Abstract

The aims of this research are to find out the effectiveness of water partition Momordicia charantia fruit to the decrease blood glucose of experimental diabetic male rats (Rattus norvegicus). This research used twenty 3 months old male rats which were adapted for 2 weeks. Rat grouped into 4 groups namely the negative control (P0), only given food and drink alone; positive controls (P1), was given only feed, drink, and streptozotocin; EP group (P2), was given food, drink, streptozotocin, and extra bitter melon 2%; water partition group was given feed, drinking water and partitions streptozotocin 50 mg / kg body weight. Streptozotocin giving conducted on days 0, giving extract bitter melon and water partition conducted from day 3 to day 20. Measurement of glucose conducted on days -1, 0, 4, 11 and 18. The obtained data were evaluated using one-way ANOVA test. The results showed that water partition Momordicia charantia fruit could decrease blood glucose levels in a dose of 50 mg / kg of body weight significantly compared with the positive control.

Abstract

Bay leaf (Syzygium polyanthum) is often used in a variety of dishes to enhance the flavor, without realizing that the leaves contain antimicrobial compounds are bactericidal. The purpose of this study was to determine the effect of immersion bay leaves infuse to the quality and durability of beef bali at room temperature laying in terms of odor, color, texture, pH, water content and water holding capacity. Test colors using a color standard ISO beef concluded by the ten panelists, test smells and textures using ten panelists, test the pH was measured using a pH meter, test the water content carried by the drying method using the oven, the next test of water holding capacity using methods Hamm. The results showed that soaking treatment bay leaf infusion with concentration (0%, 5%, 10% and 15%) was highly significant to the smell, color, and texture of beef bali, old laying on the temperature the room was highly significant on pH and water content of beef bali but did not significantly affect water holding capacity. Can be concluded that the quality and endurance of bali beef can maintained by marinated into bay leaves infuse.

Abstract

This study aims was to investigate the improvement of hepar histopathology experimental diabetic rats given moringa oleifera leaves ethanolic extract. 24 male white rats induced by a single dose STZ of 45mg/kg intraperitoneal to cause diabetes mellitus. After declared with diabetes disease, rats were divided into 6 groups consist of 1 a control group and a treatment group of 5 who were given ethanol extracts of leaves of the kelor, with a dose of 100, 200, 300, 400, and 500mg/kg, for 5 weeks. Rats then sacrificed and the hepar were taken for the histopathology preparations with Hematoxylin-Eosin staining (HE). Each preparation histopathology examined under a microscope in five visual fields at 400x magnification. The processed data is then analyzed statistically by test Non parametric Kruskal Wallis followed by Mann-Whitney test, when there is a significant difference (P<0.05). The results showed that the hepar has improved a result of the damage diabetes mellitus after given Moringa oleifera leaves ethanolic extract with a dose of 400 mg/kg.

Abstract

Penelitian bertujuan untuk mengetahui perbandingan persentase agranulosit terhadap umur bibit sapi bali betina yang dipelihara di wilayah Nusa Penida, dengan tiga rentangan umur yang berbeda yakni pedet, dara, dan dewasa. Pengambilan sampel darah dilakukan melalui vena jugularis menggunakan venoject dan langsung dilakukan pembuatan apusan darah di tempat. Preparat kemudian diamati dan dihitung di bawah mikroskop dengan pembesaran 1000 kali menggunakan metode cross sectional, jumlah limfosit dan monosit diamati serta dihitung per 100 sel leukosit. Data yang didapatkan selanjutnya dianalisis menggunakan uji sidik ragam one way anova. Hasil yang didapatkan menunjukkan adanya perbedaan persentase limfosit yang sangat nyata pada tingkatan umur pedet, dara dan dewasa. Sedangkan monosit tidak terdapat perbedaan yang nyata. Berdasarkan hasil penelitian diperoleh bahwa persentase limfosit pada umur dewasa lebih tinggi dibandingkan dengan umur pedet dan dara tetapi masih berada dalam rentangan nilai normal limfosit. Sedangkan persentase monosit memiliki nilai yang sama pada umur dewasa, dara dan pedet.

Abstract

Dexamethasone is a corticosteroid drug from the class of glucocorticosteroids that has anti-inflammation and immunosuppressant effect. This drug has therapeutic effect such as immunosuppressive and reducing the inflammatory response. Spleen is an organ of the immune system, so that when used dexamethasone in the long term and large doses can affect the spleen. To prevent dexamethason effect on spleen, provision of antioxidants is required. Vitamin E is a fat soluble vitamin that acted as antioxidant. This study used complete randomized design. Sample of this experiment were 25 male rats divided randomly into 5 groups. Negative control were given standards food and drink. Dexamethasone administrated by subcutaneous injection 0,13 mg/kg BW in positive control group, P1, P2 and P3. Vitamin E administrated orally with following P1 (100 mg/kg BW), P2 (150 mg/kg BW) and P3 (200 mg/kg BW). After 2 weeks intervention, all samples were terminated, then took the spleen to make histopatology preparation. The variables examined include hemorrhage and necrosis. From the results showed a positive control necrosis, whereas P1, P2, P3 improvements in spleen damage, but was not significant. P2 treatment showed the best results in reducing the effects of dexamethasone. From this study it can be concluded that the dexamethasone dose 0,13 mg/kg BW has histopatology of spleen changes and vitamin E dose 150 mg/kg BW can repair spleen tissue damage is better than vitamin E dose 100 mg/kg BW and 200 mg/kg BW.

Abstract

Ulcerative dermatitis is a disease that most commonly affects turtles and/or hatchlings that are kept in ponds, including in Turtle Conservation and Education Center (TCEC), Serangan. A study was conducted to determine the prevalence, pathology, and lesion location of ulcerative dermatitis in olive ridley hatchlings reared in TCEC Serangan, also the differences of the body condition index of olive ridley hatchlings with ulcerative dermatitis and healthy ones. Prevalence was determined by counting sick hatchlings compared with the total number of hatchlings at TCEC. Morphometry (straight carapace length, straight carapace width, curved carapace length, curved carapace width) and body weights were measured to determine the body condition index, and then body index differences were compared with unpaired T-Test. Skin tissue samples were processed into pathology slides and routinely staining of hematoxylin eosin (HE). The prevalence of ulcerative dermatitis in olive ridley hatchlings at TCEC was 16.2%. Crusty-yellow wound lesions of 2 mm to 2 cm in diameter were found and microscopically there were heterophils and mononuclear cells infiltration in skin dermis accompanied by erosion, parakeratosis, and necrotic material containing cell debris. Lesions were found mostly on neck area (63.04%), followed by flippers, head, skin near carapace, eyelids, and neck area with flippers combined. There was a very significant difference in body condition index between suffered hatchlings with healthy ones, where the average body condition index of suffered hatchlings were greater than healthy ones. Rearing management should be fixed.

Abstract

The aim of this study is to determine the differential granulocytes performed on neutrophils, eosinophils, and basophils in bali cattle which are raised in the highlands and lowlands in Nusa Penida. Blood samples were taken from 60 female bali cattle. Bali cattle blood sample was done randomly using multi-stage sampling. Blood collection was done through a jugular vein using a venoject. Blood smear was done using slide technique immediately, followed by fixation and sample staining using Giemsa. Differential leucocytes were counted using battlement method until 100 cells was found using 1000x magnification. Completely randomized design (CRD) was used for data analysis. The obtained data were analyzed with Independent Samples T-Test. The results showed that a type of maintenance location (highlands and lowlands) had a significant effect on neutrophil, but has not significant effect to eosinophils and basophils. The percentage of bali cattle neutrophils in the highlands is lower than in the lowlands.

Abstract

Research has been conducted to know the number bacterial isolates of Klebsiella sp. from bali cattle in terms of the level of maturity and the breeding location, also its sensitivity to the antibiotic there are ampicillin, sulfamethoxazole, and oxytetracycline. 120 bali cattle's nasal swabs consisting of calves, heifers, and adult cattle which are breeding on highland and lowland has been used as the isolates in this research. The obtained nasal swab was isolated and identified, then proceed with the sensitivity test. Bacterial of Klebsiella sp. has been found in calves were nine isolates (45%), heifers were six isolates (30%), and adult cattle were five isolates (25%). While based on the breeding location in the lowland found 13 isolates (65%) and highland found seven isolates (35%). In the sensitivity test results were obtained for the antibiotic ampicillin all isolates were sensitive (100%). sulfamethoxazole resistant four isolates (20%), intermediate two isolates (10%), and sensitive 14 isolates (70%). Oxytetracycline resistant seven isolates (35%), intermediate two isolates (10%), and sensitive 11 isolates (55%). The results showed that differences in the level of maturity and breeding location of bali cattle can affect the distribution of bacteria Klebsiella sp.

Abstract

Bali cattle eats organic and inorganic waste in Suwung Denpasar landfill as source of feed. The waste was suspected contaminated by heavy metals Pb. The aim of this study is to determine levels of heavy metals Pb and spleen histopathologic changes of bali cattle reared in Suwung Denpasar landfill. Spleen samples were collected from 5 cattle for examination of heavy metals Pb by Atomic Absorption Spectrofotometri method and histopathology preparations made by Hematoxilin-Eosin staining. The results showed the presence of heavy metal Pb contamination which varied of 1.5024 ppm, 1.5002 ppm, 2.0267 ppm, 2.0268 ppm and 0.6818 ppm. Microscopically, showed histopathological changes the form of depletion and proliferation of lymphoid cells.
