Abstract

The purpose of this research is to explore effectiveness from every part of Phyllanthus niruri such as leaves (L), fruits (F), stem (S) and combination (C) of those parts that able to inhibit Aeromonas salmonicida growth by using different methods of extraction both infundation and maceration. Experiments has been done at Budidaya Perairan Laboratory, Sriwijaya University. In vitro analysis used tetracycline 30 µg/ml as positive control, bacterial culture at 1014 CFU/ml and concentration of each part of stone breaker was 20%, 40%, 60%, 80% and 100%. The result revealed that stone breaker extract produced by infundation and maceration methods exhibited inhibitory activity against A.salmonicida. Moreover, only maceration method had bactericidal effect, in fact fruits (F) and combination (C) was the best part of P. niruri which had higher inhibition zone than control (antibiotics). It was 16,9 mm and 17,3 mm respectively. However, the extracts are recommended to cure aeromonad infection due to bacterial resistance of tetracycline already increase. Thus, some extraction methods are needed to be developed for further research with minimum inhibition zone is 20 mm.Penelitian ini bertujuan untuk mengetahui efektivitas setiap bagian Phyllanthus niruri seperti daun (L), buah (F), batang (S) dan kombinasi (C) yang mampu menghambat pertumbuhan Aeromonas salmonicida menggunakan metode yang berbeda, ekstraksi infundasi maupun maserasi. Percobaan telah dilakukan di Laboratorium Budidaya Perairan Universitas Sriwijaya. Analisis in vitro menggunakan tetra siklin 30 g/ml sebagai kontrol positif, kultur bakteri pada 1014 CFU/ml dan konsentrasi masing-masing ekstrak adalah 20%, 40%, 60%, 80% dan 100%. Hasil penelitian menunjukkan bahwa ekstrak yang dihasilkan dengan metode infundasi dan maserasi menunjukkan aktivitas penghambatan terhadap A.salmonicida. Selain itu, hanya metode maserasi yang memiliki efek bakterisidal, ternyata buah (F) dan kombinasi (C) merupakan bagian terbaik dari  P. niruri yang memiliki zona hambat lebih tinggi daripada kontrol (antibiotik). Masing-masing 16,9 mm dan 17,3 mm. Namun, ekstrak tersebut direkomendasikan untuk menyembuhkan infeksi aeromonad karena resistensi bakteri terhadap tetra siklin sudah meningkat. Oleh karena itu, perlu dikembangkan beberapa metode ekstraksi untuk penelitian selanjutnya dengan zona hambat minimal 20 mm.

Abstract

ABSTRACT The aim of this research was to analyse the toxicity and immunogenicity of ECP of non-hemolytic S. agalactiae and to evaluate ECP as a material vaccine for prevention of S. agalactiae. Toxicity to tilapia was detected in the ECP. There was a chronic  mortality pattern which was shown after post ECP injection. By intraperitoneal injection into young tilapia (± 20 g), the median lethal dose were calculated at 633,9 µg/kg bodyweight for isolate 3 and 685,4 µg/kg bodyweight for isolate 5. Cytopathic effects of ECP to tilapia’s lymphocytes were observed during 0, 30, 60 and 180 minutes. A mixture of ECP caused nuclear granulation and cytoplasmic streaming after 60 minutes. Using immunodiffusion analysis, precipitate line was demonstrated against tilapia anti-ECP sera, and there was bio-specific type anti sera against for each isolate. The ECP vaccine effective to protect tilapia from S. agalactiae was injected intraperitoneally (IP) at 2 x 105 CFU/ml. Tilapia vaccinated with ECP from isolat 3 which protein  283 µg/kg bodyweight had a RPS value 60%.  Tilapia vaccinated with ECP from isolat 5 which protein 408 µg/kg  had a RPS value 68%.   Keywords:  toxicity, immunogenicity, extracellular products, Streptococcus agalactiae, tilapia

Abstract

Ascorbic acid was one of the important nutrients for fish because it has the property as an antioxidant and imunomodulator that can improve the performance of fish. The purpose of this research is to support the production of snakehead fish (Channa striata) that have good growth and immunity. This research used a complete randomized design (CRD) consisted of 3 treatments and 3 replications. Ascorbic acid treatment consisted of P1 (0 mg.kg-1), P2 (50 mg.kg-1) and P3 (100 mg.kg-1). The results showed that the maintenance of snakehead fish (Channa striata) given ascorbic acid  with a doses of 100 mg.kg-1  for 30 days gave the best growth results and significantly different and absolute weight growth reached 1.83 g, absolute length of 18.7 mm and survival reached 95 , 83 %. For immunity performance, it can be seen from survival of fish exposed to salinity 20 ppt during 15 minute reach 73,3 % and formalin 10 % for 10 minute reach 66,7 % and disease prevalence percentage reach 16,7 %

Abstract

ABSTRACTThe purpose of this study was to determine the best fertilizer compotition that was made from technical fertilizer and waste water of cat fish farming. This experimental was conducted at Aquaculture Laboratory, Faculty of Agriculture, Sriwijaya University. It used complete randomized design (CRD) method which had five treatments and three replications, the waste water from catfish pond (P0), waste water from catfish pond + 25% technical fertilizer (P1), waste water from catfish pond + 50% technical fertilizers (P2), the waste water from catfish pond + 75% technical fertilizers (P3), the waste water from catfish pond + 100% technical fertilizers (P4). The results showed that the best maximum density of S. platensis was P3 about 3.98 g l-1, and growth rate was equal to 2.80% day-1. The most significant parameter of water quality was ammonia which decreased until 82.78%. Besides, it was more beneficial than others based on Return Cost Ratio (R/C ratio) and Benefit Cost Ratio (B/C ratio) which were 3.66 and 2.66 respectively. Keywords: Spirulina platensis, waste water pond of catfish, modified  media

Abstract

The transportation process can lead to stress which increases mortality in fish. Efforts to reduce mortality in the process of fish transportation can be conducted by minimizing the metabolic activities of fish during transportation. One of the natural substances that is used for fish transportation is tuba roots. The aim of this study is to determine the effect of different dosages of tuba root extract on the transport of catfish seeds with a closed transport system for 12 hours. This research was conducted at the Aquaculture Laboratory, Aquaculture Study Program, Faculty of Agriculture, Sriwijaya University. The research was conducted in April 2019. This study used a completely randomized design (CRD) method with four different dosage treatments (T) and three replications. The dosages of tuba root extract were: 0 ml.L-1 (T0), 0.05 ml.L-1 (T1), 0.10 ml.L-1 (T2), 0.15 ml.L-1 (T3). The parameters observed were blood glucose levels, water quality, survival of catfish after transportation and maintenance for 7 days. The results showed that T1 (0,05 ml.L-1) was gave the highest survival rate of 91.66±2.88%, with the lowest stress level indicated by blood glucose values 89±30.63. In addition, there was no further effect from the administration of tubal root extracts to protect the life of the fish after transportation and maintenance.Keywords : Tuba root, catfish, transportation

Abstract

ABSTRACT               Masculinization is a method of  sex reversal technique that can direct sexuality from female to male. The purpose of this study was to determine the effects of coconut water on masculinization in guppy by immersion administration. The study used a completely randomized design (CRD) with three treatments and three replications. After 12 days post spawning, female guppies were immersed for 8 hours.The concentrations were : 0%(P0), 20% (P1), and 40% (P2). The parameters were observed including percentage of male, survival rate and water quality (thermal, pH and kalium concentration). Baby guppies were reared until sex can be determined which were 42 days of culture. The result showed that the percentage of male guppy fish was 54.44% (P0), 77.78% (P1) and 90.00% (P2). The survival rate of  larval during maintenance was 100% for all treatments. Water quality during maintenance was 27.0-28.40C for temperature and 6.38-7.20 for pH. To sum up, this experiment proved that coconut water can be used for masculinization by immersed pregnant female broadstock and it produced 90% male when immersed with 40% of coconut water. Keywords : Coconut water, guppy (Poecilia reticulata), masculinization

Abstract

Sungsang IV Village is a village where most of the residents work as fishermen. The waters around this village have great fishery potential. The results of fish catches by fishermen are felt to be less than optimal due to a lack of knowledge about the latest technology in fishing. Fish Finder is one alternative that can be given to solve this problem because this tool can function to detect the presence of fish. Most of the residents of Sungsang IV Village don't know about the Fish Finder tool. This service activity aims to provide introduction and training regarding the use of Fish Finder to detect the presence of fish. The enthusiasm of the community is very visible from the number of training participants that exceeds the targeted number. This training is carried out by presenting material and direct practice of using the tool to make it easier for participants to understand the operating steps. The results of the evaluation of the questionnaire conducted showed that all participants felt very helpful with the training in the use of this Fish Finder tool.

Abstract

Permintaan akan kebutuhan ikan lele ukuran konsumsi terus meningkat dari tahun ke tahun. Peningkatan permintaan ini berkorelasi positif dengan adanya peningkatan produksi ikan lele. Hal ini mengambarkan bahwa budidaya ikan lele memiliki prospek yang menjanjikan jika dilakukan dengan benar. Salah satu penentu keberhasilan produksi ikan lele melalui kegiatan budidaya adalah aplikasi teknologi untuk meningkatkan kelangsungan hidup ikan dan mempercepat pertumbuhan ikan budidaya. Pengabdian ini dilakukan dengan tujuan untuk meningkatkan produktivitas kegiatan budidaya ikan lele di Desa Pulau Semambu melalui peningkatan kelangsungan hidup dan pertumbuhan ikan lele dengan aplikasi penambahan prebiotik ekstrak ubi jalar pada pakan. Kegiatan ini dilaksanakan pada bulan Agustus-Oktober 2019 di Unit Pembenihan Rakyat (UPR) Fish Under Crew Pulau Semambu, Kabupaten Ogan Ilir, Provinsi Sumatera Selatan. Metode kegiatan pengabdian yang dilaksanakan berupa transfer teknologi yang terdiri dari beberapa tahapan yaitu memberikan penyuluhan, pelatihan/peragaan berupa demonstrasi langsung serta dilakukannya kegiatan pendampingan berupa bimbingan selama 6 bulan atau dua kali siklus produksi ikan lele. Pendampingan secara langsung dapat dijadikan pedoman teknis budidaya yang baik dan benar oleh petani pembudidaya sehingga menambah keterampilan dan kemampuan petani pembudidaya yang pada akhirnya dapat meningkatkan produksi budidaya ikan lele di Desa Pulau Semambu. Pengabdian telah dilakukan dan terlihat masyarakat memberikan respon positif dan dengan antusias mengkuti penyampaian materi dan demonstrasi yang diberikan tim pengabdian, meningkatnya pengetahuan dan keterampilan petani ikan lele di UPR Fish Under Crew dalam membuat prebiotik ubi jalar serta pemberian pakan prebiotik pada pakan ikan lele. The demand for consumption size catfish continues to increase from year to year. This increase in demand is positively correlated with an increase in catfish production. This illustrates that catfish farming has promising prospects if done properly. One of the determinants of the success of catfish production through aquaculture activities is the application of technology to increase fish survival and accelerate the growth of cultured fish. This service is carried out to increase the productivity of catfish farming activities in Pulau Semambu Village by increasing the survival and growth of catfish by adding prebiotic sweet potato extract to feed. This activity was carried out in August-October 2019 at the Fish Under Crew People's Hatchery Unit (UPR) on Semambu Island, Ogan Ilir Regency, South Sumatra Province. The method of service activities carried out is in the form of technology transfer, which consists of several stages, namely providing counselling, training/demonstrations in the form of direct demonstrations and conducting mentoring activities in the form of guidance for six months or twice a catfish production cycle. Direct assistance can be used as technical guidelines for good and correct cultivation by cultivating farmers to increase their skills and abilities, which in turn can increase catfish farming production in Pulau Semambu Village. The service has been carried out, and it is seen that the community gives a positive response and enthusiastically follows the delivery of material and demonstrations given by the service team, increasing the knowledge and skills of catfish farmers at UPR Fish Under Crew in making sweet potato prebiotics and providing prebiotic feed on catfish feed.

Abstract

Starfruit contains active ingredients such as flavonoids, phenols, antioxidants and vitamin C so that it can be used as an immunostimulant in catfish farming activities. Motile Aeromonas Septicemia (MAS) is one of the obstacles that can cause catfish farming losses. This study aims to determine the effect of differences in timing of feeding containing starfruit juice to protect catfish from MAS disease. The study was designed using a completely randomized design (CRD) consisting of 6 treatments (P) in the form of feeding duration containing starfruit for 7 days (P1), 14 days (P2), 21 days (P3), 28 days (P4). and 35 days (P5) and control of feeding without star fruit juice (P0). The research was carried out at the Aquaculture Laboratory, Sriwijya University. The dose of star fruit juice mixed in the feed is 300 mL.Kg-1 while the catfish used is 8-12 cm in size. The challenge test used Aeromonas hydrophilla bacteria with a density of 108 CFU.mL-1 which was injected as much as 0.5 mL.tail-1 intramuscularly. The results showed that administration containing starfruit juice for 28 days provided the best protection. Based on the hematological test the day before and before infection, the delta decreased the hematocrit by 3% and the delta decreased the erythrocyte count by 3,103 cells.mL-1. In addition, live feed after infection was 100%, prevalence was 17.78% and there was no effect of giving starfruit juice on growth and feed efficiency.

Abstract

Fish Under Crew, a group of fish farmers located at Pulau Semambu, Indralaya Utara, Ogan Ilir. It rears several fish commodities including catfish. The knowledge of fish farmers about best aquaculture practices are still low and cost production become main problem for them. Therefore, community service held on Aquaculture Department, Sriwijaya University conducted an alternative solution to solved its problem by providing information and pilot culture about fasting and refeeding with probiotic in catfish. The community service was carried out in July-August 2021. The fasting interval was one day then refeeding with commercial pellet enriched by probiotic. Day of culture was 30 days and it compared with batch control which was daily fed without probiotic. Prior and after community service, the fish farmers were interviewed about their knowledge. During fish rearing, the fish farmers also learnt about fish rearing parameters such as growth, survival rate, feed conversion ratio (FCR) and water quality. In the end of culture, fish farmers stated that they were willing to apply the technology.
