Abstract

Tanaman obat sebagai antifertilitas belum banyak digunakan, maka perlu alternatif  obat antifertilitas  terutama pada pria, yaitu beluntas (Pluchea indica). Beluntas dapat digunakan sebagai obat antifertilitas dalam taraf uji pre-klinik (Susetyarini, 2011). Penelitian ini merupakan penelitian lanjutan tentang tanin yang dapat menurunkan potensi fertilisasi spermatozoa tikus (Susetyarini, 2010). Pengkajian tannin daun beluntas dalam mempengaruhi spermatogenesis belum pernah dilakukan, maka perlu dikaji tahapan spermatogenesis  terutama spermiogenesis. Spermiogenesis merupakan proses pembentukan spermatid menjadi spermatozoa. Spermatozoa berperanan dalam proses fertilisasi.  Penelitian ini  bertujuan untuk mengetahui pengaruh tannin daun beluntas terhadap jumlah  sel spermiogenesis  tikus putih jantan dengan berbagai waktu pengamatan. Jenis penelitian yang digunakan eksperimen dengan perlakuan kelompok kontrol (tanpa pemberian tannin), kelompok perlakuan pemberian tannin sebanyak 0,8 ml dengan ke tikus putih jantan dewasa yang diulang 3 kali. Waktu pengamatan 49+3 hari, 49+16 hari, 49+26 hari, 49+36 hari dan 49+49 hari. Tikus putih setiap waktu pengamatan dimatikan dan diambil organ testis serta dibuat preparat histology dengan metode Humason Data dianalisis dengan menggunakan anava dan uji lanjut Duncan. Hasil analisis data menunjukkan bahwa  p<0,05 berarti ada pengaruh pemberian tannin beluntas dengan berbagai waktu pengamatan terhadap jumlah spermatid dan spermatozoa tikus putih jantan. Jumlah spermatid dan spermatozoa  terbanyak pada saat waktu pengamatan 49+3 hari dan jumlah spermatid dan spermatozoa terendah pada saat waktu pengamatan 49+49 hari. Hasil penelitian ini dapat digunakan sebagai sumber belajar pada matakuliah Embriologi dan Reproduksi Hewan.  Kata Kunci : spermiogenesis, tanin daun beluntas, sumber belajar

Abstract

Abstract- Genus Hibiscus is one of genuses in Family of Malvaceae which is spread out through seed, and its generative reproductive organ is flower. One part in flower are female gametophyte (pistil) and male gametophyte (stamen) which produce pollen. Plants that different species in same genus typically has different pollen. Variation in pollen morphology can be used as one of the value in taxonomy. This aims of research are investigating the variation of pollen’s layer morphology of some Hibiscus species.  The method of current study is descriptive. Pollen which were investigated were from three species and one cultivare of Hibiscus, such as Hibiscus rosa-sinensis Mary Forbes, Hibiscus rosa-sinensis L. pink, Hibiscus rosa-sinensis L. yellow, Hibiscus tiliaceus L., dan Hibiscus schizopetalus Hook. f. The method which was used to observe pollen by SEM (Scanning Electron Microscope) with magnification 200x, 400x, and 1000x. Pollen has several characteristics which can be observed, such as aperture, pollen shape, pollen size, and type of exine ornamental. The analysis of data used descriptive qualitative.The result of this research showed that pollen some species of Hibiscus showed some variation pollen characteristics. P/E index of pollen 0.93 to 1.00, with the result the type of pollen shape are oblat spheroidal. Pollen grain size between 100 µm to 175 µm, but it have same type are permagna. The type of exine ornamental were echini, with different type, the lengthiest of echini is 20.6 µm in Hibiscus tiliaceus L., the withiest distance 40 µm of echini is Hibiscus rosa-sinensis L. yellow. Keywords: Genus Hibiscus, pollen, SEM

Abstract

Abstract - Beluntas leaves contain a variety of active compounds, one of the active compounds, namely tannins. Beluntas leaf tannins can reduce sperm fertilization potential of white rats (Susetyarini, 2010a) and decrease the number of white rat spermatogenic cells (Susetyarini, 2012). However, in these studies have not examined the role of tannins in changing levels of arginine cement white male rats. This aims, administration beluntas leaf tannins in influencing the levels of arginine cement white male rats. This type of research is the experiment with the following treatments: a control group (non-treated) and group treated leaf tannins beluntas. Giving beluntas leaf tannins administered orally as much as 0.8 ml (an effective dose of tannins in inhibiting fertilization of previous research) to white rats once daily for 49 +3, 49 +16, 49 +26, 49 +36, 49 +49 days. Observations were made according to treatment, white rat vas deferens dissected and taken to obtain cement rats. Analysis of the amino acid arginine levels using HPLC. Data were analyzed using ANOVA. The results showed that the effect of tannin There beluntas leaves the amino acid levels arginin white male rats (p <0.05). The control group without treatment giving the average levels of the amino acid arginine 1375.5 mg/100g and the group treated leaf tannins beluntas showed no decrease in the levels of the amino acid arginine of white male rats. The results of the study can be used as a learning resource for students of biology for Animal Reproduction course. Keywords:  Beluntas leaf tannins, levels of arginine cement, white male rats
