Abstract

Brucellosis in cattle is a disease caused by Brucella abortus due to the reduction in livestock population caused by abortion, stillbirth, weak birth, infertility and sterility. Brucella abortus has several potentialvirulence factors, i.e. virB11 gene that encodes VirB11 protein is an important virulence factor acts as an ATPase for assembling organelles when the bacteria replicate, helping to complete the bacterial cycle and agress to another cells. The aim of this study are to re-identification Brucella abortus and detect virB11 gene as encoding of B. abortus VirB11 protein in local isolates from Pinrang, NTT, strain vaccines S19 and RB51. The isolates Brucella abortus were re-cultured in Brucella agar base and re-identification is followed by microscopic with Gram staining and biochemical tested with urease, citrat, indol and TSIA test. virB11gene was detected with PCR method. The PCR result showed virB11 gene have DNA band 720 bp.virB11 gene are present in local isolates from Pinrang, NTT, strain vaccines S19 and RB51.

Abstract

Tujuan dari penelitian ini adalah untuk mengetahui tingkat cemaran bakteri Salmonella sp. pada susu segar dari peternakan sapi perah di kecamatan licin kabupaten banyuwangi. Sampel susu segar di ambil pada april 2021 sebanyak 16 sampel dari KPSP Ijen makmur di kecamatan Licin Kabupaten Banyuwangi. Sampel diambil sebanyak 25 ml dari masing-masing peternak. Hasil yang di dapat setelah melakukan isolasi dan identifikasi menggunakan media Salmonella Shigella Agar (SSA), dan dilanjutkan uji identifikasi menggunakan TSIA dan Urea didapatkan  dari ke 16 sampel semua dinyatakn negatif bakteri Salmonella sp.. Hal ini dapat terjadi karena para peternak sapi perah di Kecamatan licin sudah mulai menyadari pentingnya kebersihan kandang dan sanitasi kandang yang baik sehingga tingkat cemaran bakteri Salmonella sp. dapat dicegah.

Abstract

The study was aimed to know the prevalence and level of gastrointestinal worm investation of local chicken (Gallus domesticus) in Kramat Village, district of Bangkalan, Madura East Java province. The samples used in the study were 100 faeces of kampung chicken. using sedimentation and floating method. Faeces samples were tested, than followed by microscopically test to identify the worm is species. The result showed that in 38 out of 100 samples could be indentified some worm species as followed : Capillaria sp.14 (36,8%), Railletina 7 (18,4%), Hymenolepis 16 (42,1%) and Heterakis gallinarum 1 (2,6%). Using Mann Whitney U-test could be differentiated the level of the infection with an average of 45.53 in cock, while in hen with an average of 55.47. There was no significancy (p>0,05) on the level of worm gastrointestinal infection of different sex of kampung chicken.

Abstract

Antibiotic resistance in animals and humans has become a global problem that needs attention. The use of antibiotics in inappropriate on food-producing animals can lead to resistance many of the pathogenic bacteria to the various types of antibiotics, one of which is the Escherichia coli (E. coli) which produces extended spectrum β-lactamase (ESBL). The aim of this study was to isolate and identified ESBL- E. coli isolate from dairy cow rectal swabs in Sendang, Tulungagung district using the Vitek-2 method. The number of rectal swab samples used in the present study was 50. The result of the study showed that from all of the samples could be isolated and indentified E. coli, based on the colony characteristics on EMBA and biochemical test. Based on the double disc synergy test method using antibiotic disc amoxicylyn-clavulanate, ceftriaxone, aztreonam, ceftazidime and cefotaxime, 10 isolates could be identify els ESBL- E. coli. furthermore 3 out of 10 isolates DDST positives were confirmed ESBL- E. coli using Vitek-2 method.

Abstract

This study aimed to evaluate the Total Plate Count and total Staphylococcus aureus count of beef sold in wet markets in Mulyorejo sub-district below the National Standard Indonesia (SNI 7388:2009) about maximum limit of microbial contamination in food or not. Total of twenty four samples of beef purchased from traditional markets of Tempurejo, Krempyeng Yamuri, Pacar Keling, and Menur in Mulyorejo sub-district Surabaya were examined by Total Plate Count using pour plate method. The sample was also cultured in Mannitol Salt Agar. The colony suspected to be S. aureus were taken for identification. The identification of S. aureus consists of isolation in Mannitol Salt Agar, Gram staining, catalase test, and coagulase test. Total plate count result showed that four samples were exceeding the National Standard of Indonesia SNI 7388:2009 or 1x106 CFU/g and the rest were below the maximum Total Plate Count in SNI. The highest Total Plate Count result was 1,9x106 CFU/g and the lowest was 7,8x104 CFU/g. The result of identification showed that 100% samples examined were contaminated by S. aureus with the highest result was 2,9x104 CFU/g and the lowest result was 4,3x103 CFU/g or exceeding the SNI 7388:2009.

Abstract

This study aimed to detect the presence of eae gene in E. coli from chicken meat sold at the traditional market Surabaya. Identification of bacteria was carried out in several stages, inoculated on pre-enrichment media using Buffered Peptone Water (BPW), planting on Levine’s Eosin Methylene Blue Agar (L-EMB) media, suspected colonies on L-EMB were subjected to Triple Sugar Iron Agar (TSIA) and Indol test for the confirmation of E. coli. The confirmed E. coli isolates were tested by Polymerase chain reaction (PCR) to determine the presence of the eae gene. The results showed that E. coli could be identified in 28 out of 100 (28%) chicken meat samples, and eae gene was detected in three E. coli isolates (11%).
