Abstract

Community Service Activities (PENMAS) were held in August 2018 in Kapas, Bojonegoro with a target audience of SMKN 4 communities, totaling 107 people, consisting of students majoring in Cooking, members of Dharma Wanita, Chefs from 2hotels, several UMKM Food and Beverages around the school, teachers and partisipants from other schools. The activity was also attended by the Principal, the Head of the Education Office who witnessed the signing of the MOU between the PENMAS Team and the school. One of the Departments managed by SMK 4 is Tata Boga with thelearning outcome to prepare entrepreneurs or businessmen of food products. As a vocational school that has held the ISO 9001 2008 certificate, the school is obliged to prepare graduates according to the demands of their competence. Based on preliminary studies, information was obtained that there were several problems related to food consumption and production and government regulationin the case of traded food businesses. The training materials provided in this training were: Food business production regulation (1), How to make good food (2),Ingredients additional allowable food (3), Prohibited food additives and substitute alternatives (4), Good packaging methods and techniques for food and beverage (5), Halal Assurance System (SJH) and Halal food product Certification Process (6) and Simple marketing for food business (7). Based on the evaluation of the pre-test and post-test resulted that after the training participants' understanding showed an increasing trend. Three topics that showed a significant increase in understanding were: Requirements for clean and healthy food (1), Introduction of food coloring logos (2), and Examples of halal food ingredients (3). PENMAS activities will be followed up with intensive coaching activities for students of SMKN4 which are included in the group of beginner beverage food businesses.

Abstract

The single-component CPM tablet mostly used sodium tartrazine as the yellow coloring agent. Sodium tartrazine is soluble in solvents used to extract CPM from tablet and suspected interference CPM determination especially after forced degradation for stability indication testing of CPM tablets. This study aimed to develop a selective, accurate and precise method for determination and stability testing of chlorpheniramine maleate (CPM) in the presence of tartrazine in the tablet. A µBondapak® C18 column (3.9 x 300 mm, 10 µm) with diode array detector was used for separation. The mobile phase was a mixture of methanol and 0.2% triethylamine (90:10) with a flow rate of 2 mL/minutes. The validated HPLC method was used for CPM determination in tablet samples that had been forced degraded using dry heat at 105oC, UV radiation of 254 nm, hydrolysis with 1N NaOH, 1N HCl and oxidation using 5% H2O2. The HPLC chromatogram showed that CPM split into chlorpheniramine (CP) and maleic acid (MA). Resolution (Rs) among CP and the other analytes especially with the products resulting from the forced degradation by heat, UV radiation, HCl, and H2O2 were good. The CPM hydrolysis using NaOH caused the CP not completely separated from the degradation product due to tailing or overlapping peaks. The proposed HPLC method was valid for the determination of CPM in tablets containing tartrazine. Even though the stability-indicating method was inadequate especially for the result of the CPM hydrolysis process using NaOH.

Abstract

Background: Sodium benzoate and sodium cyclamate are foods additives generally used as preservative and artificial sweeteners, respectively. Monitoring of sodium benzoate and sodium cyclamate need appropriate analytical method. Objective: The main objective of this study was to gain the optimum conditions for simultaneous determination of sodium benzoate and sodium cyclamate in soft drink by High Performance Liquid Chromatography. Methods: Measurement of the additives was performed in µ-Bondapak RP18 column using methanol:phosphate buffer of pH 4 (42:58) as mobile phase with flow rate of 1 mL/minute. Results: It has shown that correlation coefficients (r) of sodium benzoate and sodium cyclamate was 0.9997 and 0.9991 respectively. Recoveries of the two analyst were acceptable. Conclusions: Five out of nine samples tested contained sodium cyclamate higher than the recommended concentration. All sodium benzoate containing samples were within the recommended concentration.

Abstract

Pendahuluan: Madu adalah cairan alami dengan konsistensi kental dan rasa manis yang dikumpulkan oleh lebah madu dari sari bunga atau bagian lain tumbuhan. Produk madu yang dipasarkan harus memiliki nomor registrasi yang dikeluarkan oleh Badan Pengawas Obat dan Makanan Republik Indonesia (BPOM RI) dan memenuhi persyaratan mutu yang ditetapkan oleh Standar Nasional Indonesia (SNI) 01-3545-2013. Nomor registrasi produk madu diawali dengan kode ML, MD, TR dan P-IRT. Perbedaan kode awal pada nomor registrasi menunjukkan katagori produsen pengolah produk madu. Tujuan: Tujuan penelitian ini adalah mengetahui mutu produk madu yang diregistrasi dengan katagori berbeda mengacu persyaratan SNI 01-3545-2013. Metode: Dalam penelitian ini mutu 4 sampel produk madu dibandingkan dengan acuan parameter SNI untuk madu. Setiap sampel produk madu mewakili satu kode awal nomor registrasi. Nama sampel tersebut dipilih secara acak dari beberapa nama produk madu dengan kode awal nomor registrasi yang sama. Parameter SNI yang diuji meliputi: organoleptis, kadar air, kadar abu, kandungan padatan tidak larut air, keasaman, kadar  5-hydroxymethyl-furfuraldehyde (HMF), aktivitas enzim diastase, cemaran logam (Pb,Cd, Hg, As), kadar batas kloramfenikol, cemaran mikrobiologi, kadar gula pereduksi dan sukrosa. Hasil: Resume hasil penelitian ini adalah: sampel dengan kode awal MD dan P-IRT memenuhi persyaratan  parameter SNI, kecuali kadar HMF melebihi batas yang ditetapkan. Sampel dengan kode awal ML memenuhi persyaratan SNI kecuali kadar HMF dam kadar abu. Sampel dengan kode awal TR memenuhi persyaratan SNI kecuali kadar air. Kesimpulan: Semua sampel madu memenuhi 12 dari 13 persyaratan parameter mutu SNI-01-3545-2013.

Abstract

Pendahuluan: Ibuprofen (IBP) banyak digunakan sebagai obat anti piretik dan pereda nyeri, obat ini sensitif terhadap oksidasi dan suhu. Cemaran IBP yang keberadaannya dalam tablet harus memenuhi syarat uji batas adalah 4-isobutilasetofenon (IBP RC C) dan 2-(4-isobutirilfenil) asam propanoat (IBP RC J). Variasi matriks tablet seringkali membuat prosedur standar untuk analisis cemaran perlu dioptimasi karena dapat menyebabkan kinerja kromatografi yang tidak memenuhi syarat kompendia. Tujuan: Penelitian ini bertujuan untuk memperoleh kolom dan kondisi operasional KCKT yang selektif, efisien dan memenuhi persyaratan kompendia untuk pengujian IBP dan cemarannya. Metode: Penelitian ini dirancang untuk membandingkan kinerja kromatografi (resolusi, faktor ikutan dan jumlah lempeng teoritis) beberapa kolom KCKT C18 yang sesuai persyaratan United States Pharmacopoeia (USP) untuk penetapan cemaran organik dalam tablet ibuprofen. Instrumen KCKT yang digunakan adalah Shimadzu Prominence dengan detektor Photo Diode Array (PDA). Kondisi analisis optimum yang diperoleh diaplikasikan untuk analisis 3 macam sediaan tablet IBP. Hasil: Kondisi optimum diperoleh melalui kolom C18-C 4,6 x 250 mm, 5 µm,  menggunakan fase gerak larutan asam kloroasetat 1% pH 3,0 : asetonitril (45:55, v/v), laju alir 1,5 mL/menit, suhu kolom ambient, detektor PDA      254 nm, volume injeksi 10 µL dengan waktu analisis 11 menit. Diperoleh puncak-puncak yang simetris dengan kinerja kromatografi yang memenuhi persyaratan USP. Kesimpulan: Metode yang telah dioptimasi berhasil diaplikasikan pada 3 sediaan tablet yang beredar di pasaran dan dapat digunakan untuk pengujian rutin dalam rangka pengawasan kualitas produk obat tablet ibuprofen.

Abstract

Background: TLC-contact bioautography is one of an effective method for identification antibiotics, by which many antibiotics could be identification and determination simultaneously. Objective: To evaluate kanamycin sulfate in injection preparations based on its inhibitory activity against Escherichia coli ATCC 8739 as test organism. Methods: Sample and standard solutions were spotted onto TLC silica gel 60 F254 plate and developed in 10% potassium dihydrogen phosphate solutionas as mobile phase. The TLC-contact bioautography method was  validated according to USP guidelines by considering specificity, LOD, LOQ, linearity, accuracy and precision parameters. Results: The TLC-contact bioautography method was found to be high sensitivity with LOD of 0.75 µg and LOQ 2.31 µg. Linearity range of 100-350 µg/mL with r = 0.9993 and linear regression equation was y = 0.0019x + 0.0338. The recovery obtained from addition of blank samples by three different concentrations of kanamycin sulfate standard was 101.40% ¬+ 2.02%. The precision of the method was good with coefficient of variation 0.080%. The TLC-contact bioautography method was supported by determination of kanamycin sulfate potency ratio in the injection preparation and kanamycin sulfate standard using 3-3 design. Random block design obtained the potential for kanamycin sulfate in injection preparations compared to kanamycin sulfate standard was 100.6%.  Conclusion: The TLC-contact bioautography for kanamycin sulfate in injection preparations could be applied to the quality control analysis of the investigated drugs.

Abstract

Introduction: Various studies have shown the benefi cial effects of green tea, not only on cardiovascular diseases but also on type 2 diabetes.Methods: In this study, the preparation of green tea water extract has been standardized to (-)-epigalocatechin gallate (EGCG), the major component of green tea. The role of green tea water extract on blood fl uidity and diabetes diseases has been studied in 13 Fructose-Fed Rat (FFR). The rats were given high fructose diet ad libitum for one week and then combination with green tea water extract every day for 6 days.Results: The results show, green tea water extract can reduces 100 μL blood passage times of wistar rat signifi cantly (p<0.01) by Micro-Channel Array Flow Analyzer (MC-FAN) instrument. Green tea water extract also had strong effect in reducing abdominal fat (p<0.05), blood glucose level (p<0.01) and body weight (p<0.01).Conclusion: These results suggest that green tea water extract may has benefi cial effects for the treatment of diabetes and reduce blood viscosity.

Abstract

Purwodadi sub-district has a surplus of dairy milk with a production of 25 tons of milk per day per KUD. There are 4 milk industry in Purwodadi sub-district as suppliers of PT Nestle. The problem is, dairy products are not entirely absorbed by the market, so it is necessary to look for breakthroughs in the distribution of dairy products that have an average price of IDR 8,000-13,000 per liter. Diversification of milk into fermented milk products will increase prices by five times, so that it is expected to increase people's income, because not only is the product variant increasing, but the market is also increasing. The result of the training shows that all participants experience a significant increase in knowledge, indicated by higher post-test result compared to the pre-test result. Another result known is that this probiotic-fermented milk can be used as a medium for a source of economic improvement for the family. This is proven when yogurt made by the community of Purwodadi has been sold among them as an additional drink during community meetings. Apart from that, this training will create a climate that enables the potential of the Youth Organization (Karang Taruna) and Family Welfare Program (PKK) to develop, namely to build group creative power by encouraging, motivating, and raising awareness of the potential of the village and efforts to develop it.

Abstract

Wonorejo village in Batu city is known as a vegetable and fruit producing area. Another popular product of Batu mostly is fresh dairy milk and only a small amount of processed products while the cow milk collected in KUD Batu has reached 16-17 tons per day. The problem was that the target community had no experiences in processing cow milk using probiotic microbes. The purpose was transfering of science and technology about the production of probiotic milk combined with fruit juice. This program was specifically directed to increase the empowerment of Kelompok Wanita Tani "Wonoasri" in Wonorejo, in developing of fruity- probiotic dairy products to increase the diversification of products with higher economic value. The method included workshop and technical assistance, which were carried out in a serie of steps: 1) exploring the knowledge of target by surveys and pre test, 2) lecturing and discussion, 3) demonstrating how to prepare and characterize the probiotic milk products, 4) training the preparation of product, 5) product characterization and packaging assistance, 6) evaluation. The results of this program were the target community gained increase in knowledge and experience, and they could produce fruity probiotic milk. In addition, we expected that they would increase their understanding and enthusiasm to appreciate the potency of their territory in order to improve the nation's competitiveness. abstrakDesa Wonorejo di kota Batu dikenal sebagai daerah penghasil sayur dan buah. Produk populer lain dari Batu adalah susu segar dan sejumlah kecil produk olahan.  Susu sapi dikumpulkan di KUD Batu. Masalahnya adalah bahwa komunitas target tidak memiliki pengalaman dalam memproses susu sapi menggunakan mikroba probiotik. Tujuannya adalah mentransfer ilmu pengetahuan dan teknologi tentang produksi susu probiotik yang dikombinasikan dengan jus buah. Program ini diarahkan untuk pemberdayaan Kelompok Tani Wanita "Wonoasri" di Wonorejo, dalam mengembangkan produk susu buah-probiotik untuk diversifikasi produk dengan nilai ekonomi lebih tinggi. Metode tersebut termasuk lokakarya dan bantuan teknis, dalam serangkaian langkah: 1) mengeksplorasi pengetahuan target dengan survei dan pra-tes, 2) ceramah dan diskusi, 3) menunjukkan bagaimana mempersiapkan dan mengkarakterisasi produk susu probiotik, 4) pelatihan persiapan produk, 5) karakterisasi produk dan bantuan pengemasan, 6) evaluasi. Hasil program ini adalah komunitas sasaran memperoleh peningkatan dalam pengetahuan dan pengalaman, dan mereka dapat menghasilkan susu probiotik buah. Selain itu, kami berharap bahwa mereka akan meningkatkan pemahaman dan antusiasme mereka untuk menghargai potensi wilayah mereka untuk meningkatkan daya saing bangsa.

Abstract

Roselle (Hibiscus sabdariffa L.) contains cyanidin-3-rutinoside, delphinidin, delphinidin-3-monoglucoside, cyanidin-3-monoglucoside, cyanidin-3-sambubioside, cyanidin-3,5-diglucoside may inhibit the growth of Extended-Spectrum-Beta-Lactamase (ESBL) Escherichia coli, but there is no research reported the determination of MIC of Roselle (Hibiscus sabdariffa L.) extract powder against ESBL E.coli ATCC 6110 and ATCC 5949 and their potency ratio compared to meropenem. This study aimed to determine the MIC of Roselle flower extract powder on the growth of ESBL E. coli ATCC 6110 and ATCC 5949 and determine the potency ratio compared to meropenem. This study used two methods in determining MIC, namely the agar diffusion method and the dilution method with Nutrient Agar media which was incubated at 37 ±1°C for 24 hours, while the determination of the potency ratio was carried out by diffusion method with the same media which was incubated at 37 ±1°C for 24 hours. The results obtained were diameter of inhibition zone (mm) which were then observed and analyzed to calculate the potency ratio. The results showed that the MIC of Roselle flower extract powder was obtained at a concentration of 12,500 ppm by diffusion method and at a concentration of 3,125 ppm by dilution method with 24 hours incubation at 37 ± 1o C and the potency ratio of Roselle flower extract powder compared to meropenem was 89.7% and 97.97% against ESBL E.coli ATCC 6110 and ESBL E.coli ATCC 5949 respectively.
