Abstract

Endosymbiont can be interpreted as microbes that live in colonies in the internal tissues of other higher organisms without causing adverse effects directly on the its host. Endosymbiont organisms have a huge potential to be exploited as a new natural products that are useful in medicine, agriculture, and industry. This research is focused on the antibacterial potential. Ascidians Didemnum moll speciments were collected from Manado waters. The fungal isolates were purified followed by screening their antibacterial activity towards Staphylococcus aureus and Pseudomonas aeruginosa. In this study we isolated 3 fungal specimens Acremonium sp. (ADMB-1, 2 and 3). Three isolates showed potential antibacterial activity. Together with the fungal isolates, we also isolated Bacillus galacosidilyticus from the same host. This bacterial isolate also show antibacterial potency.

Abstract

Abstract: Callyspongia sp. is one of the kinds of sea sponge which is hardly used instead of its potential in order to be used as antibacterial, anticancer, and also antifungi.The objective of the research is to observe the effect of antibacterial of endosymbiont fungi of Callyspongia sp. to Pseudomonas aeruginosa and Ecshericia coli. Method: Method chosen in order to test the antibacterial effect is by pasting the myselia of endosymbiont fungi on the subject. The result of the combination, then, is observed as a measure of zone of resistance in endosymbiont fungi test effect. Result: The result of the observation presents two kinds of endosymbiont fungi, isolated from the sea sponge Callyspongia; Black Myselium Fungi and Brown Myselium. The diameter’s average of zone of resistance in Brown Myselium Endosymbiont Fungi to Pseudomonas aeruginosa is 16,8 mm; the same amount gained to Eschericia coli, while the Black Myselium presents no zone of resistance. Conclusion: Brown Myselium Endosymbiont Fungi has the ability to resist the growth of Pseudomonas aeruginosa and Eschericia coli.Keywords: antibacterial, endosymbiont fungi, Callyspongia sp. sea sponge.)Abstrak: Callyspongia sp. merupakan salah satu jenis spons yang berpotensi sebagai antibakteri, antikanker, dan antijamur yang belum banyak dimanfaatkan. Tujuan: untuk mengetahui ada tidaknya efek antibakteri jamur endosimbion spons laut Callyspongia sp. terhadap bakteri Pseudomonas aeruginosa dan Eschericia coli. Metode: Metode yang digunakan untuk uji antibakteri yaitu dilakukan dengan cara menempelkan miselia jamur endosimbion pada media agar kombinasi yang telah dioleskan bakteri uji untuk mengamati dan mengukur zona hambat yang dihasilkan jamur endosimbion. Hasil: Dari penelitian yang dilakukan, diperoleh dua jenis jamur endosimbion yang diisolasi dari spons laut Callyspongia sp., jamur endosimbion miselium hitam dan jamur endosimbion miselium coklat. Rata-rata diameter zona hambat jamur endosimbion coklat terhadap bakteri Pseudomonas aeruginosa 16,8 mm dan terhadap Eschericia coli 16,8 mm, sedangkan jamur endosimbion hitam tidak memiliki zona hambat. Simpulan: Jamur endosimbion coklat memiliki aktivitas menghambat pertumbuhan bakteri Pseudomonas aeruginosa dan Eschericia coli.Kata kunci: antibakteri, jamur endosimbion, spons laut Callyspongia sp.

Abstract

Abstract: Endophytic fungi is a species of fungi which lives within plant tissues including leaf’s tissues that also harmless for the host plant itself. Isolates of endophytic fungi have a potent antibacterial capability with broad spectrum. The aim of this research is to know whether there is an antibacterial effect of endophytic fungi which isolated from Avicennia marina leaf towards Staphylococcus aureus and Shigella dysenteriae. The method that is used for the antibacterial activity test, done by putting mycelia of endophytic fungi in combination media that has been smeared with Staphylococcus aureus and Shigella dysenteriae. Based on research that has been done, it obtained two types of endophytic fungi isolated from mangrove plant, Avicennia marina leaf. Both endophytic fungi have potential for inhibiting Staphylococcus aureus and Shigella dysenteriae, but endophytic fungi with white mycelium has a better antibacterial effect than endophytic fungi with black mycelium and positive control.Keywords: antibacterial, endophytic fungi, mangrove avicennia marinaAbstrak: Jamur endofit merupakan jamur yang hidup di dalam jaringan tanaman salah satunya yaitu dalam jaringan daun dan tidak membahayakan tanaman tersebut. Isolat-isolat jamur endofit memiliki daya antibakteri yang poten dengan spektrum luas. Penelitian ini bertujuan untuk mengetahui ada tidaknya efek antibakteri jamur endofit daun Avicennia marina terhadap bakteri Staphylococcus aureus dan Shigella dysenteriae. Metode yang digunakan untuk uji antibakteri yaitu dilakukan dengan cara menempelkan miselia jamur endofit pada media agar kombinasi yang telah dioleskan bakteri uji. Dari penelitian yang dilakukan, diperoleh dua jenis jamur endofit yang diisolasi dari daun Mangrove Avicennia marina. Kedua jamur endofit memiliki potensi dalam menghambat kedua bakteri uji tetapi jamur endofit miselium putih memiliki efek antibakteri yang lebih baik dibandingkan dengan jamur endofit miselium hitam dan kontrol positif.Kata kunci: antibakteri, jamur endofit, daun avicennia marina

Abstract

Abstract: Endophytic Fungi are microorganisms that live in the plant tissue systems such as seeds, leaves, flowers, twigs, stems, and roots. Endophytic fungi used in this study was isolated from Mangrove Rhizophora apiculata leaves. R. apiculata is widely grown in coastal areas. This study aimed to determine whether there was an antibacterial effect of endophytic fungi that was isolated from leaves of Rhizophora apiculata against the sample bacteria Pseudomonas aeruginosa and Staphylococcus aureus. This study was conducted in the Pharmacology and Therapeutics Laboratory of Sam Ratulangi University by using experimental methods. The results showed two types of fungi, tusky white endophytic fungi and white filamentous endophytic fungi, isolated from Mangrove Rhizophora apiculata leaves. Both types of fungi showed the presence of bacterial growth inhibition, but the white filamentous endophytic fungi had greater antibacterial effect than the tusky white endophytic fungi.Keywords: endophytic fungi, antibacterial, rhizophora apiculata leavesAbstrak: Jamur endofit merupakan mikroorganisme yang hidup di dalam sistem jaringan tumbuhan seperti biji, daun, bunga, ranting, batang, dan akar. Jamur endofit yang digunakan pada penelitian ini merupakan hasil isolasi dari daun mangrove Rhizophora apiculata. Tanaman mangrove R.apiculata ini banyak tumbuh di kawasan pesisir pantai. Penelitian ini bertujuan untuk mengetahui apakah terdapat efek antibakteri jamur endofit yang diisolasi dari daun Rhizophora apiculata terhadap bakteri uji Pseudomonas aeruginosa dan Staphylococcus aureus. Penelitian ini dilakukan di Laboratorium Farmakologi dan Terapi Universitas Sam Ratulangi Manado dengan menggunakan metode eksperimental. Hasil penelitian ini didapatkan dua jenis jamur yaitu jamur endofit berwarna putih gading dan jamur endofit berwarna putih berserabut yang diisolasi dari daun mangrove Rhizophora apiculata. Kedua jenis jamur menunjukan adanya daya hambat pertumbuhan kedua bakteri uji, namun jamur endofit berwarna putih berserabut memiliki efek antibakteri lebih besar dari pada jamur endofit berwarna putih gading.Kata kunci: jamur endofit, antibakteri, daun rhizophora apiculata

Abstract

Title (Bahasa Indonesia): Senyawa antibakteri spons, Agelas tubulata dan Phyllospongia sp., dari Perairan Teluk Manado, terhadap pertumbuhan beberapa strain bakteri Agelas tubulataand Phyllospongiasp. are marine spongethat contain active compoundsthat have potential to beantibacterial, anticancer and antifungal which have not been used. This study was aimed to examine the sponge extracts which inhibitbacterial growth, i.e. Pseudomonas aeruginosa, Proteus mirabilis, Staphylococcus aureus, Escherichia coli, S. saprophyticus andtoanalyze the active fraction of the antibacterial component using bio-autographic method. Agelas tubulataand Phyllospongiasp. were collected from Manado Bayand method used in the study was Agar Diffusion Method. Results showed that A. tubulataand Phyllospongiasp. extract had antibacterial activity against all 5 bacteria tested. A. tubulataextract had inhibition zone withbroader spectrum than that of Phyllospongiasp. extract, while Phyllospongiasp. has specifity forP. mirabilis.Agelas tubulata danPhyllospongia sp. merupakan spesies spons laut yang mengandung senyawa aktif yang berpotensi antibakteri, antikanker dan antijamur yang belum banyak dimanfaatkan. Penelitian ini bertujuan untuk menguji ekstrak spons laut yang menghambat pertumbuhan 5 bakteri yaitu Pseudomonas aeruginosa, Proteus mirabilis, Staphylococcus aureus, Eschericia colidan Staphylococcus saprophyticus dan menganalisis fraksi aktif antibakteri melalui teknik bioautografi. Spesies spons laut A. tubulata dan Phyllospongiasp. diambil dari Perairan Pantai Malalayang Manado. Metode yang digunakan yaitu metode difusi agar. Hasil dari penelitian ini, yaitu ekstrak A. tubulatadan Phyllospongiasp. mempunyai aktivitas terhadap 5 bakteri uji. EkstrakA. tubulatamemiliki daya hambat dengan spektrum yang lebih luas dibandingkan ekstrak Phyllospongiasp.sedangkanekstrak Phyllospongia sp.memiliki spesifitas terhadap bakteri P. mirabilis.

Abstract

Abstract: Endophytic fungi could be used as an alternative antibiotic because it produced bioactive compounds, which are developed as basic ingredients for medicine such as antibiotic, antioxcide, anticancer. Endophytic fungi can be isolated from Rhizome of Alpinia galanga L. which is abundant in Indonesia. The aim of this study was to find the inhibiting zone of endophytic fungi rhizome of Alpinia galanga L. to the growth of Staphylococcus aueus and Escherichia coli. This was an experimental laboratory study using modified Kirby-Bauer well diffusion technique in Marine Pharmaceutical and Biology Molecular laboratory of FPIK faculty in Sam Ratulangi University. Endophytic fungi were produced by Rhizome of Alpinia galanga L. which are cultured in carbohydrate-rich media. Then, it produced various fungi isolate, which is being used for testing its bioactiy to Staphylococcus aureus and Escherichia coli. The results showed that only 2 isolates of endophytic fungi that have inhibitory and the range of the average value of inhibition zone endophytic fungi rhizome of Alpinia galanga L. against Staphylococcus aureus 19 mm to 21.3 mm, while the Escherichia coli 21.3 mm to 22.3 mm. Conclusion: Endophytic fungi Rhizome of Alpinia galanga L. could inhibit the bacterial growth of Staphylococcus aureus and Escherichia coli.Keywords: Staphylococcus aureus, Escherichia coli, endophytic fungi Abstrak: Jamur endofit salah satu sumber bahan baku obat yang memproduksi senyawa bioaktif potensial dalam menghasilkan efek antibiotik, antikanker, antioksidan. Jamur endofit dapat ditemukan pada tanaman Lengkuas Alpinia galanga L. yang banyak terdapat di Indonesia. Penelitian ini bertujuan untuk mengetahui daya hambat jamur endofit terhadap Staphylococcus aureus dan Escherichia coli. Jenis penelitian ialah eksperimental laboratorik dengan modifikasi Kirby-Bauer sumuran di Laboratorium Biologi Molekular dan Farmasetika Laut FPIK-UNSRAT. Jamur endofit yang dihasilkan dari rimpang Lengkuas Alpinia galanga L. yang dikultur dalam media kaya karbohidrat menghasilkan berbagai isolat jamur yang kemudian diuji bioaktivitasnya terhadap Staphylococcus aureus dan Escherichia coli. Hasil penelitian mendapatkan hanya 2 isolat jamur endofit yang memiliki daya hambat dan kisaran nilai rata-rata zona hambat jamur endofit rimpang Lengkuas Alpinia galanga L. terhadap Staphylococcus aureus 19 mm sampai dengan 21,3 mm, sedangkan terhadap Escherichia coli 21,3 mm sampai dengan 22,3 mm. Simpulan: Jamur endofit rimpang lengkuas Alpinia galanga L. dapat menghambat pertumbuhan Staphylococcus aureus dan Escherichia coli. Daya hambat jamur endofit lebih besar pada Escherichia coli daripada Staphylococcus. Kata kunci: Staphylococcus aureus, Escherichia coli, jamur endofit

Abstract

Abstract: This study was aimed to evaluate the antibacterial effect of Chromodoris dianae on E.coli and S. aureus. This was an experimental study. Samples of Chromodoris dianae was taken from Bunaken waters by diving. Extract of Chromodoris dianae was obtained by using maceration technique with 96% etanol. Antibacterial activity of this extract was tested by using the Kirby-Bauer method. The results showed that the mean diameter of the inhibition zone of E. coli was 22.3±1.5 mm and of S. aureus was 23.0±1.0 mm; both were were less than of ciprofloxacin as the positive control repeated for three times. Conclusionn: Chromodoris dianae has antibacterial effects on the growth of Escherichia coli and Staphylococcus aureus.Keywords: Chromodoris dianae, Staphylococcus aureus, Escherichia coli Abstrak: Penelitian ini bertujuan untuk melihat adanya efek antibakteri dari Chromodoris dianae terhadap bakteri E.coli dan S. aureus. Jenis penelitian ialah eksperimental. Sampel Chromodoris dianae diambil dari perairan Bunaken dengan cara menyelam. Ekstrak Chromodoris dianae dibuat dengan cara maserasi menggunakan etanol 96%. Pengujian aktivitas antibakteri dilakukan dengan menggunakan metode Kirby-Bauer. Hasil penelitian menunjukkan adanya zona hambat terhadap E. coli dan S. aureus dengan rerata diameter 22,3±1,5 mm dan 23,0±1,0 mm, yang lebih kecil daripada rerata diameter kontrol positif siprofloksasin pada tiga kali pengulangan. Simpulan: Chromodoris dianae memiliki efek antibakteri terhadap pertumbuhan bakteri Staphylococcus aureus dan Escherichia coli.Kata kunci: Chromodoris dianae, Staphylococcus aureus, Escherichia coli

Abstract

Abstract: Srikaya leaf contains terpenoid, polyphenol, alkaloid, and flavonoid that can potentially be an antibacterial. This study was aimed to obtain the potency of srikaya leaf extract against Staphylococcus aureus (ATCC25923) and Escherichia coli (ATCC11229). This was an experimental laboratory study using the Kirby-Bauer modified well diffusion technique in the Phytochemistry and Microbiology Laboratory of MIPA Faculty at Sam Ratulangi University. Srikaya leaf extract was obtained by using ethanol maceration technique. The concentrations of the extract were as follows: 50%, 25%, and 12.5%. Ciprofloxacin was used as the positive control while CMC as the negative one. The results showed that CMC did not have any inhibition zone around the well. Ciprofloxacin showed the largest mean diameters of inhibition zones: 35.78 mm against E.coli and 36.55 mm against S.aureus. The mean diameters of inhibition zones of Srikaya leaf extract 50% were 9.13 mm against E.coli and 13.78 mm against S.aureus. The mean diameters of inhibition zones of Srikaya leaf extract 25% were 7.8 mm against E.coli and 13.55 mm agaisnt S.aureus. Meanwhile, the mean diameters of inhibition zones of srikaya leaf extract 12.5% were 7.05 mm against E.coli and 11.31mm agaimst S.aureus. Conclusion: Srikaya leaf extract could potentially inhibit the growth of S.aureus and E.coli. The srikaya leaf extract could inhibit S.aureus more effectively than E.coli.Keyword: antibacterial, srikaya leaf extract, Staphylococcus aureus, Escherichia coli  Abstrak: Daun Srikaya mengandung terpenoid, fenolik, alkaloid, dan flavonoid yang berpotensi sebagai antibakteri. Siprofloksasin memiliki diameter zona hambat yang paling besar. Rerata diameter zona hambat yang dihasilkan oleh siprofloksasin ialah 35,78 mm terhadap bakteri E.coli dan 36,55 mm terhadap S.aureus. Rerata diameter zona hambat ekstrak daun srikaya 50% ialah 9,13 mm terhadap E.coli dan 13,78 mm terhadap bakteri S.aureus. Rerata diameter zona hambat ekstrak daun srikaya 25% ialah 7,8 mm terhadap E.coli dan 13,25 mm terhadap S.aureus. Rerata diameter zona hambat ekstrak daun srikaya 12,5% ialah 7,05 mm terhadap E.coli dan 11,31 mm terhadap S.aureus. Abstract

Abstract: Mangrove has been known long to have many benefits. One of the mangrove species that believed to have antimicrobial effects is Sonneratia alba. The objective of the study was to evaluate any antibacterial effect of the endophytic fungi isolated from the roots of Sonneratia alba toward the growth of Staphylococcus aureus and Eschericiae coli, using the combination method of PDA (Potato Dextrose Agar) and NA (Nutrient Agar). From the result we found two types of endophytes. Black and white colored fungus obtained from Sonneratia alba roots. Both fungi have the effect to inhibit the growth of Staphylococcus aureus and Eschericiae coli bacterial tested.Keywords: endophyte, Sonneratia alba, antibacterial, staphylococcus aureus, escherichiae coli.Abstrak: Tumbuhan bakau sejak lama telah diketahui memiliki banyak khasiat. Salah satu jenis tumbuhan bakau yang diyakini memiliki efek antimikroba adalah Sonneratia alba. Penelitian ini dilakukan bertujuan untuk melihat adanya efek antibakteri dari jamur endofit yang diambil dari akar tumbuhan bakau Sonneratia alba terhadap pertumbuhan bakteri Staphylococcus aureus dan Eschericiae coli. Metode penilitian yang digunakan dengan cara kombinasi PDA (Potato Dextrose Agar) dan NA (Nutrient Agar). Hasil penelitian di dapatkan dua jenis miselia jamur endofit. Jamur endofit berwarna hitam dan berwarna putih diperoleh dari akar tumbuhan bakau Sonneratia alba. Kedua jenis jamur ini memiliki efek daya hambat pada bakteri yang diujikan yaitu Staphylococcus aureus dan Escherichiae Coli.Kata kunci: jamur endofit, Sonneratia alba, antibakteri, staphylococcus aureus, escherichiae coli.

Abstract: Flowers and leaves of jasmine (Jasminum sambac) have chemical substances that contain phytoconstituents such as alkaloids, glycosides, saponins, terpenoids and flavonoids. Flavonoid, an antioxidant, plays some importnat roles in wound healing due to their antibacterial effect. This study aimed to determine the effectiveness of jasmine leaf extract on incision wound healing in rabbits. This was an experimental study using three rabbits as test subjects. Wounds of 0.3 cm depth, 5 cm length, and 0.5 cm widened at both ends of the wound were made on the right and left sides of the back. Incised wound on the left side was treated with jasmine leaf extract ointment meanwhile the wound on the right side was untreated and was covered only with sterile gauze. Wounds were observed for two weeks to evaluate the changes of macroscopical length. The results showed that wounds treated with jasmine leaf extract ointment shrank and their edges united with the surrounding tissues faster than the untreated wounds. Conclusion: Jasmine leaf extract ointment accelerated incised wound healing in rabbits. Keywords : wound healing, incision wounds, jasmine leaves Abstrak: Bunga dan daun melati (Jasminum sambac) memiliki kandungan kimia yang berpotensi farmakologi seperti alkaloid, glycosid, saponin, terpenoid dan flavonoid. Flavonoid berpotensi sebagai antioksidan dan mempunyai kerja yang dibutuhkan dalam proses penyembuhan luka seperti antibakteri. Penelitian ini bertujuan untuk mengetahui efektivitas ekstrak daun melati pada penyembuhan luka insisi kelinci. Jenis penelitian ini eksperimental menggunakan tiga ekor kelinci sebagai hewan uji. Punggung kanan dan kiri kelinci diinsisi sedalam 0,3 cm dengan panjang 5 cm yang dilebarkan 0,5 cm pada kedua ujung sisi luka. Luka punggung kiri diberi salep ekstrak daun melati, luka punggung kanan tidak diberi perlakuan hanya ditutup kasa steril. Pengamatan selama dua minggu untuk melihat perubahan panjang luka secara makroskopik. Hasil penelitian mendapatkan bahwa luka yang diberi salep ekstrak daun melati lebih cepat mengecil serta memiliki permukaan luka yang rata dan menyatu dengan kulit sekitar dibandingkan luka yang tidak diberi salep ekstrak daun melati. Simpulan: Salep ekstrak daun melati memiliki efek untuk mempercepat penyembuhan luka insisi kelinci.Kata kunci: penyembuhan luka, luka insisi, daun melati
