Abstract

Study on isolation of catechin from gambier (Uncaria gambier (Hunter). Roxb) for pharmaceutical and cosmetic uses has been conducted.Raw materials of gambier were collected from three different areas including Andalas University Medicinal Plant Garden, Lima Puluh Kotaand Siguntur Pesisir Selatan, West Sumatera. Catechin was isolated from gambier using two methods, namely purification and pre nonpurification. Several evalutions on quality of the obtained catechin was done including descriptions of the catechins, solubility, colorreactions, thin layer chromatography, yield, drying shrinkage, melting point and maximum uptake levels of catechins. The best results forpharmaceutical and cosmetic uses are gambier originated from Siguntur with pre purification method which obtained 56.3% yield and 96.17%levels of catechins.

Abstract

Telah dilakukan penelitian mengenai pengaruh cara pengeringan terhadap perolehan kadar ekstraktif, senyawa fenolat dan aktivitas antioksidan dalam daun dewa (Gynura pseudochina (L.) DC.). Cara-cara pengeringan yang diuji adalah: pengeringan angin pada suhu kamar, pengeringan oven pada suhu 40 oC, pengeringan oven pada suhu 60 oC, pengeringan oven microwave dan sampel segar sebagai kontrol.  Hasil penelitian menunjukkan bahwa pengeringan tumbuhan segar menyebabkan berkurangnya perolehan ekstraktif, kadar senyawa fenolat dan aktivitas antioksidan. Cara-cara pengeringan memberikan pengaruh yang berbeda nyata (P < 0,05) terhadap perolehan ekstraktif, kadar senyawa fenolat dan aktivitas antioksidan. Di antara cara-cara pengeringan yang diuji, perolehan ekstraktif tertinggi diberikan oleh pengeringan oven microwave, sedangkan kadar senyawa fenolat tertinggi dan aktivitas antioksidan yang tertinggi diberikan oleh pengeringan angin pada suhu ± 25  oC.

Abstract

Indonesia is one of the largest tea producer and exporter in the world. However, the European Union has issued Commission Regulation (EU) No.1146/2014 by setting the MRL (maximum residue limit) of anthraquinone in tea at 20 μg/kg, significantly limiting the amount of tea export from Indonesia to the European Union. Viewed from the side of the danger, especially for synthetic anthraquinone that is potential effect of human health then performed an analysis to determine the presence of anthraquinone in Indonesian tea. To find out anthraquinone concentrations in various tea products, anthraquinone was developed and validated analysis method using LC-MS/MS. anthraquinone was extracted from tea sample using dichlormethane with a soxhlet. Black tea and green tea samples were obtained from one of the tea plantation companies.The validated method was used for the determination of anthraquinone residues in black tea and green tea samples. Anthraquinone residues was detect in all of the test samples.

Abstract

Optimization studies out on isolation of catechin from gambier and pasta for the pharmaceutical and marker compounds have been carried. Gambier gambier and paste were obtained from the Medicinal Plant Garden Andalas University, Siguntur and Lima Puluh Kota. Isolation method used was non-purification method, pre-purification for gambier and fractination for pasta. The analysis performed included catechin solubility, melting point, maximum absorption, thin-layer chromatography, loss on drying, ash content, yield and determination of levels of catechin. Product which has the highest levels of catechins, the determination was continued by analysis of UV spectra and NMR. The best results for pharmaceuticals derived from Siguntur with pre-purification methods which resulted in yield 56.3% with 96.17% catechin content, whereas for a marker compound was obtained from gambier Siguntur pasta with fractionation method which yielded 12.13% with 97.96% of catechin level.

Abstract

dentification of antioxidative compounds in God leaves [Gynura pseudochina (Lour.) DC] has been performed. The dried leaves were extracted by reflux consecutively with hexane, chloroform and methanol. Each of extract was concentrated using rotary vacuum evaporator. DPPH method was used to examine the antioxidant activity of the extract. The methanolic extract exhibited the strongest antioxidative activity. Then the methanolic extract was isolated by preparative paper chromatography method. The isolated compounds were screened for their antioxidative activity by DPPH method and characterized by chemical and spectroscopic methods. Results indicated that the compound 3’,4’,5,7-tetrahydroxy flavonol (quercetin) was the most active as antioxidant and can be used as marker in standardization of God leaves or its extract.

Abstract

Telah dilakukan pembuatan kokristal katekin-nikotinamida dan evaluasi kelarutannya dalam pelarut air. Kokristal dibuat dengan dua metode, yaitu rekristalisasi dengan penguapan pelarut etanol 96% menggunakan rotary evaporator (metode I) dan penguapan pelarut pada suhu ruang (metode II). Karakterisasi kokristal katekin-nikotinamida dilakukan dengan difraksi sinar-X, analisis termal DTA (Differential Thermal Analysis), FT IR (Fourier Transform Infra Red), dan SEM (Scanning Electron Microscopy), serta uji kelarutan dalam pelarut air menggunakan orbital shaker selama 24 jam. Penetapan kadar katekin dilakukan dengan metode KCKT (Kromatografi Cair Kinerja Tinggi) menggunakan fase gerak air-asetonitril-metanol-etil asetat-asam asetat glasial (89:6:1:3:1). Hasil difraktogram sinar-X pada kokristal metode I menunjukkan terbentuknya padatan amorf, sedangkan pada metode II menunjukkan terbentuknya fase kristalin baru (kokristal). Pada analisis termal menggunakan DTA terjadi perubahan titik lebur pada kokristal metode I maupun metode II yang berbeda dengan titik lebur katekin dan nikotinamida yang menunjukkan terbentuknya campuran eutektik. Pada analisis FT IR, baik kokristal metode I maupun metode II terjadi pergeseran bilangan gelombang dari spektrum katekin dan nikotinamida yang menunjukkan terbentuknya ikatan hidrogen antara gugus hidroksi fenol dari katekin dengan gugus amida dari nikotinamida. Dari hasil SEM, terlihat habit kristal dari kokristal metode I maupun metode II berbeda dibandingkan dengan habit kristal katekin maupun nikotinamida yang menunjukkan terbentuknya kokristal. Namun kelarutan kokristal katekin-nikotinamida metode I (132,17 mg/100 mL) maupun metode II (131,09 mg/100 mL) tidak berbeda nyata dengan kelarutan katekin (124,58 mg/100 mL). Kata kunci: kokristal, katekin, nikotinamida, kelarutan, KCKT.

Abstract

Extract of god leaves [Gynura pseudochina (L.) DC] has been characterized by using the high performance liquid chromatography method (HPLC). Dried god leaves were extracted by 70% ethanol and followed by fractionation successively with hexane, ethyl acetate, butanol and water. Fractions of ethyl acetate, butanol and water were dissolved in methanol respectively and injected into HPLC system which is optimized previously. Profiles of respective fractions showed a number of specific peaks that can be used to identify and control the quality of god leaves extract.   ABSTRAK Karakterisasi ekstrak daun dewa [Gynura pseudochina (L.) DC] telah dilakukan dengan menggunakan metode kromatografi cair kinerja tinggi (KCKT). Daun dewa kering diekstraksi dengan etanol 70% dan kemudian difraksinasi berturut-turut dengan heksana, etil asetat, butanol dan air. Fraksi-fraksi etil asetat, butanol dan air dilarutkan dalam metanol dan diinjeksikan ke sistem KCKT yang telah dioptimasi sebelumnya. Pola KCKT masing-masing fraksi menunjukkan puncak-puncak khas yang dapat dipakai untuk identifikasi dan pengendalian mutu ekstrak daun dewa.

Abstract

ABSTRACT : 4,5-dihydro-4-hydroxy-5-methyl-1H-pyran-1-one (2), angiopteroside (3) and well known stigmast-5-en-3--ol (1) have been isolated from the leaves of  Sumatran King fern Angiopteris evecta G. Forst HOFFM. and investigated for their antimicrobial activities towards some human pathogenic bacteria. Angiopteroside (3) showed potential activities to inhibit the growth of Bacillus subtilis. Keywords: Angiopteris evecta, 4,5-dihydro-4-hydroxy-5-methyl-1H-pyran-1-one,  Angiopteroside, antibacterial.
