Abstract

The study of anti-inflammatory activity and isolation of the methanolic extract, hexane and ethyl acetate fractions of the leaves of puwarkincung (Nicolaia speciosa Horan) have been done. Isolation was carried out by extraction, fractionation and multiple column chromatographicmethod, followed by recrystallization technique. Two compounds were isolated and labeled as F4 and S3. F4 was derived from n-hexanefraction as yellowish-white crystals (20 mg) with mp. 55–57 oC and S3 from the ethyl acetate fraction (15 mg) as yellow crystals with mp.68–69 oC. Anti-inflammatory activity of methanolic extract using Paw Edema method showed that the dose 250 and 1000mg/kgbw wereactive as anti inflammatory agents.

Abstract

Tangkahan Conservation Forest in Karo County, North Sumatera has high biodiversity of endemic herbs. Many species of the wild herbs are well known used as traditional medicine not only by local people but also by people out of the area. The methanol extract of the medicinal wild herbs in Tangkahan Conservation Forest, Karo County to relief skin diseases caused by bacteria and fungi never been studied medically. The antimicrobial activity leave extract of the medicinal herbs to pathogenic  microorganisms are studied. The leaves extract of kembu-kembu (Callicarpa candicans), rintih bulung (Piper muricatum), cep-cepan (Castanopsis costata), and sereh kayu (Eugenia grandis), has antimicrobial to bacteria (Bacillus sp., Escherichia coli, Serratia marcescens, Staphylococcus aureus) and yeast (Candida albicans). Toxicity assay of these plants by brine shrimp method using Artemia salina indicates that cep-cepan dan sereh kayu have lethal concentration higher than kembu-kembu and rintih bulung.

Abstract

ABSTRACT Knowledge and arts of the traditionally use of medicinal plant are valuable assets of Indonesian culture, which should be documented and preserved. Simultaneously, the plant that are traditionally used for health care should be investigated, particularly regarding their chemical constituents, biological activities and their safety, as well. This study aimed to inventory of medicinal plants used by people from Kampung Adat Urug, a Sundanese traditional village. This community still practices their traditional way of life, including how to maintain health and to treat illness. This study began with an ethnobotanical survey using Participation Observatory Method (POM) and interviewing the selected traditional healers. Data analysis was carried out by calculating Use Value (UV), Relative Frequency of Citation (RFC), and Relative Importance (RI) of plant species. The study revealed 29 medicinal plants used by Kampung Urug community. Five medicinal plants (Ixora salicifolia, Scleria levis, Hippobroma longiflora; Pterocarpus indicus and Pothos junghuhnii) showed high level of UV, RFC, and RI. Those species were found to be potential in treating various neglected diseases, women health care during pregnancy, postpartum illness, and maintaining health care in general. Besides, there were four plants extracts, each from Barringtonia acutangula; Pterocarpus indicus; Selaginella cf. lana and Pothos junghuhnii which showed potent inhibition against Staphylococcus aureus. The phytochemical screening of the above extracts showed various major constituents, particularly phenolics and flavonoids. Abstract

In continuation of our study of Sumatran plants and the bioactivity of their constituents, Stereocaulon halei Lamb (Stereocaulaceae) was extracted and isolated to obtain atranorin (1), lobaric acid (6), and methyl-β-orcinol carboxylate (9). We also prepared methanolic extracts of some Sumatran medicinal plants, i.e. Alpinia denticulata, Alpinia malaccensis, Alpinia submutica, Curcuma zedoaria, Centella asiatia, Mangifera indica, Piper crocatum, Mussaenda frondosa, together with previously isolated compounds, i.e. crocatin A (2) and crocatin B (3) from P. crocatum, curcumin (4) from Curcuma longa, demethoxymatteucinol (5) from Sphaerostephanos polycarpus, mangiferin (7) from M. indica, methyl caffeate (8) from Balanophora elongata, tiliroside (10) from Guazuma ulmifolia, and usnic acid from Usnea sp (11). These extracts and compounds were tested for their cytotoxicity against MCF-7 and HSC-3 cell lines by MTT assay (3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide). It was found that the IC50 value of atranorin (1), lobaric acid (6), methyl-β-orcinol carboxylate (9), and methanolic extract of A. submutica towards the MCF-7 cell line was 208.20 µM, 172.05 µM, 382.60 µM and 70.95 µg/mL, respectively. The IC50 value of lobaric acid (6) and methyl-β-orcinol carboxylate (9) against the HSC-3 cell line was 88.92 µM and 260.09 µM, respectively. The other extracts did not show significant cytotoxicity.

Abstract

Balanophora elongata (Balanophoraceae) is a tropical parasitic flowering plant 9 cm in height. Four known phenyl acrylic acid derivatives, methyl caffeate (1), caffeic acid (2), 1,6-di-O-caffeoyl-β-D-glucopyranose (3), and coniferin (4), were isolated from this plant. Structural elucidation of the isolated compounds was determined by IR, LC-ESI-MS, 1D and 2D NMR.  Extracts and isolated compounds were tested toward some standard human pathogenic bacteria using the agar disk diffusion method. Their inhibition zones were compared to that of chloramphenicol as positive control. Compound 1 showed inhibition toward Streptococcus mutans, while compound 3 and 4 inhibited Staphylococcus aureus.
