Abstract

Background Neoplastic lesion in colorectal could be adenoma and colorectal carcinoma (CRC). An invasion in muscularis mucosa is a criteria for colorectal carcinoma. Diagnosis is formed with histopatologic interpretation. In colorectal-biopsy preparation interpretation, determining limited invasion is sometimes difficult so that it needs other diagnostic tools. The aim of this study was to evaluating the difference of Ki-67 mean expression in colorectal adenoma and CRC. Methods This study was cross-sectional study on colorectal adenoma and CRC. Cases were diagnosed by colonoscopy-biopsy between 2011 and 2012. It did re-interpretation of HE preparation of adenoma or colorectal carcinoma diagnosis and determination of the degree of differentiation of those adenoma and colorectal carcinoma. Immunohistochemical Ki-67 staining was interpreted with light microscope. The percentage of the stained tumour cells was counted. The difference of mean in Ki-67 expression of colorectal adenoma and CRC was analyzed with t-test with significance level at p

Abstract

Background Cyclooxygenase-2 (COX-2) overexpression clearly plays an important role in the pathogenesis of breast cancer. The relationship between COX-2 expresion with histological grade and estrogen receptor (ER) in invasive carcinoma NST of the breast remains uncertain. The aim of this study was to evaluate the correlation between expression of COX-2 and ER with histology grade of invasive carcinoma NST of the breast. Methods This is observasional analitik cross-sectional study, the histology grade was assessed on by hematoxillin eosin, COX-2 and ER expression was assessed by immunohistochemistry (IHC) staining. The correlation between COX-2 and ER expression with histological grade were analysed by Pearson Chi-Square. Results 60 samples were studied, the youngest case was 28 years old and the oldest case was 79 years old. According to histological grade we found the case of low grade were 27 cases (45%) and high grade were 33 cases (55%). COX-2 expression positif was found in 53.4% and was correlated to histological grade (p=0.045), however between ER expression and histological grade, and COX-2 expression and ER expressionere not correlated. Conclusion COX-2 expression has correlation with histological grade, and COX-2 expression was not correlated with ER expression, and ER expression was not correlated with histological grade invasive carcinoma NST of breast. Key words : COX-2,ER, histologic grade, invasive carcinoma NST of Breast.

Abstract

Primary oral malignant melanoma is a very rare case (0.2-8%), and because of it’s hidden location and asymptomatic, it was often found at a late stage. This kind of case was never been reported in the past 10 years in Denpasar. Malignant melanoma, especially the pigmented variant, was easily diagnosed from histopathology specimen. But many variations occurred, and could be mistaken with nevus (nevoid melanoma) or mesenchymal spindle cell tumour, so immunohistochemistry examination is needed to confirmed it. In certain cases, cell block from FNAB aspirate material could help the patient to spent a lower cost to get a diagnostic material, considering the possibility of using immunohistochemistry procedure from cell block material. The case that we reported here was clinically, Radiologic, FNAB, histopathologically and immunohistochemistry for HMB45 and S100 from cell block was confirmed the primary malignant melanoma of right gingival mandible with a late stage. Key words : cell block, FNAB, histopathology, immunohistochemistry, malignant melanoma

Abstract

Background Cervical cancer is the third most common type and the fourth most deadly cancerian women which remains a significant health problem worldwide especially in developing coumtry. Ki-67 is a prototype protein in nuclear which influence cell proliferation rate, hence it can be used as one of parameter to evaluate progressivity of tumor cells in carcinoma cases as the cervical squamous cell carcinoma (SCC). This study aims to determine the relationship between clinical staging of cervical SCC and histopatological grading of the cervical SCC with the expression of Ki-67. Methods This was an observational study with a cross sectional design. Sixty cervical squamousa cell carcinoma archive histopathological slide samples (45 biopsy specimens and 15 radical hysterectomy tissues) were collected for this study from the Anatomical Pathology Department of RSUP dr. Mohammad Hoesin Palembang (January 2008 to July 2013). The samples were stained with Ki-67 antibody and analyzed with respect to patient’s clinical stage and histopatological grade. Results The positivity of Ki-67 expressions were more common in the age of ≤ 50 year-old group, in the non-keratinizing type, in the late clinical staging (stage IIIB) and in the moderatelly differentiated histopathological grade. High scores of the Ki-67 expressions (Ki-67>20%) was predominantly found in the late clinical stage and moderatelly differentiated histopathological grade cases. Statistically, there was a significant relationship (p=0.034) with a strong correlation (r=0.714) between the Ki-67 ecpression and the clinical stage of cervical SCC, but not histopathological grade. Conclusion The higher clinical staging of cervical SCC, the higher expression of Ki-67. Key words: cervical squamous cell carcinoma, clinical staging, histophatological grade, Ki-67.

Abstract

Background Cervical cancer is the second most common cancer in female in the world. In Bali, in 2008, cervical cancer is the second most common malignancy after breast cancer. This cancer has known to develop from precancerous lesions, cervical intraepithelial neoplasia (CIN).There is difficulty to determine grade of dysplasia of the uterine cervix and to predict wheather the lesion has potency of progression. The expression of P16INK4a has correlation with degree of cervical dysplasia and carcinoma. The aim of this study is to evaluate expression of P16INK4a in squamous cell carcinoma (SCC) of cervix uteri and CIN1, CIN2, CIN3. Methods A cross sectional analytic study was performed on 60 samples that divided into 10 CIN1, 10 CIN2, 10 CIN3, and 30 SCC. Samples were tooken from block paraffin archive from Private Laboratory in Denpasar and Anatomical Pathology Laboratory Sanglah Hospital, during January 1st 2011 until Juny 30th 2013. Immunostaining was performed to determine the expression score of P16INK4a. Expression of P16INK4a was determined by for scoring system base on staining grade and intensity. The result was analyzed by Kruskal Wallis test with level of significance α at p

Abstract

Background β-catenin is an important protein in cellular adhesion and signal transduction. In unstimulated condition, β-catenin only appears on the cellular membrane. Altered expression of β-catenin has been associated with progressiveness and metastatic process of malignancy in human. The aim of this study was to evaluate the expression of β-catenin on oral squamous cell carcinoma (OSCC) and also to assess its different expression in low grade and high grade lesions based on Bryne grading system. Methods This study was conducted on 2 groups of OSCC which included 20 cases of low grade and 20 cases of high grade. Immunohistochemistry staining of β-catenin was used to identify the difference of its expression in cell membrane, cytoplasm, and nuclei on invasive tumor front. Results The expression of β-catenin on cell membrane, cytoplasm, and nuclei showed significant difference between low and high grade OSCC (p=0.000; p=0.005; and p=0.035, respectively). There has not been any significant association between β-catenin expression with age, sex, and tumor location. Conclusion Expression of β-catenin could showed morphological change the OSCC. Key words : β-catenin, Bryne grading, oral squamous cell carcinoma (OSCC).
