Abstract

Nilem fish (Osteochilus hasselti) is favored by the community because of its chewy meat texture, good taste, and relatively cheap price, so it has the potential to be developed. However, the growth of nilem fish is slow therefore it needs to be given an immunostimulant to increase its growth. Biofloc is known as a cultivation system that can maintain water quality and provide sufficient floc for additional fish feed. This study aims to determine the effect of using turmeric flour in feed to increase the growth of nilem fish reared in the biofloc system. The study used a completely randomized design (CRD), with 3 treatments and 1 control each repeated 4 times. The treatments were supplementation of turmeric flour through feed at a dose of 1.125 g (B), 0.75 g (C), and 0.375 g (D) kg-1 feed and control (A) in cultivation with a biofloc system. The study lasted for 90 days with feeding 3%/kg of biomass weight given 2 times a day, in the morning and evening. Parameters observed in this study were weight (WG) and length growth (LG), daily growth rate (SGR), feed conversion ratio (FCR) and feed efficiency (EF). The data collection method was carried out by weighing and measuring the weight and length of the fish and the amount of feed consumed. The data obtained were collected and analyzed using SPSS 2.0.The results showed that supplementation of turmeric flour through feed reared in the biofloc system could increase weight and length growth, daily growth rate, feed conversion ratio, and feed efficiency. The optimal dose was obtained in the treatment of 1.125 g kg-1 feed (B), proving that supplementation of turmeric flour through feed has the potential to be applied to fish culture using the biofloc system.

Abstract

Air tanah mempunyai manfaat yang penting untuk kehidupan manusia. Salah satu manfaat air tanah dapat digunakan sebagai media budidaya perikanan. Tujuan penelitian adalah untuk menganalisis kesesuaian kualitas air tanah untuk media budidaya ikan lele di Desa Karangsari, Kecamatan Kembaran, Kabupaten Banyumas. Metode penelitian ini menggunakan metode survei. Populasi penelitian adalah air tanah yang berada di Desa Karangsari. Pengambilan sampel menggunakan teknik purposive random sampling. Sampel air tanah  sebanyak 3 titik pengambilan sampel yaitu titik I (lahan tegalan), titik II (lahan pemukiman) dan titik III (lahan rerumputan). Data penelitian meliputi parameter kualitas airtanah, yaitu temperatur air, pH, oksigen terlarut (dissolved oxygen), amonia (NH3), dan nitrit (NO2). Pengumpulan data dilakukan dengan cara pengukuran parameter kualitas air secara in situ dan di laboratorium. Pengukuran parameter kualitas air secara in situ meliputi temperatur air, pH, dan oksigen terlarut, sedangkan  pengukuran parameter kualitas air di laboratorium meliputi amonia dan nitrit. Temperatur air tanah diukur menggunakan thermometer. pH air tanah diukur menggunakan pH meter. Dissolved Oxygen (DO) air tanah diukur menggunakan DO meter. Pengukuran ammonia dan nitrit air tanah dilakukan dengan metode spektrofotometer. Pengolahan data dilakukan menggunakan tabulasi data. Analisis data menggunakan matching data, dengan mencocokkan kualitas air tanah dengan standar kelayakan kualitas air untuk budidaya ikan lele menurut BBPBAT (Balai Besar Pengembangan Budidaya Air Tawar). Hasil penelitian menunjukkan bahwa semua parameter kualitas airtanah pada titik II (lahan pemukiman) dan titik III (lahan rerumputan) sesuai dengan standar kelayakan untuk budidaya ikan lele, namun pada titik I (lahan tegalan) tidak sesuai dengan standar kelayakan, karena kadar ammonia airtanah melebihi ambang batas yang ditentukan menurut BBPBAT.

Abstract

Ikan nila (Oreochromis niloticus) merupakan salah satu komoditas unggulan perikanan yang seiring dengan meningkatnya permintaan di pasaran, ssehingga produktivitasnya harus dipacu terus menerus dengan berbagai teknolgi akuakultur sistem intensif. Penelitian ini bertujuan untuk mengetahui pengaruh pemberian ekstrak bawang putih melalui pakan dalam meningkatkan pertumbuhan ikan nila yang dipelihara dalam sistem bioflok. Penelitian dilaksanakan pada bulan Maret sampai bulan Juni 2021 di Laboratorium Basah Kolam Bioflok. Penelitian menggunakan metode eksperimen berupa Rancangan Acak Lengkap dengan 3 perlakuan 1 kontrol serta 3 kali ulangan. Parameter yang diamati berupa parameter pertumbuhan meliputi pertambahan berat dan panjang, laju pertumbuhan spesifik (spesific growth rate/SGR), konversi pakan (food convertion ratio/FCR), dan efisiensi pakan serta jumlah flok yang terbentuk dalam ember kultur. Parameter pendukung berupa kualitas air yang meliputi suhu dan pH. Data dianalisis menggunakan ANOVA (Analysis of Variance) pada taraf kepercayaan 95%, dilanjutkn dengan uji DMRT (Duncan Multiple Range Test). Data kualitas air dianalisis secara deskriptif. Penambahan serbuk bawang putih (Allium sativum) dalam pakan untuk meningkatkan pertumbuhan ikan nila (O. niloticus) hanya berpengaruh nyata pada pertambahan panjang dan SGR, sedangakan pertambahan berat dan FCR serta efisiensi pakan tidak berbeda nyata. Dosis optimum yang mampu meningkatkan pertumbuhan ikan nila yaitu pada P3 22,5 g/kg pakan. Berdasarkan penelitian ini, pemberian serbuk bawang putih melalui pakan berpotensi meningkatkanpertumbuhan ikan nila yang dipelihara dalam sistem bioflok.

Abstract

Lele dumbo merupakan komoditas ikan air tawar yang mempunyai nilai kompetitif dibanding ikan air tawar lainnya sehingga banyak diminati masyarakat. Tujuan penelitian ini untuk mengetahui pengaruh pemberian ekstrak rumput laut cokelat (Padina sp.) yang dicampur dengan vitamin C dalam pakan terhadap persentase hematokrit dan leukokrit serta differensial leukosit pada lele dumbo (Clarias gariepinus). Penelitian ini menggunakan metode eksperimen dengan rancangan acak lengkap, masing-masing 4 perlakuan dan 3 kali ulangan individu. Perlakuan dalam penelitian ini meliputi dosis P1 (3000 mg ekstrak rumput lau cokelat/kg pakan), P2 (2250 mg ekstrak rumput laut cokelat/kg pakan dan 750 mg vitamin C/kg pakan), P3 (1500 mg ekstrak rumput laut cokelat/kg pakan dan 1500 mg vitamin C/kg pakan), dan P4 (750 mg ekstrak rumput laut cokelat/kg pakan dan 2250 mg vitamin C/kg pakan). Parameter utama berupa persentase hematokrit, leukokrit, dan differensial leukosit, sedangkan parameter pendukung berupa kualitas air yang meliputi suhu, pH, dan DO. Data dianalisis menggunakan uji Analyisis of Variance (ANOVA) dengan tingkat kepercayaan 95%, apabila hasil analisis tersebut menunjukkan adanya perbedaan yang nyata maka dilanjutkan dengan menggunakan uji DMRT (Duncan Multiple Range Test) pada taraf kepercayaan 95%. Hasil penelitian menunjukkan bahwa pemberian ekstrak rumput laut cokelat (Padina sp.) yang dicampur dengan vitamin C berpengaruh nyata (P<0,05) terhadap hematokrit dan differensial leukosit. Nilai hematokrit tertinggi dicapai pada P3 sebesar 48,33%, diferensial limfosit tertinggi pada P3 sebesar 81,29% dan diferensial monosit tertinggi pada P2 sebesar 10,77%. Hasil penelitian menunjukkan bahwa pemberian ekstrak rumput laut cokelat (Padina sp.) yang dicampur dengan vitamin C dalam pakan sebesar 1500 mg ekstrak rumput laut cokelat/kg pakan dan 1500 mg vitamin C/kg pakan merupakan dosis  terbaik dalam meningkatkan imun non-spesifik.

Abstract

Fish farming is well developed because many people like to consume fish. The main problem in cultivation is an attack of a disease. Generally, the disease in fish is MAS (Motile Aeromonas Septicemia) which is caused by Aeromonas hydrophila. Prevention of diseases in fish can be done by increasing immunity through the addition of multivitamins in feed. This study aims to determine the ability of multivitamins against non-specific immune responses in African catfish (C. gariepinus) as well as to know the optimum dose. The study used an experimental method with a completely randomized design using 3 treatments, 1 control, and 4 individual replications. The treatment using a multivitamin through feed with a dose of T1 = 2 g kg-1, T2 = 2.5 g kg-1, T3 = 3 g kg-1 and T0 = control. The parameters observed were phagocytic activity, and differential leukocytes of monocytes and lymphocytes. Data were analyzed using Analysis of variance (ANOVA) at the 95% confidence level. If there is a real difference, then proceed with the Duncan Multiple Range test Test (DMRT) at a 95% confidence level. The results showed that the addition of multivitamins through feed on African catfish had a significant effect (p<0.05) on phagocytic activity and differential leukocyte of monocytes, but had no significant effect (p> 0.05) on differential leucocytes of lymphocyte. The optimum dose for increasing non-specific immune responses in African catfish is T2 at a dose of 2.5 g kg-1 of feed. Based on research, the addition of multivitamins through the feed can potentially be used in fish farming to increase immunity.

Abstract

Ikan mas (Cyprinus carpio L.) merupakan  ikan air tawar yang  banyak dibudidayakan di Jawa Barat dan memiliki harga jual yang relatif tinggi sehingga banyak dibudidayakan secara intensif. Tingginya padat tebar dan konsumsi pakan pada budidaya intensif menyebabkan penurunan kualitas air akibat timbunan sisa pakan dan  ekskresi ikan, menimbulkan stress sehingga ikan mudah terserang penyakit. Penyakit yang sering menyerang ikan air tawar adalah Motile Aeromonas Septicemia (MAS), disebabkan oleh Aeromonas hydrophila. Untuk mengatasinya, pada umumnya dilakukan pengobatan dengan antibiotik, namun dapat mengakibatkan resistensi bakteri sehingga perlu alternatif lain. Salah satu alternatif adalah penggunaan ß-glucan dari ekstraksi ragi roti Saccaromyces cerevisiae untuk meningkatkan imunitas ikan. Penelitian ini bertujuan untuk mengetahui pengaruh penggunaan β-glucan terhadap respon imun non-spesifik pada ikan mas. Penelitian ini menggunakan metode eksperimen dengan rancangan acak lengkap (RAL) terdiri atas 5 perlakuan dan 3 ulangan. Dosis ß-glucan yang digunakan 0 g, 2,5 g, 5 g, 7,5 dan 10 g per kg pakan. Parameter yang diamati yaitu diferensial leukosit meliputi persentase limfosit, monosit dan neutrofil serta aktivitas fagositosis. Analisis data menggunakan Analisis of Varians (ANOVA) dengan tingkat kepercayaan 95% dilanjutkan dengan uji Duncan Multiple Range Test (DMRT) untuk melihat pengaruh antar perlakuan. Hasil penelitian menunjukkan bahwa penambahan ß-glucan melalui pakan berpengaruh signifikan terhadap persentase limfosit, peresentase neutrofil dan aktivitas fagositosis namun tidak berpengaruh signifikan terhadap persentase monosit. Dosis optimum untuk meningkatkan respon imun non-spesifik ikan mas yaitu 5 g/kg pakan.

Abstract

Fucoidan merupakan senyawa polisakarida sulfat yang diekstrak dari rumput laut cokelat Padina sp. Fucoidan diketahui mempunyai beberapa bioaktivitas, salah satunya sebagai imunostimulan. Ikan gurami (Osphronemus gouramy) ukuran + 100 gr diberi perlakuan fucoidan yang dicampur kedalam pakan dengan dosis 0,2 gr kg-1 pakan (P1), 0,4 gr kg-1 pakan (P2) , 0,6 gr kg-1 pakan (P3) serta kontrol (Po) masing-masing dengan 3 kali ulangan. Hasil penelitian menunjukkan, fucoidan dari ekstrak rumput laut cokelat Padina sp. secara signifikan dapat meningkatkan respon imun non-spesifik terhadap persentase hematokrit, leukokrit dan superoksida anion pada ikan gurami. Perlakuan P3 (0,6 gr kg-1) merupakan dosis optimum yang dapat meningkatkan respon imun non-spesifik ikan, menunjukkan fucoidan dapat digunakan dalam budidaya ikan gurami.Kata kunci: fucoidan, Padina sp., ikan gurami, imun non-spesifik

Abstract

The main problem in the cultivation of African catfish is a disease MAS (Motile Aeromonas Septicemia) Aeromonas hydrophila bacteria attack. In addition, other problems that also resulted in the successful cultivation of this fish is the high price of fish feed and the cost of production for the procurement of the feed. Needs to be studied the use of feed raw materials that utilize industrial waste as well as the use of vaccines in controlling disease MAS. This study aims to apply feed made from 3 waste, namely chicken feathers, pulp, and trash fish, then add the vaccine (vaccine feed) and tested in the field. Field test conducted in the village of Bangsa, District of Kebasen, Banyumas. This study uses a completely randomized design (CRD) with 3 treatments and 5 replications. The treatment consisted of P1 = vaccine feed 10 days; P2 = vaccine feed 15 days, and P3 = no vaccine feed (controls). Feed was given as much as 5% of the weight of fish / day. Research using plastic tarp length x width x height = 80 x 60 x 60 cm. African catfish which used a length of 10-13 cm with a weight of 7.3 to 21.1 g. Fish reared for 8 weeks. Parameters measured were immune response such as antibody titer, weight gain and length, specific growth rate (SGR), as well as survival. Parameters measured were supporting water quality parameters include water temperature, pH, and dissolved oxygen. Data were analyzed using Analysis of Variance (ANOVA) and Duncan Multiple Range Test (DMRT test) with a test level of 5%. The results showed that the vaccine could increase the production of antibody titers (P <0.05) compared to controls. The use of artificial feed of chicken feathers, pulp, and trash fish are added the vaccine (vaccine feed) can be applied in the field.Keywords: Aeromonas hydrophila, Banyumas, field test, vaccine feed

Abstract

Abstract - The purpose of this study is to examine fermentation of chicken feather meal with Bacillus licheniformis B2560 to improve the quality of fish feed ingredients. The study was conducted using an experimental method to completely randomized design (CRD) 4 treatments, 3 replications, ie P0 = non- fermented ; P1 = 5 ml inoculum fermentation with B. licheniformis B2560 ; P2 = 10 ml inoculum fermentation with B. licheniformis B2560 ; and P3 = 15 ml inoculum fermentation with B. licheniformis B2560, respectively for chicken feather meal as much as 2 g .Parameters measured were the levels of protein and organoleptic parameters that support, in the form of the physical properties of chicken feather meal, include color, texture, and smell. Data were analyzed using a protein content analysis of variance (ANOVA) with a confidence level of 95 %, followed by Duncans Multiple Range Test Test (DMRT) with a confidence level of 95 % , while for the data results of the organoleptic qualitatively analyzed descriptively.The results showed that the P0 treatment was significantly different (P < 0.05) with P1, P2, and P3, and between treatments P1, P2, and P3 respectively were significantly different (P < 0.05). The treatment gives the best results is the treatment that P1 has the highest protein content of 84.08 % with a change in the physical properties of white -yellow (color), soft (texture), and less typical sting (smell). Keywords :Bacillus licheniformis B2560, fermentation, the quality of raw materials, chicken feather meal, Fish feed

Abstract

The aims of this research were to evaluate the eficacy of intra- and extracellular products vaccines of Aeromonas hydrophila to control Motile Aeromonas Septicemia (MAS) in catfish (Clarias sp.). Catfish with 10-13 cm of total length were used for the experiment with three treatments in five replicates. The treatments were vaccinations with (1) A: intracellular product of A. hydrophila; (2) B: extracellular product of A. hydrophila; (3) without vaccination (control). Vaccination was conducted by intramuscular injection of 5 µg protein/fish (each fish was injected with 0.1 ml of vaccine). Booster was conducted one week after vaccination. The challenge test was conducted two weeks after booster. The results showed that the survival rate, Relative Percent Survival (RPS), and antibody titer of A treatment was significantly different (PA. hydrophila (A treatment) reached 85.33 and 82.26%, respectively. The highest antibody titer was 1843.2 which was obtained by the A treatment. The results suggested that intracellular product vaccine of A. hydrophila was more effective than extracellular product vaccine.
