Abstract

Rotifer (Brachionus rotundiformis), sejenis zooplankton asal Sulawesi Utara telah dikaji sebagai produsen senyawa antibioaktif. Tujuan penelitian ini adalah mengembangkan metode kultur rotifer dengan pemberian mikroalga lokal pada beberapa salinitas sebagai induksi produksi senyawa bioaktif; mendapatkan ekstrak kasar senyawa bioaktif dari rotifer dengan pelarut metanol; dan mengevaluasi aktivitas antimikroba pada beberapa bakteri pathogen. Dari rotifer hasil kultur ini telah diekstrak senyawa bioaktif, dan senyawa ini telah diuji aktivitasnya pada 3 jenis bakteri pathogen yaitu Bacillus subtilis, Eschherichia coli dan Vibrio cholerae. Sebagai antibiotik pembanding digunakan tetrasiklin dan amoksisilin. Ada 2 strain yang dicobakan, yaitu strain Manembo-nembo yang pernah dideteksi mengandung senyawa sitotoksik, dan strain Minanga yang belum pernah diuji apakah mengandung senyawa bioaktif atau tidak. Masing-masing strain dikultur pada 3 macam salinitas yaitu 4, 20 dan 40 ppt untuk strain Minanga, dan 40, 50 dan 60 ppt untuk strain Manembo-nembo. Jenis pakan berupa Nannochloropsis oculata yang dikultur dalam medium Hirata dalam kondisi kultur baku di laboratorium. Ekstraksi bioaktif dilakukan dengan metoda baku menggunakan pelarut methanol. Ternyata ekstrak kasar senyawa dari rotifer strain Minanga dan strain Manembo-nembo dengan pakan N. oculata yang dikultur pada beberapa kadar salinitas, semuanya terbukti memiliki aktivitas antibakteri terhadap E. coli, B. subtilis, dan V. cholerae. Respons bakteri terhadap senyawa aktif yang dicobakan secara umum terlihat berbeda menurut jenis. Ekstrak dari rotifer yang dikultur pada salinitas tinggi (40 ppt) dengan pakan N. oculata menunjukkan aktivitas antimikroba paling tinggi. Bakteri yang paling rentan terhadap ekstrak senyawa dari rotifer yang diberi pakan N. oculata adalah B. subtilus .Kata kunci: rotifer, Nannochloropsis oculata, bakteri patogen, antimikroba

Abstract

Basil leaf (O. basilicum L) could be developed as a herbal supplement  due to its phenolic compounds which have antioxidant activity. Extract of this plant would be more effective as a natural antioxidant if it is coated with nanochitosan. Nanochitosan was prepared from the parrot fish (scarus sp) scale and chitosan of crab’s shell by gelatin ionic method. The purpose of this study was to determine  the effects of the nanochitosan coating on phenolic content of basil leaves. The parameters tested were phenolic content of  the basil leaf extracts and nanochitosan coated basil leaf extracts. The results show that the  size’s average of nanochitosan  from fish scales were in 657,4 nm. Phenolic compound was not detected in both the fishscale and crab’s shell derived chitosan. In the other hand, phenolic compound of the basil leaf extract  showed the presence of phenol content of 34.38 mg/kg. In basil leaf extracts coated with nanochitosan from fish scales and nanochitosan of crab’s shell showed a phenolic content of 39.28 mg / kg and 57.44 mg/kg respectively. This suggests that synthesis of phenolic compound was induced when the extract of basil leaves was coated by nanochitosan.Keywords: basil leaves, chitosan, nanoparticles, phenolic compounds

Abstract

An increasing production of avocado in Indonesia  has resulted in abundant seeds as wastes. Avocado’s seeds contain potential antioxidant compound, such as phenolic compound, which could be exploited for human consumption. Utilization of seeds as functional food could be more efficient by coating with a certain substance. Chitosan is a bio-compatible and non-toxic compound, and could be used as coating agent in the form of nanochitosan. The purpose of this study was to evaluate the content of  phenolic-compound in extract of avocado’s seed with and without coated by nanochitosan. In this study, the extract of avocado’s seed was coated with  Parrot fish scale (Scarus sp) derived nanochitosan, compared with nanochitosan from crab’s shell. Nanochitosan was prepared from chitosan by gelation ionic method. The results show that avocado’s seed extract has 44.89 mg/Kg of phenolic content,  much lower than the content in the extract coated with nanochitosan of fish and crab shell, which were 84.08  and 113,67 mg/Kg, respectively.   Therefore, coating with nanochitosan was proven increasing the phenolic compound in the extract of avocado’s seed.  Keywords: avocado, nanochitosan, phenol

Abstract

Konsumsi ikan laut sangat diminati oleh masyarakat Indonesia, termasuk di Sulawesi Utara. Berlimpahnya restoran seafood yang menyediakan ikan laut sebagai menu utamanya dan penjualan ikan laut di pasar turut meningkatkan produk samping dari ikan seperti sisik yang kurang dimanfaatkan secara optimal. Untuk meningkatkan nilai guna dari sisik ikan laut sebagai suatu produk sampingan perikanan, maka perlu adanya riset untuk mengetahui komposisi kimia yang terkandung dalam sisik ikan laut sebagai upaya menyelidiki potensi sisik untuk dapat dijadikan sumber biomaterial yang dapat berguna bagi manusia. Penelitian ini menggunakan sisik ikan yang berasal dari perairan laut yaitu ikan kakatua, kakap merah, napoleon, salem, dan sahamia. Sejumlah 50 gram sisik dari setiap jenis ikan laut yang dikumpul dianalisis proksimat untuk mengetahui kadar-kadar kimia di dalamnya seperti air, abu, lemak, protein, dan karbohidrat. Kandungan sisik ikan laut yang sudah dikeringkan secara umum adalah air 11%, abu 39%, lemak 5%, protein 30%, dan karbohidrat 15%.

Abstract

Program pengabdian kepada masyarakat dengan Program Pengembangan Produk Unggulan Daerah ini telah digagas untuk pemberdayaan masyarakat nelayan melibatkan 2 kelompok mitra Nelayan Bersehati dan Viva Cipta Mandiri di Desa Tiberias Poigar Bolaang Mongondow, dengan tujuan menghilirisasi hasil riset untuk pemberdayaan mitra masyarakat pesisir, meningkatkan mutu produk hasil penerapan teknologi, meningkatkan daya tarik produk unggulan kepada masyarakat luas, mempercepat difusi teknologi dan manajemen masyarakat perguruan tinggi ke masyarakat industri. Untuk tahun pertama 2017 ini, hasil-hasil riset dihilirasi dalam bentuk penerapan teknologi pengemasan dingin dan pengasapan ikan dengan aplikasi kitosan, dengan metode pelaksanaan pelatihan dan pendampingan dalam hal teknis, manajemen dan pemasaran. Luaran kegiatan yang telah dicapai, antara lain peningkatan nilai asset berupa fasilitasi instalasi pengasapan ikan secara terkontrol. Peningkatan nilai laba dengan pendampingan penerapan teknologi pengemasan dingin terhadap produk ikan segar yang dipasarkan mencapai sekitar 20%. Kegiatan pendampingan dalam penerapan teknologi pengasapan ikan telah diunggah ke youtube https://www.youtube.com/watch?v=xaPK4iX4yTE dengan judul video “Pengasapan Ikan Fufu”. Aplikasi nanokitosan pada produk ikan asap telah pula diujicobakan. Produk yang dihasilkan telah dianalisis kadar proksimatnya dan siap untuk diberi label.

Abstract

The concentrations and the distribution of organotin (OT) compounds and booster biocides in sediment and biological samples from Indonesia, Malaysia, Thailand and Vietnam are reviewed. Basically, the concentrations of tributyltin (TBT) in these countries were within the levels that have been reported in developed countries. However, the concentrations of triphenyltin (TPT) and booster biocides were lower than the reported values. Concerning the spatial distribution of antifouling biocides, higher concentrations of OTs were detected in the industrial area and fishing ports. Among ASEAN countries, the highest concentrations of TBT and TPT were observed in Bitung, Indonesia, where many ships from Southeast Asia visit and is moored in the port. The ratio of detected alternative biocide differed from country to country. Especially, many booster biocides were detected and their concentrations were high in Vietnam.

Abstract

This study examined the diversity and abundance of zooplankton in Lembeh Strait, North Sulawesi. Plankton were sampled monthly from September 2004 to February 2005, collected at four stations using a plankton net of 300 pm meshsize with vertical tow of 10 m water column from the bottom. Environmental factors i.e pH, salinity, current velocity, and dissolved oxygen (DO) were measured in situ at the time of plankton sampling. In general, the environmental conditions of Lembeh Strait were in optimal range for zooplankton. There was a similar pattern in the abundance of zooplankton among stations. Crustacea (mainly Copepoda) was the dominant genera among stations, followed by Ciliata and Tunicata. In all stations, the most abundance taxa (72.3%) was Crustacea (21 genera), mainly copepod (18 genera). There were 7 other classes of holoplankton: Ciliata (2 genera), Rhisopoda (1 genus), Hidrozoa (1 genus), Polychaeta (2 genera), Sagitoedea (1 genus) and Tunicata (2 genera). Meroplankton (larva) were only represented by 5 genera. The most common genera were calanoid copepods, such as Calanus spp., Undinula spp., Eucalanus spp., Rhincalanus spp., Paracalanus spp., and Acartia spp. Diversity, evenness and dominance indices showed that Serena Island, a relatively uncontaminated area, was more productive than the other three stations which were considered as relatively contaminated area with anthrophogenic sources.

Abstract

In order to characterize the distribution pattern of rotifers Brachionus spp. in North Sulawesi, sample collections have been conducted at four locations, two at east coast and other two at west coast of North Sulawesi peninsula, which are connected to Maluku and Sulawesi Seas, respectively. At each location, a 40 p.m mesh size plankton net with a 0.3 m opening diameter was horizontally towed at three sites to represent coastal water, estuary and brachkiswater pond. Sampling periods were in January and August 2005 to represent west monsoon and east monsoon, respectively. Each sampling was performed twice a day during high and low tide. Some environmental factors were also measured in situ during the sampling. Three species of Brachionus were found during the survey, Brachionus rotundiformis, B. caudatus and B. quadridentatus. Their abundances were significantly different, both between locations and sampling sites. However, among the similar habitat such as between Manembo-nembo and Minanga brachkishwater ponds, B. rotundiforms abundance was not significantly different. This species was the most abundant compared to the other two species, in average 1984,4 ind/m3, while the other species was as low as 26,1 ind/m3 and 212,9 ind/m3 for B. caudatus and B. quadridentatus, respectively. The abundance of rotifers in northern areas connected to Molluccas Sea is higher compared to those in the southern areas connected to Sulawesi Sea. Distribution of B. caudatus is very similar to those of B. rotundiformis, while B. quadridentatus appears to have its specific pattern. Results of this study suggest that the abundance and species composition of the rotifers reflect trophic status of the aquatic ecosystem in the area. It is still awaiting future studies to elaborate the impacts of the trophic gradients in horizontal distribution of rotifers. Therefore, it is necessary to conduct a long term monitoring with emphasis on the structure of rotifer assemblages in a variety of sites subjected to environmental perturbations, including anthropogenic contaminant inputs.

Abstract

Uji toksisitas tributiltin secara akut telah dicobakan pada kopepoda tropis Apocyclops sp. yang diisolasi dari tambak Manembo-nembo Bitung, Sulawesi Utara. Kopepoda dikultur dalam kondisi laboratorium (25-27oC, 30 ppt dan tanpa penerangan) dengan pemberian mikroalga Nannochloropsis oculata sebagai pakan. Semua individu kopepoda yang digunakan sebagai hewan uji berasal dari sepasang induk jantan dan betina. Kopepoda untuk eksperimen tributiltin (TBT) diberi perlakuan dalam air laut dan selama eksperimen tidak diberi pakan, dan larutan stok TBT-Cl dilarutkan dalam aseton. Pengaruh starvasi (tanpa pemberian pakan) dan aseton diamati sebelum uji toksisitas TBT dilakukan. Setiap eksperimen, 10 kopepoda dewasa (5 jantan dan 5 betina) dari satu kohort dimasukkan ke dalam cawan petri (diameter 3 cm) berisi masing-masing 10 ml air laut. Ternyata perlakuan tanpa pemberian pakan tidak mempengaruhi kopepoda selama periode eksperimen. Dalam uji toksisitas TBT, hanya 3 individu yang dapat bertahan sampai akhir eksperimen (8 jam) walaupun dengan konsentrasi terendah (0.0001 ng.l-1). Kebanyakan individu telah mati sebelum 8 jam diekspos ke konsentrasi TBT 0.01 ng.l-1. Pada konsentrasi TBT yang lebih tinggi (0.1 dan 1 ng.l-1), tingkat kelulusan hidup kopepoda hanya 50% dalam waktu kurang dari satu jam, sedangkan kopepoda yang sisa masih hidup semuanya sebelum mati jam ke-4 yang diberi perlakukan. Dalam uji toksisitas ini, semua konsentrasi yang dicobakan ternyata lebih kecil dari rata-rata konsentrasi TBT di alam (10 ng.l-1). Kisaran konsentrasi TBT yang lebih lebar masih perlu diuji-cobakan untuk mengklarifikasi efek akut TBT agar dapat diperoleh konsentrasi untuk uji toksisitas secara kronis.
