Abstract

Ethyl p-methoxycinnamate, major ingredient of Kaempferia galanga rhizome, have been reported not only has analgesic – anti inflammatory activities like NSAIDs which inhibited cyclooxygenase, but also inhibit tumor cell proliferation in specimen of mouse epidermis. Therefore, it will be interesting to carry out  synthetic studies on the derivates of ethyl  p-methoxycinnamate and searching their citotoxic activity on WiDr cell. We wish to report of structure modification on carboxyl moiety of  ethyl p-methoxycinnamate  and  evaluation on their citotoxic activity  on WiDr cell. Isolation of ethyl p-methoxycinnamate from Kaempferia galanga rhizome was carried out by percolation with ethanol 96% as solvent. Hydrolysis of ethyl p-methoxycinnamate in basic condition was performed to obtain p-methoxycinnamic acid. Preparation of some thiourea derivates of ethyl  p-methoxycinnamate was carried out  by microwave irradiation. Citotoxicity assay was carried out by MTT method for 48 h.   Modification  of  carboxyl  group  of  ethyl  p-methoxycinnamate to its thiourea form could be carried out by microwave irradiation gave; (E)-3-(4-methoxyphenyl)-N-(phenylcarba- mothioyl)acrylamide (50%); (E)-3-(4-methoxyphenyl)-N-(4-methoxyphenylcarbamothi- oyl)acrylamide (26%) and (E)-3-(4-methoxyphenyl)-N-(4-methylphenylcarbamothioyl) acrylamide (54%), yield calculated for 2 step from the acid chloride. All compounds showed no citotoxic effect on WiDr cell at 48 h incubation.

Abstract

Community Service Activities (PENMAS) were held in August 2018 in Kapas, Bojonegoro with a target audience of SMKN 4 communities, totaling 107 people, consisting of students majoring in Cooking, members of Dharma Wanita, Chefs from 2hotels, several UMKM Food and Beverages around the school, teachers and partisipants from other schools. The activity was also attended by the Principal, the Head of the Education Office who witnessed the signing of the MOU between the PENMAS Team and the school. One of the Departments managed by SMK 4 is Tata Boga with thelearning outcome to prepare entrepreneurs or businessmen of food products. As a vocational school that has held the ISO 9001 2008 certificate, the school is obliged to prepare graduates according to the demands of their competence. Based on preliminary studies, information was obtained that there were several problems related to food consumption and production and government regulationin the case of traded food businesses. The training materials provided in this training were: Food business production regulation (1), How to make good food (2),Ingredients additional allowable food (3), Prohibited food additives and substitute alternatives (4), Good packaging methods and techniques for food and beverage (5), Halal Assurance System (SJH) and Halal food product Certification Process (6) and Simple marketing for food business (7). Based on the evaluation of the pre-test and post-test resulted that after the training participants' understanding showed an increasing trend. Three topics that showed a significant increase in understanding were: Requirements for clean and healthy food (1), Introduction of food coloring logos (2), and Examples of halal food ingredients (3). PENMAS activities will be followed up with intensive coaching activities for students of SMKN4 which are included in the group of beginner beverage food businesses.

Abstract

Abstract: The use of medicinal plants to maintain or overcome health problems has long been carried out by worldwide community. Along with the development of knowledge, data about the existence of undesirable effects or side effects associated with the use of medicinal plants that are not appropriate are obtained. This research is conducted to determine public knowledge about the safety several medicinal plants use. The study was conducted with a questionnaire technique in Lombok Praya, Indonesia. The results show that knowledge about the safety of the use of medicinal plants in breastfeeding mothers obtains the correct answer by 48%, knowledge about the parts of medicinal plants that are dangerous for consumption by 39% and knowledge about medicinal plants that can increase health risks by 21%. Whereas, the knowledges for the use of medicinal plants in pregnant women and the use of medicinal plants that are not right obtains very good results with correct answers of 79% and 81%. Based on further interviews, these good results are obtained because they are supported more by traditional use of the plant that has already existed in the area. Public understanding of the safety of some medicinal plants in Lombok Praya is still lacking. It is necessary to add knowledge about the use of medicinal plants that requires special attention, especially on plants that are often used by people in the area. Local health agencies need to disseminate information on the safety of medicinal plants that is wider and more sustainable. Keywords: Community, medicinal plant, safety, public understanding.

Abstract

Pendahuluan: Salah satu usaha meningkatkan aktivitas asam p-kumarat adalah meningkatkan sifat lipofiliknya. Reaksi benzoilasi gugus fenol asam p-kumarat merupakan salah satu strategi meningkatkan sifat lipofilik. Iradiasi microwave merupakan salah satu alternative metode untuk meningkatkan efisiensi dalam reaksi organik. Selain memiliki kelebihan mempercepat reaksi kimia, penggunaan iradiasi gelombang mikro juga dapat menyebabkan kerusakan struktur kimia produk maupun terjadinya reaksi samping lainnya. Tujuan: Mendapatkan kondisi optimum reaksi sintesis asam 4-benzoiloksisinamat. Metode: asam 4-benzoiloksisinamat disintesis dengan cara mereaksikan asam p-kumarat, benzoil klorida dan piridin sebagai katalis. Reaksi benzoilasi dilakukan dengan iradiasi gelombang mikro pada daya 180, 270, dan 360 Watt. Karakterisasi asam 4-benzoiloksisinamat dilakukan menggunakan alat  spektrometer UV-Vis, FT-IR dan 1H–NMR. Hasil: Persentase hasil yang diperoleh pada daya 180, 270, dan 360 Watt berturut-turut adalah sebagai berikut 49,3%, 58,5%, and 47,7%. Terdapat perbedaan yang signifikan persentase hasil yang diperoleh pada daya 270 Watt dengan persentase hasil pada daya 180 Watt dan 360 Watt, tetapi tidak ada perbedaan bermakna persentase hasil pada daya 180 Watt dan 360 Watt. Persentase hasil terbesar diperoleh selama enam puluh detik pada daya 270 Watt, yaitu 61,4%. Jika reaksi dilakukan lebih dari enam puluh detik, terjadi kerusakan struktur senyawa target. Kesimpulan: Kondisi optimum sintesis asam 4-benzoiloksisinamat menggunakan iradiasi gelombang mikro adalah selama enam puluh detik pada daya 270 Watt.

Abstract

Background: Urea derivatives showed good analgesic activity compared to diclofenac sodium.  The addition of the allyl group to the thiourea and 4-tertiary-butylbenzoyl chlorideis expected to provide a better analgesic effect. Objective: The research aimed to synthesize 1-allyl-3-(4-tertiary-Butylbenzoyl) Thiourea and determine its analgesic activity in mice (Mus musculus). Methods: The synthesis was carried out by a modified Schotten-Baumann reaction, via nucleophilic substitution reaction of allylthiourea on 4-tertiary-butylbenzoyl chloride.  A writhing test was performed to observe analgesic activity in the test compound.  Confirmation of the structure of pure 1-allyl-3-(4-tertiary-Butylbenzoyl) Thiourea was obtained through UV, IR, 1H-NMR, and 13C-NMR data. Results: The compound showed better pain inhibition activity compared to diclofenac sodium, with ED5019,018 mg/kg BW. Conclusion: The compound 1-allyl-3-(4-tertiary-butylbenzoyl) thiourea showed better analgesic activity than diclofenac sodium.

Abstract

Background: Cinnamic acid and its derivatives have been widely studied for their efficacy because of the pharmacological effect on good health and well being. Microwave irradiation is more time effective to synthesize than conventional heating method because it conducts heat faster and shortens the reaction time.  Objective: This study aimed to synthesize 4-hydroxy-3-methoxycinnamic acid using microwave irradiation and its antiplatelet activity by blood clotting time method. Methods: Synthesis of 4-hydroxy-3-methoxycinamic acid with malonic acid and 4-hydroxy-3-methoxybenzalehyde as a starting material using ammonium acetate catalyst with microwave irradiation (960 Watt, 4 minutes). The synthesis results were tested for purity by thin-layer chromatography, a melting point determination and structure identification (UV-Vis, infrared, and proton NMR spectrometry). The antiplatelet activity test consisted of a negative control group CMC-Na, a positive control acetosal, cinnamic acid, and 4-hydroxy-3-methoxycinnamic acid, each group consisted of 3 different doses, namely 0.0037 mmol/Kg (I), 0.0069 mmol/Kg (II) and 0.0139 mmol/Kg (III). Results: Synthesis of 4-hydroxy-3-methoxycinamic acid had a yield percentage of 30.55%. The test results showed that the 4-hydroxy-3-methoxycinnamic acid compound has antiplatelet activity with an ED30 value of 1.3080 mg/Kg BW and antiplatelet activity comparable to acetosal. Conclusion: 4-hydroxy-3-methoxycinamic acid can be synthesized by microwave irradiation and had antiplatelet activity 1.7 fold greater than cinnamic acid.

Abstract

Abstract. The independence of food supply in Indonesia is supported by the abundant diversity of natural ingredients from vegetable, animal and mineral sources. Compared to drugs, dependence on food products in other countries is relatively lower, because the fabrication requires facilities and infrastructure that can be fulfilled domestically. The regulatory system managed by the Republic of Indonesia Drug and Food Control Agency (BPOM-RI) has been implemented to ensure that food products consumed by the public are of high quality, nutritious, safe, halal, and affordable and with a taste that satisfies user communities. Circular-worthy products must at least meet the registration requirements of halal aspects, maximum microbial limits and chemical contamination. The growth of Micro, Small and Medium Enterprises (MSMEs) is also facilitated by the government. Dharma Boga is one of the Small and Medium Enterprises (UKM) in Klangon sub-district, Bojonegoro which is engaged in food production for more than 10 years. Some problems related to registration, halal certification and handling of production, especially food additives that meet requirements, has been communicated to the Community Service Team (PENMAS) Faculty of Pharmacy, Airlangga University (FFUA). Alhamdulillah, PENMAS activities with training materials on how to produce and register good and halal food have been carried out with satisfactory results, although it still needs to be followed up with a partnership for the MSME revitalization program in the future. Of the three parameters used to measure participants' understanding of training material, the average obtained more than 30% showed an increase. For the introduction of the halal logo and the dangers of formaldehyde, 100% of the message is understood. The training involved experts and stakeholders in the authority of the East Java Indonesian Ulama Council and the teaching staff and the Airlangga University Faculty of Pharmacy Testing Service Unit.         Abstrak. Kemandirian penyediaan pangan di Indonesia didukung oleh diversitas bahan alam yang melimpah baik dari sumber nabati, hewani, maupun mineral. Dibandingkan obat, ketergantungan produk pangan pada negara lain relatif lebih rendah, karena fabrikasinya membutuhkan fasilitas dan infrastruktur yang dapat terpenuhi di dalam negeri. Sistem regulasi yang dikelola oleh Badan Pengawas Obat dan Makanan Republik Indonesia (BPOM-RI) telah diberlakukan untuk menjamin produk makanan yang dikonsumsi masyarakat berkualitas, bergizi, aman, halal, dan terjangkau serta dengan rasa yang memuaskan selera masyarakat. Produk yang laik edar minimal harus memenuhi persyaratan registrasi dari aspek halal, batas maksimum mikroba dan cemaran bahan kimia. Tumbuh kembangnya Usaha Mikro, Kecil dan Menengah (UMKM) juga difasilitasi oleh pemerintah. Dharma Boga adalah salah satu Usaha Kecil Menengah (UKM) di kecamatan Klangon, Bojonegoro yang bergerak dibidang produksi makanan selama lebih dari 10 tahun. Beberapa permasalahan terkait registrasi, sertifikasi halal dan penanganan produksi terutama bahan tambahan makanan yang memenuhi persyaratan, telah dikomunikasikan kepada Tim Pengabdian kepada Masyarakat (PENMAS) Fakultas Farmasi Univeritas Airlangga (FFUA). Alhamdulillah kegiatan PENMAS dengan materi pelatihan cara produksi dan registrasi pangan yang baik serta halal telah dilaksanakan dengan hasil yang memuaskan, walaupun masih perlu ditindaklanjuti dengan jalinan kerjasama untuk program revitalisasi UMKM di masa mendatang. Dari tiga parameter yang digunakan untuk mengukur pemahaman peserta terhadap materi pelatihan, rerata yang diperoleh lebih dari 30% menunjukkan peningkatan. Untuk pengenalan logo halal dan bahaya formalin, 100% peserta sudah paham. Pelatihan melibatkan pakar dan pemangku kewenangan dari Majelis Ulama Indonesia Jawa Timur dan para staf pengajar serta Unit Layanan Pengujian Fakultas Farmasi Universitas Airlangga.

Abstract

P2Y12 is a platelet receptor that is involved in ADP signal transduction and is an attractive target for antithrombotic drugs. The side effects of antithrombotic drugs are not pleasant for the patient, so research into the development of new antithrombotic agents is still necessary. Evaluation of absorption, distribution, metabolism, elimination, and the toxicity profile of candidate drugs is an important step in drug development. The aim of this study was to predict the potency of ferulic acid (FA) and its derivatives (FA1-24) as antiplatelet drugs by a docking study on the P2Y12 receptor (PDB ID: 4PXZ) and their ADMET performance. The docking study was performed using Molegro Virtual Docker, version 5.5. ADMET prediction of FA was conducted using the pkCSM online tool. The results of the in silico study showed that FA-19 had the lowest MolDock score (MDS), which means that this compound is predicted to have the greatest activity. FA-19 is also predicted to be practically non-toxic. It is expected that FA-19 will have good intestinal absorption and is similarly distributed in the intestine and in the blood plasma. Its penetration in the blood-brain barrier is moderate but does not inhibit the CYP2D6 and CYP3A4 enzymes.
