Abstract

Resistance test strains of transgenic potatoes transformed with RB gene to late blight (Phytophthora infestan) in KP Pasirsarongge, Cianjur. Potato late blight caused by Phytophthora infestans (P. infestans) (Mönt.) de Barry continues to be one of the most important crop diseases of all time. Genetic engineering of potato using RB gene for resistant plant to this disease is the most effective and environmental friendly to prevent widespread of late blight. This research aims to perform resistance of transgenic potato lines containing RB gene to lateblight (P. infestans) in Pasirsarongge, Cianjur field trial station. The first generation of transgenic lines were planted on polybag containing soil:manures using randomized complete block design. Tested plant inoculation was done naturaly from inoculum source from border row (Granola) that has been planted at one month before. The symptom was observed at one month after planting and damage scoring was done every three days for five times. Twenty two transgenic lines of tested plant showed various resistance respond to late blight (P. infestans) attack. Three transgenic lines showed highly resistance to late blight (P. infestans) were lines 11, 24, and 25, one transgenic line has resistant level was line 6.

Abstract

Virulence of various Metarhizium spp. (Deuteromycotina: Hyphomycetes) isolates on cocoa pod borrer pest, Conopomorpha cramerella Snell. (Lepidoptera: Gracillariidae). In Indonesia, Conopomorpha cramerella Snell. (Lepidoptera : Gracillariidae) is a serious pest of cocoa. Entomopathogens such as Metarhizium spp. (Deuteromycotina: Hyphomycetes) can be used to control this pest as an alternative to chemical insecticides. The purpose of this research was to study the virulence of isolates of Metarhizium spp. to pupae of C. cramerella. Nine isolates of Metarhizium spp each with 107 conidia/ml rate were used in the experiment. Pupal mortality was recorded daily. The results showed that isolate MetApCi from chili rhizosphere had the highest virulence causing 96.67% mortality of C. cramerella pupae and with an LT50 of 3.04 days.

Abstract

Diversity and phylogeny of fruit fly (Diptera: Tephritidae) in South Kalimantan based on morphology and molecular (RAPD-PCR and DNA sequencing). Seven species of fruit fly was known by morphological identification. The fruit flies were found from  trapping with methyl eugenol and fruit collecting at all study sites in South Kalimantan. The results showed that as much as 17  plants were infected by fruit fly. Dendrogram based on morphological identification analyzed by using UPGMA with MEGA 4 program consisted in a group consisting of 5 sub-groups. Bactrocera carambolae and Bactrocera papayae of morphology were still a closely related fruit fly at 0.935. Whereas, based on RAPD result analized by UPGMA using 20 character of DNA based, showed that out of seven species consisted 2 groups, 1st group were B. umbrosa,  B. occipitalis and sub-group of B. latifrons. The second group consists of sub-groups B.carambolae, B. papaya, sub-group B. albistrigata and B. cucurbitae. The results of dendrogram from sequencing DNA fruit fly analysis comprised one of group and three sub-groups. The first sub-groups were B. papayae, B. carambolae, B. occipitalis, B.latifrons. The second subgroup were B. cucurbitae and B. umbrosa. While B. albistrigata separate but still one group with another fruit flies. The results of DNA sequencing showed that there were a homology of the seven species of the fruit fly i.e at 83 base pair / bp (C), 101 bp (T), 265 bp (G), 420 bp (A), 432 bp (T), 600 bp (A ). The length of the base pair for B. occipitalis, B. cucurbitae, B. albistrigata, B. carambolae, B. papayae, B. latifrons were respectively 615, 898, 570.969, 898 and 615 bp. The results of morphological analysis and RAPD methods showed difference in the distribution of groups and sub-groups. But based on morphologycal and DNA identification seven species of fruit flies found were all same as the genebank.

Abstract

Effect of Pseudomonas fluorescens P60 on pathological and physiological quality and growth of rice IR 64 seedlings. The research objectives were (1) detection and identification of seed-borne pathogens of IR 64 rice, (2) testing Pseudomonas fluorescents P60 in inhibiting the in vitro growth of seed-borne pathogens colonies, (3) testing P. fluorescents P60 for pathological and physiological seed quality, and (4) testing P. fluorescents P60 on the growth of seedlings in the greenhouse. The results showed that some seed-borne pathogens can be found both on farmers’ IR 64 rice and factory’s; they were Aspergillus flavus, Alternaria padwickii, Pseudomonas glumae, and P. syringae. Application of P. flourescens P60 was able to inhibit the in vitrogrowth of colonies of all seed-borne pathogens, except P. syringae. Related to pathological quality, the effect of P. flourescens P60 on percentage of seed-borne pathogens attack did not significantly different from that of benomil but smaller than distilled water. On the physiological quality of seeds, treatment of P. flourescens P60 has the same effect with benomil and distilled water, with germination rate was more than 80%. In the greenhouse study,treatment of seed immersion time in P. flourescens P60 suspension showed that the effect of immersion time as long as15 minutes and 25 minutes on seedling height, root length, and seedling dry weightdid not significantly different. were. However, 25 minutes immersion time resulted in fresh seedling weight and root dry weight higher than that of 15 minutes immersion time.

Abstract

Identification of Globodera rostochiensis using differential clones. Potato cyst nematode, Globodera rostochiensis, is a relatively new pathogen in Indonesia that becomes a constraint to potato production. To manage the parasite effectively, it is very important to identify the pathotype of G. rostochiensis populations. Therefore, this research was carried out to identify G. rostochiensis pathotypes. Four G. rostochiensis samples, consisting of three samples from East Java and one sample from Central Java, were identified using a set of differential clones. The result showed that G. rostochiensis samples from East Java were new pathotype, whereas the sample from Central Java was Ro1 pathotype.

Abstract

Screening and identification of endophytic bacteria to control bacterial leaf blight disease on Shallot. The experiment was conducted in Laboratory and Green House, from January to June 2012. Laboratory experiment consisted of three steps: (1) isolation of endophytic bacteria from healthy onion roots, (2)In planta /screening of endophytic isolates capable of reducing bacterial leaf blight disease, and (3) molecular identification of potential endophytic isolates. Treatments of in planta test were arranged in Completely Randomized Design. Collected isolates were tested for their capability in controlling bacterial leaf blight disease on shallot. The variables observed were disease incidence, disease severity, and shallot yield. The results showed that out of 82 isolates successfully isolated, 56 isolates (68.29%) were Gram positive, and 26 isolate (31.71%) were Gram negative. All isolates were HR negative and pathogenicity negative. Six endophytic isolates showed better performance in inducing resistance and increasing onion yield. Based on 16S rRNA sequence the six isolates were Bacillus cereus strain P14 , Bacillus cereus strain Se07, Bacillus sp H1, Bacillus sp SJ1 and Serratia marcescens strain PPM4.

Abstract

The effectiveness of polar and non-polar fractions of Ageratum conyzoides L. to control peanut rust disease and phytochemical screenings of secondary metabolites. Peanut rust disease caused by Puccinia arachidis is one of the important diseases which causes yield loss more than 50%. An alternative control which is more environmentally friendly can be conducted by applying botanical fungicides extracted from weeds. This study aimed to obtain effective concentrations of polar and non-polar fractions of roots, leaves, and flowers of Ageratum conyzoides as well as to investigate secondary metabolites in each extract. A spore germination test was conducted to determine the effectiveness of each extract. An application of 5% polar fraction of the leaves resulted 98±2% ungerminate spores which was not different with the application of 5% polar fraction of the roots which resulted 95±2% ungerminate spores. The use of a polar solvent extracted more diverse secondary metabolites compared with that of a non-polar solvent. Phytochemical screening tests on the three extracts showed that the leaf polar fraction contained more diverse secondary metabolites as compared with the others. The polar fraction of the leaves contained alkaloids, flavonoids, tannins, saponins, and terpenoids. A further test to confirm the existence as well as to separate these compounds with thin layer chromatography showed that alkaloids, flavonoids, saponins, and terpenoids separated into 7, 9, 6, and 8 spots, respectively. These secondary metabolites may be responsible to inhibit the spore germination of P. arachidis.

Abstract

Effectiveness of three botanical insecticides against the papaya mealybug Paracoccus marginatus and their safety to the predatory Curinus coeruleus larvae. Preparations of Tephrosia vogelii (Tv) leaf extract (0.5% and 1% w/v), Annona squamosa (As) seed extract (0.5% and 1%), and Cinnamomum multiforum (Cm) leaf essential oil (1% and 2%), and their mixtures (Mix-1: Tv 0.25% + As 0.25% + Cm 0.5%; Mix-2: Tv 0.5% + As 0.5% + Cm 1%) were tested for their effectiveness on third-instar nymphs of Paracoccus marginatus by spraying the test materials on undetached papaya leaves and by direct spraying on the test insects using Potter spray tower. Tv extract was also applied on the test insects placed on undetached papaya leaves. The safety test was done by direct spraying of the test materials on the predatory Curinus coeruleus larvae using Potter spray tower. Tv and As extracts at a concentration of 1% each are potential to be used for the control of P. marginatus. Spraying of T. vogelii extract on the test P. marginatus nymphs placed on papaya leaves was more effective than spraying of the test materials on papaya leaves or direct spraying on the test insects only. The treatment with Cm essential oil required twice the concentration of Tv and As extracts to obtain the same level of effectiveness. The treatment with Mix-2 caused lower P. marginatus mortality than the sum of mortality caused by its components applied separately. Nonetheless, the three botanical insecticides and their mixtures were safe to C. coeruleus larvae. On the other hand, although the synthetic insecticide imidacloprid (neonicotinoid), included in the study for comparison, was effective against P. marginatus, it was also toxic to the predatory C. coeruleus larvae so its use should be avoided or limited.

Abstract

Utilization of chitosan to control Bean common mosaic virus (BCMV) on yard long bean. Bean common mosaic virus (BCMV) is one of important viruses infecting yard long bean in Indonesia. One of efforts to control its infection is by utilizing chitosan. Thus, the aim of the conducted research was to test the effectiveness of chitosan in controlling BCMV on yard long bean. Concentration of chitosan tested were 0.1% and 1.0% and it was applied as seed treatment (PB), leaf spraying before (SB) and after (ST) mechanical inoculation of BCMV. Incubation period, disease incidence and severity, peroxidase enzyme activity, and BCMV titer were measured as observation parameters. Incubation period of all treated plants were longer as compared with untreated control plants. Symptoms of treated plants at concentration 0.1% (SB and ST) showed mild to severe symptom and leaf malformation, while at concentration 1.0% showing milder symptoms and some of tested plants remain symptomless. Treated plants showed lower peroxidase enzyme activity and significantly lower BCMV titer as compared with that of untreated control plants. The lowest to highest percentage of virus inhibition exhibited by ST0.1, SB0.1, PB0.1, PB1, SB1 and ST1 treatments with virus relative inhibition level ranged 75.94%-86.07% and severity inhibition level ranged 39.55%-64.63%.

Abstract

Compatibility test of four Trichoderma spp. Isolates and in vitro inhibition ability on several plant pathogens. In vitro descriptive compatibility research was carried out to know the compatibility among Trichoderma spp. isolates and their inhibition ability toward several plant pathogens. Four Trichoderma spp. isolates used were ginger, shallot, banana, and pineapple isolates; while the pathogens used were pathogenic fungi (Fusarium, Colletotrichum, Phytophthora, and Sclerotium), bacteria (Ralstonia), and nematode (Meloidogyne dan Globodera). Observation was done toward inhibition zone between Trichoderma spp. isolates, colony radial growth, mycelial dry weight, and nematode mortality. Result of the research indicated that the four Trichoderma spp. isolates were compatible and no growth inhibition was observed. The inhibition ability of all Trichoderma spp. isolates varied and the ginger isolate had the highest inhibition ability and mortality on all fungal species and the nematodes, while on the pathogenic bacteria there was no inhibition.
