 mg/kg. After 2 hours of admin-istration showed the highest frequency compared to all groups. However, the ethanol extract could not reduce the mount-ing and intromission latency as low as sildenafil citrated treated group. Present findings provide preliminary evidence of aphrodisiac properties from the ethanol extract of porcupine tail meat.

Abstract

Indonesia memiliki keanekaragaman hayati yang tinggi baik hewan, tumbuhan, maupun mikroorganisme. Diaporthe sp. merupakan jamur endofit yang hidup pada tumbuhan gambir. Jamur ini dapat memproduksi metabolit sekunder yang bernama (+)-2,2’-Episitoskirin A. Senyawa tersebut memiliki kemampuan sebagai antimikrobia terutama terhadap Staphylococcus aureus. Staphylococcus aureus merupakan bakteri penyebab infeksi terbesar di dunia. Asia merupakan benua dengan kejadian methicillin-resistant S. aureus (MRSA) tertinggi. Penelitian ini bertujuan untuk mengetahui efek dari (+)-2,2’-Episitoskirin A terhadap hati mencit yang terinfeksi S. aureus. Penelitian ini menggunakan 30 ekor mencit jantan. Mencit dibagi menjadi enam kelompok perlakuan. Perlakuan pertama merupakan kontrol negatif (tanpa perlakuan), kelompok kedua sebagai kontrol positif (perlakuan S. aureus kelompok ketiga diinjeksi S. aureus dan dicekok (+)-2,2’-Episitoskirin A dosis 50 mg/kg BB (P1), kelompok keempat diinjeksi S. aureus dan dicekok (+)-2,2’-Episitoskirin A dosis 100 mg/kg BB (P2) dan kelompok kelima diinjeksi dengan S. aureus kemudian dicekok (+)-2,2’-Episitoskirin A dosis 200 mg/kg BB (P3). Hewan uji dinekropsi dan dikoleksi hatinya untuk histologi. Hasil pemeriksaan histopatologis memperlihatkan adanya peradangan pada hati semua perlakuan kecuali kontrol negatif. Dosis efektif (+)-2,2’-Episitoskirin A yang dapat mengurangi tingkat peradangan akibat infeksi S. aureus adalah 50 mg/kg BB.

Abstract

Fungal endophytes have been known as sources for bioactives with high chemical structure variability. This study aimed to screen extracts of some endophytic fungi associated with plants from Enggano Island as antioxidant and antibacterial using thin layer chromatography (TLC) bioautography method. Antibacterial ativity was performed against Escherichia coli and Staphylococcus aureus. Seventeen out of 22 extracts showed growth inhibition activity against both bacteria, while 16 extracts were active as DPPH free radical scavenging agents. Further determination on IC50 value of four prominent extracts revealed that fungal endophytes AK3018-1, FC-1, KC-4, and SO-3 have IC50 value of 85, 84, 704, and 347 ?g/ml, respectively in DPPH radical scavenging method. Extracts of fungal endophytes CR-3, CS-2, and SM-2 showed prominent antibacterial activity among other extracts, indicated by wide clear white zone around the spot. Further evaluation on minimum inhibitory concentration (MIC) of those three extracts by microdilution method showed that CR-3, CS-2, and SM-2 have MICs value of 512, >512, and 64µg/ml, respectively against S. aureus.Fungal endophytes AK3018-1, FC-1, KC-4, SO-3, CR-3, CS-2, and SM-2 were respectively isolated from Dioscorea bulbifera tuber, Fibraurea chloroleuca twig, Knema cinerea twig, Smilax odoratissima stem, Cryptocarya sp. twig, Calophyllum soulattri twig, and Smilaxmacrophylla stem.

Abstract

Ellianol 70% and water exlracls of Alpinia spp. i.e. Alpinia zemmbei. A. kaisumadai. A. malaccensis and A. officinarum were examined for their impact in in-vitro phagocytosis activity and capacity of mouse (Mus musculus) peritoneum macrophage induced by Staphylococcus epidermidis.The extract concentrations used in this experiment were 0: O.I: 1.0: 10: 100: and 1000 µg/ml,Imboost (Echinacea pwpurea extract) 1000 µg/ml was used as positive control while distilled water as negative control.The assay results showed that all of the extracts were active to promote phagocytosis activity and capacity of macrophage cells.The phagocytosis acitivity and capacity were increased by increasing extract concentration, and ethanol extract showed better activity than water extract. Alpinia officinarum and A. kaisumadai extracts reveal better phagocytosis activity and capacity than others. Activity and capacity of phagocytosis of each concentration was significantly (P<0.05) different each other as well as with negative control. There is significant difference among each extracts and positive control at 1000  µg/ml

Abstract

Pathogenic bacterial resistance to antibacterial agents used in the treatment of recent formal systems have undergone significant increase. It is necessary to get a source of new antibacterial agents to overcome resistance. Endophytic fungi have been known as one of the organisms that have the ability to produce a variety of bioactive metabolites. In this study, 26 endophytic fungi have been isolated from various parts of Cinnamomum burmannii plant. The results of antibacterial activity screening of endophytic fungal extracts byTLC bioautography method without elution showed that as many as 18 extracts inhibited the growth of Staphylococcus aureus InaCC-B4 with the diameter of inhibition zone (DIZ) were in the range of 5.00 to 16.00 mm. On the other hand, the number of extracts that could inhibit the growth of Escherichia coli InaCC-B5 were 24 extracts with the DIZ were in the range of 4.00 to 28.00 mm. A detailed analysis of eluted TLC-bioautography method showed the chemical compounds of extracts that were responsible for antibacterial activity.

Abstract

Latar Belakang Kelainan refraksi merupakan salah satu penyakit mata yang dapat menyebabkan kebutaan. Miopia merupakan salah satu bentuk kelainan refraksi. Data WHO tahun 2010 didapatkan prevalensi miopia di dunia sebesar 27% dan 2,8% untuk miopia tinggi. Faktor risiko terjadinya miopia adalah faktor keturunan dan faktor lingkungan. Diperlukan studi tentang faktor risiko dalam upaya pencegahan miopia.Tujuan Mengetahui dan menganalisis perbedaan prevalensi dan faktor-faktor risiko yang berhubungan dengan miopia miopia pada anak sekolah di perkotaan dan pinggiran kotaMetode Penelitian ini merupakan penelitian analitik observasional dengan pendekatan studi crossectional. Subjek penelitian adalah siswa SD kelas 4-6, dengan jumlah 59 di pinggiran kota dan 75 di perkotaan. Data dikumpulkan dengan cara wawancara menggunakan kuesioner. Uji statistik yang dilakukan adalah uji Chi-square.Hasil Prevalensi miopia di perkotaan 56%, sedangkan di pinggiran kota 28,8%. Didapatkan hasil yang signifikan antara jarak membaca buku (p=0,011), dan aktivitas di luar ruangan pada hari libur (p=0,002) dengan miopia di daerah perkotaan. Di daerah pinggiran kota didapatkan hasil yang tidak signifikan antara faktor-faktor yang diteliti dengan miopia. Didapatkan hasil yang signifikan antara letak geografis dengan miopia (p=0,002)Simpulan Prevalensi miopia lebih tinggi di perkotaan dibandingkan dengan piggiran kota. Faktor yang berhubungan dengan miopia di perkotaan adalah tingkat pendidikan orang tua, status ekonomi, jarak membaca buku, dan aktivitas di luar ruangan pada hari libur. Tidak terdapat hasil yang signifikan antara faktor-faktor dengan miopia di pinggiran kota. Terdapat hubungan antara letak geografis dengan miopia.

Abstract

Bisanthraquinones (+) - 2,2'-epicytoskyrin A and (+) -1,1'bislunatin produced by the endophytic fungus Diaporthe sp. GNBP-10 showed potent antibacterial activity on in-vitro test and have the opportunity to become new antibiotics candidates. The aspects of safety and toxicity of drug candidates have to be examined before applying to human. This study was conducted to determine the safety aspects of the compounds through acute oral toxicity testing in mice (Mus musculus). Acute toxicity of (+) - 2,2'-epicytoskyrin A and (+) - 1,1'-bislunatin evaluated by the method of Up and Down Procedure with limit test at a dose of 2000 mg / kg. Results of acute toxicity test showed that the LD50 of (+) - 2,2'-epicytoskyrin A and (+) - 1,1'-bislunatin were of 1638.87 mg / kg and > 2000 mg / kg respectively. Administration of (+)- 2,2'-epicytoskyrin A resulted in increased miliari multifocal hepatitis, fatty degeneration and necrosis of liver cells, and the renal tubule epithelial degeneration. Administration of (+) - 1,1'-bislunatin at a dose of 2000 mg / kg resulted in multifocal accumulation of inflammatory cells in the liver and degeneration of cells in the islets of Langerhans although not resulting in death. The administration of those compounds indicated the changes in the organs, but based on the UN/ECE classification of LD50 value showed that (+) 2,2'- epicytoskyrin A and (+) -1,1'-bislunatin included as low acute toxicity substance.

Abstract

Studies on the therapeutic use of insects and insect products have been neglected compared to the use of other animals or plants.This study aims to determine the antibacterial and antioxidant potential of ants extracts. Preliminary study related to antibacterial and antioxidant screeningof 17 extracts of ant colonies that belongs to 8 species were performed by Thin Layer Chromatography (TLC)-Bioautography. Antioxidant activity was measured by DPPH free radical scavenging method. The antibacterial activity was done against three pathogenic bacteria, i.e Bacillus subtilis InaCC B-1, Staphylococcus aureus InaCC B-4 and Escherichia coli InaCC B-5 were performed by non-eluted TLC-autobiography assay. The minimum inhibitory concentration (MIC) and IC50 values of DPPH radical scavenging activity of active extracts were determined by microdilution in 96-well microplate. The results showed 6 extracts active against B.subtilis, 5 extracts active against S.aureus. The lowest MIC value was 512 µg/ ml.  Ten extracts had the antioxidant activity with various IC50 values. The extracts of ants might be used as bioactive resources for antibacterial and antioxidant.

Abstract

Abstrak Penelitian ini di latarbelakangi oleh kemampuan bercerita anak yang masih didominasi dengan kegiatan membaca dan menulis membuat kemampuan bercerita kurang terasah. Oleh karena itu, untuk mengembangkan kemampuan bercerita diperlukan stimulus untuk melatih kemampuan bercerita anak dengan menggunakan mind mapping. Tujuan dari penelitian ini yaitu untuk membuktikan ada tidaknya pengaruh metode mind mapping terhadap kemampuan bercerita pada anak kelompok di TK Ar-Rahmah Surabaya. Penelitian ini menggunakan pendekatan penelitian kuantitatif dengan jenis penelitian Quasi Experimental Design dan design penelitian Nonequivalent Control Group Design. Subjek penelitian adalah kelompok B1 sebagai kelompok Eksperimen sebanyak 16 anak dan B2 sebagai kelompok kontrol sebanyak 16 anak. Teknik pengumpulan data antara lain dengan menggunakan observasi, tes, dan dokumentasi. Teknik analisis data menggunakan uji Mann Whitney U Test. Dari hasil perhitungan diperoleh Uhitung &gt; Utabel (64,5&lt;75), Berdasarkan analisis data tersebut dapat dinyatakan bahwa ada pengaruh menggunakan metode mind mapping secara signifikan terhadap kemampuan bercerita pada anak kelompok B TK Ar-Rahmah Surabaya. Therefore, to develop the ability to tell the necessary stimulus to train the child's ability to tell by using mind mapping. The purpose of this study is to prove the existence of mind mapping methods influence on the ability to tell a group of children in kindergarten Ar-Rahmah Surabaya. This study uses a quantitative research approach to the type of research Quasi-Experimental Design and design research Nonequivalent Control Group Design. The subjects were a group B1 as many as 16 children experiment group and the control group B2 as many as 16 children. Data collection techniques such as by using observation, testing, and documentation. Data were analyzed using the Mann Whitney U Test. From the calculation Uhitung&gt; Utabel (64.5 &lt;75), Based on the analysis of these data it can be stated that there is the effect of using mind mapping methods significantly affect the ability to tell the children in group B TK Ar-Rahmah Surabaya.
