Abstract

University of Riau Forest, Riau Province is one of distribution area of ferns (Pteridophytes). This study aimed to identify fern species at University of Riau Forest and examined the DNA band pattern based on M13 DNA marker. Samples were collected from the field using exploration method and were then identified. DNA isolation was carried out using DNAeasy Plant Mini Kit (Qiagen), and amplified based on PCR method using DreamTaq Green PCR Master Mix (2X) (Thermo Scientific) with M13 DNA Marker. The inventorisation result showed a total of 26 fern species belong to 8 families were identified in this study. The electrophoresis result of PCR product gave the variaous DNA band pattern ranged from 200 to 2000 bp. DNA band pattern using M13 primer can be used to support the taxonomical data of fern taxa

Abstract

Kasturi orange (Citrus microcarpa Bunge.) is a medicinal plant that contains chemical compounds from secondary metabolites. Limitations of the seed and slow rate of growth is a challenge in the Kasturi orange plant propagation. Propagation of shoots in the kasturi orange can be produced quickly with shoots induction in vitro. This study aims investigate the influence and determine the concentration of BAP in inducing shoots from nodes kasturi orange explants in vitro. This research was conducted at the Integrated Laboratory, Department of Biology, Faculty of Mathematics and Natural Sciences, The University of Riau from March - September 2020 using completely randomized design (CRD) which consists of 6 (six) treatments than is control BAP (1,2,3,4,5 mg / L). Results showed BAP had a significantly affect the number of shoots and shoot length. The treatment of 1 mg / L BAP produces the highest number of shoots that 2.50. The highest callus growth was in the treatment of 5 mg / L BAP. The shoot appears fastest time is 12.00 day after planting (DAP) contained in the treatment of 2 mg/L BAP.

Abstract

Kurma (Phoenix dactylifera L.) merupakan tanaman tahunan yang hidup di wilayah iklim tropis seperti di Negara Timur Tengah dan Afrika Utara. Perluasan budidaya tanaman kurma secara tradisional masih dibatasi oleh kemampuan tanaman untuk menghasilkan bibit baru dalam jumlah banyak, seragam, dan dalam waktu singkat. Perbanyakan pada kurma dapat diproduksi cepat dengan inisiasi biji secara in vitro. Penelitian ini bertujuan untuk mengetahui pengaruh dan menentukan konsentrasi terbaik GA3 tunggal dan kombinasi GA3+BAP terhadap perkecambahan dari eksplan biji (Phoenix dactilyfera L.) secara in vitro. Penelitian ini dilaksanakan di Laboratorium Terpadu, Jurusan Biologi, Fakultas Matematika dan Ilmu Pengetahuan Alam Universitas Riau dari bulan Mei–September 2020 dengan menggunakan Rancangan Acak Lengkap (RAL) yang terdiri atas 10 (sepuluh) perlakuan yaitu kontrol, GA3 tunggal (2,5; 5; 7,5 mg/l) dan dikombinasikan dengan BAP (1 mg/l dan 2 mg/l). Hasil penelitian menunjukkan pemberian GA3 tunggal dan dengan BAP berpengaruh nyata terhadap waktu muncul akar dan panjang akar dari eksplan biji kurma (Phoenix dactilifera L.). Pada perlakuan 2,5 mg/l GA3 adalah konsentrasi terbaik untuk waktu muncul akar tercepat yaitu 5,50 HST. Panjang akar terbaik di antara semua perlakuan pada perlakuan 5 mg/l sebesar 6,36 cm.
