Abstract

Dendeng is a traditional food results by the public Rote Ndao. Method for making dendeng by drying using sunlight as well as using palm sugar as the main ingredient pickling as well as the addition of spices. This study aims to determine the effect of the use of palm sugar in the manufacture of Dendeng Ikan. The processing is done in the District Lobalain(Rote Ndao). Methods of use of the sugar palm consisting of 4 treatments, KO (without giving sugar palm), K1 (10% concentration of palm sugar), K2 (20% concentration of palm sugar), K3 (30% concentration of palm sugar), K4 (40% concentration palm sugar). 4 times the storage of dendeng with the observation that the 5th day (P1), 10th day (P2), 15th day (P3), 20th day (P4). The quality of dendeng observed included presentations lost water content after processing, testing organoleptic and microbiological quality in terms of Total Plate Count (TPC). The test results of water content during storage of dendeng showed that, giving palm sugar concentration of 40% in the processing of fish dendeng, has a water content lower than the palm sugar concentration of 10%, 20% and 30%. During storage, less than the 15th day dendeng with palm sugar concentration giving 40% meet the Standar Nasional Indonesia moisture content, while the dendeng with the provision of palm sugar concentration of 30% has less storage than day-to-5 that meets the water content of the Standar Nasional Indonesia. The test results showed that the organoleptic, Giving palm sugar concentration levels in the processing of fish dendeng affect the appearance, consistency / texture, aroma and taste of dendeng. During storage in certain storage time of each treatment dendeng change the appearance, consistency / texture, aroma and taste. The test results TPC (Total Plate Count) of dendeng showed that, giving the concentration of sugar palm sugar palm 40% in the processing of dendeng, has a value of TPC (Total Plate Count) is lower than the concentration of sugar palm 10%, 20%, and 30 %. During storage, only on day 5 dendeng with palm sugar concentration giving 40% to abide by the TPC (Total Plate Count) of Standar Nasional Indonesia.

Abstract

Fish as a source of nutrients for the content of high value and high protein, omega 3, minerals and vitamins A and D for humans as well as human resources for the economy. Various species of tuna can be found in almost all waters of NTT, as a migration destination of the various species of tuna. Various species of marine fish known as intermediate host of different species of parasites, including nematodes Anisakis Anisakiasis which is the causative agent in humans. Anisakiasis can occur in humans after consumption of raw or undercooked fish infected by Anisakis sp, particularly in East Nusa Tenggara, the information about the fish Anisakis infection has not been reported and not well documented. Because it was in doing this research, conducted by collecting larvae of nematodes of the two species of tuna (Katsuwonus pelamis) tuna (Auxis thazard) from the sale of fish Pasir Panjang Kota Kupang. Organs are examined, namely the abdominal cavity, internal organs and muscle tissue. The larvae of nematodes be identified morphologically showed Anisakis sp, then separated for the calculation of Anisakis larvae. Parasites in fiksasi in 70% alcohol, followed by coloring Carmine, and the calculation of Anisakis larvae. The morphology, Anisakis sp. can be distinguished by other parasites by their ventriculus and mukron are clearly visible on Anisakis sp. under a stereo microscope. Anisakis sp . Found from the swordfish and fish skipjack all of them including Anisakis type i characterized by the presence of boring tooth on the tip of the anterior and mukron the percentage. Where as prevalence of the parasite Anisakis sp. on skipjack (Katsuwonus pelamis) that of 50 samples that were observed, as many as 8 samples were infected with Anisakis sp. with a prevalence of 16% value. While the tuna (Auxis thazard) of the 50 samples was observed, which infected 10 samples with grades prevalence of 20%.

Abstract

Salmonellosis is one of foodborne disease. Milkfish is very susceptible to contamination of Salmonella Sp bacteria. Which can be transmitted to humans. Microbiological testing is one of the indicators on the examination of contamination. The main factor causing the contamination could be the presence of pathogenic bacteria found in foodstuffs. The purpose of this study was to detect contamination of Salmonella Sp. In milkfish (Chanos chanos) in Sidoarjo Fish Market using checking indicator using media for SSA and TSIA. The results showed that there were 20 samples from a total of 42 milkfish taken from the fish market was positive for Salmonella sp bacteria. This positive detection indicates the height of its contamination that can lead to the transmission of disease in humans.

Abstract

A study to identification the microscopic structure of broiler meat by histological methods, has been carried out. The meat broiler samples were collected from musculus iliotibialis cranialis has been taken from Kupang supermarket. The tissue samples were fixed, dehydrated and embedded in paraffin and 5 μm sections. Harris-Haematoxilin-Eosin staining method, using to identified of histological structure. Microscopic analysis was performed using a binocular light microscope (100X). The study showed that, histological structure of broiler meat was composed by longitudinally and transverselly skeletal muscle. The nuclei in the periphery of the cell, there are dense connective tissue, fat, with small blood vessels. The skeletal myofibers diameter of muscle is 6,1 ± 0,8 μm. We observed for the presence of endomysium, perimysium and epimysium.

Abstract

Penyakit Mulut dan Kuku (PMK) adalah salah satu penyakit penting yang menginfeksi hewan sapi, kambing, domba dan babi serta beberapa jenis hewan liar. Penyakit ini penting secara ekonomi karena selain mengakibatkan angka mortalitas yang tinggi pada hewan muda, penurunan produksi susu maupun bahan asal hewan lainnya serta dapat mengakibatkan pembatasan perdagangan internasional bagi negara yang terinfeksi PMK. Selain dampak langsung dari penurunan produksi peternkan dan pembatasan perdagangan internasional, wabah PMK juga memberikan dampak yang serius bagi aspek sosial ekonomi dan industri pariwisata. Sampai saat ini, penyakit ini menyebar luas di Amerika selatan, Asia dan Africa. Mengingat arti pentingnya penyakit ini dan dampaknya secara global, maka penting untuk menyusun langkah strategis pencegahan dan eradikasi penyakit ini. Tulisan ini akan membahas beberapa langkah strategis penting yang dapat dimplementasikan dalam program eradikasi PMK khususnya melalui kegiatan-kegiatan yang berbasis teknologi molecular mulai dari penyiapan vaksin, tes diagnostic sampai kegiatan monitoring status penyakit.

Abstract

The aimed of this research was to know the incidence of carrion chicken sales in Surabaya traditional markets. The samples used in this study were chicken meats that had physical characteristics resembling carrion chicken. Chicken meat samples were taken from 20 traditional markets in Surabaya consisting of 5 traditional markets in North Surabaya, 5 traditional markets in East Surabaya, 5 traditional markets in south Surabaya, and 5 traditional markets in West Surabaya. Each traditional market had taken 10 samples of chicken meat each weighing 100 grams coming from different seller. The methode of carrion chicken test that used in this reseach was Malachyte Green Test by Warris method. The results showed that there were no positive sample of carrion chicken in North Surabaya, 9 positive samples of carrion chicken in West Surabaya, 24 positive samples of carrion chicken in South Surabaya, and 14 positive samples of carrion chicken in East Surabaya. The incidence of carrion chicken sales in Surabaya traditional markets showed that there were 47 positive carrion chickens or 23.5% of 200 samples taken.

Abstract

The aim of this study was to investigate the effect of Siwalan Palm Juice as a natural extender for maintaining boar sperm quality. Semen ejaculates collected from three Duroc and Landrace boars were macroscopically (volume, colour, odor, pH and consistency) and microscopically (motility, concentration and abnormality) evaluated. Semen samples fulfilled all criteria for semen characteristic (motility > 70%, concentration 200 x 106 cells /ml and abnormality ≤ 20%). Antibiotics were added in this semen samples. This study used completely randomized design with 6 treated groups, each group repeated three times. K0 (control): fresh semen; K1: Siwalan juice + Landrace boar semen; K2: Coconut water + Landrace boar semen; K3: NaCL + Landrace boar semen; K4: Siwalan juice + Duroc boar semen; K5: Coconut water + Duroc boar semen; K6: NaCL + Duroc boar semen. This semen samples were stored under room temperature at 220C. The result shows that Duroc and Landrace boar spermatozoa can be stored in Siwalan Palm Juice extender for up to 24 hours. It can concluded that Siwalan Juice could be used as one of natural extender for maintaining boar sperm quality under room temperature at 220C.

Abstract

Sopi is a traditional drink from East Nusa Tenggara, made through a fermentation process that is derived from palm trees (Borassus flabellifer L.) which also has the potential to be used as an antimicrobial agent. The purpose of this study was to identify the antimicrobial activity of Sopi against pathogenic bacteria. Identification of the antimicrobial activity conducted in two stages, testing the alcohol content and acidity from sopi; and bioassay testing using pathogenic bacteria Salmonella Typhimurium and Salmonella Enteritidis. The results showed that the Sopi has antimicrobial activity against pathogenic bacteria Salmonella Typhimurium and Salmonella Enteritidis. Results of this research may be basic research to the development of Sopi as an antimicrobial agent.

Abstract

The purpose of this study was to determine the genetic variation of P.multocida in Yogyakarta with other overseas P. multocida strains. Samples were obtained from necropted intensive pig farming with clinically pneumonia symptoms, specific pulmonary lessions and then followed by histopathological examination and b iochemical characterization of the isolates. Then followed by histopathological examination and biochemical characterization of the isolates. Moleculartest performed by DNA extraction using the QIAGEN QIAamp DNA minikit, amplification of the 16S rRNA gene using forward primer 5' GGA GTG AAC TGC AGC TAA TAC C 3', and reverse primer 5' GTA GGT AAG CTT CGC GTT GTT G 3', electrophoresis, purification and sequencing. Macroscopic and histopathological examination results were analyzed descriptively. Sequencing r esults were analyzed by multiple alignment with other Pasteurella spp. taken from GenBank using the Clustal W software, subsequently analyzed using Neighbour Joining and Maximum Parsimony method that exist in program MEGA version 5.1 The results showed gen etic distance based on 765 nucleotides of 16S rRNA gene of P. multocida isolates from lungs of bronchopneumonia of swine in Yogyakarta at 0%. Filogram based on the nucleotide sequence showed a high similarity between P. multocida isolates from Yogyakarta a nd other isolates from USA, Germany, China, Europe, and Hungaria.

Abstract

Diabetes mellitus (DM) is a metabolic disorder which damage the beta cells (β) which led to the destruction of langerhans of pancreas gland. Diabetes mellitus is characterized by increased blood glucose levels and progressive changes to the structure of the histopathology of the pancreas. The purpose of this study was to determine the effect of ethanol extract of sambiloto leaf ( Andrographis paniculata Nees) on pancreatic histopathology of Alloxan induced diabetic mice. This research is an experimental research design by Post Test Randomized Control Group Design Experim ental animals used were mice ( Mus musculus) 12 sample of mice ±2 weeks old were adapted for 5 days. Mice were divided into 4 groups, namely, negative control, positive contro l (3 days of alloxan injection) and diabetic mice treated with the ethanol extract of sambiloto leaf with 2 different doses (doses I =2,2mg/kg and dosesII=4,4mg/kg) for 14 days. On day15, mice were necropsy and pancreas were collected for further Hematoxiline Eosin (HE) coloration. The results showed that DM alloxan induced type 1 mice (Mus musclus), the pancreas characterized by the occurrence of necrosis, langerhans islet was not bounded clear and breakthe linkage of the langerhans islet with a cinar while on ethanol extract therapy of sambiloto leaf dose 2,2 mg/kg and 4,4 mg/kg in mice DM type I showed the results an improvement on the langerhans islet. Based on these results, it can be concluded that a dose of 2,2 mg/kg and 4,4 mg/kg is effective dose to repair the structure of langerhans islet of mice Type I DM, but it is more adv isable to us a therapeutic dose of 4,4 mg/kg because it changes the structure of Langerhans islet.
