Abstract

Research to determine species diversity, classification and identification of key species and the distribution of macroalgae in coastal waters Sekotong, West Lombok has been done. The study was conducted by survey method, specimen collection and identification. Survey and sampling carried out in five locations in the waters of Sekotong: West Sekotong, Gili Nanggu, Gili Sudak, Gili Gede, and Bangko-Bangko. Based on the results of the inventory and identification is known that in the waters of Sekotong there are 61 species of macroalgae. Rhodophyta is the largest division with the highest species diversity, while Phaeophyta has the lowest number of species. Rhodophyta consists of 25 species, 17 genera and 11 families. Chlorophyta consists of 21 species, 11 genera and10 families. Phaeophyta consists of 15 species, 7 genera and 4 families. Key to the identification of species of macroalgae in Sekotong has been prepared based on morphologic characters. There are differences in the composition and species richness in the five beaches that have been surveyed. West Sekotong beach is area with the highest diversity of macroalgae at Sekotong, while the lowest diversity was found in the waters of Gili Nanggu

Abstract

Ganoderma merupakan salah satu jenis jamur makroskopis yang menjadi  kekayaan hutan Indonesia, sementara laju pengrusakan hutan yang tinggi dapat mengancam kelestarian biota ini. Penelitian ini bertujuan untuk mengetahui jenis-jenis Ganodermataceae dari berbagai kawasan hutan Pulau Lombok. Pengambilan sample jamur  dilakukan di enam kawasan hutan yaitu hutan Kerandangan, Pusuk, Nuraksa Sesaot, Lemor, Pergasingan Sembalun dan Gunung Tunak. Penelitian ini dilakukan dengan menggunakan metode jelajah, yaitu membagi daerah penjelajahan menjadi 2-3 jalur penjelajahan dan dilakukan dengan teknik visual sensus. Identifikasi Ganoderma didasarkan pada karakteristik makroskopik, lalu dicocokkan dengan buku-buku identifikasi(profile matching). Hasil penelitian diperoleh 9 spesies Ganoderma, yaitu G. applanatum, G. lucidum, G. adspersum, dan 6 isolat Ganoderma belum teridentifikasi.  Ganoderma applanatum ditemukan tersebar dihampir semua kawasan hutan dipulau Lombok, terkecuali TWR Nuraksa Sesaot dan Bukit Pergasingan Sembalun.Kata kunci: makrofungi, Ganoderma, Pulau Lombok

Abstract

A research has been conducted to optimize the rate of aeration and initial cell aggregates weight in the production of ajmalicine in Catharanthus roseus cell culture in bioreactor. Catharanthus roseus culture were grown in Zenk medium with the addition of 2.5 x 10"6 M NAA (naphthalene acetic acid) and 10s M BAP (benzyl amino purine). Cell aggregates were subcultured two times before transferring 20 and 30 g/fw of cell aggregates into bioreactor, respectively, and aerated with the rate of 0.2460 L/min and 0,3405 L/min, respectively. The pattern of ajmalicine production in bioreactor were observed every 3 days for 24 days.Qualitative and quantitative analysis were conducted using HPLC connected to Cromatopac CL-7APlus. The results showed that the cell aggregates and medium contain ajmalicine. The highest concentration was obtained in combination of 30 g/fw and 0.3405 L/min aeration compare to 20 g/fw - 0,246 L/min, 20 g/fw- 0,3405 L/min, as well as 30 g/fw - 0,2460 L/min. The highest ajmalicine content in cell aggregates was obtained on the 12"d days (79.23ug/g) whilst in medium was obtained in the 18th days (981.15 ug/L).

Abstract

Masyarakat Hindu di Desa Jagaraga di Kabupaten Lombok Barat merupakan komunitas yang masih melakukan pengobatan tradisional menggunakan tumbuh-tumbuhan. Hal-hal terkait kearifan lokal pemanfaatan tumbuhan sebagai bahan obat perlu digali sebelum tergeser oleh perubahan yang selalu dinamis di masyarakat, salah satunya melalui kajian etnobotani. Tujuan penelitian ini adalah untuk mengidentifikasi jenis-jenis tumbuhan obat dan manfaatnya, mengetahui signifikansi kultural jenis-jenis tumbuhan obat bagi masyarakat setempat, serta kearifan lokal masyarakat terkait pemanfaatan tumbuhan sebagai bahan obat-obatan. Penelitian ini bersifat deskriptif eksploratif dengan metode kualitatif dan kuantitatif. Analisis kuantitatif didasarkan atas perhitungan Reported Use (RU), Index of Cultural Significance (ICS), dan Fidelity Level (FL) masing-masing jenis tumbuhan obat. Pengumpulan data dilakukan melalui observasi, wawancara, dan dokumentasi, dengan pemilihan narasumber berdasarkan metode purposive sampling dan snowball sampling. Penelitian dilakukan pada lima dusun yang merupakan komunitas Hindu di Desa Jagaraga, Lombok Barat. Berdasarkan hasil penelitian diperoleh 21 spesies yang tergolong dalam 17 famili dan 20 genus. Lima jenis dengan nilai IC) tertinggi berturut-turut adalah Jatropha curcas L., Curcuma longa L., Kaempferia galanga L., Allium cepa L., dan Musa paradisiaca L. Secara umum, terdapat berbagai nilai kearifan lokal masyarakat terkait dengan pemanfaatan, pengelolaan, dan pemeliharaan jenis-jenis tumbuhan berkhasiat obat, dimana hal ini berkontribusi pada kelestarian tumbuhan dan lingkungan di sekitar wilayah kajian.

Abstract

Abstrak: Kegiatan ini bertujuan untuk meningkatkan pengetahuan dan keterampilan mitra pengabdian tentang teknik kultur jaringan tanaman. Kegiatan pelatihan dibagi menjadi 2 sesi yaitu sesi pertama, penyampaian materi tentang konsep dasar kultur jaringan, kebutuhan peralatan dan bahan, media, zat pengatur tumbuh, kondisi lingkungan tumbuh, tahapan kultur jaringan dan macam-macam teknik kultur in vitro. Sesi kedua, praktek teknik kultur jaringan di laboratorium meliputi pengenalan laboratorium,alat dan bahan beserta fungsinya, multiplikasi (perbanyakan) tunas anggrek in vitro, kultur kalus, kultur embrio dan kultur mata tunas dengan sistem single node. Kegiatan pengabdian pelatihan teknik dasar kultur jaringan dapat membantu meningkatkan pengetahuan dan keterampilan peserta tentang kultur jaringan. Hal ini dapat dilihat dari antusiasme peserta selama kegiatan dan meningkatnya persentase pemahaman peserta menjadi 54,5% (tingkat pengetahuan baik), 36,4% peserta dengan tingkat pengetahuan cukup, dan 9,1% peserta dengan tingkat pengetahuan kurang.Abstract: This activity aims to improve the knowledge and skills of partners about plant tissue culture techniques. The training activity is divided into 2 sessions, namely the first session, delivering material on the basic concepts of tissue culture, equipment and material requirements, media, growth regulators, growing environmental conditions, tissue culture stages, and various in vitro culture techniques. In the second session, the practice of tissue culture techniques in the laboratory included introducing the laboratory tools and materials and their functions, multiplication (propagation) of orchid shoots in vitro, callus culture, embryo culture, and organ culture using a single node system. Basic technique training of plant tissue culture can help increase participants' knowledge and skills about tissue culture. This can be seen from the participants' enthusiasm during the activity and the increase in the percentage of participants' knowledge to 54,5% (good level of knowledge), 36,4% of participants with a sufficient level of knowledge and 9,1% of participants with a low level of knowledge.

Abstract

The purpose of this study was to determine the variation of the stem anatomical structure (i.e. cortex: sclerenchyma, phloem; xylem: trachea, rays, interxylary phloem and wood parenchyma; and pith) of Gyrinops (Thymelaeaceae) in Sulawesi. The method of this research was descriptive exploration. Stem agarwood sample came from several regions areas of Sulawesi which were collected by Agarwood Research Center Laboratory, Faculty of Mathematics and Natural Sciences, University of Mataram. The slide methods were used fresh slides. The results of this research showed there were 1 species of Gyrinops decipiens  and 3 species of Gyrinops sp., with various anatomical characters in G. decipiens  and Gyrinops sp. in terms of the position, diameter and percentage of pith thickness; cortical sclerenchyma pattern and arrangement of wood parenchyma. There were also variations in cortical thickness; diameter, frequency and arrangement of trachea; frequency of rays; diameter, and interxylary phloem frequency; pith diameter; and the position of Ca-Oxalate in Gyrinops sp. 3 with Gyrinops sp. 1 and 2, while Gyrinops sp. 2 with Gyrinops sp. 1 there were found 5 variations such as the diameter, frequency and arrangement of trachea; arrangement of rays and interxylary phloem length.

Abstract

The purpose of this study was to determine the potential and concentration of Sargassum cristaefolium extract as a natural cytokinin in tissue culture media of Dendrobium antennatum Rchb.f. This study is experimental with a completely randomized design, using several extract concentrations compared with the positive control (BAP 1.5 ppm) and negative control (MS0 media). Extract concentrations used 5 ppm, 10 ppm, 15 ppm, 20 ppm, and 25 ppm. Based on analysis of variance (ANOVA), the effect of Sargassum cristaefolium extract on the growth media significant on all parameters. Sargassum cristaefolium extracts caused different responses at certain levels of concentration. Extract concentration of 10 ppm was able to initiate the highest number of shoots and leaves compared to other extract concentrations, where as the concentration 20 ppm was able to accelerate and increase root growth.

Abstract

Lingkungan Jorong merupakan lingkungan Kelurahan Kelayu Jorong merupakan salah satu kelurahan Kota Selong dengan jumlah penduduk sekitar 2000 jiwa. Sebagian besar penduduk di lingkungan Jorong profesi sebagai petani. Kondisi ini menyebabkan Lingkungan Jorong sangat potensial sebagai penghasil sampah baik sampah dari hasil pertanian dan peternakan, sampah rumah tangga maupun sampah lainnya. Saat ini di lingkungan Jorong berkembang wisata aquatik berupa kolam pemandian yang mulai ramai dikunjungi oleh masyarakat dari luar Kelurahan Kelayu Jorong. Hal ini sangat potensial menghasilkan sampah organik maupun non-organik dari makanan yang dikonsumsi oleh pengunjung. Kegiatan Pengabdian pada Masyarakat ini dilaksanakan di areal Perguruan NW Jorong yang meliputi Madarasah Tsanawiyah (MTs) dan Madrasah Ibtidaiyah (MI). Kegiatan ini telah dilaksanakan pada bulan November 2018. Dalam kegiatan ini telah dilakukan pendekatan yaitu  ceramah dan pemutaran audio visual tentang manajemen pengelolaan sampah yang meliputi pengetahuan jenis-jenis sampah, akibat-akibat yang ditimbulkan oleh sampah, metode pembuangan sampah, serta pengolahan sederhana sampah menjadi produk yang bisa dimanfaatkan. Selain itu telah dilaksanakan demonstrasi yang meliputi desain penampung sampah berdasarkan jenisnya dan tata laksana pengumpulan sampah, serta pengolahan samapah sederhana menjadi produk yang bisa dimanfaatkan. Terbatasnya waktu serta kondisi lingkungan yang tidak memungkinkan akibat bencana alam gempa yang menimpa pulau Lombok akhir Juli-September 2018 menyebabkan tidak semua target kegiatan terpenuhi terutama pada fase pemanfaatan sampah organik dan uji coba penggunaaan produk pupuk organic untuk penanaman sayur organik.

Abstract

The aim of this research is to determine the comparative anatomy of the stem of Gyrinops versteegii (Thymelaeaceae) in Sumbawa Island. Stem samples of G. versteegii were used from collections of the Agarwood Center Laboratory, Faculty of Mathematics and Natural Science, University of Mataram that were taken from Sumbawa Island. This research used permanent slides with freehand section and maceration methods. Observation data consisted of cortex tissues, xylem, pith, and Ca-Oxalate crystals. The results showed that there were four provenances of G. versteegii on Sumbawa Island, namely provenance Pantai, Hajumee, Kayuk Pisak, and new provenance (Tartar provenance). Comparison of anatomy between the stems of G. versteegii provenance Pantai with other provenances had differences in periderm thickness, the pattern of tissue sclerenchyma, interxylary phloem thickness, cortical parenchyma diameter, number of rows of wood parenchyma tissue, tracheid diameter, pith cell diameter, Ca-oxalate shape. Provenance Hajumee with Tartar and Kayuk pisak differed in trachea length, trachea diameter, and the length of the tracheids, while between the provenances of Tartar and Kayuk pisak differed in the frequency of the trachea, the length of the fiber tracheids, the arrangement of the pith radius, and the location of the pith.

Abstract

Grynops verstegii is a non-timber forest product commodity that has a very high economic value. People know this tree as the agarwood tree. In Lombok, the Beringin provenance is one of the leading provenances, so it is very important to conserve its germplasm. This study aims to find alternatives for gaharu development through seed production using in vitro culture techniques. This research was carried out in July 2021 at the Tissue Culture Unit of the Immunobiology Laboratory, University of Mataram. In this experiment there were 8 media treatments with different concentrations of BAP (Ssitokini) and NAA (auxin). The results showed that callus growth occurred on MS 1, MS 3, MS 4, MS 6, and MS 7 media. However, the best growth was obtained in MS 1 treatment with a combination of 1.0 ppm BAP and 0.5 ppm NAA. While most of the samples experienced browning and contamination.
