Abstract

The Present study was designet to investigate the influence of vanadyl sulphate towards GLUT-4 protein activities in skeletal muscle tissue in streptozotocin induced diabetic mice. Twenty five mice divided into five groups i.e. placebo group, diabetic group, and three treatment groups based on vanadyl's doses (5,30 or 100 mg/kg BW, respectively). Diabetic mice model was induced by twice intraperitoneal administration of streptozotocin. The first dose of streptozotocin is 100 mg/kg that were inject in the first day and then dose 50 mg/kg, in the day 14th. Diabetes occured on 21st day after streptozotocin injection and it was shown by increasing blood glucose level from 151.4 ± 25.1 mg/dL to 237.1 ± 33.0 mg/dL. Administration of vanadyl sulphate at the dose of 5,30 or 100 mg/kg BW was significantly reduces blood glucose concentration (p<0,001). The muscular tissue were harvested on the day 28th and stained using routine histology staining, hematoxylin-eosin for morphological qualitative analysis and immunohistochemical examination to observe the activities of GLUT-4 protein in skeletal muscle. The result showed that vanadyl sulphate restore atrophic condition of muscular cells and inhibit cell necrosis in muscular tissue. On immunohistochemical examination, vanadyl sulphate might increased the GLUT-4 protein activities in skeletal muscle in streptozotocin-induced diabetic mice Keywords : vanadyl sulphate, streptozotocin, diabetes mellitus, GLUT-4 Abstrak Penelitian ini bertujuan untuk mengetahui pengaruh vanadil sulfat terhadap aktivitas protein GLUT-4 pada otot skelet mencit yang menderita diabetes mellitus akibat induksi streptozotocin. Sebanyak 25 ekor mencit dibagi menjadi 5 kelompok, yaitu kelompok kontrol, diabtes dan perlakuan yang diberi vanadil sulfat dengan dosis 5,30 dan 100 mg/kgBB. Keadaan diabetes diinduksi dengan pemberian streptozotocin dosis 100 mg/kgBB pada hari pertama dan 50 mg/kgBB pada hari ke -14. Pada hari ke-21 terjadi peningkatan kadar glukosa dari 151,4 ± 25,1 mg/dL menjadi 237,1 ± 33,0 mg/dL. Pemberian vanadil sulfat selama 7 hari akan menurunkan kadar glukosa darah secara signifikan (p<0,001). Otot skelet diambil pada hari ke-28 dan dipreparasi dengan hematoksilin-eosin untuk diamati histologinya dan secara imunohistokimia untuk pengamatan aktivitas protein GLUT-4. Hasil pengamatan menunjukkan pemberian vanadil sulfat akan memperbaiki keadaan atropi dan menghambat nekrosis pada sel otot skelet. Pengamatan secara imonohistokimia menunjukkan bahwa vanadil sulfat dapat meningkatkan aktivitas GLUT-4. Kata Kunci: vanadil sulfat, streptozotocin, diabetes mellitus, GLUT-4

Abstract

Dianella nemorosa Lam. (Liliaceae) is a medicinal plant traditionally used in Papua for treatment of cancer, injury, fracture, inflammation and also for antiseptic. The aim of this study was to evaluate antipriliferative activities of methanol extract of D. nemorosa leaves against HCT+116 (colorectal cell line), C2C12 (adherent mouse myoblast cell line) and 293A (Primary embryonal human kidney transformed with human adenovirus type 5 DNA). Powder of leaves was extracted using methanol. Antiproliverative activities were determined by cell proliferation reagents WST-1 assay for 1 h, 2h and 4h after 72h incubation. The result showed that methanol extract of D.nemorosa leaves showed remarkable antiproliferatiove activities against HTC-116 cell line with IC50 values of 199.31 μg/ml (1h), 197.87 μg/ml (2h) and 161.12 μg/ml (4h). The activities against C2C12 cell line resulted in the IC50 values of 405.51 μg/ml (1h), 435.12 μg/ml (2h) and 394.38 μg/ml (4h), while the IC50 values for 293A cell line were 580.81 μg/ml (1h), 442.21 μg/ml (2h) and 366.74 μg/ml (4h), respectively. Those results indicated that methanol extract of D.nemorosa leaves posses potential antiproliferative activities against HCT-116, C2C12 and 293A cell line. Further study is necessary to investigate the inhibitory mechanism of methanol extract of D. nemorosa leaves on HCT-116, C2C12 and 293A cell lines.Keywords : Dianella nemorosa Lam, cancer cell, HCT-116, C2C12, 293A

Abstract

Bioactivies of n-hexane extract and crude alkaloid extract from stem bark of Actinodaphne macrophylla (Blume) Nees var angustifolia Coordinator. & Valeton (Lauraceae) have been investigated. The plants of Lauraceae family are known to contain active compounds such as alkaloids, lactones, fatty acids, terpenes and essential oils. Phytochemical screening of the bark exhibited that it contains triterpenoids, alkaloids and tannins. N-hexane extract was prepared by maceration and the crude alkaloid extract was prepared by maceration in dichloromethane after the powdered bark was previously moistened with 25% NH4OH. The bioactivity assays involved the test of toxicity against brine shrimp larvas (Artemia salina Leach), cytotoxicity against Murine P-388 cells and antimalarial against Plasmodium falciparum. The results indicated that n-hexane extract was active against shrimps larvas with LC50 value of 85.11 ppm, and against P.falciparum with IC50 values of 1.824 ppm, but inactive against Murine P-388 cells with IC50 values of 43.0 ppm, while the crude alkaloid extract was active against shrimp larvas with LC50 value of 36.31 ppm, against Murine P-388 cells with IC50 values of 5.2 m ppm, and against P.falcifarum with IC50 value of 0.1 ppm.Keywords : Lauraceae, Actinodaphne, bioactivity, alkaloids, extract.

Abstract

The protective effects of isolated compounds of ethylacetate fraction of surian's leaf (Toona sureni BL Merr) towards endothelial cell dysfunction on hypercholesterolemia have been investigated. Hypercholesterolemia was induced by atherogenic cocktail for the duration of 60 days. The protective effect was determined by Endothelia Derivate Relaxing Factor 9EDRF/NO) concentration. The result showed that isolated compounds oe ethyl acetate fraction of surian's leaf was able to protect the endothelia cell damage in hypercholesterolemia that can be seenn by increasing the bioavailability of NO serum (P<0.05).Keywords : flavonoid, cocktail, hypercholesterol and nitrogen mono oxide (EDRF)

Abstract

Bruguiera gmnorhiza is one of mangrove plant that has not been widely studied as potential anticancer compounds. Previous research noted that the methanol extract of the bark of B.gymnorhiza are cytotoxic towards HeLa cells. The purpose of this study was to determine the compounds in the chloroform fraction and cytotoxic activity in HeLa cells. Stem bark of B. gumnorhiza was extracted with methanol, further fractionation by partitioning method, using of the solvent h-hexane and chloroform. Fraction of chloroform was evaporated, the tested cytotoxic activity against HeLa cells by MTT method and apoptotic induction by double staining method. Faction levels are used for 500, 250, 125, 62.5 and 31.25 mg/mL with 24-hour incubation time. Subsequent fractions were identified by GC-MS method. The result showed that the chloroform fraction contains 21 compounds, 6 in the form of fatty acids and terpenoids that have anticancer cytotoxic activity (IC50) of 134 ug/mL with apoptotic mekanism.Keywords: Apoptosis, Bruguiera gymnorhiza, cytotoxic, Fractionation, HeLa cell

Abstract

Sponge-derived fungi are source of natural bioactive compounds. The aim of this research is to test the anti-bacterial and anti-mitotic activities from marine sponge-derived fungus. The sponge-derived fungus (strain TID.24041021-2) has been successfully isolated from sponge (strain TID.24041021-2) collected from the sea around Tidung Island. The morphology of this fungus has been identified and classified in the genus Wangiella sp. This fungus was cultivated in PDA with salinity 25 ppm for 21 days at room temperature, extracted with acetone, partitioned with ethyl acetate, and tested the anti-bacterial activity against Bacillus subtilis, Escherichia coli, and Staphylococcus aureus, and also tested the anti-mitotic activity using root tip cells of Allium cepat. The results showed that crude extract of ethyl acetate could inhibit the growth of B. subtilis 91.30-2.30 mm), E.coli (1.50-3.03 mm) dan S.aureus (2.80-5.0 mm) bacteria. Also, antimitotic activity from crude extract of ethyl acetate showed that there is chromosome abnormalities of 0.76 and 1.38 in extract concentration of 20% and 25%, respectively.Keywords : Sponge-derived fungi, anti-bacterial, anti-mitotic, chromosome abnormalities.

Abstract

The degradation of a plastic film containing polymer syntheticpolystyrene (PS) and biopolymer poly(3-hydroxybutyrate)[P(3HB)] and poly (3-hydroxybutyrate-co-3-hydroxy valerat)[P(3HB-co-3HV)] in various composition in aquous solution by in-vitro and in-vivo has been carried out. The testing was conducted based on immersion test in aquadest, river water, pH 4 solution, 6,8 and 10. Rate degradation measured use linear regression equality and parameter is time degradation 50% (t 50%), time degradation 95% (t 95%) and indicated by the slope (k). The result showed that, degradation of a plastic film containing PS and P(3HB) in pH 6 solution occurred faster, followed by ratio PS/P(3HB) 95:5, PS/P(3HB) 90:10, PS/P(3HB) 85:15 and PS/P (3HB) 80:20, in contrast degradation of pure PS (100:0) not occured. It is also observed that the biodegradation plastic film containing PS/P(3HB-co-3HV) in river water ratio PS/P(3HB-co-3HV) 95:5, PS/P (3HB-co-3HV) 90:10 and PS/P (3HB-co-3HV) 85:15, in contrast degradation of pure PS (100:0) not occurred.Keywords : biodegradation, polystyrene, poly(3-hydroxybutyrate), poly (3-hydroxybutyrate-co-3-hydroxyvalerat), plastic film.

Abstract

Antibacterial activity tests were conducted on the leaf fraction of Aka lambuang (Merremia peltata (L.)Merr.) by dilution method and microplate reader towards Staphylococcus aureus and Escherichia coli. The results showed that activity performed at the minimum inhibitory concentration of 500 ppm and 1000 ppm respectivelyKeywords :  Merremia peltata, antibacterial, microplate reader.
