Abstract

ABSTRACTThe use of tetraploid banana in triploid banana breeding has been conducted through 4x x 2x crossing and/or its reciprocals, 2x x 4x crossing. However, the availability of tetraploid banana is naturally very limited, therefore artificially induced tetraploid banana is needed. The present research is aimed to generate tetraploid banana plants by crossing triploid banana ?Goroho? as female parent with wild diploid banana, Musa acuminata var. nakaii Nasution as male parent, and to characterize the tetraploid hybrid. 94 crossings were made by pollinating stigma of Pisang Goroho with polen of M. acuminata var. nakaii. Embrios resulted from hybridization were in vitro rescued and propagated. Hybrid plantlets were then acclimatized in the green house and seedlings were planted in the field for two cycles of banana reproduction. Ploidy identification, morphology and molecular characteristics were also conducted for two cycles of banana reproduction. Results of the experiment showed that 9 hybrid seeds were produced from 94 pollinations. However, only 2 hybrid embryos grew and developed into shoots and from two shoots only one shoot was survived. The one shoot were then in-vitro propagated to produce 21 hybrids planted in the field. After being evaluated for two plant reproductive cycles, all hybrid plants were tetraploids (2n=4x=44). The hybrid plants had drooping leaves, bigger fruit diameter compared to both parents, and blunt fruit tip. DNA (RAPD) profiles of tetraploid hybrid showed that 37.5% of the bands were from female parent, 37.5% from male parents, and 25% from both parents. The tetraploid hybrid ?GRNK? is expected to be useful in triploid banana breeding in the future.Key Words: Hybrid banana, tetraploid, Goroho, Musa acuminata Colla var. nakaii Nasution

Abstract

Taraxacum officinale Weber ex F. H. Wigg. is a herbaceous medicinal plant species belonging family Asteraceasewhich has apomictic and poliploid characteristics. Multiplication of shoots using tissue culture was used to obtaindefine high quality seedlings, uniform, stable or free of diseases. However, changes in chromosome number canoccur in regenerated plants. The research aim was to determine the chromosome number of T. officinale plants regeneratedfrom culture in vitro using explants of roots, petioles and leaf blades. Therefore, selection of regenerantscan be done in order to find out transplants which have high yield of secondary metabolites. Analysis of chromosomenumber from root tips samples was conducted using 24 plantlets regenerated from root, 27 plantlets regeneratedfrom leaf blade, 21 plants regenerated from petiole and 102 roots of grown seeds using orcein squash method.The results showed that germinating seeds (control) and regenerated plants had variation in chromosome number.The range of chromosome numbers from regenerated plants were 2n=8-39, and cells with diploid number (2n = 2x= 16) was as most observed. The range numbers in germinated seeds were 2n=10-38, and cells with triploid number(2n = 3x = 24) was as most observed. This results proved that variation in numbers of chromosome was caused byapomixis and poliploid characteristics of the parent plant regenerated to their regenerants.Keywords : Taraxacum officinale. Abstract

ABSTRACTThe objective of this research was to study molecular markers were linked to upland rice tolerance of Situ Patenggang x B11930F-TB2 against Al using SSR method. The plant materials were 36 lines of Situ Patenggang x B11930F-TB2 with categories tolerant, moderate, and susceptible to acid soil based on selection in East Lampung. IR6008032 line and ITA variety used as comparative control of tolerant and susceptible. The results showed that 40 primer pairs were applied to 36 lines and allegedly 3 primer pairs that are: RM205, RM257 and RM247 linked to Al tolerance. The markers location analysis on rice chromosome listed as follows: RM205 and RM257 were located on chromosome 9 and RM247 located on chromosome 12 with genetic distance 93,12 cM, 772.62 cM and 13,05 cM, respectively.Keywords: SSR marker, upland rice, aluminium tolerance, ultisols

Abstract

The road across the forest can cause the diversity exchange in the forest ecosystem. Distribution of the exotic speciescaused by the road are the main factor affecting the extinction of the native plants. The forest corridor in HalimunSalak National Park is separated by the road which is connecting five villages in surroundings. The aims of the researchwere to determine the response of diversity and abundance of the understory plants to the road existence, toobtain the effect of the road to the habitat condition, and to identify the exotic plants and its relation to the road.Vegetations were observed by transect sampling system, 5 transects of 150 m length were placed in forest corridorside along the road. In each transect, there were 12 sampling plots (1m x 1m) placed in distance (from the road): 0,5, 10, 15, 20, 30, 40, 45, 60, 90, 120, and 150 m from the road. The distance of each transect was approximately100 m and each transect were placed in the corridor area having forest buffer about 100 m distance. There were 117plant species including 9 exotic species. Plant community analysis using Squared Euclidean Distance (SED) showedthat road side area to 5 m in distance showed the different composition of the understory plant to inside the forest.Exotic plants and grass dominated in the area close to the road. Canopy cover in the road side to 10 meter to theforest was relatively opened than inside the forest. Plant diversity analysis on both of all species and local speciesusing Canonical Correspondence Analysis (CCA) showed that species diversity of understory plants was notsignificantly affected by the distance from the road. Nevertheless, distance from the road was a main factorinfluencing the exotic species, while distance of 0 m showed the highest exotic plants diversity.Keywords: Corridor, exotic plant, Halimun Salak National Park, road, squared euclidean distance, understory plant

Abstract

Beberapa Pengaruh Antropogenik pada Sosioekologi pada Monyet ekor Panjang (Macaca fascicularis) di Pulau Lombok, Indonesia. Survey distribusi monyet ekor panjang dilakukan di P. Lombok selama 2001-2009 dan dari 37 kelompok, satu kelompok jantan muda dan 3 individu soliter yang terdata dari 27 lokasi sekitar 63% monyet dijumpai pada areal karena terpengaruh oleh aktivitaskehidupan manusia, seperti hutan sekunder, kebun buah-buahan, hutan persembahan dan tempat rekreasi. Besarnya kelompok monyet cenderung menjadi lebih besar di area semi buatan dibandingkan dengan kelompok yang hidup liar (Liar= 7.08, buatan =19.04, X2 = 5.4763, df = 1, P= 0.01928). Faktor-faktor ketergantungan pada manusia yang menyediakan sejumlah pakan ternyata mempengaruhi tingkah lakunya sedangkan penebangan hutan dan pengubahan alih fungsi menjadi kawasan lain selain hutan akan memicu monyet terkesan menjadi hama tanaman.Kata Kunci: Monyet kra, Macaca fascicularis, Pulau Lombok, Anthropogenik

Abstract

ABSTRACTExtraction of Antibacterial Compound from Diatom Chaetoceros gracilis WithDifferent Methods. Diatom is phytoplankton which is commonly found in off shore.Chaetoceros produce antibacterial which inhibit some bacteria. This research was donein 3 steps. At first, Chaetoceros gracilis was cultivated in temperate controlled roomwith lighting 24 hours. The culture was harvested on 14 days, then it was dried andweighted. The second step, biomass was disrupted by different method such as sonicator,glass beads, and undisrupting. Then the biomass was maserated, filtrated, and evaporated.The crude extracts were tested to pathogen bacterial. The third step, the extraction wasconducted using hexana (non polar solvent), ethyl acetate (semi polar solvent), andmethanol (polar solvent). The crude extracts were tested to the pathogenic bacteria.The result showed that the produce cell disruption antibacterial activity by sonicator.The biggest inhibition zone was obtained by hexana but produced lower yield.Key words: Chaetoceros gracilis, growth, extraction, disrupting, antibacteria
