Abstract

ABSTRACTColletotrichum musae (Berk and Curt.) Arx. is a pathogen that causes anthracnose disease in bananas. Biological control is an alternative because it is friendly to the environment and reduces the negative impact of synthetic pesticides. The objective of this study was to obtain yeast isolates from bananas potentially used as biocontrol agents. The research was conducted at the Laboratory of Faculty of Agriculture Faculty of University of Tanjungpura Pontianak from September 2017 to February 2018. All the obtained yeast isolates were tested as antagonistic potential against C. musae through dual culture method and viability test of conidia C. musae. The hypovirulent test of yeast isolates that have potential as biocontrol agents are carried out on cucumber. The isolation result obtained 9 isolates of yeast. Based on antagonistic test, yeast isolates had inhibition between 53,42-78,67%. Viability test of conidia C. musae on media that has been shed in yeast cells as much as 106 cells/mL one day earlier showed that conidia still germinate 97.5-100%. Calculation of Severity Index of Disease in hypovirulent test for all isolates of yeast showed 0%. Keywords: antagonist, Colletotrichum musae, hypovirulen, spore germination, yeast

Abstract

ABSTRACTRice (Oryza sativa L.) is one of the world's major food crops. Its used as food most for almost all Indonesian people. Productivity of rice plants tend to fluctuate, one of them caused by Pyricularia oryzae. It caused as leaf blast or blast disease on rice. The aim of this research is to get the rice phyllosphere bacteria which possibly have potential as the biocontrol agent to P. oryzae. This research was conducted at Plant Disease Laboratory Faculty of Agriculture, University of Tanjungpura Pontianak. The study was divided into three term, which were leaf sampling, microbial isolation and antagonism testing. There were found 21 isolates of rice leaf filosphere bacteria, there were 12 isolates have capability to decrease growth of pathogens, and 14 isolates had chitinolytic activity. The 5 isolates as a superior biocontrol agent were not as plant pathogenic based on HR test om tobacao.Keywords: Antagonism, Exploration, Fhyllosphere, Chitinolytics.

Abstract

ABSTRACTColletotrichum musae (Berk. and Curt.) Arx. is a pathogen that causes anthracnose on bananas. Biocontrol agents is one of the alternatives to reduce negative impact of pesticide. This study aims to explore the phylosphere bacteria in bananas which are potential as antagonists to C. musae. The study was carried out at the Laboratory of Plant Diseases, Faculty of Agriculture, University of Tangjungpura Pontianak from November 2017 to April 2018. The bacterial isolates obtained were tested for antagonistic potential against C. musae by dual culture, conidial viability test C. musae and ability to lyse the hyphae. Hypersensitivity testing of bacterial isolates which have the potential as biological agents carried out on tobacco plants. The results of isolation of phylosphere bacteria obtained 16 bacterial isolates. Based on the testing, 3 isolates NPOB1, BPNB1 and BPNB3 have great potential as a antagonists to C. musae. According to antagonistic test, bacterial isolates had an inhibition between 51,84%-65,18%. The inhibition of conidia germination was 4,85%-39,4%. The ability of chitinolytic enzymes produced was 12,43%-31%. Sixteen phylosphere bacteria isolates were not plant pathogenic based on HR tests on tobacco leaves. Abstract

Pertumbuhan populasi serangga tidak selalu sama, hal ini dikarenakan perkembangan populasi dipengaruhi oleh lingkungan dan makanan. Penelitian ini bertujuan untuk menetukan tanaman inang yang sesuai untuk perkembangan populasi P. citri dan D. brevipes dan membedakan perkembangan P. citri dan D. brevipes pada masing-masing tanaman inang. Penelitian ini dilaksanakan di Laboratorium Pestisida Fakultas Pertanian Universitas Tanjungpura Pontianak selama 5 bulan, yaitu bulan Juli sampai bulan November 2017. Penelitian ini menggunakan 2 jenis kutu putih yaitu P. citri dan D.brevipes. Empat tanaman inang yang digunakan adalah bibit jeruk manis (Citrus sinensis.L.), tunas kentang (Solanum tuberosum L.), buah labu kuning (Cucurbita moschata Duch.), dan buah golden mama (Cucurbita moschata). Hasil penelitian menunjukkan ukuran tubuh imago P. citri dan D. brevipes yang dipelihara pada buah labu kuning lebih panjang dan lebar dibandingkan pada buah golden mama, bibit jeruk dan tunas kentang. Rata- rata populasi P. citri yang dipelihara pada empat tanaman inang yaitu buah labu kuning 295,2 ekor,bibit jeruk 120,4 ekor, tunas kentang  37 ekor, dan buah golden mama 168,5 ekor. Rata- rata populasi D. brevipes yang dipelihara pada empat tanaman inang yaitu buah labu kuning 425,8 ekor, bibit jeruk 29 ekor, tunas kentang 84,2 ekor, dan buah golden mama 143,5 ekor. Laju pertumbuhan populasi P. citri dan D. brevipes pada empat tanaman inang menunjukkan pertumbuhan yang paling cepat dipelihara pada buah labu kuning dengan koefesien regresi pada P. citri yaitu 66,18 dan D. brevipes yaitu 83,42. Hasil penelitian menunjukkan tanaman inang yang sesuai untuk perkembangan populasi P. citri dan D. brevipes yaitu buah labu kuning karena laju pertumbuhan populasi paling cepat dan ukuran tubuh paling besar.

Abstract

 Blast is important disease of rice, and it caused by Pyricularia oryzae Cav. Synthetic fungicide in plant disease control may have negative impacts, so another alternative is needed. Yeast can used as a biocontrol agent. This research aims to investigate the potential of the yeast from leaf phyllosphere of rice paddy as biocontrol agent against P. oryzae. This research  was consisted of yeast exploration, isolation and filtrate antagonistic inhibition essay to P. oryzae, and yeast growth respon due to temperature. Ten isolates successfully obtained and tested as antagonist to P. oryzae. Five isolates, YLP05; YLP07; YLP08; YLP09; YLP10, have percentage inhibition more than 30% at 3 dai (day after incubation) dan more than 50% at 7 dai, but their filtrate had not shown any inhibition activity.  The five isolates had been formed colony on 1st day at 30°C.  Based on diameter and percentage of forming colony, the optimal temperature is different for each yeast isolates. Four isolates which can still grow at 20-40oC those are YPL05, YLP07, YLP09, and, YLP10.

Abstract

ABSTRACT               Chili pepper is one of horticultural crop that is quite important in Indonesia, it has a high economic value. Chili pepper plant productivity tends to fluctuate due to Fusarium wilt disease. This study aims to suppress the development of F. oxysporum disease in chili pepper plant using compost inoculated by Trichoderma spp. which de Decomposers and as biological agent. This research was conducted at Plant Disease Laboratory Faculty of Agriculture, University of Tanjungpura Pontianak. The experiment ased a randomized block design with 4 treatments: (Trichoderma spp. that has inoculated capability as dissolving cellulosa), (Trichoderma spp. Isolates as antagonists, and mixed two isolates Trichoderma spp. cellulose solvent with antagonist), and Control (EM4 as decomposer). This research is divided into two stages, on nursery and planting in the field. Trichoderma both as decomposer and antagonist treatment can suppress disease incidence at nursery 14.21%. composting give Trichoderma spp. has ent been able to stop the development Fusarium attacks on chili papper although the plant can still produce the fruit.

Abstract

ABSTRACTPyricularia oryzaeis one of the important pathogen in rice. Blast disease can decreasing the yield of rice production in various countries in the world. This study aims to determine diease incidence and disease severity of blast at Desa Sungai Itik, both in leaves and  panicles. This research was conducted from July to December 2017. The research was conducted in Desa Sungai Itik Kecamatan Sungai Kakap Kabupaten Kubu Raya. The variables used were disease severity and disease insidence both on leave and panicle.Blast disease was found both at leave and panicles. The disease severity of leaf between 51,85% - 59,25% and  at panicle 48,87% - 71.35%. There was a positive correlation between leaf blast and neck blast.Keywords: Disease Insidence, Disease Severity, Correlation, Pyricularia oryzae, Rice.

Abstract

Penelitian ini bertujuan untuk mempelajari daya adaptasi dan produksi beberapa varietas padi unggul baru pada sawah tadah hujan. Penelitian dilaksanakan di Desa Sungai Dungun Kecamatan Sungai Kunyit Kabupaten Mempawah Provinsi Kalimantan Barat berlangsung dari Desember 2015 sampai April 2016. Penelitian menggunakan rancangan acak kelompok. Perlakuan dalam penelitian terdiri dari 5 jenis varietas padi yakni: A (Inpari 30), B (Inpari 32), C (Inpari 33), D (IPB 3S) dan E (IPB 4S). Masing-masing perlakuan diulang sebanyak 5 kali dan setiap unit percobaanterdiri dari 5 tanaman sampel. Variabel yang diamati dalam penelitian ini meliputi: tinggi tanaman, jumlah anakan, persentase anakan produktif, panjang malai, bobot 1000 butir, jumlah gabah per malai, persentase gabah bernas per malai, jumlah biji total per rumpun, berat gabah per petakan. Hasil penelitian menunjukan bahwa pertumbuhan dan produksinya yang ditanam tidak optimal dibandingkan dengan deskripsinya yang disebabkan oleh faktor genetik dan factor lingkungan. Kendala yang dihadapi saat melakukan penelitian yaitu serangan hama tikus yang mengakibatkan 3 varietas padi tidak dapat dipanen.     Kata kunci: adaptasi, padi, pertumbuhan, produksi, sawah tadah hujan, varietas unggul baru.

Abstract

Karakter morfologi padi lokal beras hitam varietas Senakin dan Beliah sangat diperlukan. Selain memberi identitas, mengumpulkan sifat-sifat unggul yang terdapat pada padi lokal beras hitam dapat dimanfaatkan sebagai program perbaikan varietas. Penelitian ini bertujuan untuk menentukan karakter morfologi fase generatif padi lokal beras hitam varietas Senakin danBeliah yang ditanam di tanah sawah. Penelitian dilaksanakan di kebun percobaan Fakultas Pertanian Universitas Tanjungpura Pontianak selama 67 hari,mulai 14 November 2016 sampai dengan 19 Januari 2017.Penelitian menggunakan pola rancangan acak lengkap (RAL), terdiri dari 2 (dua) perlakuan varietas, Senakin dan Beliah. Setiap perlakuan diulang 8 (delapan) kali, dan setiap ulangan terdiri dari 6 (enam) sampel tanaman. Variabel pengamatan ini adalah waktu berbunga, warna ujung gabah, ekor/bulu ujung gabah, keluarnya malai, tipe malai, tinggi tanaman, umur tanaman, jumlah malai per rumpun, persentase anakan produktif, jumlah gabah per malai, bobot 1000 butir, warna kulit ari berasdan pengamatan lingkungan. Hasil penelitian menunjukan bahwa karakter morfologi dari kedua varietas padi lokal beras hitam varietas Senakin dan Beliah ini memiliki kesamaan pada waktu berbunga, warna ujung gabah berwarna putih, ekor/bulu ujung gabah tergolong indica, tipe malai antara kompak dan sedang, tinggi tanaman, umur tanaman,persentase anakan produktif, dan jumlah gabah per malai. Karakter yang berbeda kedua varietas adalah keluarnya malai, jumlah malai per rumpun, bobot 1000 butir dan warna kulit ari beras. Abstract

Antraknosa merupakan penyakit penting pada cabai. Penyakit disebabkan oleh Colletotrichum sp. Pengendalian menggunakan agens hanyati adalah pilihan yang dapat digunakan untuk menggantikan penggunaaan fungisida kimia  sintetik. Penelitian ini bertujuan untuk mengkaji potensi kamir yang berasal dari filosfer buah cabai sebagai antagonis terhadap Colletotrichum sp. Penelitian ini meliputi eksplorasi kamir, isolasi Colletotrichum sp, uji antagonis, uji filtrat kamir terhadap Colletotrichum sp.. Kamir yang didapatkan dari filosfer buah cabai adalah sebanya 8 isolat, dan semuanya bersifat antagonis terhadap Colletotrichum sp, dengan kisaran antara 47% sampai dengan 53% Uji filtrat dilakukan menggunakan 3 isolat kamir dengan daya  antagonis lebih dari 50% yaitu : isolat Y3, Y6, dan Y8. Hasil uji filtrat kamir menunjukkan isolat Y3 dapat menghambat perkecambahan spora Colletotrichum sp, lebih besar dibandingkan dengan Isolat Y6 dan Y8 Kata kunci : agens hayati, fungisida kimia sintetik, uji filtrat
