Abstract

The purpose of this study was to find out the diversity index of molluscs (gastropods & bivalves) based on mangrove degradation level in Tobea Island of Muna Regency. The data were collected from May until July 2017, in Wangkolabu municipal area of Tobea sub-district, using observation method and direct observation in the research location to find out the index of ecology of molluscs such as index of diversity (H'), evenness (E), density (N), dominance (D), and dispersion pattern (Id) from both gastropod and bivalve classes based on mangrove degradation level. Direct identification in the field is applied to the type of mangrove vegetation and mollusc organisms that have been known by it’s scientific name. But if not, then the samples are taken and the documentation is then done further analysis at the Laboratory of Biology Unit of Ecology and Taxonomy of FMIPA UHO with reference to the guide book of Introduction of Mangrove in Indonesia and book Snail & Shell Indonesian. Then for water environment parameters in the form of water and substrate samples are further analyzed at Biology Laboratory of UHO Forensic and Biomolecular Unit using Miller triangle fraction method and APHA standard method. Statistical analysis is done using Excel Stat to determine the major components affecting the ecological index of molluscs in degraded mangrove areas. The results showed four species mangroves in Wangkolabu Village are Rhizophora mucronata, Rhizophora apiculata, Bruguiera gymnorrhiza, and Sonneratia alba and that mangrove degradation on the coast of Tobea Island, Wangkolabu village, Tobea sub-district has varied values for good mangrove criteria (Density/RDi = 1.600 individuals/hectare), medium mangrove (Density/RDi = 1.175 individuals/hectare), and mangrove broken/rare (Density/RDi = 375 individuals/hectare). Besides that, Cerithium spp., Nerita costata, Terebralia sulcata, and Clypeomorus sp., type gastropods is able to adapt to mangrove forests that have moderate density (RDi= 1.000–1.500 individuals/hectare), with mangrove substrate types in the form of sandy loam that has soil organic content (BO = 3,18%). While the gastropod type Telescopium telescopium prefers low density mangrove forests (RDi < 1.000 individuals/hectare) with the type of substrate in the form of clay containing organic matter (BO = 2,35%).  Based on the results of the study it is expected that the need for mangrove forest management in the study area for the sustainability of aquatic biota, especially molluscs that can be developed as conservation areas by looking at the level of mangrove damage through non-fish aquaculture activities (shellfish) for the welfare of coastal communities and islands small in Southeast Sulawesi.

Abstract

The study was conducted to assess the caracteristic of cytochrome C oxidase subunit I (COI) gene on wild honey bee Apis cerena, and detection of barcode sites from these gene. A total fifteen individual A. cerena were collected from Hoga Island, Southeast Sulawesi. Genomic DNA was extracted from torax, then amplified by PCR method and than sequenced. Sequencing result characterized their nucleotide and amino acid content. The results showed that 595 nucleotides at the 5' end of COI gene of A. cerena very conserved at the most of the sites. Nucleotide dominated by thymine and adenine bases (± 70%). There are 25 barcoding sites for A. cerena. There are two of these sites are diagnostics for A. cerena from of the Hoga Island. From of 595 basepairs nucleotide were prediction encode 198 amino acid, and only eleven percent of the these are varied between species. Amino acid 91th (treonine) and 103th (asparagin) were diagnostics amino acid for A. cerena from Hoga Island, Southeast Sulawesi. Molecular phylogeny reconstructed based on both nucleotide and amino acid sequence placing apis cerena from Hoga Island, Souteast Sulawesi is closely related to Apis cerena from Genbank source.

Abstract

Kata kunci: Vegetasi mangrove, zonasi, struktur, komposisi  This research was aimed to describe the structure, dominance of mangrove vegetation, zonation,  and natural regeneration at Kaledupa Island of Wakatobi National Park. Sampling of vegetation used line transect and plot methods.  Data of mangrove vegetation was obtained through observation of each plot at all growth phases (trees, poles, sapling and seedling) covering species name, number of individual of each species, and size of the breast height diameter of stem. Data of water inundation at high tide was measured at each observation plot. Observation was conducted for 30 days. Dominance of mangrove vegetation was determined by importance value parameter, zonation by relative density parameter and natural regeneration by seedling density. Results of this research found that mangrove community at Kaledupa Island at trees stratum were dominated by species Bruguiera gymnorrhiza, poles stratum was dominated by species Rhizophora mucronata, and at sapling and seedling stratum were dominated by species Ceriops tagal. Mangrove zonation at Kaledupa Island consisted of four zones, that are R. mucronata, R. apiculata, C. taga, and C. decandra zone. Height of seawater inundation is controlling factor of formation of mangrove zonation at Kaledupa Island. Species C. tagal  and C. decandra had high level of natural regeneration, while species R.mucronata, R. apiculata, B. gymnorrhiza, Xylocarpus granatum, Sonneratia alba, and Avicennia marina had low natural regeneration.
