Abstract

ABSTRACTStudy of Reproduction Aspects of Endemic Fish Bonti-bonti (Paratherina striata)in Lake Towuti, South Sulawesi. Bonti-bonti, is one of endemic fishes in Lake Towuti andLake Mahalona. Bonti-bonti has been utilized by people around the lake for consumption inthe form of 6esh fish, dry fish, and salty fish; as ornamental fish; and as raw material foranimal feed. It should be protected from decreasing of fish population due to increasingexploitation and habitat quality changes. The objective of this research is to know thereproduction aspects of this endemic fish, as basic data for conservation. This research wasconducted in Lake Towuti, South Sulawesi from May 2006 to April 2007 at five stations.Samples were collected using experimental gillnet sized 0.625, 0.75, 1.0, and 1.25 inches.The result shows that testis and ovary was found each in one organ during a period of gonadadevelopment. Gonada maturity, was observed histologically oscured at five maturity stages.The range of egg diameters were from 0.01 to 1.50 mm. The fust gonada maturity period ofmale and female fish were reached at 5.53 and 6.26 cm of total length, respectively. Male andfemale fish gonada maturity (50%) were at 16.78 and 14.61 cm total length, respectively.Gonada maturity index value of male and female were 0.01-3.96% and 0.02-6.77%. Thereproduction ability is related to fecundity which have an effect on number ofjuveniles. Therange of fish fecundity were 818-6,051, which were devided kom female with 11.30-18.33cm total length. Bonti-bonti fish is partial spawner type.Key words : Reproduction aspects, endemic fish, Paratherina striata, and Lake Towuti

Abstract

ABSTRACTPotency of Hypertolerance of Calopogonium mucunoides, Centrosema pubescensand Cajanus cajan . Grown on Gold Mine waste Media Contaminated by Cyanideand Mercury. Cleaning up contaminated environment by using green technology ofphytoremediation is urgently needed in the future. These plants will be utilized ashyperaccumulators for cleaning up the contaminated sites. The results of plant screeningshowed that some plant species, which grow in the contaminated areas, indicated hightolerance and potentially effective in accumulating cyanide and mercury in their rootsand above ground portions. Those plants are Calopogonium mucunoides and Centrosemapubescens. This research aims to examine the potency of three candidate plants i.eCalopogonium mucunoides, Centrosemapubescens and Cajanus cajan, that were grownunder three different media i.e tailing waste of PT ANTAM contaminated by cyanide,public mine or PET1 waste that were contaminated by mercury and non contaminated topsoil as a control. The results showed that all of the species were able to grow undercontaminated media and showed a considerable tolerance by indicated high cyanide andmercury accumulation in their shoot and root. Among those three plants, Centrosemapubescens showed the highest level of cyanide and mercury accumulation i.e 3.29 mg I- CN (in the shoot), 34.72 mg I- Hg (in the shoot) and 17.47 mg lk Hg (in the root),followed by Calopogonium mucunoides i.e 14,97 mg I- CN (in the root). Concentrationratio of CN and Hg accumulation in shoot/root of Centrosemapubescens was >I indicatingthat this plant, according to the definition can beconsidered as a hyperaccumulator.Key Words: Calopogonium mucunoides, Centrosemapubescens, Cajanus cajan, mercury,cyanide, waste

Abstract

The Effect of Anorganic- Iron Supplementation on The Blood Cell of Humpback Grouper Fish (Cromileptes altivelis ) Infected by Kbrio parahaemolyticus This experiment was conducted to find the immunity response of humpback grouper (Chromileptes altivelis) correlated to the anorganic-iron supplement in feed. The blood cell was analyzed as the infection response of F5brioparahaemolyticus. The fish weight 6.9 5 0.22 g/ind. was cultured in 60x40~50cm aquarium and stocked at density 10 ind. / aquarium. After the fish fed by inorganic-iron supplementation at 0,25,50 and 100 mg/ kg for 68 days, then infected by J! parahaemolyticus. The blood cell showed the 50 and 100 mg Felkg inorganic-iron supplement enhanced erythrocytes, hematocrite, hemoglobin, lymphocytes and neutrophil after the 24hand the 72ndh ours infection. Total leucocytes backed to normal after the 7Pd infection and the index of phagocytic was higher with the 100 mg Fe/kg of iron supplementation. 1 Key words: iron (Fe), humpback grouper, feed, bacteria and blood cell.

Abstract

ABSTRACTPurifikasi Parsial dan Karakterisasi Enzim Kitin Deasetilase yang Dihasilkan Bacillus thermoleovorans LW-4-11. Kitin deasetilase yang dihasilkan dari bakteri Bacillus thermoleovorans LW-4-11 telah dimurnikan menggunakan amonium sulfat 70% diikuti dengan perlakuan panas (suhu 70°C selama 1 jam). Kemurnian enzim yangdipisahkan dari medium fermentasi meningkat 4.28 kali dengan aktivitas spesifik sekitar 43.7 mUImg. Enzim memiliki suhu dan pH optimum masing-masing 80°C dan 6.0 dalam substrat 0-hydroxyethylated chitin (glycol chitin). Kitindeasetilase ini relatif tahan panas dengan waktu-paruh sekitar 30 menit pada suhu 80 "C. Enzim dihambat oleh ion Li, ZnZ+, MnZ+, Co2 and Ni pada konsentrasi 1 mM, tetapi diaktifkan oleh EDTA 1 mM.Kata kunci: purifikasi parsial, karakterisasi, kitin deasetilase, B. thermoleovorans LW-4-11
